Introduction: Oxidative stress has been assumed to contribute to the pathophysiology of schizophrenia. Increased oxidative stress is the result of either an increased production of free radicals or a depletion of the endogenous antioxidants. Objective: To assess the levels of oxidative stress and antioxidant status in schizophrenia. Materials and Methods: This observational study was carried out in the department of Pharmacology, Bangabandhu Sheikh Mujib Medical University from September 2013 to January 2015. Ninety three schizophrenia patients were enrolled as study group and 30 healthy indivuduals were taken as control group. The peripheral levels of several molecules associated with oxidative stress namely malondialdehyde (MDA), glutathione (GSH) and anti-oxidant status like plasma levels of ascorbic acid (vitamin C) and α-tocopherol (vitamin E) in 93 patients with schizophrenia and 30 healthy participants were assessed. Results: Study found that the schizophrenia group presented substantially higher levels of oxidative stress than the control group, as revealed by elevated quantities of the pro-oxidant MDA (6.3±0.5μmol/L in study group and 2.1±0.5μmol/L in control group), decreased levels of the antioxidants GSH (0.6±0.2mg/gm of Hb in study group and 2.1±0.5mg/gm of Hb in control group), plasma α-tocopherol and ascorbic acid. Results found were highly significant (p=0.001). Conclusion: In schizophrenia there are increased level of oxidative stress and decreased level of the antioxidants. Background: Oxidative stress plays an important role in the development of metabolic changes in malaria patients. During infection RBCs are exposed to continual oxidative stress. The univalent reduction of oxygen results in a series of cytotoxic oxygen species such as O2-, H2O2, OH•. Objective was to determine the level of oxidative stress in patients suffering from malaria.Methods: The present study was conducted on 551 malaria patients and 211 age-sex matched controls, in department of Biochemistry, C U Shah Medical College, Surendranagar, Gujarat from April 2012 to May 2013.  In stage-I, day-1 malaria patient’s v/s control group, In stage-II, day-3 v/s day-1 after anti-malarial treatment and in Stage-III day-3 v/s day-1 after anti-malarial + antioxidant treatment.Results: The mean erythrocytic activity of SOD, CAT, GST were decreased (0.71±0.25EU, 9.9±2.4μmol/sec, and 11.7±3.9 U/gmHb% respectively), mean level of GSH and MDA were increased (42.1±6.06gm/Hb%, 10.9±2.83 respectively) significantly (P<0.001) as compared to control group. In the follow up study with anti-malarial treatment the mean levels of erythrocytic GSH and MDA (28.7±7.54gm/Hb% and 8.08±1.95nM/L) decreased significantly (P<0.001 and P<0.01 respectively), whereas mean activity of erythrocytic enzymes like SOD, CAT and GST (0.99±0.15 EU, 15.8±2.68μmol/sec and 22.5±5U/gmHb%) were increased significantly (P<0.001) as compared to day-1.Conclusions: Erythrocytic antioxidant enzymes, GSH and MDA may be considered to be reliable biochemical markers for diagnostic and therapeutic potential in malaria.

Introduction Retinopathy of prematurity (ROP) is the leading cause of blindness in newborn babies worldwide. The benefit of anti-oxidant was investigated for ROP cases by assessing its effect on the oxidative stress of the tissues. Glutathione is a primary endogenous in human body and its supplementation has been discovered for its benefits towards some ocular diseases. This study aims to understand the effect of glutathione on oxidative stress marker, serum Malondialdehyde (MDA), in ROP rat models.Materials and methods This was an experimental study with post test only controlled group design. Sixteen Wistar rats that met our study criterias were divided into two groups, study group and control group. The study group were exposed to 95% oxygen for 4 hours / day followed by normoxic laboratory condition for 20 hours. Glutathione 1.5 mg / day were injected intramuscularly to rats in study group. The control group was exposed to 95% oxygen followed by normoxic laboratory condition with the same manner, and did not received glutathione. This cycle was repeated for 14 days. Both groups were settled in a room temperature settings on days 15-22. Serum sampel was collected from retroorbital vein. The malondialdehyde level was analyzed using MDA analyser kit.Results MDA level was found significantly higher in study group compared to control group (546.99 ng/ml vs 201.51 ng/ml, respectively, p 0,001).Conclusion Our study demonstrated a higher MDA levels in ROP rat models given glutathione injection compared to the control group.

Background: Epilepsy is a serious clinical condition characterized by recurrent seizures. Oxidative stress plays an important role in the etio-pathogenesis of epilepsy. Measurements of serum thiol and disulfide levels were used to evaluate the antioxidant status of the body. Objective: The aim of this study was to determine serum levels of thiol and disulfide in epileptic pediatric patients. Methods: Ninety patients, 54 epilepsy and 36 controls were included in the study. Serum levels of native thiol total thiol and disulfide were measured and disulfide/native, disulfide / total thiol and native thiol/ total thiol ratios were calculated. Hence, the ratios of disulfide/ native thiol, disulfide / total thiol and native thiol/ total thiol were calculated. Results: Serum levels of native thiol, total thiol and disulfide were significantly lower in the epilepsy group than the control group. The ratio of disulfide/native thiol and disulfide / total thiol were significantly higher in the study group than the control group. As well as, the native thiol / total thiol ratio was lower in the epilepsy group than the control group. Native thiol, total thiol and disulfide were significantly lower in the epilepsy group who were taking anti-epileptic drugs than those who were not taking anti-epileptic drugs. Conclusion: We demonstrated a meaningful relationship between oxidative stress markers and epilepsy in pediatric patients.

Background: The anti-oxidant and the per oxidation product levels both are increased in pregnancy. Much recent works has focused on the role of oxidative disturbance in the preeclampsia and eclampsia. Preeclampsia remains a leading cause of maternal and fetal morbidity and mortality.Methods: This prospective study was conducted in upper India sugar exchange and maternity hospital, department of Obstetrics and Gynecology, GSVM Medical College, Kanpur in Collaboration with department of pathology, GSVM Medical College, Kanpur (UP), India. All the patients selected for the present study, both from control and study group was in detail with regard the clinical history, general examination, local examination, basic investigation and Specific Investigation, Super Oxide Dismutase (SOD). Blood samples would be collected with the informed consent from control as well as study group for assessment of antioxidant status by determining levels of superoxide dismutase.Results: The mean age and BMI of control as well as study group is almost same. The incidence of pre-eclampsia and eclampsia in primipara was 29.31 and in multiparous it was 9.52 in all groups. It implies that 95% cases will have improvement in SOD value with mean difference in improvement of at least 0.297807. There is statistically significant difference in mode of delivery by LSCS in study group A as compared to study group B and control in PIH patients. The incidence of complications is significantly higher in Study Group A as compared to control group and Study Group B. There is statistically significant difference in the incidence of preterm birth in study group A as well as Study Group B as compared to control. However, there was no statistically significant difference in the incidence of preterm birth after giving intervention to the study group. When cut off value of SOD enzyme as < 0.578U/mg of protein, sensitivity is 66.07% and specificity as 51.85%.Conclusions: Vitamin-c supplementation will only prevent PIT in PIH with already reduced antioxidant status.

Using hyperbaric oxygen (HBO) therapy as an in vivo oxidation model, we investigated the effect of a diet enriched in ascorbic acid (AA) on HBO-induced oxidative stress. Volunteers (n 46) were allocated to the AA-rich diet group or the control group. Blood samples were collected at the basal time, after the 1-week diet before and immediately after the HBO treatment, and 1 week after the HBO treatment. AA level, total antioxidant status (TAS), hydroperoxides (HP), lymphocyte DNA oxidation and DNA repair capacity were assessed. The expression of genes involved in oxidative stress was evaluated in lymphocytes and the protein activity of the modulated genes was determined in the plasma. The AA level and the antioxidant status of plasma were increased by AA-rich food consumption. HBO exposure did not affect the AA levels or TAS, but induced HP formation in the control group. The lymphocytes isolated from dietary-supplemented subjects were resistant to ex vivo DNA oxidation, showing an increased DNA repair capacity compared with controls. A difference in gene expression pattern was observed between the groups. AA-rich foods provide dual protection against oxidative stress, enhancing plasma antioxidant levels and stimulating genes involved in cell detoxification.

Introduction: Fever is the most common presenting complain for which children are brought to the paediatrician.Physical methods are widely used in treating febrile children, tepid sponging being commonly practiced in hospitals along with antipyretics. The objectives of this study were to compare the effectiveness of tepid sponging and antipyretic drug versus antipyretic drug alone in febrile children.Material and Methods: This was a Randomized controlled trial done in JSS Medical College and Hospital. All children under the age of 6 months to 12 years, admitted with axillary temperature of >99oF were included in the study. A total of 500 children were included over two years study period. Children with recorded axillary temperature of >990F were randomized into control and study group by computer generated randomisation. Children in the control group received only paracetamol (15mg/kg) at 5 minutes and combined group received paracetamol and tepid sponging at five minutes. Axillary temperature was monitored every 15 minutes for a period of 2 hours in both the groups.Results: There is no significant difference in reduction of temperature between the two groups by the end of two hours. Children in combined group had a higher level of discomfort than those in only antipyretic group.Conclusion: Tepid sponging does not add to the efficacy of paracetamol in antipyresis and that addition of tepid sponging to antipyretic, results in additional discomfort for the child. This study, therefore, endorses the view that antipyretic alone without tepid sponging should be the modality of therapy in children with fever.

Congenital missing teeth (OMIM #106600) is the most common dental abnormality. The aim of the study was to evaluate the effects of tooth agenesis on the total mandibular length, length of the mandibular body and alveolar process, and the mandibular anteroposterior position. The material was obtained from the Department of Orthodontics, Medical University of Warsaw. The study group included 116 patients aged 9–18 years with a congenital absence of at least two permanent tooth buds in the maxilla and/or mandible (mean: 6.2 teeth missing/patient). All patients were Caucasians: 68 (59%) females and 48 (41%) males. The control group included 115 patients without tooth agenesis matched with the age and gender of the study group. A cephalometric analysis was performed, and it was focused on assessing anteroposterior mandibular measurements. This assessment was based on 17 measurements (12 linear and 5 angular). Statistical analysis of the cephalometric measurements between the study group and the control group showed significant changes regarding selected mandibular measurements. Tooth agenesis does not affect the total length of the mandible and the length of the mandibular body, but it might reduce the length of the mandibular arch length and result in a more retrusive mandibular position.

The aim of the study was to detect significant biochemical changes in antioxidant status during acute period of ischemic stroke in its various pathogenic subtypes in patients aged 45–74years. The most important results were blood levels of ascorbic acid, malondialdehyde and free/ oxidized glutathione ratio. Cardioembolic stroke was associated with: the reduced ascorbic acid level of 62–74% comparable to control group; 5-times increased malondialdehyde level at the end of the first week of the disease; and also 50% decreasing of cell antioxidant capacity comparable to control group and other stroke subtypes. In lacunary stroke minimal changes in antioxidant status were measured, possibly because of small size of brain damage. Positive clinical dynamics was followed by decreasing of free glutathione levels. It could be related with its previous significant expenditure in process of cell recovery in penumbra area. In case of favorable outcome all of the markers return to the levels which were comparable to control group. The differences in the dynam-ics of the cells antioxidant capacity and consumption of native antioxidants, the time from the onset of the disease, the severity of the clinical picture, depending on the stroke subtype, were revealed. Further research of this problem may help to create new methods of diagnostics and selective pharmacological correction of this pathology.

AbstractBackground and objectives: Acute kidney injury (AKI) is a clinical condition associated with a degree of morbidity and mortality despite supportive care, and ischemia/reperfusion injury (I/R) is one of the main causes of AKI. The pathophysiology of I/R injury is a complex cascade of events including the release of free oxygen radicals followed by damage to proteins, lipids, mitochondria, and deranged tissue oxygenation. In this study, we investigated whether the antioxidant ascorbic acid would be able to largely prevent oxidative stress and consequently, reduce I/R-related injury to the kidneys in terms of oxygenation, inflammation, and renal failure. Materials and methods: Rats were divided into three groups (n = 6/group): (1) a time control group; (2) a group subjected to renal ischemia for 60 min by high aortic occlusion followed by 2 h of reperfusion (I/R); and (3) a group subjected to I/R and treated with an i.v. 100 mg/kg bolus ascorbic acid 15 min before ischemia and continuous infusion of 50 mg/kg/hour for 2 h during reperfusion (I/R + AA). We measured renal tissue oxidative stress, microvascular oxygenation, renal oxygen delivery and consumption, and renal expression of inflammatory and injury markers. Results: We demonstrated that aortic clamping and release resulted in increased oxidative stress and inflammation that was associated with a significant fall in systemic and renal hemodynamics and oxygenation parameters. The treatment of ascorbic acid completely abrogated oxidative stress and inflammatory parameters. However, it only partly improved microcirculatory oxygenation and was without any effect on anuria. Conclusion: The ascorbic acid treatment partly improves microcirculatory oxygenation and prevents oxidative stress without restoring urine output in a severe I/R model of AKI.

Mercury is a harmful toxic pollutant, which has hepato-nephrotoxic, hematotoxic, genotoxic and neurotoxic, effects. The aim of the study was to evaluate the protective efficacy of wheatgrass on mercuric chloride (HgCl2) induced oxidative stress and associated complications in rat model. Albino rats were divided into four groups (three rats per group). Group I normal control group. Group II oxidative stressed group received mercuric chloride (0.5 mg/kg/day). Group III only received wheatgrass extract (100 mg/kg/day), whereas Group IV received wheatgrass (100 mg/kg/day) after one hour, followed by mercuric chloride (0.5 mg/kg/day) for 30 days. The results of the study showed that wheatgrass supplementation significantly decreased the HgCl2 induced elevated oxidative stress parameters Plasma Malondialdehyde (MDA) content, Plasma membrane redox system (PMRS), Advanced oxidation protein products (AOPP), simultaneously elevated lipid profile (Total Cholesterol, Triglycerides, Low-density lipoprotein (LDL), liver enzymes as, Plasma Alkaline phosphatase (ALP), Aspartate aminotransferase (AST), and Alanine aminotransferase (ALT), Serum Urea, and Creatinine levels in rats. In addition, wheatgrass treatment improved the antioxidant status in terms of intracellular Reduced Glutathione (GSH), Ferric reducing antioxidant power (FRAP) and 2, 2- diphenyl -1- picrylhydrazyl (DPPH). Therefore it can be concluded that wheatgrass has great potential to diminish the stress-mediated complications and improve the antioxidant status.
