Plant viruses pose a serious threat to agricultural production systems worldwide. The world's population is expected to reach the 10-billion mark by 2057. Under the scenario of declining cultivable land and challenges posed by rapidly emerging and re-emerging plant pathogens, conventional strategies could not accomplish the target of keeping pace with increasing global food demand. Gene-editing techniques have recently come up as promising options to enable precise changes in genomes with greater efficiency to achieve the target of higher crop productivity. Of genome engineering tools, clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated (Cas) proteins have gained much popularity, owing to their simplicity, reproducibility, and applicability in a wide range of species. Also, the application of different Cas proteins, such as Cas12a, Cas13a, and Cas9 nucleases, has enabled the development of more robust strategies for the engineering of antiviral mechanisms in many plant species. Recent studies have revealed the use of various CRISPR-Cas systems to either directly target a viral gene or modify a host genome to develop viral resistance in plants. This review provides a comprehensive record of the use of the CRISPR-Cas system in the development of antiviral resistance in plants and discusses its applications in the overall enhancement of productivity and nutritional landscape of cultivated plant species. Furthermore, the utility of this technique for the detection of various plant viruses could enable affordable and precise in-field or on-site detection. The futuristic potential of CRISPR-Cas technologies and possible challenges with their use and application are highlighted. Finally, the future of CRISPR-Cas in sustainable management of viral diseases, and its practical utility and regulatory guidelines in different parts of the globe are discussed systematically.

Cytoplasmically transmissible agents causing diseases of plant pathogenic fungi characterized by reductions in pathogenicity, ability to form sexual and asexual spores, spore viability and growth rate, are often associated with the presence of one or more specific segments of virus-like double-stranded RNA (dsRNA). In Italy, hypovirulent dsRNA-containing strains of the chestnut blight fungus, Endothia ( Cryphonectria ) parasitica , have become predominant in many areas where blight is no longer a serious problem. dsRNA-containing strains of other pathogens, with various degrees of debilitation, survive in natural populations but have not become predominant or resulted in any great reduction in disease. Examples include the Dutch elm disease fungus, Ophiostoma ( Ceratocystis ) ulmi , and the wheat take-all fungus, Gaeumannomyces graminis var. tritici . Successful biological control of such pathogens could probably be achieved, however, if methods could be developed to suppress the loss of dsRNA that occurs during the sexual and other stages of their life cycles, and to suppress the vegetative incompatibility reactions that reduce the cytoplasmic transmission of dsRNA. Systemic infection with attenuated strains of plant viruses can protect plants from later infection by virulent strains of the same or closely related viruses. Despite some notable successes, e.g. control of citrus tristeza and tomato mosaic viruses, such ‘cross-protection’ has not been widely applied because of the cost and difficulty of application, and caution about the widespread distribution of infectious agents in the environment. These problems could be overcome if cross-protection could be achieved by the expression of a single viral gene rather than infection with intact virus, and consideration of possible mechanisms of cross protection suggests novel ways of producing virus-resistant plants.

Abstract Background The cotton aphid, Aphis gossypii Glov. (Hemiptera: Aphididae), is a major insect pest on a wide range of plants that causes high damage and transmits plant viruses. This study was carried out to evaluate an indigenous isolate, Beauveria bassiana (Bb), and extracts of 5 plant species: Psiadia penninervia, Pulicaria crispa, Euryops arabicus, Salvia officinalis, and Ochradenus baccatus against A. gossypii, as individual and combined treatments to estimate their compatibility under laboratory conditions. Also, the antifungal activity of these plant extracts against B. bassiana was evaluated. Results LC50 value was 8.64 × 104 spores/ml of Bb against A. gossypii, while LC50 values of the tested 5 plant extracts on A. gossypii were 103.64, 879.92, 747.90, 783.28, and 262.42 μg/ml for P. penninervia, P. crispa, E. arabicus, S. officinalis, and O. baccatus, respectively. Both P. penninervia and O. baccatus extracts had the highest antifungal activities against Bb and were significantly different from the other 3 plant extracts. After 24 h of treatment with the combination of Bb and each extract, no effect for these combinations on A. gossypii mortality was recorded. Meanwhile, 5 days after treatment, the combined treatments between Bb and each plant extract achieved a significant increase in mortality than that of the single treatment with Bb or plant extract, except for P. penninervia extract, which did not achieve a significant mortality increase when combined with B. bassiana than that of its single treatment. Conclusion P. penninervia extract was not compatible with B. bassiana, but the other tested 4 plant extracts were compatible with B. bassiana. These 4 plant extracts could be used to control aphids in combinations with B. bassiana. Further laboratory and field investigations are needed to examine the effects of these plant extracts on other insect pests or associated beneficial insects.

India is the second largest producer of rice in the world and it is the most important staple food grain. All India Coordinated Rice Improvement Project (AICRIP) was initiated with objective of conducting multi-location trials to identify suitable genotypes of high yield potential along with appropriate crop management practices. Since its inception AICRIP contributed significantly in meeting the growing demand both within and outside India. Significant progress has been achieved through AICRIP in terms of varietal release thereby increasing the crop productivity and also meeting the food and nutritional security. This paper makes a sincere effort in bringing out the significant achievements/milestones achieved under the AICRIP program and also gives a few directions for widening the areas under AICRIP.

Background:Monoterpenes are the main constituents of the essential oils obtained from plants. These natural products offered wide spectra of biological activity and extensively tested against microbial pathogens and other agricultural pests.Methods:Antifungal activity of 10 monoterpenes, including two hydrocarbons (camphene and (S)- limonene) and eight oxygenated hydrocarbons ((R)-camphor, (R)-carvone, (S)-fenchone, geraniol, (R)-linalool, (+)-menthol, menthone, and thymol), was determined against fungi of Alternaria alternata, Botrytis cinerea, Botryodiplodia theobromae, Fusarium graminearum, Phoma exigua, Phytophthora infestans, and Sclerotinia sclerotiorum by the mycelia radial growth technique. Subsequently, Quantitative Structure-Activity Relationship (QSAR) analysis using different molecular descriptors with multiple regression analysis based on systematic search and LOOCV technique was performed. Moreover, pharmacophore modelling was carried out using LigandScout software to evaluate the common features essential for the activity and the hypothetical geometries adopted by these ligands in their most active forms.Results:The results showed that the antifungal activities were high, but depended on the chemical structure and the type of microorganism. Thymol showed the highest effect against all fungi tested with respective EC50 in the range of 10-86 mg/L. The QSAR study proved that the molecular descriptors HBA, MR, Pz, tPSA, and Vp were correlated positively with the biological activity in all of the best models with a correlation coefficient (r) ≥ 0.98 and cross-validated values (Q2) ≥ 0.77.Conclusion:The results of this work offer the opportunity to choose monoterpenes with preferential antimicrobial activity against a wide range of plant pathogens.

Eleven published articles (4 reviews, 7 research papers) are collected in the Special Issue entitled “Organelle Genetics in Plants.” This selection of papers covers a wide range of topics related to chloroplasts and plant mitochondria research: (i) organellar gene expression (OGE) and, more specifically, chloroplast RNA editing in soybean, mitochondria RNA editing, and intron splicing in soybean during nodulation, as well as the study of the roles of transcriptional and posttranscriptional regulation of OGE in plant adaptation to environmental stress; (ii) analysis of the nuclear integrants of mitochondrial DNA (NUMTs) or plastid DNA (NUPTs); (iii) sequencing and characterization of mitochondrial and chloroplast genomes; (iv) recent advances in plastid genome engineering. Here we summarize the main findings of these works, which represent the latest research on the genetics, genomics, and biotechnology of chloroplasts and mitochondria.

Our previous study reported that fresh produce grown in aquaponic and hydroponic systems can pose potential food safety hazards due to an accidental introduction of contaminated fish and cross-contamination between the systems. In this study, we examined the effects of plant species and age on the likelihood and level of internalization of Shiga toxin-producing Escherichia coli (STEC) in aquaponic and hydroponic systems. Four plant species, basil (Ocimum basilicum L. cv. Genovese), cilantro (Coriandrum Sativum L.), lettuce (Lactuca sativa cv. Cherokee), and kale (Brassica oleracea var. sabellica), received root damage treatment as seedlings before transplanting or mature plants at three weeks after transplanting by cutting off 1-cm tips of one-third of the roots. Enrichments and selective media were used for the isolation, and presumptive positive colonies were confirmed by PCR for the presence of stx1 gene in plant tissues, recirculating water, and fish feces collected at four weeks after transplanting. In hydroponic systems, STEC was found neither in the solution nor in the roots and leaves of all four plant species, possibly through improved sanitation and hygiene practices. However, consistent with our previous findings, STEC was found in the water, on the plant roots, and in the fish feces in aquaponic systems, even after thorough sanitation prior to the study. Regardless of plant age, STEC was internalized in the roots of all plant species when the roots were damaged, but there was no difference in the degree of internalization with STEC among plant species. STEC was present in the leaves only when seedlings received root damage treatment and were grown to maturity, indicating that root damage allows STEC to internalize in the roots within a week, but a longer period is required for STEC to internalize into the leaves. We concluded that root damage on seedlings can cause the internalization of E. coli O157:H7 in the edible parts of leafy vegetables and herbs in soilless production systems.

Potable and good-quality drinking water availability is a serious global concern, since several pollution sources significantly contribute to low water quality. Amongst these pollution sources, several are releasing an array of hazardous agents into various environmental and water matrices. Unfortunately, there are not very many ecologically friendly systems available to treat the contaminated environment exclusively. Consequently, heavy metal water contamination leads to many diseases in humans, such as cardiopulmonary diseases and cytotoxicity, among others. To solve this problem, there are a plethora of emerging technologies that play an important role in defining treatment strategies. Phytoremediation, the usage of plants to remove contaminants, is a technology that has been widely used to remediate pollution in soils, with particular reference to toxic elements. Thus, hydroponic systems coupled with bioremediation for the removal of water contaminants have shown great relevance. In this review, we addressed several studies that support the development of phytoremediation systems in water. We cover the importance of applied science and environmental engineering to generate sustainable strategies to improve water quality. In this context, the phytoremediation capabilities of different plant species and possible obstacles that phytoremediation systems may encounter are discussed with suitable examples by comparing different mechanistic processes. According to the presented data, there are a wide range of plant species with water phytoremediation potential that need to be studied from a multidisciplinary perspective to make water phytoremediation a viable method.

AbstractThe purpose of this article was to give a comprehensive review of the published research works on biological control of different fungal, bacterial, and nematode plant diseases in Iran from 1992 to 2018. Plant pathogens cause economical loss in many agricultural products in Iran. In an attempt to prevent these serious losses, chemical control measures have usually been applied to reduce diseases in farms, gardens, and greenhouses. In recent decades, using the biological control against plant diseases has been considered as a beneficial and alternative method to chemical control due to its potential in integrated plant disease management as well as the increasing yield in an eco-friendly manner. Based on the reported studies, various species of Trichoderma, Pseudomonas, and Bacillus were the most common biocontrol agents with the ability to control the wide range of plant pathogens in Iran from lab to the greenhouse and field conditions.

The use of plant species to stabilize and accumulate trace elements in contaminated soils is considered of great usefulness given the difficulty of decontaminating large areas subjected to mining for long periods. In this work, the bioaccumulation of trace elements is studied by relating the concentrations in leaves and roots of three plants of Mediterranean distribution (Dittrichia viscosa, Cistus salviifolius, Euphorbia pithyusa subsp. cupanii) with the concentrations of trace elements in contaminated and uncontaminated soils. Furthermore, in the case of D. viscosa, to know the concentration of each element by biomass, the pool of trace elements was determined both in the aerial part and in the roots. The bioaccumulation factor was not high enough in any of the species studied to be considered as phytoextractors. However, species like the ones studied in this work that live on soils with a wide range of concentration of trace elements and that develop a considerable biomass could be considered for stabilization of contaminated soils. The plant species studied in this work are good candidates for gentle-remediation options in the polluted Mediterranean.

Sulphur plays crucial roles in plant growth and development, with its functions ranging from being a structural constituent of macro-biomolecules to modulating several physiological processes and tolerance to abiotic stresses. In spite of these numerous sulphur roles being well acknowledged, agriculture has paid scant regard for sulphur nutrition, until only recently. Serious problems related to soil sulphur deficiencies have emerged and the intensification of food, fiber, and animal production is escalating to feed the ever-increasing human population. In the wake of huge demand for high quality cereal and vegetable diets, sulphur can play a key role in augmenting the production, productivity, and quality of crops. Additionally, in light of the emerging problems of soil fertility exhaustion and climate change-exacerbated environmental stresses, sulphur assumes special importance in crop production, particularly under intensively cropped areas. Here, citing several relevant examples, we highlight, in addition to its plant biological and metabolism functions, how sulphur can significantly enhance crop productivity and quality, as well as acclimation to abiotic stresses. By this appraisal, we also aim to stimulate readers interests in crop sulphur research by providing priorities for future pursuance, including bettering our understanding of the molecular processes and dynamics of sulphur availability and utilization in plants, dissecting the role of soil rhizospherical microbes in plant sulphur transformations, enhancing plant phenotyping and diagnosis for nutrient deficiencies, and matching site-specific crop sulphur demands with fertilizer amendments in order to reduce nutrient use inefficiencies in both crop and livestock production systems. This will facilitate the proper utilization of sulphur in crop production and eventually enhance sustainable and environmentally friend food production.
