Nowadays, there is a growing interest for foods with a lower sugar content and rich in fiber and biologically active substances. The main purpose of this study was to prepare flakes from non-traditional pigmented cereals (Oryza sativa, Chenopodium quinoa, and Eragrostis tef) and to analyze their fibre, sugar, and in vitro digestibility values. Regarding phenolic antioxidants (soluble, soluble conjugated, and insoluble bound fractions), their content and antioxidant activity were measured using spectrophotometry and high performance liquid chromatography (HPLC) methods. Hydrothermally treated grains resulted in flakes with higher total dietary fibre contents (11.1–24.4%), quinoa and teff flakes were rich in maltose (up to 42.0 mg/g). Non-traditional flakes had lower in vitro digestibility, but conversely, they exhibited the highest phenolic contents corresponding with the highest antioxidant activity values (up to 2.33 mg Gallic acid equivalent/g of total phenolic content and 1.59 mg Trolox equivalent/g for 2,2-diphenyl-1-picrylhydrazyl (DPPH) in case of brown teff). Among free phenolics, the main contributors to an antioxidant activity were p-coumaric, o-coumaric, and gallic acids (r > 0.8186); among the soluble conjugated fractions, they were epigallocatechin, epicatechin, caffeic, and vanillic acids (r > 0.5935); while caffeic, protocatechuic, and ferulic acids (r > 0.5751) were the main contributors among the insoluble bound phenolics.

Fruits contain a number of useful substances including antioxidants. Their bio-accessibility after passing through the digestive tract is of primary importance when considering their benefits. In this respect, we investigated the effect of in vitro digestion on the phytochemicals of eight fruit juices. Freshly prepared juices from pomegranate, orange and grapefruit were used as well as commercially available juices from cherry, black grapes and aloe vera, blackberry and chokeberry, and two types of chokeberry and raspberries. Spectrophotometric and HPLC methods were used in order to analyse the sugar content, the total phenolic (TPC) and flavonoid contents (TFC), anthocyanins, phenolic acids and antioxidant activity. Principle component analysis was used to explain the differentiation among the types of fruit juice. Sugar recovery variation was between 4–41%. The bio-accessibility of TPC ranged from 13.52–26.49% and of flavonoids between 24.25–67.00%. The pomegranate juice and the juice of black grapes and aloe vera kept 58.12 and 50.36% of their initial anthocyanins content, while for the other samples less than 1.10% was established. As a result, a maximum of 30% remaining antioxidant activity was measured for some of the samples, but for most this was less than 10%. In conclusion, fruit juices are a rich source of biologically active substances, but a more detailed analysis of food transformation during digestion is needed.

The nutraceutical interest in quinoa (Chenopodium quinoa Willd.) seeds is associated with the presence of macronutrients, micronutrients, minerals, vitamins, and polyphenols. In particular, polyphenols contribute to the health-promoting effects of this food crop, and their levels are influenced by environmental conditions. Production of quinoa is recently being explored in temperate climate areas, including Italy. The aim of this research was to assess the profile of bioactive compounds in seeds of two quinoa varieties, Regalona-Baer and Titicaca, grown in northern Italy, compared to that of seeds of those varieties grown in Chile and Denmark, respectively. High-performance liquid chromatography-diode array detector (HPLC-DAD) analysis of phenolic acid and flavonoid profiles, both in their free and soluble conjugated forms, showed that the main differences between Regalona grown in Chile and Italy were for the free vanillic acid and daidzein contents, while the two Titicaca samples mainly differed in quercetin derivative levels. The total phenolic index was comparable in Titicaca and Regalona, and only a slight decrease in this parameter was found in seeds of the two varieties grown in Italy. The in vitro antioxidant activity of seed extracts, evaluated by means of three different assays, indicated that it correlated with flavonol (quercetin derivative) levels. In conclusion, the results indicate that, although environmental conditions alter the polyphenolic profile and biological activities, it is possible to grow good-quality quinoa in northern Italy.

The demand for organic production is increasing worldwide. The willowherb, grown in an organic way, contributes greatly to the idea of a healthier society and clean land. Willowherb is widespread in the world and has high polyphenols, carotenoids, and antioxidant properties. The purpose of this work was to investigate the influence of solid-phase fermentation (SPF) under different conditions on the variation of polyphenols and carotenoids in the organic leaves of willowherb. The leaves were fermented for different periods of time: 24, 48, and 72 h; and in aerobic and anaerobic conditions. The evaluation of polyphenols and carotenoids was completed using high-performance liquid chromatography (HPLC), and antioxidant activity was measured with spectrophotometric method. Hierarchical cluster analysis was used to describe differences in biologically active compounds between willowherb samples. The experiment showed that the highest quantities of total phenolic acids and flavonoids were determined after 24 h under aerobic SPF, but the amountof total carotenoids was higher after 72 h anaerobic SPF, compared to control. Not-fermented willowherb leaves had a lower antioxidant activity. compared to fermented leaves. In conclusion, SPF can be used to change polyphenol and carotenoid quantities in organic leaves of willowherb.

The aim of this study was to determine the best extraction conditions of phenolic compounds present in açaí, blueberry and goji berry fruits using the response surface methodology (RSM). The phenolic compounds profile by high-performance liquid chromatography, antioxidant activity was also determined. A factorial 23 design was used to analyze the effect of the solvent (ethanol and water), time (30 and 60 min) and temperature (30 °C and 60 °C) on the extraction of total phenolic compounds (TPC) and activity antioxidant (AA). The variables time and temperature had a positive effect on antioxidant activity (AA) in their highest levels, 60 min and 60 °C, respectively. The solvent ethanol 800 g/mL was more efficient in TPC extracting with AA in all matrices. Rutin was present in high amounts in blueberry and goji berry, and the myricetin in açaí. The açaí showed higher in vitro antioxidant activity when extracted at 60 °C for 60 min. The high correlation coefficient (0.98) of global response (GR) showed that we can find out single and global response in research with multiple dependent variables.  The GR analysis indicated the highest values of the TPC and AA when the fruits were extracted at 60°C for 60 min using ethanol as solvent and it was very useful for simplifying and improving the phenolic compounds extraction performance.

Objective: The present investigation focuses on the use of Cardiospermum halicacabum L. in their phytochemical and biological activities. Methods: In this study, in vivo stem and in vitro callus ethanolic extracts of C. halicacabum were tested for their phytochemical attributes by qualitative method, Fourier transform infrared (FTIR), antioxidant, antibacterial, and bioactive compound properties. The bactericidal activity of the in vivo stem and in vitro callus extract has been evaluated in both Gram+ve and Gram-ve microorganisms using the disk diffusion method. Results: The highest frequency (78%) of well developed, dark green organogenic callus was induced from stem explant on Murashige and Skoog (MS) medium supplemented with 0.7 mg/l 2,4-Dichlorophenoxyacetic acid (2, 4-D) and 0.5 mg/l benzyl adenine (BA). The results of FTIR spectra confirmed the presence of functional groups in wild stem and in vitro callus extract of C. halicacabum with various peaks. The total phenolic content in ethanolic extract of in vivo plant and in vitro callus was 80.46 mg gallic acid equivalent (GAE)/g dry weight and 76.4 mg GAE/g dry weight, respectively. The highest percentage of tannins was measured at 78.03 in wild stem ethanol extracts followed by 75.22 in callus extract. The antioxidant activity of 2,2-diphenyl-2- picrylhydrazyl (DPPH) ethanol extract was found to be 206.54 μg/ml. IC50 values of the stem extracts of C. halicacabum are 306 μg/ml and 286 μg/ml in callus extract, respectively. Antibacterial activity of the ethanol extract was higher for Staphylococcus aureus (S. aureus) with a 17 mm zone of inhibition. Conclusion: The present investigation recommended that the callus ethanolic extract function as a good source of biologically active compounds and natural antioxidants.

The article presents Dara on the content of biologically active compounds (BAC) and the total phenolic antioxidants activity (TPA) evaluated for leaves and inflorescences of eight species from the Rosaceae family: Padus avium, Malus baccata, Alhemilla vulgaris, Potentilla inquinans, Filipendula vulgaris, F. palmata, Sanguisorba officinalis, S. alpina. It was determined that leaves and inflorescences contain flavonols, catechins, tannins, carotenoids, saponins, pectin substances. Usually, woody plants tend to accumulate less BAC and TPA than herbs. The highest level of flavonols and tannins was registered in inflorescences of F. vulgaris (12.25% and 45.02 %). The content of pectin substances was quite high, with the biggest share of protopectins (4.4-10.65%). The maximum quantity of catechins (4.76%) was found in the leaves of F. palmata, saponins – in the inflorescences of M. baccata (20.77%). The highest content of carotenoids was found in the leaves of plants, most notably in P. inquinans (1.00 mg/g). The highest antioxidant activity was recorded for water-ethanol extracts from inflorescences (up to 7.77 mg/g) and leaves (2.11 mg/g) of F. vulgaris and A. vulgaris plants due to increased content of phenolic compounds, in particular flavonols and tannins, compared to the other species.

Thein vitroantioxidant and antibacterial assays against clinically isolatedStreptococcus suisandStaphylococcus intermediusof the extracts prepared by decoction and ethanolic reflux of different parts of Chettaphangki (Cladogynos orientalisZipp. ex Span), including the leaves, roots, and stems, using 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging assay and disc diffusion method were conducted. Quantitative analysis of total phenolic and total flavonoid contents in the extracts using spectrophotometric methods was also performed. Finally, phytochemical screening by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC) was conducted. Leaf ethanolic reflux extract (100 g) contained the highest total phenolic and total flavonoid contents of 7.21 ± 0.28 μg gallic acid equivalent (GAE) and 11.51 ± 2.02 μg rutin equivalent (RE), respectively. Chettaphangki extracts promoted low antioxidant activity with EC50values in the range of 0.27–0.48 mg/mL. Extracts and fractions from the roots and stems of this plant promoted low to intermediate antibacterial activity againstS. intermediuswith the inhibition zones between 7 and 14 mm. The chromatographic data suggested that the leaf extracts ofC. orientaliscontained rutin while the root and stem extracts contained scopoletin and chettaphanin I. Rutin promoted strong antioxidant activity while chettaphanin I showed low antibacterial activity againstStaphylococcus intermedius.

Fireweed has recently been recognized as a plant with high antioxidant potential and phenolic content. Its leaves can be fermented to prepare an infusion with ideal antioxidant activity. The aim of this study was to investigate and to determine the influence of solid-phase fermentation of different durations on the variation of polyphenols in the leaves of fireweed. Laboratory experiments were conducted in 2017–2018. The leaves of fireweed, naturally growing, were fermented for different periods of time: not fermented (control) and fermented for 24 and 48 h. The evaluation of polyphenols and antioxidant activity in leaves was performed using high- performance liquid chromatography (HPLC). Additionally, principal component analysis was used to characterize differences in bioactive compounds between fireweed samples fermented at different durations. Solid-phase fermented leaves were characterized by higher contents of oenothein B, quercetin and benzoic acid but had lower contents of quercetin-3-O-rutinoside, luteolin and chlorogenic and gallic acids. Antioxidant activity in short- (24 h) and long-term (48 h) fermentation (compared to control) gave the highest level of regression in 2017, but in 2018 the effect was observed only with short-term fermentation and control. In conclusion, solid-phase fermentation can be used to modulate biologically active compounds in fireweed leaves.

ABSTRACTCyanobacteria are natural enormous sources of various biologically active compounds with great contributions in different industries. This study aimed to introduce molecular and biochemical characterization for four novel cyanobacterial isolates obtained from Egyptian wastewater canals. Besides, In vitro biological activity of these isolates and their potential ability to take up nutrients and heavy metals from wastewater were examined. The obtained accession numbers were KY250420.1, KY321359.1, KY296359.1 and KU373076.1 for Nostoc calcicola, Leptolyngbya sp, Nostoc sp, and Nostoc sp, respectively. The isolate Leptolyngbya sp (KY321359.1) showed the lowest identity (90%) with other deposited sequences in database. While the isolate Nostoc sp (KU373076.1) showed the highest total phenolic content as well as the highest levels of caffeic, ferulic and gallic acids. Consequently, it appeared the highest antioxidant scavenging activity. All cyanobacterial isolates revealed potent ability to take up nutrients and heavy metals from wastewater. Generally, this study provides a taxonomic and molecular evidence for four novel cyanobacterial isolates with antioxidant activity and potent phycoremediation ability.
