Fucus vesiculosus L. is a common coastal brown seaweed associated with various benefits to human health due to its phenolic content and nutrients and is used as food through different methods of consumption. This study aims to evaluate the influence of the seaweed’s gender and growth stage on different types of biological activities as well as its chemical constitution and elements present. Akin to food preparation, aqueous extracts of the seaweed were prepared at 25 °C (salad) and 100 °C (soup). Biological activities were determined by measuring total phenol content (TPC), antioxidant activity and inhibition of acetylcholinesterase (AChE). Liquid Chromatography High Resolution Mass Spectrometry (LC-HRMS/MS) was used for compound identification, and elemental analysis was carried out by using Total Reflection X-ray Fluorescence Spectrometry (TXRF). Older females and males had higher TPC compared to the new ones at 100 °C. Antioxidant activity depended on the extraction temperature but was higher for the youngest male at 100 °C. AChE inhibitory activity was higher for older males at 25 °C, but at 100 °C it was higher for older females. Primary metabolites and various phloroglucinol were the main compounds identified. Additionally, since this seaweed is often harvested in estuarine systems with high anthropogenic impacts, its safety was evaluated through the evaluation of the sample’s metal content. The heavy metals detected are within the limits established by various regulating entities, pointing to a safe food source.

In this paper, ultrasonic cellulase extraction (UCE) was applied to extract polyphenols from passion fruit. The extraction conditions for total phenol content (TPC) and antioxidant activity were optimized using response surface methodology (RSM) coupled with a Box-Behnken design (BBD). The results showed that the liquid-to-solid ratio (X2) was the most significant single factor and had a positive effect on all responses. The ANOVA analysis indicated quadratic models fitted well as TPC with R2 = 0.903, DPPH scavenging activity with R2 = 0.979, and ABTS scavenging activity with R2 = 0.981. The optimal extraction parameters of passion fruit were as follows: pH value of 5 at 30 °C for extraction temperature, 50:1 (w/v) liquid-to-solid ratio with extraction time for 47 min, the experimental values were found matched with those predicted. Infrared spectroscopy suggested that the extract contained the structure of polyphenols. Furthermore, three main polyphenols were identified and quantified by HPLC. The results showed the content of phenolic compounds and antioxidant activity of the optimized UCE were 1.5~2 times higher than that determined by the single extraction method and the Soxhlet extraction method, which indicates UCE is a competitive and effective extraction technique for natural passion fruit polyphenols.

Fucus vesiculosus is a brown macroalgae used in food and generally considered safe to be consumed, according to EU Directive (EC 258/97). The aim of this study is to analyze the effect of food preparation on F.vesiculosus of different origins on what concerns its chemical constituents and final bioactivities. The aqueous extract of the seaweeds were obtained at different temperatures, similar to food preparation and then purified by SPE. The compound identification was carried out by Liquid Chromatography High Resolution Mass Spectrometry (LC-HRMS/MS) and algae extracts microstructure were observed by Scanning Electron Microscopy (SEM). The activities were determined by using antioxidant activity, inhibition of acetylcholinesterase (AChE) and 3-hidroxi-3-methyl-glutaril-CoA (HMG-CoA) reductase (HMGR) together with Caco-2 cells line simulating the intestinal barrier. The activity of AChE and the HMGR were inhibited by the extracts giving IC50 values of 15.0 ± 0.1 µg/mL and 4.2 ± 0.1 µg/mL, respectively and 45% of the cholesterol permeation inhibition. The main compounds identified were phlorotannins and peptides derivatives. The mode of preparation significantly influenced the final bioactivities. Moreover, the in vitro results suggest that the preparation of F. vesiculosus as a soup could have hypercholesterolemia lowering effect.

SummaryFucoidan is a highly complex sulfated polysaccharide commonly extracted from brown seaweed. In addition to their many biological activities, fucoidans have recently been demonstrated to inhibit or increase coagulation at different concentration ranges. Their structural features, i.e. molecular weight (Mw), Mw distribution, degree of sulfation, monosaccharide composition, and different linkages, are known to affect these activities. Therefore, structure-activity relationship (SAR) analysis of fucoidan is crucial for its potential use as a procoagulant. In this study, Fucus vesiculosus (F.v.) fucoidan was fractionated by charge and size as well as over- and desulfated to different degrees to yield preparations with various structural properties. The fractions’ pro- and anticoagulant activities were assessed by calibrated automated thrombography (CAT) and activated partial thromboplastin time (aPTT) assays. Binding to and inhibition of the anticoagulant protein tissue factor pathway inhibitor (TFPI) and the ability to activate coagulation via the contact pathway were also investigated. This paper discusses the impact of charge density, size, and sugar composition on fucoidan’s pro- and anticoagulant activities. Fucoidan requires a minimal charge density of 0.5 sulfates per sugar unit and a size of 70 sugar units to demonstrate desired procoagulant activities for improvement of haemostasis in factor VIII/factor IX-deficient plasma.

The objective of this study was to investigate the efficiency of phenolic extraction and antioxidant activity from sesame cake using water extraction method and to evaluate the possibility of employing microwave irradiation to improve the extraction yield. The result showed that extraction temperature had major influence on total phenolic content and antioxidant activities of the extracts, whereas extraction time was found to be insignificant. The optimum extraction condition recommended were 90oC for 30 min in this research. Furthermore, microwave pre-treatment at 120 s could have significantly positive influence on the overall extraction yield, especially the total phenolics and antioxidants based on FRAP assay. Therefore, the obtained results suggest that sesame aqueous extracts could be a source of antioxidants with more feasible applications in food as well as other industries.

Objective: This study aimed to determine the stability of microspheres of Sargassum plagyophyllum (brown seaweed) after preparation using spraydrying with maltodextrin DE 10–15 and during drying and storage.Methods: Aqueous extracts of brown seaweed were formulated into microspheres using maltodextrin DE 10–15 as a coating agent. For increasing thestability of polyphenol compounds, spray drying was performed with an inlet temperature of 110°C. Four microsphere formulations were producedusing maltodextrin DE 10–15 at concentrations of 0%, 5%, 10%, and 15%. The resulting microspheres were then characterized in the assessments ofmoisture contents, particle size distributions, pH, total phlorotannin contents, and antioxidant activity, and surface morphology was analyzed usingscanning electron microscope analyses.Results: Powders that were produced with 0% and 15% maltodextrin were more stable at 4°C±2°C than at 28°C±2°C and 40°C±2°C. At the lowesttemperature, phlorotannin contents were maintained in powders that were prepared with 15% maltodextrin but were decreased by 10% in powdersthat were prepared without maltodextrin.Conclusion: Maltodextrin DE 10–15 is a suitable coating agent for dry formulations of S. plagyophyllum powder and maintained stability during spraydrying at 110°C and during storage for 2 months at 4°C±2°C.

In Mexico, mistletoes have several applications in traditional medicine due to the great variety of compounds with biological activities that have not been characterized to date. The goals of the present study are to analyze the composition of minerals and phytochemical compounds in Mexican mistletoes Phoradendron bollanum and Viscum album subs. austriacum qualitatively and quantitatively, identify the compounds using HPLC-MS, and assess the antimicrobial potential in phytopathogenic microorganism control. Mineral content was evaluated with X-ray fluorescence. Three types of extracts were prepared: ethanol, water, and aqueous 150 mM sodium chloride solution. Characterization was carried out using qualitative tests for phytochemical compound groups, analytical methods for proteins, reducing sugars, total phenol, flavonoids quantification, and HPLC-MS for compound identification. The antimicrobial activity of mistletoe’s liquid extracts was evaluated by microplate assay. K and Ca minerals were observed in both mistletoes. A qualitative test demonstrated alkaloids, carbohydrates, saponins, flavonoids, tannins, and quinones. Ethanolic extract showed flavonoids, 3845 ± 69 and 3067 ± 17.2 mg QE/g for Phoradendron bollanum and Viscum album subs. austriacum, respectively, while aqueous extracts showed a total phenol content of 65 ± 6.9 and 90 ± 1.19 mg GAE/g Phoradendron bollanum and Viscum album subs. austriacum, respectively. HPLC-MS identified largely hydroxycinnamic acids and methoxycinnamic acids. Clavibacter michiganenses was successfully inhibited by aqueous extract of both mistletoes.

The purpose of this study was to determine the effects of convective air-drying at different temperatures (35, 50, 60 and 75°C) on the color of Bifurcaria bifurcata (BB) seaweed powders obtained after milling, the antioxidant activity and polyphenolic and carbohydrate content of the aqueous extracts obtained by ultrasound-assisted extraction. BB seaweed powders exhibited significant color differences between powders obtained from BB dried at 35ºC (yellowish-green) and 50–75 °C (brown). High air drying temperature (above 60ºC) significantly reduced the total polyphenolic, carbohydrate content and scavenging activity of aqueous extracts of BB. Objective: This study focuses on the antioxidant potential of Djibouti brown seaweed and their phenolic contents. Methods: We evaluated the antioxidant potential by DPPH method (1,1-diphenyl-2-picrylhydrazyl) and their phenolic contents of seven Djibouti seaweed: Cytoseira myrica, Padina pavonica, Sargassum fluitans, Sargassum ilifolium, Sargassum sp, Turbinaria triquetra and Turbinaria turbinata. Also, we searched the secondary metabolites of these seaweeds. Results: We obtain a higher antioxidant activity at 60,7±0,9 % and a higher phenolic content at 199,01±0,5 μg equivalent phloroglucinol (PGE)/g dry matter for Padina pavonica. A good linear correlation (R2 = 0,898) is observed between the antioxidant activity and the phenolic content of the seaweed studied. Also, two Padina pavonica collected in two different locations have different biochemical concentrations and antioxidant activity, suggesting the influence of the marine environment on the biosynthesis of secondary metabolites and the biological activities of seaweed. The present study shows the presence of tannins, saponosides, flavonoids and steroid-terpenes. Conclusion: The species studied show interesting antioxidant activities and can be consumed to prevent oxidative stress.

Nirwana Beach is one of the tourist destinations in Padang West Sumatra, but almost every day littered by brown seaweed that washed up along the coast. Very unfortunate, the people around still consider this waste and do not know the usefulness and benefits. Brown seaweed has been widely studied. The content consist of alginate, fukosantin and secondary metabolites namely fucoidan. Fucoidan has been made into a supplement as antioxidant, anticoagulant and antithrombotic, antiviral, anticancer, antidiabetic, immunomodulating, antiinflammatory, antilipidemic and antifertilization. The purpose of the study was to determine the antioxidant activity of polyphenol extract from brown seaweed. The brown seaweed was extracted using aquades by heating and without heating, then the extract was evaporated with a rotary evaporator up to a volume of 50 ml extract and then analyzed the total phenol and its antioxidant activity test. The results showed extraction with heating had higher total phenol content than without heating. The total phenol of brown seaweed extract with heating is 669.33 mg galic acid/g, without heating 352.5 mg of gallic acid/g and on seaweed 538.5 mg gallic acid/g. The antioxidant activity of brown seaweed extract is very strong ie 4 ug/ml with heating and 16.6 ug/ml without heating.

Background: Bougainvillea x buttiana has long been used as a Mexican conventional medicinal drug to cure different sicknesses. Objective: In this work, the consequences of ethanol combination and temperature extraction for phenolic compound recovery with the biological activity in Bougainvillea x buttiana extracts were investigated. Materials and Methods: The phenolic recovery from Bougainvillea x buttiana was determined by comparing the effects of ethanol 0%, 50-100% (v/v) and extraction temperature (26, 45 and 64°C). The total phenolic content was investigated using spectrophotometric methods. Antioxidant activity from different Bougainvillea x buttiana extracts was evaluated by determining their potential to scavenge the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical. The chemical compositions and cytotoxic effect of extracts were determined using GC-MS techniques and the crystal violet technique, respectively. Results: Our results confirmed that all extractions with different concentrations and temperatures had significant effects over the total phenolic contents and antioxidant activity (DPPH) of the extracts. The best recuperation of the total phenol content was observed in extracts with 60% ethanol at 26°C (201.08 mgEGA/g dry extract). Extracts with 100% ethanol at 26°C showed the highest antioxidant activity (IC50 was 286.75 μg/mL). GC-MS revealed the presence of various phytochemical contents with polar and non-polar properties. The phenolic compounds were investigated as to the structure, quantity and position of the hydroxyl groups on the benzene ring. Conclusion: The ethanolic extract of Bougainvillea x buttiana was confirmed to have high phenolic content and contain pharmacologically active compounds, with potent antioxidant effects and low cytotoxic effects.
