Photochemotherapy is one of the most interesting current therapeutic approaches for the treatment of melanoma. Different classes of naturally occurring phytochemicals demonstrated interesting photoactive properties. The aim of this study was to evaluate the photocytotoxic potential of two Cachrys species from Southern Italy: C. sicula and C. libanotis (Apiaceae). The enriched-coumarin extracts were obtained from aerial parts through both traditional maceration and pressurized cyclic solid-liquid (PCSL) extraction using Naviglio extractor®. Qualitative and quantitative analyses of furanocoumarins were performed with GC-MS. The photocytotoxic effects were verified on C32 melanoma cells irradiated at a dose of 1.08 J/cm2. The apoptotic responses were also assessed. Moreover, phenolic content and the in vitro antioxidant potential were estimated. Xanthotoxin, bergapten, and isopimpinellin were identified. All the samples induced concentration-dependent photocytotoxic effects (IC50 ranging from 3.16 to 18.18 μg/mL). The C. libanotis sample obtained with Naviglio extractor® was the most effective one (IC50 = 3.16 ± 0.21 μg/mL), followed by C. sicula sample obtained with the same technique (IC50 = 8.83 ± 0.20 μg/mL). Both Cachrys samples obtained through PCSL induced up-regulation of apoptotic signals such as BAX (Bcl2-associated X protein) and PARP (poly ADP-ribose polymerase) cleavage. Moreover, these samples proved to be more photoactive, giving a greater upregulation of p21 protein in the presence of UVA radiation. Obtained results suggest that investigated species could be promising candidates for further investigations aimed to find new potential drugs for the photochemotherapy of skin cancer.

Olax scandens Roxb. (family Olacaceae) available in throughout tropical India. The current study, aerial parts of different concentrates of Olax scandens was evaluated for its in-vitro antioxidant potential by FRAP assay taking ascorbic acid as the standard and estimation of total phenolic content and flavonoids content. The IC50 value was originated that methanolic concentrates of Olax scandens are more efficient in antioxidant activity by FRAP methods compared EA & PE concentrates. The methanolic concentrates of Olax scandens & ascorbate exhibited antioxidant potential possessing IC50 207µg/ml & 50µg/ml by Ferric reducing ability Power assay. The methanolic and EA concentrates of Olax scandens showed the total phenolic content (14.426 ± 0.032, 4.128 ± 0.025) respectively, and ৒lavonoids content (11.526 ± 0.054, 3.682 ± 0.042) respectively. Invitro antioxidant studies show methanolic concentrates of Olax scandens have better antioxidant activity as well as a higher content of total phenolic and flavonoids content. These results indicate that aerial parts of methanolic concentrates Olax scandens could serve as a natural antioxidant, which may be useful in preventing free radical-induced diseases.

Postprandial hyperglycemia is a prime characteristic of diabetes mellitus and has been a focus in the therapy for diabetes. One of the therapeutic approaches which involve decreasing hyperglycemia aims at inhibiting the enzyme α-amylase. The leaves of T. populnea were studied for the presence of amylase inhibitors. The fractions obtained by successive fractionation using solvents of varying polarity were studied for the presence of primary and secondary metabolites. The total phenolic content of the different fractions was determined by HPLC and was correlated with their amylase inhibitory potential. Similarly, the protein content of the extracts was also estimated to understand the nature of the inhibitor present. This study shows that the leaves of T. populnea were effective in inhibiting α-amylase, thereby proving to be potential hyperglycemic agents.

In this study <em>Amaranthus cruentus</em> plants were grown under controlled optimal conditions (28/21 °C) for three months and then subjected to cold (14/7 °C) and hot (33/40 °C) temperatures. We investigated the influence of these temperature regimes on the metabolite profile of the leaves through analyses of data by TLC, HPLC and GC-MS spectrometry. The phytotoxic potential of a methanol-water (MW) and dichloromethane (DCM) extract from the aerial parts were examined through in vitro screening of germination and growth of lettuce and pepper. The optimal extracts displayed the highest diversity of secondary metabolites, and the highest total phenolics and flavonoids content. Through TLC and HPLC analysis the significantly lower phenolic content in the hot temperature treated samples was confirmed. A wide range of metabolites were detected in the DCM extracts through GC-MS analyses. The phytotoxicity of both the MW and DCM extracts were demonstrated, as germination and growth of pepper and lettuce were significantly inhibited, indicating the presence of more than one allelochemical compound which may affect the allelopathic activity of <em>A. cruentus</em> during changes in environmental temperatures.

Steam-distilled aerial parts of Ravensara Aromatica and Cinnamomum camphora from Madagascar and Leptospermum scoparium from New Zealand have been subjected to qualitative and quantitative analysis by means of GC techniques. This allowed the elucidation of conflicting data present in the available literature for these species. Also, the biological activity in vitro was evaluated by measuring MICs and GIZs.

ABSTRACT The trachea delivers inhaled air into the lungs for gas exchange. Anomalies in tracheal development can result in life-threatening malformations, such as tracheoesophageal fistula and tracheomalacia. Given the limitations of current therapeutic approaches, development of technologies for the reconstitution of a three-dimensional trachea from stem cells is urgently required. Recently, single-cell sequencing technologies and quantitative analyses from cell to tissue scale have been employed to decipher the cellular basis of tracheal morphogenesis. In this Review, recent advances in mammalian tracheal development and the generation of tracheal tissues from pluripotent stem cells are summarized.

Artemisia absinthium L. is a medicinal plant largely used in traditional medicine. The aim of this study was to estimate the content of polyphenols, and flavonoids compounds and also to evaluate the antioxidant and the anti-ulcer activities of the Aqueous extract from Artemisia absinthium L. aerial parts. The Folin-Ciocalteau and AlCl3 methods were applied in order to quantify the polyphenolic and flavonoids contents, respectively. However, DPPH   method was used to evaluate the in vitro antioxidant activity. Quantitative analysis of the yield and phenolic content of the aqueous extract of Artemisia absinthium showed that the yield of the aqueous extract was 19.32% and its phenolic content was 58.66 ± 2.16 μg GAE / mg dry extract for polyphenols and 6.85 μg QE / mg dry extract for the flavonoids. The antioxidant activity of the plant extract evaluated by the DPPH test is very important (IC50=45.48±0.37 µg/ml). Treatment of mice with the aqueous extract of Artemisia absinthium at a dose of 400 mg / kg significantly reduced the ulcerogenic effect of ethanol on the gastric wall with an estimated protection rate of 91%. These findings suggest that Artemisia absinthium L.  aqueous extract possessed good antiulcer and antioxidant potentials. This supports the traditional claims of this plant in folklore medicine. Inorganic particulate material of diverse types is present in the ambient and occupational environment, and exposure to such materials is a well recognized cause of some lung disease. To investigate the interaction of inhaled inorganic particulates with the lung it is necessary to obtain quantitative information on the particulate burden of lung tissue in a wide variety of situations. The vast majority of diagnostic and experimental tissue samples (biopsies and autopsies) are fixed with formaldehyde solutions, dehydrated with organic solvents and embedded in paraffin wax. Over the past 16 years, I have attempted to obtain maximal analytical use of such tissue with minimal preparative steps. Unique diagnostic and research data result from both qualitative and quantitative analyses of sections. Most of the data has been related to inhaled inorganic particulates in lungs, but the basic methods are applicable to any tissues. The preparations are primarily designed for SEM use, but they are stable for storage and transport to other laboratories and several other instruments (e.g., for SIMS techniques).

SummaryWe evaluated the in vivo anti-metastatic activity of recombinant Ancylostoma caninum Anticoagulant Peptide (rAcAP), a potent (Ki = 265 pM) and specific active site inhibitor of human coagulation factor Xa originally isolated from bloodfeeding hookworms. Subcutaneous injection of SCID mice with rAcAP (0.01-0.2 mg/mouse) prior to tail vein injection of LOX human melanoma cells resulted in a dose dependent reduction in pulmonary metastases. In order to elucidate potential mechanisms of rAcAP’s anti-metastatic activity, experiments were carried out to identify specific interactions between factor Xa and LOX. Binding of biotinylated factor Xa to LOX monolayers was both specific and saturable (Kd = 15 nM). Competition experiments using antibodies to previously identified factor Xa binding proteins, including factor V/Va, effector cell protease receptor-1, and tissue factor pathway inhibitor failed to implicate any of these molecules as significant binding sites for Factor Xa. Functional prothrombinase activity was also supported by LOX, with a half maximal rate of thrombin generation detected at a factor Xa concentration of 2.4 nM. Additional competition experiments using an excess of either rAcAP or active site blocked factor Xa (EGR-Xa) revealed that most of the total factor Xa binding to LOX is mediated via interaction with the enzyme’s active site, predicting that the vast majority of cell-associated factor Xa does not participate directly in thrombin generation. In addition to establishing two distinct mechanisms of factor Xa binding to melanoma, these data raise the possibility that rAcAP’s antimetastatic effect in vivo might involve novel non-coagulant pathways, perhaps via inhibition of active-site mediated interactions between factor Xa and tumor cells.

Pergularia daemia belongs to the family Asclepiadaceae, known to have anticancer, anti-inflammatory activity. Aim of the present study was to evaluate qualitative and quantitative phytochemical and antioxidant properties of ethanolic extracts of leaf, stem and root parts of P. daemia . Preliminary phytochemical analysis and in vitro antioxidant properties were evaluated by standard methods. The qualitative phytochemical analysis of P. daemia showed presence of flavonoids, tannins, alkaloid, phytosterol, carbohydrate, phenol, saponin, glycosides, terpenoids, steroids proteins and reducing sugars. Quantitative analysis showed polyphenol, flavonoid, flavonone, flavone and flavonol in P. daemia leaves, stem and root in considerable quantity. The in vitro antioxidant activity of P. daemia clearly demonstrated that leaf, stem and root parts have prominent antioxidant properties and was effective in scavenging free radicals.
