Aims: To optimize the culture media of 0288 Tricholoma matsutake and to identify it according to its rDNA ITS sequence. Methods: Fruit body, collected from Heilongjiang province, is used as the material, and the mycelium is isolated and radiated under ultraviolet. Then the radiated mycelium is cultured in five different media and its morphology of fungus colony and growth rate are investigated. After that, the rDNA ITS sequences of 0288 and the natural fruit body of T. matsutake are analyzed and identified by PCR and sequencing. Results: The radiated mycelium grew faster in formula A culture medium than others and its average growth rate is up to 0.575mm/d. Results also showed that the homology of the rDNA ITS sequence is almost up to 99% between 0288 and the natural fruit body. Through this experiment, the best growth condition of 0288 is obtained and its fruit body is induced as pure cultural product. It would lay strong foundation for the artificial domestication of the wild T. matsutake.

AbstractThe aim of the study was to investigate the potential of microalgal cultivation on anaerobic liquid digestate as a growth medium. The two methods of liquid digestate treatment including centrifugation and distillation and the two algal strains (Chlorella vulgaris and Arthrospira platensis) were compared. Additionally, the volume of the liquid digestate used to prepare the culture medium constituted from 10 to 50% of the medium volume. The study demonstrated that the highest C. vulgaris and A. platensis biomass productions of 2490 mg TS/L and 2990 mg/L, respectively, were obtained by adding 50% of distilled digestate to a growth medium. Regarding centrifuged liquid digestate, only 10% dilution was required to obtain the maximum final biomass concentration. A. platensis removed 81.1% and 66.4% of the total nitrogen from medium prepared on distilled and centrifuged digestate, respectively, while C. vulgaris ensured 64.1% and 47.1% of removal, respectively. The phosphorus removal from both culture media was higher than 94.2% with A. platensis, while it was 70.4% from distilled and 87.4% from centrifuged media with C. vulgaris. The study confirmed a great potential of microalgal biomass production on anaerobic liquid digestate with a high treatment efficiency of digestate.

Biomass such as agriculture waste and urban waste are enormous potency as energy resources instead of enviromental problem. organic waste can be converted into energy in the form of liquid fuel, solid, and syngas by using of pyrolysis technique. Pyrolysis process can yield higher liquid form when the process can be drifted into fast and flash response. It can be solved by using microwave heating method. This research is started from developing an experimentation laboratory apparatus of microwave-assisted pyrolysis of biomass energy conversion system, and conducting preliminary experiments for gaining the proof that this method can be established for driving the process properly and safely. Modifying commercial oven into laboratory apparatus has been done, it works safely, and initial experiments have been carried out, process yields bio-oil and charcoal shortly, several parameters are achieved. Some further experiments are still needed for more detail parameters. Theresults may be used to design small-scale continuous model of productionsystem, which then can be developed into large-scale model that applicable for comercial use.

The minimum size of coral transplants, Acropora formosa, was assessed to support their survival and growth. For this, 150 coral fragments of different sizes (5, 10, 15 cm) were transplanted close to the donor colony. Their survivorship and growth were observed for 12 months. At the end of the observation time, 90% of 15 cm-transplanted coral fragments survived, while the others (10cm and 5 cm) did 86% and 82% respectively. The average growth rate of 5 cm-coral fragments was 0.860 cm/month, while 10 and 15 cm-fragments were 0.984 cm/month and 1.108 cm/month respectively. One-way ANOVA showed that there was significant difference (p<0.05) among the three (5, 10, 15 cm) transplant initial sizes in which the longest fragment size tended to survive longer than the smaller one.  However, the smaller transplants grew better than the bigger one, 10.318 cm/year (206%) for 5 cm-transplant, 11.803 cm/year (118%) for 10 cm-transplant, and 13.299 cm/year (89%) for 15 cm-transplant, respectively.

In the production of astaxanthin from Haematococcus pluvialis, the process of growing algal biomass in the vegetative green stage is an indispensable step in both suspended and immobilized cultivations. The green algal biomass is usually cultured in a suspension under a low light intensity. However, for astaxanthin accumulation, the microalgae need to be centrifuged and transferred to a new medium or culture system, a significant difficulty when upscaling astaxanthin production. In this research, a small-scale angled twin-layer porous substrate photobioreactor (TL-PSBR) was used to cultivate green stage biomass of H. pluvialis. Under low light intensities of 20–80 µmol photons m−2·s−1, algae in the biofilm consisted exclusively of non-motile vegetative cells (green palmella cells) after ten days of culturing. The optimal initial biomass density was 6.5 g·m−2, and the dry biomass productivity at a light intensity of 80 µmol photons m−2·s−1 was 6.5 g·m−2·d−1. The green stage biomass of H. pluvialis created in this small-scale angled TL-PSBR can be easily harvested and directly used as the source of material for the inoculation of a pilot-scale TL-PSBR for the production of astaxanthin.

Mortality and growth rates of trees in various crown classes and size classes were analyzed from 40-year permanent plot records of slope and ravine forest dominated by chestnut oak (Quercusprinus L.) and northern red oak (Quercusrubra L.). Average 5-year mortality rates for suppressed trees ≥2.5 cm dbh of chestnut oak and red oak in the slope forest were 26 and 45%, respectively. None of the suppressed red oaks survived the 40-year period, compared with 14% of the chestnut oaks and 33% of the red maples (Acerrubrum L.). Mortality of oak trees in the intermediate crown class was less than half that of suppressed trees, but still much higher than that of maples and birches on the tracts. Survival was reasonably high for oaks as long as the top of the crown was receiving direct sunlight, but the expected 40-year survival rate of red oaks in such a position is only 20%, with an average growth rate of 1.0 mm in diameter per year. Curves and equations expressing average mortality and growth rates at various levels of competition are presented for each species.

Cocoa is commodities third largest foreign exchange earner after oil palm and rubber. In 2012 production totaled 833 310 tonnes of cocoa with an average growth of 1.56%. There are two major challenges in the development of the national cocoa industry, the increase in quantity (production and productivity) and quality. To support these efforts the role of research and development is needed to ensure that the procedures adopted are appropriate and fit. This study aims to study the role of research and development and other supporting factors in addressing the problems of the national cocoa. The study is in the form of survey respondents which are R&D institutes, universities and cocoa processing industry. The results show that there is a problem of national cocoa industry in all aspects, here the cultivation aspect is the biggest problem. The next aspects are mentoring, institutional, business scale, quality, policy, processing of intermediate products and processing of downstream products. The study also shows that there is still a lot of research results that can not be utilized by the cocoa industry, the rest has to be applied even if but only for a small scale so that the less significant. In the small and medium scale of cocoa enterprises, the equipment used is generally low capacity so inefficient.
