Ginseng is an ancient herb widely consumed due to its healing property of active ginsenosides. Recent researchers were explored to increase its absorption and bioavailability of ginsenosides at the metabolic sites, due to its pharmacological activity. The purpose of this study was to investigate the isolation and characteristics of components obtained by a shorter steaming cycle (seven cycles) of white ginseng to fermented black ginseng, using a novel strain of Aspergillus niger KHNT-1 isolated from fermented soybean. The degree of bioactive of Rg3 increased effectively during the steaming process, and biotransformation converted the color towards black along active ginsenosides. Glycol moiety associated with C-3, C-6, or C-20 underwent rapid biotransformation and hydrolysis, such as Rb1, Rb2, Rc, Rd → Rg3, F2, and was converted to CK. Dehydration produces Rg3 → Rk1, Rg5. Rh2 → Rk2; thus, converted fermented black ginseng was solvent-extracted, and the isolated components were identified by TLC, HPLC, and quantification by LCMS. The unique composition obtained during this process with Rk1, Rg3, Rg5, and CK is nontoxic to HaCaT cell line up to 200 ug/mL for 24 h and was found to be effective in B16BL6 cell lines, in a dose- and time-dependent manner. Thus, it is a suitable candidate for nutraceuticals and cosmeceuticals.

Black ginseng (BG) has better health benefits than white ginseng. The intake of BG changes the levels of metabolites, such as amino acids, fatty acids, and other metabolites. However, there is no research on the effect of BG extract intake on the metabolic profile of dog serum. In this study, serum metabolic profiling was conducted to investigate metabolic differences following the intake of BG extracts in beagle dogs. The beagle dogs were separated into three groups and fed either a regular diet (RD, control), RD with a medium concentration of BG extract (BG-M), or RD with a high concentration of BG extract (BG-H). Differences were observed among the three groups after the dogs ingested the experimental diet for eight weeks. The concentrations of alanine, leucine, isoleucine, and valine changed with the intake of BG extracts. Furthermore, levels of glycine and β-alanine increased in the BG-H group compared to the control and BG-M groups, indicating that BG extracts are associated with anti-inflammatory processes. Our study is the first to demonstrate the potential anti-inflammatory effect of BG extract in beagle dogs. Glycine and β-alanine are proposed as candidate serum biomarkers in dogs that can discriminate between the effects of ingesting BG-H.

Delineation of factors which affect wound healing would be of immense value to enable on-time or early healing and reduce comorbidities associated with infections or biochemical stress like diabetes. Plasma gelsolin has been identified earlier to significantly enable injury recovery compared to placebo. This study evaluates the role of rhuGSN for its antioxidant and wound healing properties in murine fibroblasts (3T3-L1 cell line). Total antioxidant capacity of rhuGSN increased in a concentration-dependent manner (0.75-200 μg/mL). Cells pretreated with 0.375 and 0.75 μg/mL rhuGSN for 24 h exhibited a significant increase in viability in a MTT assay. Preincubation of cells with rhuGSN for 24 h followed by oxidative stress induced by exposure to H2O2 for 3 h showed cytoprotective effect. rhuGSN at 12.5 and 25 μg/mL concentration showed an enhanced cell migration after 20 h of injury in a scratch wound healing assay. The proinflammatory cytokine IL-6 levels were elevated in the culture supernatant. These results establish an effective role of rhuGSN against oxidative stress induced by H2O2 and in wound healing of 3T3-L1 fibroblast cells.

ABSTRACT A number of seaweed species are used as traditional foods and medicine in different parts of the world, including Asian countries. However, very few data on the anti-melanogenic effect of seaweed have been published. Undaria pinnatifida (Dolmiyeok), a brown alga, is a traditional food in Jeju Island, the southern regions of the Korea peninsula. In this study, ethylacetate extracts of U. pinnatifida (UPE) were examined for their anti-melanogenic potentials. Our results supports the finding that UPE down-regulated melanin content in a dose-dependent pattern. To clarify the target of UPE action in melanogenesis, we performed Western blotting for tyrosinase and microphthalmia-associated transcription factor (MITF), which are key melanogenic enzymes. UPE inhibited tyrosinase and MITF expressions in a dose-dependent manner. These results indicate that treatment with UPE significantly inhibits the melanogenesis in B16 cells, and may be effective in the whitening agent for the skin

Background: Pectin is a heterogeneous polysaccharide mainly present in citrus fruits and has different biological activities.Objective: High molecular weight Citrus Pectin and modified citrus pectin (MCP) were tested for their cytotoxic, anti-proliferative, and anti-oxidant activity.Methods: The cytotoxicity of pectin was studied against HaCaT cell line (human keratinocyte cell line) using Trypan blue method and LDH-cytotoxicity assay. Anti-proliferative activity was assayed using a WST-1 proliferation kit. Antioxidant activity was determined using the DPPH scavenging assay.Results: MCP and Pectin both reduced the viability of HaCaT cells in a dose dependent manner; however MCP was found to be more cytotoxic than high molecular weight citrus pectin since it had a lower IC50 (300ug/ul). At non-cytotoxic concentrations, the viability of cells decreased with increase of concentration of MCP as determined by the WST-1. MCP exhibited a higher antioxidant effect than pectin (SC50 at a concentration range between 2 and 4mg/ml).Conclusion: This study suggests that MCP exhibits a stronger cytotoxic and anti-proliferative effect on HaCaT cell line than pectin. The most probable explanation of this observation is the different effect due to the variable molecular weight and exposed side-chains of MCP and high molecular weight citrus pectin. Keywords: Cytotoxic, Anti-proliferative, Pectin, MCP, HaCaT cell line

Black ginseng exhibits numerous pharmacological activities due to higher and more diverse ginsenosides than unprocessed white ginseng. The ginsenoside derivatives have been investigated in order to determine their chemical structures and pharmacological activities. We found a peak which was increased 10-fold but unidentified in the methanol extracts of a black ginseng product. The unknown peak was tracked and identified as linoleic acid rather than a ginsenoside derivative using liquid chromatography–tandem mass spectrometry (LC-MS/MS) and nuclear magnetic resonance (NMR) spectroscopy. NMR analysis confirmed no presence of conjugated linoleic acids. Ginsenoside profiles and linoleic acid contents in black ginseng products were quantified using LC-MS/MS. Linoleic acid content was more directly proportional to the number of applied thermal cycles in the manufacturing process than any ginsenosides.

Introduction and Aim: Flacourtia indica (Burm.f.) Merr. is a member of the family Flacourtiaceae. This herb was used to treat a range of ailments and served as the foundation for ethnomedicine. Despite its enormous medical importance, the plant has received very little research with only a few antibacterial and pharmacological studies. The plant's roots, in particular, are understudied.  In this study, the antioxidant and antifungal activities of methanol (MeOH) root extract of F. indica was tested in vitro.   Materials and Methods: Antifungal activity was assessed against human fungal pathogens, Aspergillus niger (MTCC 404) and Trichophyton mentagrophytes (MTCC 7687). The natural antioxidants in roots were studied using five different antioxidant methods, including 2, 2-diphenyl-1-picrylhydrazyl (DPPH), nitric oxide scavenging (NOS), reducing power assay (RPA), measuring total phenolic content (TPC) by Folin-Ciocalteu Reagent (FCR).   Results: When compared to Aspergillus niger, the root extract has strong antifungal activity against Trichophyton mentagrophytes (MTCC 7687), generating a broader zone of inhibition (MTCC 404). Clotrimazole is a common antifungal medicine that can be used as a positive control in this investigation. The antioxidant activity of MeOH root extract increased in a dose-dependent manner, and it had outstanding NOS and antioxidant properties.   Conclusion: The results conclude that, F. indica appears to be a promising herbal option for the treatment of a variety of fungal and bacterial infections, with high antioxidant capacity.

Arum cyreniacum is an important member of the family of Araceae because of its bio-activities. Hence this work aimed to establish a link between Arum cyreniacum and its uses as bio-control against plant pathogenic fungi which had never hitherto been established. This work was carried out to evaluate the activity of the aqueous extracts of tubers, leaves, and flowers of Arum cyreniacum against three different types of pathogenic fungi, Fusarium solani, Rhizopus microspores and Aspergillus niger. The antifungal activity of the aqueous extracts of Arum cyreniacum was determined by poisoned food technique. The results showed that Arum cyreniacum had an inhibitory effect in a dose-dependent manner on Fusarium solani, Rhizopus microspores, while Aspergillus niger was resistant to all extracts. However, the great inhibition activity against tested fungi was associated with increasing concentrations of the aqueous extracts of Arum cyreniacum. Data in this work indicated that the use of Arum cyreniacum could be a valid alternative for bio-control of plant pathogenic fungi.

In the food industry and herbal markets, it is critical to control the quality of processed Panax ginseng products. In this study, ultra-performance liquid chromatography coupled to quadrupole time of flight mass spectrometry (UPLC-QTOF/MS)-based metabolomics was applied for the quality evaluation of white ginseng (WG), tae-geuk ginseng (TG), red ginseng (RG), and black ginseng (BG). Diverse metabolites including ginsenosides were profiled by UPLC-QTOF/MS, and the datasets of WG, TG, RG, and BG were then subjected to multivariate analyses. In principal component analysis (PCA), four processed ginseng products were well-differentiated, and several ginsenosides were identified as major components of each product. S-plot also characterized the metabolic changes between two processed ginseng products, and the major ginsenosides of each product were found as follows: WG (M-Rb1, M-Rb2, M-Rc, Re, Rg1), TG (Rb2, Rc, Rd, Re, Rg1), RG (Rb1, Rb2, Rc, Rd, Re, Rg1), and BG (Rd, Rk1, Rg5, Rg3). Furthermore, the quantitative contents of ginsenosides were evaluated from the four processed ginseng products. Finally, it was indicated that the proposed metabolomics approach was useful for the quality evaluation and control of processed ginseng products.
