Multidrug resistance is a leading public health challenge that is causing a significant increase in mortality and morbidity. If antimicrobial resistance (AMR) remains unsolved, it may cause 10 million deaths every year. Along with a public health concern, it is also a financial concern that would cause 2-3.5% reduction in Gross Domestic Product (GDP) and a 100 trillion USD loss to the world. One of the ways to combat AMR is to discover new antibiotics.  This study was aimed to evaluate the antibiofilm and antibacterial potencies of essential oil of Nigella sativa. Standard microbiological guidelines (CLSI) were used for the identification and antibiogram of selected strains of MRSA. Moreover, a time-kill assay of MRSA against Thymoquinone extracted from Nigella Sativa was also performed. Five strains, including four MRSA strains from implants related infections and one standard strain ATCC 25923, were examined. GC-MS identified components of essential oil of Nigella Sativa. Thymoquinone and p-cymene, major compounds of essential oil, were subjected to antibacterial and antibiofilm activities. Thymoquinone revealed strong inhibitory activities against MRSA strains. Zone inhibition measured 22 to 44 mm, and MIC values ranged from 26 to 43 Ul/mL. Thymoquinone also exhibited strong antibiofilm activity against biofilm producer MDR strains of Staphylococcus aureus.

Methicillin-resistant Staphylococcus aureus (MRSA) is one of the most complicated problems in infection control and disease prevention. Imported strains or group of similar strains called clones were introduced from other states within Australia and also from other countries. Western Australia (WA) MRSA clones known to be the most predominant clones circulating in Western Australia (WA). However, MRSA strains originally from other states and countries outside Australia (imported) have increased in number in the last decades. The aim of this study was to examine the epidemiology of CA MRSA clones in Western Australia for the past 11 year’s period (2003-2013). There were 10 predominant clones of CA-MRSA circulating in WA with WA MRSA-1 (32,82%), Queensland (22,67 %) and WA MRSA-2 (22,67 %) clones as the top three predominant clones. The percentage of PVL positive CA-MRSA clones such as Queensland (Qld) clone has been increasing significantly (R2=0,67) from 2,38 % in 2002 to 25,69% in 2013. In contrast, the percentages of two major PVL negative clones, WA MRSA-1 and WA MRSA 2, have been decreasing nearly by half from 52,38 % to 27,71 % and 30,95 % to 16,59 % respectively in the last 11 years. PVL positive clones have been introduced in WA from other states and overseas in the last eleven years. Due to the virulence factor of PVL toxin, the emergence of PVL positive CA-MRSA should be considered as public health concerns.

Mammarenavirus genus groups viruses causing human haemorrhagic diseases, including the New World (NW) Junín virus (JUNV), and the Old World (OW) viruses Lassa (LASV), among others. The high mortality and morbidity rates associated to pathogenic mammarenaviruses, the absence of vaccines and the constant threat of new emerging species, make these viruses a public health concern in endemic areas. Autophagy is a widely-known intracellular metabolic pathway involved in maintaining the cellular homeostasis in response to several stress conditions.

Background High blood pressure is an important public health concern and the leading risk factor for global mortality and morbidity. To assess the implications of this condition, we aimed to review the existing literature and study the factors that are significantly associated with hypertension in the Pakistani population. Methods We conducted several electronic searches in PubMed, ISI Web of Science, PsycINFO, EMBASE, Scopus, Elsevier, and manually searched the citations of published articles on hypertension from May 2019 to August 2019. We included all studies that examined factors associated with hypertension regardless of the study design. To assess the quality of the research, we used the Newcastle-Ottawa Quality Assessment Scale. We also conducted meta-analyses using the DerSimonian & Laird random-effects model to collate results from at least three studies. Results We included 30 cross-sectional and 7 case-control studies (99,391 participants country-wide) in this review and found 13 (35.1%) to be high-quality studies. We identified 5 socio-demographic, 3 lifestyle, 3 health-related, and 4 psychological variables that were significantly associated with hypertension. Adults aged between 30–60 years who were married, living in urban areas with high incomes, used tobacco, had a family history of hypertension, and had comorbidities (overweight, obesity, diabetes, anxiety, stress, and anger management issues) were positively associated with hypertension. On the other hand, individuals having high education levels, normal physical activity, and unrestricted salt in their diet were negatively associated with hypertension. Conclusion We found several socio-demographic, lifestyle, health-related, and psychological factors that were significantly (positively and negatively) associated with hypertension. Our findings may help physicians and public health workers to identify high-risk groups and recommend appropriate prevention strategies. Further research is warranted to investigate these factors rigorously and collate global evidence on the same.

ABSTRACT Infections due to methicillin-resistant Staphylococcus aureus (MRSA) are present worldwide and represent a major public health concern. The capability of PCR followed by high-resolution melt (HRM) curve analysis for the detection of community-associated and livestock-associated MRSA strains and the identification of staphylococcal protein A (spa) locus was evaluated in 74 MRSA samples which were isolated from the environment, humans, and pigs on a single piggery. PCR-HRM curve analysis identified four spa types among MRSA samples and differentiated MRSA strains accordingly. A nonsubjective differentiation model was developed according to genetic confidence percentage values produced by tested samples, which did not require visual interpretation of HRM curve results. The test was carried out at different settings, and result data were reanalyzed and confirmed with DNA sequencing. PCR-HRM curve analysis proved to be a robust and reliable test for spa typing and can be used as a tool in epidemiological studies.

In the recent era, diabetes mellitus has emerged as one of the significant threats to public health and this situation demands the attention of the researchers and allied stakeholders. Dietary regimens using functional and nutraceutical foods are gaining wide range of acceptance and some traditional medicinal plants are of considerable importance. The main objective of this instant study was to explore the antidiabetic potential ofNigella sativafixed oil (NSFO) and essential oil (NSEO). Three experimental groups of rats received diets during the entire study duration, that is, D1(control), D2(NSFO: 4.0%), and D3(NSEO: 0.30%). Experimental diets (NSFO & NSEO) modulated the lipid profile, while decreasing the antioxidant damage. However, production of free radicals, that is, MDA, and conjugated dienes increased by 59.00 and 33.63%, respectively, in control. On the contrary, NSFO and NSEO reduced the MDA levels by 11.54 and 26.86% and the conjugated dienes levels by 32.53 and 38.39%, respectively.N. sativaoils improved the health and showed some promising anti-diabetic results.

The emergence of Panton-Valentine leukocidin (PVL)-positive methicillin-resistantStaphylococcus aureus(MRSA) is a public health concern worldwide. PVL is associated with community-associated MRSA and is linked to skin and soft tissue infections (SSTIs). However, PVL genes have also been detected in health care-associated (HA) MRSA isolates. The diseases associated with PVL-positive HA-MRSA isolates and the distributions of PVL-encoding bacteriophages in HA-MRSA have not been determined. In this study, a total of 259 HA-MRSA strains isolated between 2009 and 2012 in China from inpatients with SSTIs, pneumonia, and bacteremia were selected for molecular typing, including staphylococcal cassette chromosomemectyping, multilocus sequence typing, and staphylococcal protein A gene typing. The PVL genes and PVL bacteriophages in the MRSA isolates were characterized by PCR. Among the tested MRSA isolates, 28.6% (74/259) were PVL positive. The high prevalence of PVL-carrying HA-MRSA was observed to be associated with SSTIs but not with pneumonia or bacteremia. The PVL-positive HA-MRSA isolates were colonized mainly by infective PVL phages, namely, Φ7247PVL, ΦSLT, and ΦSa2958. The distribution of PVL-carrying bacteriophages differed geographically. Our study highlights the potential risk of the emergence of multidrug-resistant HA-MRSA strains with increased virulence.

In vitro antimicrobial and antibiofilm activities of the Lippia alba essential oil and its major components (citral and carvone) against Staphylococcus aureus were investigated. Essential oils (LA1EO, LA2EO, and LA3EO) were extracted from the aerial parts of three L. alba specimens by hydrodistillation and analyzed by gas chromatography coupled to a mass spectrometer. Minimum Inhibitory Concentrations (MIC) and Minimum Bacterial Concentration (MBC) were determined by the microdilution method. For the antibiofilm assays, the biomass formation in the biofilm was evaluated by the microtiter-plate technique with the crystal violet (CV) assay and the viability of the bacterial cells was analyzed. All oils and their major components presented antibacterial activity, and the lowest MIC and MBC values were 0.5 mg mL−1 when LA1EO and citral were used. Potential inhibition (100%) of S. aureus biofilm formation at the concentration of 0.5 mg mL−1 of all EOs was observed. However, the elimination of biofilm cells was confirmed at concentrations of 1 mg mL−1, 2 mg mL−1, 2 mg mL−1, and 0.5 mg mL−1 for LA1EO, LA2EO, LA3EO, and citral, respectively. The results obtained in the present research point to the promising antibacterial and antibiofilm potential of L. alba EOs against S. aureus, a species of recognized clinical interest.
