BACKGROUND: Ficus deltoidea Jack. leaves have a great potential as traditional medicine, but the safety level of its use is still unknown. AIM: This study aimed to determine the phytochemical contents of the ethanol extract of F. deltoidea leaves and evaluate the level of safety and toxicity through acute and subchronic toxicity tests in mice (Mus musculus). METHODS: The ethanol extract of F. deltoidea leaves was determined for phytochemical contents such as alkaloids, phenolics, flavonoids, coumarin, steroids, saponins, carotenoids, and tannins. In the acute toxicity test, 10 male mice were divided into a control group and the extract treatment group with 2000 mg/kg body weight (BW) dose for 14 days to identify signs of toxicity and mortality. Meanwhile, in the subchronic toxicity test, 25 male mice were divided into control and four treatment groups with various doses (125, 250, 500, and 1000 mg/kg BW), respectively, for 28 days. The toxicological effect was evaluated by observing behavior, signs of toxicity, and changes in BW. At the end of the treatment, hematological and biochemical evaluations were also measured. RESULTS: The results showed that the ethanol extract of F. deltoidea Jack leaves qualitatively contains alkaloids, phenolic, flavonoids, coumarin, and steroids, whereas quantitatively total phenolics, flavonoids, and IC50 were 107.6583211 μg GA/mg, 175.9103641 μg CE/mg, and 103.7484 μg/mL. Moreover, 2000 mg/kg BW dose resulted in no symptoms of toxicity and mortality, indicating that the 50% lethal dose (LD50) was above 2000 mg/kg BW. Meanwhile, there were no behavioral changes, significant differences in weight changes, hematological parameters, and serum biochemistry of mice in subchronic toxicity tests. CONCLUSION: The present study shows that acute and subchronic oral administration of the ethanol extract of F. deltoidea leaves for male mice does not induce clinical symptoms of toxicity or mortality. The LD50 of the ethanol extract of F. deltoidea leaves for mice >2000 mg/kg is considered as practically non-toxic.

ABSTRACTObjective: As an antiobesity therapy, combination extracts of Detam 1 soybean and Jati Belanda will be consumed for a long time; therefore, theirtoxicities to the liver need to be investigated. To determine the effect of subchronic toxicity test of combination of ethanol extract of Detam 1 soybean(EEDS) and ethanol extract of Jati Belanda (EEJB) on liver function with parameters: Alanine transaminase (ALT), macroscopic, and histopathologicalof liver.Methods: This study was conducted on 120 Wistar rats (60 males and 60 females), 90 days (treatment group) and 120 days (satellite group). Ratswere divided into six treatment groups (3 test materials, 1 control, and 2 satellites); each group included 10 males and 10 females.Results: ALT levels of treatment groups (low dose, medium, and high), both males and females were lower than the control group (p<0.05). Thetreatment groups demonstrated a good effects effect on liver function. Liver weight of all groups showed no significant difference compared with thecontrol group (p>0.05). Results of histopathological score interpretation of male and female liver rats of low dose groups were not disturbed; middledose groups were slightly disturbed and high dose groups were damaged. Satellite high doses of male groups were disrupted, while female groupswere not.Conclusion: The combination of EEDS and EEJB has a good effect on liver function, did not lead to change organ weight and at low doses did not causerenal histopathology damage in rats after 90 days administration.Keywords: Combination of soybean Jati Belanda, Toxicity subchronic test, Function, Weight, Histopathology, Liver.

Tarap (Artocarpus odoratissimus Blanco) is one of the plants in the tropics that are consumed by dayak tribe in East Kalimantan. Toxicity tests on seeds and bark have been done but there is no data regarding the acute toxicity of Artocarpus odoratissimus Blanco seeds and flesh of fruit causing the need for acute toxicity tests. This Research to know the acute toxic effects of tarap (Artocarpus odoratissimus Blanco) seed and flesh extracts on larvae of Daphnia magna. Tarap seeds and flesh (Artocarpus odoratissimus Blanco) was taken from dayak market in Samarinda, is East Kalimantan, Indonesia. The seeds and flesh of the tarap fruit are extracted by maceration with ethanol solvent. An acute toxicity test was performed by exposing Dapnia magna larvae aged ≤ 24 hours with a solution of the experimental group and the control group for 48 hours. Toxicity test results are expressed in percentage of immobilization of larvae of Daphnia magna calculated by probit test to obtain EC50 (Half maximal effective concentration) values. Extracts are toxic if the EC50 value > 1000ppm. EC50 Ethanol extract of tarap seeds obtained values ( 3922,301 ± 324,590 ) for EC50 24h and ( 2964,498 ± 412,498 ) for EC50 48h. The EC50 value of ethanol extract from flesh of tarap fruit is ( 12224.514 ± 2186. 899 ) EC50 24h and ( 6165,235 ± 1940,006 ) EC50 48h.Ethanol extract of tarap fruit and flesh is non toxic to larvae of Daphnia magna.

<p>Penelitian ini bertujuan untuk melihat pengaruh pemberian ekstrak etanol kulit petai (Parkia speciosa Hassk) terhadap penurunan kadar glukosa darah mencit jantan yang diinduksi aloksan. Hewan percobaan dibagi atas 5 kelompok diantaranya kelompok kontrol negatif, kelompok kontrol positif,dosis I (280 mg/kgBB mencit), dosis II (560 mg/kg BB mencit), dosis III (840 mg/kg BB mencit). Penelitian dilakukan selama 21 hari. Persentase penurunan kadar glukosa darah mencit jantan setelah diberikan ekstrak etanol kulit petai pada hari ke-21 adalah dosis I (77,52 %) lebih besar dibandingkan dengan dosis II (69,5 %) dan dosis III (73,37 %). Data yang diperoleh dianalisis dengan uji Two Way Anova dengan program SPSS 17. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak etanol kulit petai untuk tiga variasi dosis menyatakan perbedaan yang bermakna secara statistik terhadap penurunan kadar glukosa darah mencit jantan.</p><p><em>Petai (Parkia speciosa Hassk) has a compound β-sitosterol and stigmasterol that have efficacy to decreased blood glucose levels. This study aimed to determine the effect of ethanol extract of petai peel for decrease blood glucose levels of male mice induced by alloxan. Experimental animals were divided into 5 groups including negative control group, positive control group, the first dose (280 mg/kg in mice), the second dose (560 mg/kg in mice), the third dose (840 mg/kg in mice). The study was conducted for 21 days. After 21 days, the result found that the percentage of blood glucose levels after the male mice given the ethanol extract of petai peel was, the first dose (77.52%) biger than the second dose (69.5%) and the third dose (73.37%). The data obtained were analyzed by Two Way ANOVA using SPSS 17. The results showed that have signicantly difference between three dose variation of ethanol extract of petai peel in blood glucose levels.</em></p>

Subchronic toxicity test is a test to detect the toxic effect that arises after the administration of the test reparation with repeated doses were given orally to the tested animal for 28 or 90 days. Leaves agarwood (Aquilaria malaccensis Lamk) is a tree from a tribe Thymeleaceae, already started popular used the farmer agarwood in Langkat as a drink that in pour. The result of an interview with the farmer agarwood explained that consume tea from the leaves agarwood of this kind of have many benefits include improve canal. To that was done the research security against the tea leaves agarwood induction taken from agriculture agarwood in Langkat, Sumatera North through test toxic subchronic oral. This study aims to determine the symptoms of toxic posed by product tea agarwood induction. This study used laboratory animals that male mice and female mice were divided into 5 groups, namely the 130, 260, 390, 520 mg/kgBW and the control group. The observation of clinical symptoms indicate the presence of toxic symptoms of weakness, changes in fur and agitated at doses of 390 and 520 mg/kgBW in male mice and female mice, the observation macropathology organs alloxan still normal the red-brown, the surface of slippery and consistency chewy. Histopathological results showed hemoglia and dilation of the blood vessels in all groups. Results showed that mice were given tea steeping agarwood induction doses ranging from 130, 260,390 and 520 mg/kgBW there are no mice died, so it can be concluded that the administration of agarwood tea steeping in mice does not cause toxic symptoms and safe for consumption.

Studies on the safety of herbal medicine are essential for the development of new drugs. The aim of this study was to evaluate the no-observed-adverse-effect-level (NOAEL) of HVC1 (Gamisamhwangsasim-tang, a 30% ethanol extract of a mixture of Pruni Cortex, Scutellariae Radix, Coptidis Rhizoma, and Rhei Rhizoma) and identify its target organs after oral administration to Sprague-Dawley (SD) rats repeatedly for 13 weeks. Three test groups were treated with HVC1 at a dose of either 500 (low-dose), 1,000 (middle-dose), or 2,000 (high-dose) mg/kg/day. Another group received high-dose HVC1 and was observed for 4 weeks following treatment to examine recovery from the effects of the extract. All treatment groups were compared to a vehicle control group. During the study, mortality, clinical signs, body weight changes, food consumption, abnormal lesions in the eye, urinary parameters, hematological parameters, blood coagulation time, blood biochemical parameters, changes in organ weight, gross findings, and histopathological changes were examined. No systemic toxicity related to HVC1 was observed in any group, and it was concluded that the NOAEL of HVC1 was 2,000 mg/kg/day. No target organ was identified.

Mumefural is a bioactive compound derived from the processed fruit of Prunus mume Sieb. et Zucc., a traditional health food; however, its safety has not been evaluated. We investigated the toxicity of mumefural through single and repeated oral administration at doses of 1250, 2500, and 5000 mg/kg in Institute of Cancer Research (ICR) mice. The acute toxicity assessment was not associated with adverse effects or death. Similarly, the subacute (four weeks) toxicity assessment did not reveal any mumefural-associated mortality, abnormal organ damage, or altered clinical signs, body weight, food consumption, or hematological parameters. However, albumin/globulin ratio and chloride ion levels were significantly increased in male mice treated with mumefural at ≥2500 mg/kg. Female mice exhibited significantly higher levels of chloride, sodium, and potassium ions, at a dose of 5000 mg/kg. Furthermore, the administration of 2500 and 5000 mg/kg mumefural decreased the absolute weight of spleen in male mice. These findings indicated that the approximate lethal dose of mumefural in ICR mice was >5000 mg/kg. No significant mumefural toxicity was observed at ≤5000 mg/kg. Our findings provide a basis for conducting future detailed studies to evaluate reproductive, neurological, genetic, and chronic toxicity of mumefural.

The herb frequently used as spices or remedies in the Indonesian community, with the seed as the most common part is kapulaga (Amomum cardamomum). According to earlier evidence, this possessed antibacterial, antifungal and several biological properties, reduced blood glucose and atherogenic parameter, and is developed as standardized herbal cures. However, the application of herbal medicine requires validating evidence of safety and effectiveness, including toxicity tests, particularly in clinical settings. The target organs in this comprised hepar, due to the role in several drug metabolism. This study aimed at discovering the safety profile of kapulaga seed extract based on the hepatic transaminase enzyme (SGOT and SGPT) level, by conducting an acute toxicity test in Winstar rats. Also, this was implemented with the OECD 420 Fixed-Dose Procedure, and the preliminary test employed 300 mg/kg BW dose followed by a maximum single quantity (2000 mg/kg BW) of kapulaga. The main test was executed by a separation into control and treatment groups of 5 rats each. Therefore, a single dose of 2000 mg/kg BW kapulaga seed extract was administered to the treatment group, while the control group received standard pellets and water ad libitum. The blood from orbital vein was acquired on day 14, and SGOT and SGPT were subsequently assessed by an enzymatic-photometric method. Also, this data was analyzed using an independent sample t-test, and the mean of SGOT in both groups were 116.92±22.35 and 98.02±16.38 (p=0.17), with 58.72±8.79 and 47.64±7.30 (p=0.06) as SGPT respectively. Therefore, there was no statistical difference, and no acute toxicity signs were discovered. Beberapa bukti sebelumnya menunjukkan bahwa kapulaga memiliki berbagai aktivitas biologis seperti antibakteri, antijamur, dan sudah dibuktikan mampu menurunkan glukosa darah dan parameter arterogenik. Bukti tersebut mendukung pengembangan kapulaga sebagai obat herbal terstandar. Penggunaan obat herbal terutama di klinik harus didukung dengan adanya bukti keamanan maupun efektivitasnya termasuk uji toksisitas. Hepar merupakan salah satu target organ dari uji toksisitas karena perannya yang penting pada metabolisme sebagian besar obat. Penelitian ini bertujuan untuk menilai profil keamanan ekstrak biji kapulaga melalui uji toksisitas akut menggunakan tikus Winstar berdasarkan kadar enzim transaminase hepar (SGOT dan SGPT). Uji toksisitas akut berpedoman pada OECD 420 Fixed Dose Procedure. Uji pendahuluan menggunakan ekstrak biji kapulaga dosis 300 mg/kg BB dan diikuti dengan dosis tinggi 2000 mg/kg BB yang diberikan secara tunggal. Uji utama dilakukan dengan membagi tikus ke dalam kelompok kontrol dan perlakuan, masing-masing kelompok terdiri atas 5 tikus. Berdasarkan hasil uji pendahuluan, uji utama menggunakan dosis tunggal 2000 mg/kg BB untuk kelompok perlakuan, sedangkan kelompok kontrol hanya mendapatkan pelet dan air secukupnya. Pada hari ke-14, darah dari vena orbital diambil, lalu kadar SGOT dan SGPT diukur menggunakan metode enzymatic-photometric. Independent sample t-test digunakan untuk menilai data rata-rata kadar SGOT dan SGPT dari kedua kelompok. Rata-rata kadar SGOT pada kelompok kontrol dan perlakuan sebesar 116,92±22,35 dan 98,02±16,38 (p=0,17), sedangkan rata-rata SGPT sebesar 58,72±8,79 dan 47,64±7,30 (p=0,06). Perbedaan rata-rata SGOT dan SGPT pada kedua kelompok tersebut secara statistik tidak bermakna dan tidak ditemukan tanda toksisitas pada semua hewan coba. Ekstrak biji kapulaga dosis maksimal 2000 mg/kg BB tidak toksik pada hepar tikus karena tidak menimbulkan tanda toksisitas maupun mengubah enzim transaminase hati (SGOT dan SGPT). Kata kunci: Amomum cardamomum, kapulaga, toksisitas akut, SGOT, SGPT

Combretum collinum root extract has been recognized long ago as traditional medicinal plant in curing several diseases among the indigenous people of Alela-land (Zuru), Kebbi State, Nigeria. This research work was carried out to evaluate the toxicological effects of Combretum collinum methanol root extract (CCME) in albino rats. Acute toxicity was performed by a fixed single oral administration at a dose of 10, 100, 1000 mg/Kg and 1600, 2900, 5000 mg/Kg. Subchronic toxicity studies of CCME was conducted at doses of 32, 63, 126 and 253 mg/Kg for 28 days. The result showed that acute administration of CCME resulted at mortality and general behavioral changes at 1000 to 5000 mg/Kg. Therefore, the estimated lethal dose (LD50) of CCME was 316.23 mg/Kg. Subchronic oral administration of CCME revealed a significant (P<0.01) decrease in body weight in rats receiving 63 to 253 mg/Kg throughout the study period compared with the control group. The results also showed a significant (P<0.01) increase in serum ALT, AST, creatinine, potassium and bicarbonate in rats administered with 126 and 253 mg/Kg of the extract. Haematological analysis of the same extract revealed a significant (P<0.01) increase in WBC, HCT, MCV, MCH, MCHC, PLT, LYM and NEUT in rats receiving 126 and 253 mg/Kg only. Histopathological examination of liver revealed severe periportal inflammation, hypertrophy, areas of hydropic changes, cancerous tumor, areas of infiltration and necrosis of the hepatic cells while the kidney showed a mild mesengial hyperplasia, compressed blood vessels, glomerular degeneration, tubular degeneration and tubular widened lumen in rats treated with 63 to 253 mg/Kg. Therefore, caution should be applied as C. collinum root extract has a low mean lethal dose and would be toxic at higher concentrations.
