Recent developments in the use of environmental DNA are opening up new horizons for the assessment of the quality of aquatic environments. These rapid and cost-effective methods, in very swift progress, will potentially offer the opportunity to identify all the taxa present in an environmental sample (water or biota) by the use of complementary markers. The produced inventories can then be used for the assessment of biodiversity and ecological quality. However, the inclusion of these new DNA-based methods in monitoring practices is not straightforward and requires harmonised actions in the coming years at national and international levels. In order to foresee and stimulate such a harmonised implementation, the European network DNAqua-Net (COST Action CA15219) brought together some of its members, experts of ECOSTAT and other environmental biomonitoring stakeholders from different European countries. Through workshops, bringing together 51 participants in 7 sub-groups in April 2020, an implementation roadmap was designed. The coordinated actions to be taken in the different countries, and the possible collaborations and steps to be taken at the EU level were identified. This presentation will give an overview of all discussions (Lefrançois et al. 2020) reflecting the diversity of situations in Europe, as well as common views. We will highlight important actions required for a successful implementation of DNA-based biomonitoring of aquatic ecosystems to the horizon of 2030.

Abstract Objective Silica gel beads have promise as a non-toxic, cost-effective, portable method for storing environmental DNA (eDNA) immobilized on filter membranes. Consequently, many ecological surveys are turning to silica bead filter desiccation rather than ethanol preservation. However, no systematic evaluation of silica bead storage conditions or duration past 1 week has been published. The present study evaluates the quality of filter-immobilized eDNA desiccated with silica gel under different storage conditions for over a year using targeted quantitative real-time polymerase chain reaction (qPCR)-based assays. Results While the detection of relatively abundant eDNA target was stable over 15 months from either ethanol- or silica gel-preserved filters at − 20 and 4 °C, silica gel out-performed ethanol preservation at 23 °C by preventing a progressive decrease in eDNA sample quality. Silica gel filter desiccation preserved low abundance eDNA equally well up to 1 month regardless of storage temperature (18, 4, or − 20 °C). However only storage at − 20 °C prevented a noticeable decrease in detectability at 5 and 12 months. The results indicate that brief storage of eDNA filters with silica gel beads up to 1 month can be successfully accomplished at a range of temperatures. However, longer-term storage should be at − 20 °C to maximize sample integrity.

E-commerce is currently a dynamically developing branch of the Russian economy. Owing to modern digital technologies, new opportunities are opening up for business, government and consumers. The development of this scope requires the adoption of balanced legal decisions, which is not always possible in the context of the emergence of new and transformation of existing e-commerce technologies. And here it is appropriate to talk about the need for legal experiments in order to create effective legal regulators in the field of electronic commerce in the future. The author outlines the main features of a legal experiment, presents arguments justifying the need for such experiments in the field of electronic commerce. The experience of the Russian Federation and a number of foreign countries in organizing and conducting such experiments is also analyzed. Based on the results of the study, it was concluded that conducting a legal experiment contributes to a more successful implementation of e-commerce technologies in the Russian legal system, increases the efficiency of adopted legal acts, the quality of activities of public authorities and business, and as a result, the achievement of the intended results.

In co-ordination with the EU-guidelines the large wastewater treatment plants in Switzerland have to be extended with enhanced nitrogen removal. Denitrification in tertiary filtration is a cost-effective alternative to extended denitrification in the activated sludge system, which needs additional reactor volume. At the wastewater treatment plant Zürich-Werdhölzli full-scale experiments of denitrification with methanol in tertiary filtration were performed during a summer and a winter campaign of 4 months each. For this purpose one of the original 22 filter cells was equipped with a methanol dosage. At temperatures of 12-15°C rates of denitrification of about 1.0 kgN m−3 d−1 are attained. After main backwashing, denitrification is significantly reduced. Frequent backwashings (several times per day) led to methanol breakthroughs due to biofilm loss. The yield coefficient YCOD was 0.4 kg CODX kg−1 CODme. In spite of methanol dosage the quality of the filter effluent was very good during normal operation in the winter campaign. Accumulation of the nitrite intermediate product was observed in summer at temperatures of 20-22°C.

This study aims to identify the dominant factors of the successful implementation of benchmarking on the performance of contracting companies and test the significance of the application of benchmarking on the performance of contracting companies. The research sample was saturated samples of 65 qualified contractor companies. The method and type of research used were correlational methods of multiple regression analysis using SPPS. The results of the study concluded that 1.Benchmarking significantly influences the performance of contracting companies in the Blitar City DPUPR; 1. The ranking of success factors for the Blitar City contractor companies in the process of implementing benchmarking (1) planning, (2) data collection, (3) acception and action and (4) analysis; 2.Benchmarking has proven to significantly improve company performance as measured by increasing (1) Corporate Finance (2) Company productivity, (3) DPUPR Consumer Satisfaction, (4) Community Satisfaction, (5) Quality of the company's construction technical personnel, (6) Satisfaction employee work, (7) Project acquisition rate in one year, (8) Effective completion of construction work, (9) Construction product quality.

The results of elements development of varietal agrotechnics of a new high-potential line 238h of winter triticale presented. It was identified that under the conditions of 2020 the most cost effective fertilizer system is an early spring application of NPK (S) (15-15-15 (10)) at a dose of 200kg/ha. The profitability level of this fertilizer was 88,9%.

Growth in telecom infrastructure and provision of modern telecom services to consumers at a cost based tariff helps growth of national economy. Modern telecoms serve as the engine of growth of national economy. Following the global trends of liberalisation and deregulation in telecoms monopolies which have thus far been providing inefficient communication at a greedily high tariffs are falling apart. Mergers in telecoms are not for increasing the size of the monopoly but to provide more efficient and cost effective services to the consumers. In Pakistan the erstwhile T&T department played a needful role at its time. Conversion of the department into a corporation and then into a company were steps necessary for following the global trends. Need now is to continue this trend further, eliminate the monopolistic approach by allowing more players in the field thus permitting the market forces to decide the provision of better quality of modern services at competitive price.

Abstract Today, failure analysis involving flip chip [1] with copper pillar bump packaging technologies would be the major challenges faced by analysts. Most often, handling on the chips after destructive chemical decapsulation is extremely critical as there are several failure analysis steps to be continued such as chip level fault localization, chip micro probing for fault isolation, parallel lapping [2, 3, 4] and passive voltage contrast. Therefore, quality of sample preparation is critical. This paper discussed and demonstrated a quick, reliable and cost effective methodology to decapsulate the thin small leadless (TSLP) flip chip package with copper pillar (CuP) bump interconnect technology.

Several methods of Deoxyribonucleic acid (DNA) extraction have been applied to extract bacterial DNA. The amount and the quality of the DNA obtained for each one of those methods are variable. The study aimed to evaluate bacterial DNA extraction using conventional boiling method followed by alcohol precipitation. DNA extraction from Gram negative bacilli was extracted and precipitated using boiling method with further precipitation by ethanol. The extraction procedure performed using the boiling method resulted in high DNA yields for both E. coli and K. pneumoniae bacteria in (199.7 and 285.7μg/ml, respectively) which was close to control method (229.3 and 440.3μg/ml). It was concluded that after alcohol precipitation boiling procedure was easy, cost-effective, and applicable for high-yield quality of DNA in Gram-negative bacteria.
