Phenylpropanoid metabolism is an important secondary metabolism pathway in plants. Lignin, as a secondary metabolite was produced by secondary metabolism pathway, plays important roles on disease resistance in plants. PAL, C4H, and CHS are key enzymes involved in the secondary metabolism pathway and critical on disease resistance. We hypothesize that the expression of key enzymes of secondary metabolism pathway will be affected by abnormal environmental conditions; therefore provide the resistance to severe environment for plant. In this study, we analysed the gene expressions of PAL, C4H, and CHS under higher temperature and infection of germ (Pseudoperonosporacubensis) in Cucumber JingChun No.4, which is highly resistant to downy mildew. The gene expressions were quantified by semi-quantitative RT-PCR. Our results showed that the expression of C4H was consistently higher and not affected by germ inoculation. However, the expressions of PAL and CHS were increased at 2 h and 25 h, respectively, after inoculating with germs. Interestingly, we found that the expressions of all these three genes were decreased with treatment of higher temperature. Our results demonstrated that PAL, C4H and CSH are important in secondary metabolism and affect the potential resistant ability of plant to various severe growing environments.

It has been previously reported that POST-applied isoxaben can effectively control established hairy bittercress. Experiments were conducted to determine the relative importance of root vs. foliar entry of POST-applied isoxaben. At a common isoxaben rate of 0.56 kg/ha, foliar-only and foliar plus soil applications provided 10.5 and 23.3% control, respectively, as determined by fresh weight reduction. In contrast, soil-only application provided 47.0% control. Hairy bittercress foliar absorption of14C–isoxaben did not exceed 15% of the amount applied after 72 h. Therefore, the comparatively less effectiveness of foliar-only applications may be attributed primarily to limited absorption. Minimal isoxaben concentration required to inhibit root growth of hydroponically grown hairy bittercress was 0.0025 mg/L. Higher concentrations were required to produce a response in the foliage. Sorption of isoxaben by pine bark rooting substrate, typical of what is used in container nursery production, exceeded 99% of amount applied after 36 h. Even with 99% sorption, the probable concentration within the aqueous phase remains sufficient to inhibit hairy bittercress root growth. Additional studies with14C–isoxaben established that approximately 35% of the root-absorbed isoxaben was translocated into the foliage. Translocation from the roots into the foliage was reduced to 16% when the experiment was repeated during environmental conditions less favorable for vegetative growth (i.e., longer day length and higher temperature). Results indicate that the control of hairy bittercress with POST-applied isoxaben is likely the result of root absorption and root-growth inhibition. Expression of phytotoxicity within the foliage is also a component, but is dependent upon the root-absorbed isoxaben being translocated into the foliage. Extent of this translocation is dependent upon plant maturity and prevalent environmental conditions.

In our study, rats were subjected to Oxymetazoline hydrochloride treatment and Rhinitis medicamentosa (RM) was formed and then autophagy gene expression levels were determined after the application of an antioxidant agent erdosteine (ED). The rats were divided into three groups; Group 1 was the control group. Group 2 (RM) and group 3 (RM+ED) rats received two spray puffs of 0.05% oxymetazoline into the nasal cavities three times daily for eight weeks. After determination of RM in the rats, the RM group were killed. The ED+RM group received 10 mg/kg of an ED suspension. At the end of seven days, these rats were also killed. All groups? hippocampus tissues were obtained for the measurement of autophagy gene expressions. In rhinitis medicamentosa group Atg5, Atg7 and Atg10 gene expressions in the left hippocampus were reduced as compared to control group (p=0.01, p>0.05, p=0.01, respectively). Also, erdosteine treatments were restored mRNA expression of autophagy genes. In right hippocampus of rhinitis medicamentosa group, Atg5 and Atg10 gene expressions was found to be down-regulated as compared to control group (p>0.05, p<0.05, respectively). Both BECN1 and ULK genes expression were found to be reduced in left hippocampus of rhinitis medicamentosa group. Erdosteine applications was restored the expression of these genes (p=0.03, p=0.03, respectively). Additionally, in right hippocampus, Erdosteine application was restored the expression of ULK gene (p=0.01). This is the first report that evaluated the expression autophagy genes in RM rat models and the changes observed after erdosteine applications.

The ring test method is used to evaluate the behavior of concrete under restrained shrinkage. Four kinds of environmental conditions (20±1°C, RH60±5%; 35±1°C, RH60±5%; 50±1°C, RH15±5%; 65±1°C, RH15±5%) were designed to investigate the effect the environmental condition on the shrinkage behavior of concrete prepared with different kinds of mineral admixtures. The results show that higher temperature and lower relative humidity can increase the risk of shrinkage cracking. The addition of different mineral admixtures increases the shrinkage of concrete at room conditions. In the case of higher temperature and lower relative humidity, the addition of mineral admixtures can delay the shrinkage cracking in the test duration.

Legumes have previously been reported with hypolipidemic effect caused by the presence of flavonoid. This study was carried out to evaluate the antioxidant and hypolipidemic effects of fermented mung bean on hypercholesterolemic mice. Blood from all mice was collected and subjected to serum lipid and liver profiles biochemical analysis and quantitative RT-PCR for atherosclerosis related gene expressions. Besides, livers were collected for antioxidant assays and histopathology evaluation. Fermented mung bean was found to reduce the level of serum lipid and liver enzyme profiles of hypercholesterolemic mice. Furthermore, liver antioxidant and nitric oxide levels were also significantly restored by fermented mung bean in a dosage dependent manner. The gene expression study indicated thatApoeandBcl2a1awere upregulated whileNpyandVwfexpressions were downregulated after the treatment. The effects of fermented mung bean were greater than nonfermented mung bean. These results indicated that fermented mung bean possessed antioxidants that lead to its hypolipidemic effect on hypercholesterolemic mice.

Background: During the third trimester of pregnancy multiple sclerosis (MS) disease activity is reduced. It is not fully understood which factors mediate this disease amelioration.Objective: To study alterations of the monocyte transcriptome during pregnancy in MS patients, using a genomewide approach to identify differentially regulated genes.Methods: Women with MS and healthy controls were longitudinally studied, including a visit before pregnancy.Results: RNA-microarray analysis was performed in six patients. We found a significant increase of CD64 (Fc gamma receptor 1a, FcgR1a) during the third trimester compared with baseline, confirmed by RT-PCR in a group of ten patients. Analysis with Ingenuity software was performed using all genes expression of which was altered at least 1.5-fold in at least five out of six patients. Major networks that were altered during MS pregnancy were: cell-to-cell signalling and interaction, immune response, and cell signalling. From the genes selected for Ingenuity analysis, seven additional candidate genes, selected for their biological interest, were tested using RT-PCR in ten patients with MS and nine controls. We found an increased expression of JAK2 and STAT1 directly postpartum in patients with MS and in controls.Conclusion: The increased CD64 expression during pregnancy is indicative of enhanced innate immune functions.

The effect of environmental conditions immediately before anthesis on potential grain weight was investigated in wheat at the experimental field of the Faculty of Agronomy (University of Buenos Aires, Argentina) during 1995 and 1996. Plants of two cultivars of wheat were grown in two environments (two contrasting sowing dates) to provide different background temperature conditions. In these environments, transparent boxes were installed covering the spikes in order to increase spike temperature for a short period (c. 6 days) immediately before anthesis, i.e. between ear emergence and anthesis. In both environments, transparent boxes increased mean temperatures by at least 3·8 °C. These increases were almost entirely due to the changes in maximum temperatures because minimum temperatures were little affected. Final grain weight was significantly reduced by higher temperature during the ear emergence–anthesis period. It is possible that this reduction could be mediated by the effect of the heat treatment on carpel weight at anthesis because a curvilinear association between final grain weight and carpel weight at anthesis was found. This curvilinear association may also indicate a threshold carpel weight for maximizing grain weight.
