The objective of this study was to investigate the antibacterial activity of primary and secondary metabolites from <em>Vibrio alginoliticus</em> isolated from sponge <em>Haliclona</em> sp. against <em>Staphylococcus aureus</em>. A descriptive method was used in this research. The antibacterial activity was analysed by paper disk method. The results showed that the primary metabolites produced by <em>Vibrio alginoliticus</em> that is in symbiosis with sponge <em>Haliclona</em> sp. were able to effectively inhibit <em>Staphylococcus aureus</em> growth with an inhibition zone diameter of 12.9 mm, while the secondary metabolites of 9.9 mm. Electrophoresis analysis of the primary metabolites showed that there were 11 protein bands which were not found in secondary metabolites. Protein bands with low molecular weights presumably had an inhibiting effect on the growth of <em>Staphylococcus aureus</em>.

ABSTRAK Kunyit (Curcuma longa L.) merupakan salah satu jenis tanaman obat yang termasuk dalam keluarga Zingiberaceae. Senyawa aktif yang terkandung dalam rimpang kunyit (Curcuma longa L.) mampu bekerja sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari ekstrak etanol rimpang kunyit (Curcuma longa L.) terhadap Staphylococcus aureus dan Pseudomonas aeruginosa. Ekstraksi dilakukan dengan cara refluks menggunakan pelarut etanol 96%. Pengujian aktivitas antibakteri dilakukan menggunakan metode difusi paper disk dengan masing-masing konsentrasi ekstrak 10%, 20%, 30%, 40% b/v. Kontrol positif yang digunakan adalah Ciprofloxacin sedangkan kontrol negatif yang digunakan adalah DMSO. Hasil skrining fitokimia ekstrak etanol rimpang kunyit mengandung alkaloid, flavonoid, fenol, tanin dan terpenoid. Berdasarkan hasil penelitian, ekstrak etanol rimpang kunyit dapat menghambat bakteri Staphylococcus aureus dan Pseudomonas aeruginosa pada konsentrasi 10%, 20%, 30%, 40% dan konsentrasi 40% merupakan konsentrasi yang memberikan diameter zona hambat terbesar terhadap kedua bakteri uji yaitu 8,63 mm dan 7,8 mm. The active compounds contained in the Curcuma longa L. rhizome can work as antibacterial. This study aims to determine the antibacterial activity of the ethanol extract of Curcuma longa L. Antibacterial activity testing was carried out using the paper disk diffusion method with each extract concentration of 10%, 20%, 30%, 40%. The positive control used was Ciprofloxacin while the negative control used was DMSO. The results of phytochemical screening of the ethanol extract of Curcuma longa L. rhizome contain alkaloids, flavonoids, phenols, tannins and terpenoids. Based on the results of the study, the ethanol extract of turmeric rhizome can inhibit Staphylococcus aureus and Pseudomonas aeruginosa bacteria at concentrations of 10%, 20%, 30%, 40% and 40% concentrations which give the largest inhibition zone diameter of the two test bacteria which is 8.63 mm and 7.8 mm. Endophytic fungi is the one of the types microbes that lives in the plant tissue. The fungus can produce secondary metabolites potential as a source of antimicrobial and anticancer. The objectives of this study was to the exploration of new antibacterial compounds derived from marine endophytic fungi isolated from coastal plant terong pungo (Solanum sp.). Eight isolates of marine endophytic fungi with different morphology were collected. Endophytic fungus TPL2 was the selected isolate based on antagonism test. The growth curve showed the stationary phase of isolate TPL was on the 9th day to 12th day. Crude extract of endophytic fungi TPL2 showed the antibacterial activity against Staphylococcus aureus, Staphylococcus epidermidis, and Pseudomonas aeruginosa with diameter of inhibition zone 4 mm, 6 mm, and 6 mm, respectively for 2 mg extract/well.

Telah dilakukan penelitian yang bertujuan untuk mengidentifikasi senyawa metabolit sekunder  dari ekstrak daun kaf menggunakan tiga jenis pelarut yakni n-heksan, etil asetat, dan metanol. Ekstrak yang diperoleh diuji aktivitas antibakterinya dengan metode difusi yang menggunakan bakteri Staphylococcusaureus dan Escherichia coli. Dari hasil pengujian fitokimia, ketiga ekstrak tersebut menunjukan bahwa sampel mengandung senyawa; alkaloid, saponin, steroid, flavonoid dan tanin. Pengujian aktivitas antibakteri  dengan metode difusi menunjukan hasil yang paling tinggi aktivitas antibakteri adalah  estrak etil asetat dengan zona hambat 35,75 mm terhadap bakteri  Staphylococcus aureus, sedangkan pada ekstrak metanol memiliki zona hambat 23,75 mm terhadap bakteri Escherichia coliResearch has been carried out aimed at identifying secondary metabolites of kaf leaf extract using three types of solvents, namely n-hexane, ethyl acetate, and methanol.The obtained extracts were tested for antibacterial activity by diffusion method using Staphylococcus aureus and Escherichia coli bacteria. From the results of phytochemical testing, the three extracts showed that the sample contained compounds; alkaloids, saponins, steroids, flavonoids and tannins. Antibacterial activity testing with diffusion method showed the highest antibacterial activity was estracethyl ethyl acetate with 35.75 mm inhibition zone against Staphylococcus aureus bacteria, whereas methanol had 23.75 mm inhibitory zone against Escherichia coli bacteria

Mekai leaves (Pycnarrhena cauliflora Diels.) (P. cauliflora). Are known to contain flavonoid compounds, tannins and phenolics that act as antibacterial agents and are used in the treatment of eye pain. Staphylococcus aureus (S. aureus) is one of the examples of bacterial diseases of eye pain and also a major cause of many infections in communities and health facilities with cases of resistance to various antimicrobial agents. The purpose of this study was to prove the antibacterial activity of mekai leaves ethanol extract against S. aureus bacteria. This research is an experimental research. The stages of this research began by extracting mekai leaves using maceration method with 96% ethanol solvent. Antibacterial activity was tested by the disc method (Kirby-Bauer) using ethanol extract concentrations of mekai leaves (EPC) 20%, 30%, 40%, 50%, 60%, 70%, and 80%, positive control using 25 μg amoxicillin and negative control using DMSO 10%. The measurement results of inhibition zones of mekai leaf ethanol extract 20%, 30%, 40%, 50%, 60%, 70% and 80% respectively were 8.32 mm, 8.32 mm, 8.32 mm, 8.67 mm, 9.00 mm, 8.67 mm, and 8.33 mm. While the positive control measurement of 25 μg amoxicillin against S. aureus is 28.67 mm and the measurement of 10% negative DMSO control does not produce inhibitory zones, so it can be concluded that the ethanol extract of mekai leaves has antibacterial activity, but the area of ​​inhibition zone produced is smaller than amoxicillin 25 µg. The results of statistical tests using Mann Whitney between negative controls with all EPC concentrations obtained significant differences with p values ​​<0.05, it can be concluded that there is antibacterial activity produced at all EPC concentrations.

Hornstedtia scyphifera var. fusiformis Holttum is endemic plant of Zingiberaceae family that distributed in Sumatera. Hornstedtia scyphifera var. fusiformis Holttum contained some secondary metabolite such as phenolic, saponin, triterpenoid and alkaloid. The biological activity of methanol, ethyl acetate, and n-hexane extracts from Hornstedtia scyphifera var. fusiformis Holttum suspended roots was tested for antibacterial and antioxydant activity. Antioxidant activity was analized by DPPH method. The antibacterial activity  was tested by used disk difussion method againts Staphylococcus aureus bacteria (gram-positive bacteria) and Escherichia coli (gram-negative bacteria). Almost all of Hornstedtia scyphifera var. fusiformis Holttum suspended roots extract were given antibacterial activity, nonetheless the biggest inhibition zone of Escherichia coli that was inhibited by n-hexane and ethyl acetate extracts which have inhibition zone 10 mm at  concentration 500 mg/L and also againts Staphylococcus aureus, the biggest inhibition by ethyl acetate and n-hexane extracts which have inhibition zone 10.30 mm at concentration 500 mg/L. Among all extracts tested, methanol extract of the possessed moderate free radical scavenging activity with IC50 = 51.89 mg/L.

Phyllidiella nigra is an organism that is suspected to have secondary metabolites because their ability to develop its self defense system by camouflage and using chemical compounds derived from their nature diet as deterrent against their predators. The purpose of this study was to isolate symbiotic bacterial derived from P. nigra, extracted and followed by, the antibacterial assays against Escherichia coli and Bacillus megaterium as well as the anti-UV assay. The results showed that the five isolates tested had an antibacterial activity with the highest average inhibition zone against E. coli DSM 498 bacteria, isolate 1 (14.67 mm), isolate 5 (14 mm), and against B. Megaterium DSM 32T bacteria, isolate 3 (13.33 mm). The three isolates which had the highest inhibition zone and P. nigra extract were tested for anti-UV assay using a UV-Vis Spectrophotometer. The results obtained isolate 3 has absorption of UV-A with the UV absorbtion maximum at λ 340 nm and P. nigra extract has absorption on UV-B radiation with UV absorption maximum at λ 290 nm. Key words: Nudibranchia, Bacteria, Anti-bacteial, Anti-UV Phyllidiella nigra merupakan organisme yang diduga memiliki metabolit sekunder karena mampu mengembangkan sistem pertahanan dirinya dengan cara kamuflase dan menggunakan senyawa kimia sebagai racun yang didapat dari makanannya. Tujuan dari penelitian ini yaitu mendapatkan isolat bakteri yang bersimbiosis dengan P. nigra, mendapatkan ekstrak dari baktri simbion, dan menguji antibakteri dan anti-UV ekstrak etil aseta bakteri simbion dengan metode difusi agar terhadap bakteri Escherichia coli dan Bacillus megaterium. Hasil penelitian didapatkan kelima isolat yang diuji memiliki aktivitas antibakteri dengan rerata zona hambat tertinggi terhadap bakteri E. coli DSM 498 yaitu isolat 1 (14,67 mm), isolat 5 (14 mm), dan terhadap baktri B. megaterium DSM 32T yaitu isolat 3 (13,33 mm). Ketiga isolat yang memiliki zona hambat tertinggi dan ekstrak P. nigra diujikan anti-UV menggunakan alat UV-Vis Spektrofotometer. Hasil yang didapat isolat 3 memiliki serapan terhadap radiasi sinar UV-A dengan puncak tertinggi pada λ 340 nm dan ekstrak P. nigra memiliki serapan terhadap radiasi sinar UV-B dengan puncak tertinggi berada pada λ 290 nm. Though essential oils exhibit antibacterial activity against food pathogens, their underlying mechanism is understudied. We extracted ginger essential oil (GEO) using supercritical CO2 and steam distillation. A chemical composition comparison by GC-MS showed that the main components of the extracted GEOs were zingiberene and α-curcumene. Their antibacterial activity and associated mechanism against Staphylococcus aureus and Escherichia coli were investigated. The diameter of inhibition zone (DIZ) of GEO against S. aureus was 17.1 mm, with a minimum inhibition concentration (MIC) of 1.0 mg/mL, and minimum bactericide concentration (MBC) of 2.0 mg/mL. For E. coli, the DIZ was 12.3 mm with MIC and MBC values of 2.0 mg/mL and 4.0 mg/mL, respectively. The SDS-PAGE analysis revealed that some of the electrophoretic bacterial cell proteins bands disappeared with the increase in GEO concentration. Consequently, the nucleic acids content of bacterial suspension was raised significantly and the metabolic activity of bacteria was markedly decreased. GEO could thus inhibit the expression of some genes linked to bacterial energy metabolism, tricarboxylic acid cycle, cell membrane-related proteins, and DNA metabolism. Our findings speculate the bactericidal effects of GEO primarily through disruption of the bacterial cell membrane indicating its suitability in food perseveration.

Acronychia crassipetala is an endemic plant species in Australia. Its phytochemistry and therapeutic properties are underexplored. The hexane extract of the fruit A. crassipetala T. G. Hartley was found to inhibit the growth of the Gram-positive bacteria Staphylococcus aureus. Following bio-activity guided fractionation, two prenylated acetophenones, crassipetalonol A (1) and crassipetalone A (2), were isolated. Their structures were determined mainly by NMR and MS spectroscopic analyses. This is the first record of the isolation and structural characterisation of secondary metabolites from the species A. crassipetala. Their antibacterial and cytotoxic assessments indicated that the known compound (2) had more potent antibacterial activity than the antibiotic chloramphenicol, while the new compound (1) showed moderate cytotoxicity.

The aim of this study was to determine the effects of various concentration ofTerminalia catappabrown leaves extract which can inhibit the growth ofStaphylococcus aureusATCC 25923 andPseudomonas aeruginosaATCC 27853. The crushed-brown leaves ofTerminalia catappawas extracted using 95% ethanol, filtered, and evaporated. The driedT. catappaextract was used to identify phytochemical content qualitatively. Total phenolic and flavonoid contents were also measured quantitatively from dried extract. The dried extracts were also dissolved in sterile aquadest and serial dilutions were prepared to final concentration of 30, 60 and 90%. A disc diffusion method was used to evaluate the antibacterial activity of various concentrations of ethanol extract of brown leaves ofT. catappa. Inhibition zone diameter was measured to determine antibacterial activity. Gentamycin sulfate and distilled water were used as positive and negative controls, respectively. Dried ethanolic extract of brownT. catappaleaves contained flavonoid, quinon, phenolic, triterpenoid, and tannin. A total of 208.722 mg gallic acid equivalent/g extract of total phenolic and 35.7671 mg quercetin equivalent/g extract of total flavonoid were also found in the dried extract. The inhibition zone diameters of ethanolic extracts ranged from 1.73 to 9.06 mm (S. aureus) and from 1.83 to 6.5 mm (P. aeruginosa). The higher concentration of extract, the wider the inhibition zone diameters for both bacteria.P. aeruginosawas more resistant to high concentrations of extract (90%) thanS. aureus. Ethanolic extracts of the brown leaves ofT. catappahad different antibacterial effects againstS. aureusandP. aeruginosa. Currently multiresistant Staphylococcus aureus is one common cause of infections with high rates of morbidity and mortality worldwide, which directs scientific endeavors in search for novel antimicrobials. In this study, nine extracts from Bidens pilosa (root, stem, flower and leaves) and Annona crassiflora (rind fruit, stem, leaves, seed and pulp) were obtained with ethanol: water (7:3, v/v) and their in vitro antibacterial activity evaluated through both the agar diffusion and broth microdilution methods against 60 Oxacillin Resistant S. aureus (ORSA) strains and against S. aureus ATCC6538. The extracts from B. pilosa and A. crassiflora inhibited the growth of the ORSA isolates in both methods. Leaves of B. pilosa presented mean of the inhibition zone diameters significantly higher than chlorexidine 0.12% against ORSA, and the extracts were more active against S. aureus ATCC (p < 0.05). Parallel, toxicity testing by using MTT method and phytochemical screening were assessed, and three extracts (B. pilosa, root and leaf, and A. crassiflora, seed) did not evidence toxicity. On the other hand, the cytotoxic concentrations (CC50 and CC90) for other extracts ranged from 2.06 to 10.77 mg/mL. The presence of variable alkaloids, flavonoids, tannins and saponins was observed, even though there was a total absence of anthraquinones. Thus, the extracts from the leaves of B. pilosa revealed good anti-ORSA activity and did not exhibit toxicity.
