Effects of Synchro-Mate B and prostaglandin F2α on estrus synchronization and fertility in beef cattle

The efficiency and precision of estrus synchronization and fertility were evaluated after prostaglandin F2α (PGF) treatment with or without pretreatment with Synchro-Mate B (SMB) in beef cattle. In the treatment group (SMB-PGF, n = 109), postpartum suckled (n = 53), and nonsuckled (n = 32) cows and virgin heifers (n = 24) received SMB for 9 d and PGF 72 h before implant withdrawal. In the control group (Control, n = 111), postpartum suckled (n = 51), and nonsuckled (n = 36) cows and virgin heifers (n = 24) were observed for estrus and bred at spontaneous estrus over a 6-d period. Those not detected in estrus were then injected with PGF and bred at induced estrus over an additional 6-d period. Synchronization rate at induced estrus was higher (95.4 vs. 84.2%, P < 0.01) and time interval from induction to onset of estrus was shorter (37.0 ± 2.1 h vs. 56.7 ± 2.8 h, P < 0.01) in the SMB-PGF group than after PGF in the Control group. Whether the time around the mean interval to estrus (precision) was ± 6, ± 12, ± 18 or ± 24 h, the synchronization rate was always higher (P < 0.01) in the SMB-PGF than in the Control group. Heifers and cows in both groups had similar estrus synchronization rates and mean intervals to estrus. Suckled and nonsuckled cows behaved similarly in their estrus response within the SMB-PGF group but not in the Control group. Our results indicate that administration of PGF 72 h before SMB implant withdrawal could be a very efficient and precise synchronization method without a negative effect on fertility in beef cattle. The aim of the present study was to evaluate the effect of the short-term progestagen treatments plus PMSG prior ram introduction on the estrus synchronization and the fertility of Ile de France ewes in the beginning of mating season. The study was carried out with 36 pure-bred ewes (aged 4-6 years) during April 2009. The ewes were divided in 3 groups (n=12 for each group): group I - vaginal sponge impregnated with 30mg FGA for six days, as at the time of placement of sponge 125 ?g cloprostenolum was put i.m At the time of removal of the sponge 250 UI PMSG was put i.m.; group II- the same as the I group, but without cloprostenolum treatment: III group - control. The teasers were introduced to ewes 24h after the sponge removal and ewes in estrus was inseminated artificially. The following parameters were studied: effect of estrus synchronization (EES) ewes in estrus was recorded twice daily for the first 6 days after sponge removal; onset, end and duration of estrus for group I and II, fertility (at first estrus) and fecundity (calculated after lambing). The data for fertility and fecundity for the control group was obtained as ewes were fertilized during 30 days from the begging of the breeding. A significant effect of both schemes to estrus synchronization was determined F=33,33, P<0,001). The EES, fertility and fecundity for group I, II and III were - 91,66%, 63,64% and 142,0%; 91,66%, 45,45% and 140,0 %; 8,33%, 91,66% and 140,0% respectively. The mean onset, end and duration of estrus was 51,27 h, 77,45 h and 27,27h for the I group and 57,82h, 87,27h and 29,45 h respectively for the II group. We conclude that the scheme applied for the first group is better to use for estrus synchronization in the beginning of mating season.

The objective of this study was to evaluate the effect of inducing oestrus in superovulation-treated Japanese Black donor cows after egg collection using a controlled internal drug release (CIDR) device and prostaglandin F2α treatments. The test animals were 50 biparous or triparous Japanese Black cows that had been treated for superovulation during the period of 75-110 days post-calving and from each of which 10 or more ova or embryos had been recovered using a uterine reflux method. In the 25 cows of the control group that received the conventional treatment, a CIDR was not introduced into the vagina, the number of days from egg harvesting to standing oestrus was recorded, and AI was performed using frozen semen. At the time of AI, 50µg of gonadotrophin-releasing hormone (GnRH; Consultan, Aska Animal Health) was administered intramuscularly. The cows that did not conceive in the first AI attempt were again given the same AI and GnRH treatments in the next oestrus cycle. In the 25 cows of the treatment group, a CIDR (CIDR 1900, Zoetis) device was inserted in the vagina after egg harvesting and was kept there for 12 days. At the time of insertion and removal of the CIDR, 150µg of d-cloprostenol formulation (Dalmazin, Kyoritsu Seiyaku) was administered intramuscularly. The number of days from egg harvest to standing oestrus was recorded, and GnRH was administered at the time of AI. The cows that did not conceive in the first AI attempt were given the same AI and GnRH treatments in the next oestrus cycle. The control and treatment groups were compared for the number of days from egg harvesting to oestrus (t-test) and the conception rate after AI (chi-square test). The mean number of days from egg collection to standing oestrus in the control group was 38.6±11.9, and the conception rate was 60% (15/25). Among the animals that failed to conceive in the first AI attempt, the second standing oestrus occurred 60.7±13.1 days after egg collection, and 70% (7/10) of the cows conceived after the second AI. In the treatment group, on the other hand, the mean number of days from egg collection to standing oestrus was 15.8±3.9, and 72% (18/25) of the cows conceived after the first AI. Among the cows that failed to conceive, the second standing oestrus occurred 37.4±4.7 days after egg harvest, and 71% (5/7) of the cow conceived after the second AI. There was a significant difference between the two groups in the number of days from egg collection to the first standing oestrus (P&lt;0.05). The results described above suggest that keeping a CIDR in the vagina for 12 days and intramuscularly administering prostaglandin F2α at the time of insertion and removal of the CIDR in superovulated cows are effective for early onset of standing oestrus after egg collection and thus can reduce the number of days to conception.

This study was initiated on 24 prepubertal Holstein x Kankrej crossbred heifers of nearly identical age (7-9 months) and body weight (130-140 kg) at University farm to evaluate the effect of high plane of nutrition on blood biochemical and minerals profile and the age at puberty. Twelve heifers were managed under routine farm feeding (control) and the rest 12 under ideal optimum feeding regime (treatment) that included extra 1 kg concentrate, 30 g min mix and ad-lib dry fodder. The body weight and ovarian ultrasonography together with blood sampling was carried out at monthly interval from 10 to 18 months of age to study the ovarian dynamics and blood biochemical changes. High plane of nutrition to growing heifers was beneficial in reducing the age of onset of puberty (by 2-3 months) compared to routine farm fed group. The mean plasma total protein and cholesterol concentrations showed a rising trend with significant variations from 10 to 16 months of age, where it got mostly stabilized indicating adult profile. The activity of enzymes GOT and GPT also rose gradually and significantly from 10 months till 14-15 months of age, and thereafter it remained more or less static till 18 months of age. The levels of both these enzymes were higher, with lower protein and cholesterol, in control than the treatment group from 15-16 months of age onwards. The mean plasma levels of both calcium and phosphorus increased gradually and significantly with advancing age till 16-17 months of age, with little higher values in supplemented than a control group. The plasma levels of zinc, iron, copper, and cobalt also showed rising trend with significant differences between 10th and 12th-14th months of age, and from 15th to 18th months of age the levels were statistically the same in all the groups with slightly higher values in the treatment group.

Abstract Prenatal transportation stress (PNS) results in calves that are more temperamental and have greater circulating concentrations of cortisol compared to control calves. The objective of this experiment was to evaluate whether PNS alters the number of pituitary corticotrophs in mature Brahman cows. We hypothesized that the increased circulating cortisol concentrations previously characterized in this bovine model is associated with developmental changes in the anterior pituitary leading to an increased number of corticotrophs. Pregnant Brahman cows (n = 48) were transported in trailers for 2-hour periods at 60±5, 80±5, 100±5, 120±5, and 140±5 days of gestation. Non-transported pregnant cows (n = 48) were designated as the Control group. Control and PNS offspring heifers were managed together under the same environmental conditions. At approximately 5 yr of age, randomly selected non-pregnant cows (Control, n = 8; PNS, n = 6) were humanely harvested and the whole pituitaries were collected. Pituitaries were weighed, fixed in paraformaldehyde, serially dehydrated with graded ethanol, embedded in paraffin blocks, and cut into 5-μm sections. Immunohistochemistry was performed to detect cells expressing adrenocorticotropic hormone (ACTH) as a marker for corticotrophs. Three comparable sections from the midsagittal plane from each animal were processed using an ovine ACTH-specific antibody (Dr. A.F. Parlow, NIDDK). Five fields of view were analyzed per section (15 fields per animal). Anterior pituitary gland weight did not differ (P &gt; 0.10) between groups (Control = 2.11 ± 0.12 g; PNS = 2.10 ± 0.15 g). The mean number of ACTH-positive cells between control (531±56 cells/section) and PNS cows (477±49 cells/section) also did not differ (P &gt; 0.10). In conclusion, the number of pituitary corticotrophs in mature Brahman cows was not affected by prenatal transportation stress and is likely not a mechanism mediating the increased circulating cortisol concentrations seen in this bovine model of fetal programming.

Background: A Kader refers to a Village Health Worker (VHW), a volunteer, which becomes one of the sources of community reference. Commonly, they sustain a mother's knowledge regarding adequate complementary feeding. However, there are still some VHW who have not possessed a health education background nor been able to be independent.Objectives: This study aimed to analyze the impact of collaborative models on VHW's competence as in knowledge, self-efficacy, attitudes, and counseling skill on the topic of complementary feeding.Materials and Methods: The design of this research is a quasi-experiment control group pretest-postest, with the retrieval of subjects using purposive sampling. Subjects were selected based on the location of the posyandu. The population in this research was VHWs in the Lembang district were 40 VHWs in each group. The treatment group was given training intervention for 1 month with a collaborative model, combining several methods into a series. The main topic was counseling and complementary feeding. The control group was given booklets and leaflets. VHW's competence was measured using questionnaires. This research was conducted in January-April 2020.Results: Statistical test results before the treatment of both groups showed no difference (p>0,05) in each variable. Two months after the intervention, there were significant differensces in the mean score of knowledge (p=0,001), attitude (p=0,001), and VHWs self-efficacy (p=0,000) in both groups. VHW counseling skills (p=0,149) until the first month there was not a significant difference. Unexpectedly, in the second month, the VHW counseling skills could not be observed due to the global COVID-19 pandemic. The mean score of each group increased significantly, but the score of the treatment group was higher than the control group.Conclusions: A collaborative model is effective when compared to only providing booklets and leaflets in increasing VHW's knowledge, attitudes, and self-efficacy but not effective yet for VHW counseling skills.

The soybean aphid Aphis glycines Matsumura (Hemiptera: Aphididae) is a primary pest of soybeans and poses a serious threat to soybean production. Our studies were conducted to understand the effects of different concentrations of insecticides (imidacloprid and thiamethoxam) on A. glycines and provided critical information for its effective management. Here, we found that the mean generation time and adult and total pre-nymphiposition periods of the LC50 imidacloprid- and thiamethoxam-treatment groups were significantly longer than those of the control group, although the adult pre-nymphiposition period in LC30 imidacloprid and thiamethoxam treatment groups was significantly shorter than that of the control group. Additionally, the mean fecundity per female adult, net reproductive rate, intrinsic rate of increase, and finite rate of increase of the LC30 imidacloprid-treatment group were significantly lower than those of the control group and higher than those of the LC50 imidacloprid-treatment group (P < 0.05). Moreover, both insecticides exerted stress effects on A. glycines, and specimens treated with the two insecticides at the LC50 showed a significant decrease in their growth rates relative to those treated with the insecticides at LC30. These results provide a reference for exploring the effects of imidacloprid and thiamethoxam on A. glycines population dynamics in the field and offer insight to agricultural producers on the potential of low-lethal concentrations of insecticides to stimulate insect reproduction during insecticide application.

Background: The incidence of burns in Indonesia progressively increases with the increase in its population and industries. From January to September 2000, 158 patients were treated in the burn unit of Dr Soetomo Hospital with a mortality rate reaching 5,8%. Burns have a direct effect in causing both local and systemic changes in the body, not occurring in other injuries. In severe burns, a hypermetabolic state can occur, which increases cardiac workload and causes muscle atrophy and other morbidities. The purpose of this study is to examine the effect of propranolol on the hypermetabolic state in severely burned patients by measuring various clinical & laboratory parameters.Methods : This is an experimental study using pre and post test control group design with the objective of assessing the treatment outcome with oral propranolol given in 15 consecutive days for burn patients involving 25%- 60% TBSA. Measurements were taken three times, on day 0, 7 and 14.Results : Obtained 16 samples divided into 2 groups. In the treatment group, there was a significant decrease in CRP levels on days 0, 7 and 14 (p <0.05). The Mid Arm Circumference variable did not obtain a significant decreasing on days 0, 7 and 14. The albumin level studied showed a significant decreasing on day 0 & 7 days with a value of p = 0.045. From the comparison between the two groups, there were significant differences in CRP levels on days 0 & 14, with the mean value of the treatment group -5.12 + 2.88 and the mean value of the control group 2.86 + 7.37, and the value of p = 0.019.Conclusions: This study successfully proved that the administration of propranolol can overcome the effects of hypermetabolism which is characterized by decreasing in CRP levels.

As a result of IDD is the occurrence of impaired child growth makes researchers interested in conducting research on "Provision of iodized salt, food counseling about the source of iodine and goitrogenic substances with urinary iodine excretion status in elementary school children". Research on IDD is often carried out in primary school-age children, aged 6-12 years because of their vulnerability to iodine deficiency. The purpose of this study was to determine the effect of iodized salt interventions and counseling patterns of iodized and goitrogenic food consumption patterns on levels of urinary yodiun excretion in families with elementary school children. Research methods: The design of this study included quasi-experimental using a specific design that is "pre and post test control group design". The study population was elementary school children with a sample size of 30 children aged 9 -12 years in each group. Data collected included the consumption of nutrients by the 24-hour recall method, the results of urine iodine examination by the spectrophotometric method. The collected data is then analyzed with an independent sample T test. The results showed there were differences in urinary yodiun excretion levels in the two groups (treatment and control), while the mean in the treatment group before intervention was 106.97 ug / L and after the intervention was 43.19 ug / L. Whereas in the control group, the level of urinary yodiun excretion before intervention was 117.30 μg / L and after the intervention was 243.19 μg / L. The mean of respondents who consumed goitrogenic sources in the treatment group before the intervention (Yes = 63%, No = 37%), after the intervention (Yes = 23%, No = 77%). Whereas in the Control group before the intervention (Yes = 56%, No = 73%), after the intervention (Yes = 23%, No = 77%). The average amount of protein consumption before treatment was 47.91 µg/L ± 6.54 and 50.15 µg/L ± 12.52 after treatment. For consumption, an increase with a mean before treatment was 89.88 µg/L ± 38.45 and after treatment was 113 µg/L ± 26. The results of the independent sample t-test showed that in the treatment group there was no significant difference between after and before the intervention (p = 0.058). Whereas in the control group there were significant differences between before and after the intervention (p = 0.002). It can be concluded that there are many factors that need to be controlled in the provision of interventions, especially the use, type of salt and goitronic as well as the method of examination of iodine analysis in urine. Background. The aim of this study was to clarify the relationship between the change in estimated glomerular filtration rate (eGFR) and urinary albumin by antihypertensive treatment.Methods. We randomized 611 treated patients with morning hypertension into either an added treatment group, for whom doxazosin was added to the current medication, or a control group, who continued their current medications. We compared the change in eGFR and urinary albumin creatinine ratio (UACR) between the groups.Results. The extent of the reduction in eGFR was significantly greater in the added treatment group than in the control group (−3.83  versus −1.08 mL/min/1.73 m2,P=0.001). In multivariable analyses, the change in eGFR was positively associated with the change in UACR in the added treatment group (β=0.20,P=0.001), but not in the control group (β=−0.002,P=0.97). When the changes in eGFR were divided by each CKD stage, eGFR was significantly more decreased in stage 1 than in the other stages in the added treatment group (P<0.001), but no differences were seen in the control group (P=0.44).Conclusion. The reduction of eGFR could be seen only in the early stage of CKD, and this treatment appeared to have no negative effect on renal function.

Abstract Chronic heat stress is considered to decrease the immune functions which makes negative effect on broiler growth performance. Here, we investigated the relationship between chronic heat stress, growth performance, and immunity in the small intestine of broilers. The study included two groups (control and heat stressed group) with eight replications per group. Ten broilers of 20-day aged were allocated in each replication. On day 35, the treatment group was subdivided into two groups based on their body weights (heavy and low body weight). Although, there was only the control and treatment group on day 28. The growth performance decreased and expression of heat shock protein 70 (HSP70), HSP60, and HSP47 increased on days 28 and 35 in the chronic heat stress group as compared with those in the control group. The expression levels of HSPs were significantly higher in the low body weight group than in the control group. The genes HSP70 and HSP60 were significantly associated with pro- and anti-inflammatory cytokines in the small intestine of the broilers of the treatment group. Thus, HSP70 and HSP60 activated the adaptive immunity in the small intestines of the broilers from the treatment group to allow adaptation to chronic heat stress environment.
