Introduction: Bacteria and their virulence factors are the main causes of apical periodontitis after endodontic treatment. The bacteria that is often found and considered as pathogens in root canals after endodontic treatment is Enterococcus faecalis. The main goal of endodontic treatment is the elimination of microorganisms and preventing re-infection after root canal treatment which can be achieved through mechanical-chemical debridement using an irrigation solution. Basil leaves (Ocimum basilicum) which is an herbal plant are thought to have antibacterial compounds that can inhibit Enterococcus faecalis. This research was conducted to determine the antibacterial potential of the n-hexane extract of basil leaves against Enterococcus faecalis ATCC 29212. Methods: This experimental laboratory study tested the effectiveness of n-hexane extract of basil leaves (Ocimum basilicum) against Enterococcus faecalis ATCC 29212 using disk diffusion method with three times of repetition. Results: The results of this study indicated that the n-hexane extract of basil leaves did not have an inhibition zone against Enterococcus faecalis ATCC 29212. Discussion: The concentration of n-hexane extract of basil leaves was not directly proportional to the diameter of the resulting inhibition zone. This is due to several factors that can affect the test results. Conclusion: The n-hexane extract of basil leaves does not have antibacterial potential effect against Enterococcus faecalis ATCC 29212.

Pendahuluan: Enterococcus faecalis (E. faecalis) merupakan bakteri yang persisten pada saluran akar gigi, sehingga dapat menyebabkan infeksi sekunder pada saluran akar gigi yang telah diobturasi. Bahan irigasi dan medikamen harus diberikan pada saluran akar gigi untuk menunjang keberhasilan perawatannya. Daun kemangi (Ocimum basilicum/O. basilicum) merupakan tanaman obat tradisional yang diketahui memiliki potensi antibakteri dan berpeluang untuk dikembangkan sebagai bahan irigasi saluran akar gigi. Penelitian ini bertujuan untuk mengetahui zona hambat ekstrak etil asetat daun O. basilicum terhadap E. faecalis ATTC 29212. Metode:Jenis penelitian eksperimental laboratori. Pengujian zona hambat ekstrak etil asetat daun O. basilicum yang diperoleh dari proses maserasi dilakukan dengan metode difusi cakram dengan menggunakan klorheksidin 0,1% sebagai kontrol positif dan dilakukan tiga kali pengulangan terhadap E. faecalis ATCC 29212. Ekstrak pekat dilarutkan menggunakan DMSO 10% dan dilakukan pengenceran dua tingkat sehingga diperoleh konsentrasi ekstrak 512.000 – 1.000 ppm. Hasil: Ekstrak etil asetat daun O. basilicum pada konsentrasi uji 512.000 ppm telah memperlihatkan zona abu-abu seluas 1 mm di luar cakram uji, sementara konsentrasi uji lainnya tidak memperlihatkan adanya penghambatan. Beragam faktor dalam penelitian ini dapat mempengaruhi hasil pengujian, diantaranya adalah kelarutan ekstrak uji dalam pelarut yang digunakan, prosedur sebelum ekstraksi, dan keterbatasan metoda pengujian. Simpulan: Ekstrak etil asetat daun O. basilicum tidak memiliki zona hambat terhadap E. faecalis ATTC 29212.Kata kunci: Ocimum basilicum, Enterococcus faecalis ATCC 29212, zona hambat. ABSTRACTIntroduction: Enterococcus faecalis (E. faecalis) is a bacteria that persist in root canals, cause secondary infection in obturated root canals. Irrigation material and medicament must be administered to the root canal to support the treatment success. Basil leaves (Ocimum basilicum/O. basilicum) is a traditional medicinal plant known with antibacterial potential and has the opportunity to be developed as a root canal irrigation agent. This study was aimed to determine the inhibition zone of the ethyl acetate extract of O. basilicum leaves against E. faecalis ATTC 29212. Methods: This study was an experimental laboratory. The inhibition zone test of the ethyl acetate extract of O. basilicum leaves obtained from the maceration process was conducted with the disc diffusion method using 0.1% chlorhexidine as a positive control and three replications E. faecalis ATCC 29212. The concentrated extract was dissolved using 10% DMSO, and two dilution levels were carried out to obtain an extract concentration of 512,000 - 1,000 ppm. Results: The ethyl acetate extract of O. basilicum leaves at the test concentration of 512,000 ppm showed a grey zone of 1 mm outside the test disc, while other test concentrations did not show any inhibition. Various factors in this study can affect the test results, including the solubility of the test extract in the solvent used, the procedure before extraction, and the limitations of the test method. Conclusion: O. basilicum leaf ethyl acetate extract had no inhibition zone against E. faecalis ATTC 29212.Keywords: Ocimum basilicum, Enterococcus faecalis ATCC 29212, zone of inhibition.

Introduction: Individuals with poor oral health have a greater risk factor for systemic diseases. Caries, periodontal disease, and root canal infections are a common dental and oral diseases caused by dominance of Streptococcus mutans, Porphyromonas gingivalis, and Enterococcus faecalis bacteria (S. mutans, P. gingivalis, and E. faecalis). An alternative way to prevent dental and oral diseases is to use herbal medicine as one of the active ingredients for mouthwash or toothpaste. One of the herbs that can be used is Beluntas leaves (Pluchea indica (L.) Less leaves). The objective of study was to analyze the antibacterial activity of ethanol extract of Pluchea indica (L.) Less leaves on the growth of Streptococcus mutans, Porphyromonas gingivalis, and Enterococcus faecalis. Methods: This research was experimental laboratory with post test only control design, using disk diffusion method. There were five concentrations (2.5, 3.5, 4.5, 5.5 and 6.5%, positive controls, and negative controls. Data analysis was performed using One Way Anova and post Hoc test. Results: The ethanol extract of Pluchea indica (L.) Less leaves has moderate-strong antibacterial activity against Streptococcus mutans, Porphyromonas gingivalis, and Enterococcus faecalis. The largest diameter of inhibitory zone in Enterococcus faecalis at a concentration of 6.5% followed by Streptococcus mutans and Porphyromonas gingivalis at the same concentration and the smallest diameter of inhibition zone in Porphyromonas gingivalis, followed by Enterococcus faecalis and Streptococcus mutans at 2.5% concentration. Conclusion: The ethanol extract of Pluchea indica (L.) Less leaves with a concentration of 2.5, 3.5, 4.5, 5.5, and 6.5% has antibacterial activity in inhibiting the growth of Streptococcus mutans, Porphyromonas gingivalis, and Enterococcus faecalis.

Startfruit (Averrhoa bilimbi L.) is one the kind of plant that is widely used as a traditional herbal medicineto overcome various diseases i.e. diabetes mellitus, cough, rheumatism, thrush, diabetes, toothache, acne, etc. The secondary metabolites of the starfruit leaves contain flavonoids and tannins, whereby these activecompounds can be used as an antibacterial. This study aimed to determine the inhibiting power of starfruit leaf ethanol extract against Staphylococcus epidermidis at concentrations of 25%, 50%, 75%, and 100%. The research was conducted at the  AKAFARMA Laboratory and the Chemistry Laboratory , Faculty of Teacher Training and Education, Syiah Kuala University from June to July 2020. The research method was laboratory experimental using the disk diffusion method. The population of starfruit leaves was obtained from Lhokseumawe  using a purposive sampling technique. Ethanol extract of starfruit leaves with concentrations of 25%, 50%, 75%, and 100%. The diameter of the inhibition zone of the ethanol extract  from the starfruit leaves at  concentrations of 100%, 75%, 50%, and 25% were 15 mm, 12 mm, 11 mm and 10 mm, respectively. It can be concluded that  ethanol extract of starfruit leaves can inhibit the growth  of  Staphylococcus  epidermidis  with  a  maximum inhibitory concentration  of  100%,  which is classified  as a strong category.

The leaves of kenikir are known to have many benefits, one of which can be used as medicine. Kenikir leaves are reported to have antibacterial activity against the bacteria Salmonella typhi (S.typhi). S. typhi is a bacterium that causes typhoid fever. Typhoid fever is still an endemic disease in Asian countries, especially Indonesia. Typhoid fever became the third largest disease in Indonesia in 2010. The study related to the antibacterial activity of kenikir leaf fraction to S. typhi has never been reported. Therefore, it is necessary to conduct research related to the antibacterial activity of extracts and leaf fractions of kenikir to compare their potential against S. typhi. The study was conducted using the disk diffusion method using 5 series of concentrations namely 5%, 10%, 15%, 20%, and 30%. Tests were carried out on methanol extract, hexane, ethyl acetate, and methanol-water fraction. The results showed that highest inhibition zone diameter is 7,75 mm which is reached by methanol extract. The order of the highest inhibiting zone diameter are extract, hexane, ethyl acetate, and methanol-water fraction.

Genus Mentha presents group of plants which are the most studied in family Lamiaceae. Aboveground parts are used for different purposes in pharmacy, food industry or confectionery. Most important is natural product extracted from leaves - essential oil (EO). The aim of presented experiment was to demonstrate different chemotype and compare antibacterial activity of two Mentha species EO. Plant samples were obtained from various environments – from Slovakia and from Italy. Dominant compounds were determined by GC/MS. The results showed high amount of menthol and menthone in tested Slovak peppermint EO. On the other hand, carvone and 1,8-cineole were determinate as dominant compounds in Italian spearmint EO. The antimicrobial activity of the EO was investigated by disc diffusion and broth micro dilution methods. EO was evaluated for their antibacterial activity against 7 microorganisms: Enterobacter cloacae, Salmonella spp., Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus, Streptococcus pyogenes. The determination results of antibacterial activity by agar disk diffusion method ranged from 7 to 14 mm of the growth inhibition zone. MIC of tested mint EO varied from 0.625 to 2.5 μg/mL. In addition, both EO showed relatively the same antibacterial activity against the selected Gram-negative bacteria. However, there is a variation in the antibacterial activity against Gram-positive bacteria.

ABSTRACT We have evaluated a disk diffusion method to determine the activities of five drugs against 50 strains of dermatophytes and to assess the influence of the culture medium (antibiotic medium 3, high-resolution medium, and RPMI) on the inhibition zone diameters (IZD). There were no differences among the medium/drug combinations, except for itraconazole-RPMI, which showed the narrowest IZD.

The number of appointments necessary to treat infected root canals is one of the most controversial issues in endodontics. This study evaluated, in dogs, the response of the periradicular tissues to the endodontic treatment of infected root canals performed in a single visit or in two visits, using different interappointment dressings. Periradicular lesions were induced by inoculating Enterococcus faecalis in the root canals. After confirming that a periradicular lesion developed, the root canals were treated within one or two visits, using either ozonized oil or calcium hydroxide in camphorated paramonochlorophenol (CMCP) as an intracanal medication. After 6 months, the animals were sacrificed and the specimens were processed for histological and histobacteriological analysis. The root canals treated in a single visit showed a success rate of 46%. When a calcium hydroxide/CMCP-based interappointment intracanal medication was used, 74% of the cases were categorized as success. In cases where ozonized oil was used as the intracanal medication, a success rate of 77% was observed. These results of the present study demonstrated that the two-visit treatment offered a higher success rate compared to one-visit therapy. In addition, ozonized oil may potentially be used as an intracanal medication.

The aims of this research were to study the inhibition activity of essential oil and crude extract of nutmeg seed (Myristica fragrans and M. fattua) on growth of Xanthomonas campestris which is pathogen to broccoli (Brassica oleracea var. italica), to find out the best concentration of those substances to prevent X. campestris and to compare the inhibition activity of both substances on growth of X. campestris. The essential oil of nutmeg seed (M. fragrans and M. fattua) was obtained by Stahl distillation, while crude extract of the nutmeg seeds was extracted by methanol method. The method of this research was the disk diffusion method on nutrient agar. Paper disk was prestreaked on agar medium with X. campestris to be tested. Each petri disk was placed 5 paper disk dropped with the essential oil and the crude extract of the nutmeg seed in concentrations of 100%, 10%, 1% respectively, while for the comparison, the bactericide Agrept 0,2% was used. The parameters measured were the inhibition zone from the essential oil and the crude extract of the nutmeg seeds. The result of the analysis indicated that the essential oil of M. fragrans effectively inhibited X. campestris starting at concentration of 1%, the essential oil of M. fattua effectively inhibited X. campestris at concentration of 10% and the crude extract of M. fragrans effectively inhibited X. campestris at concentration of 100%. The crude extract of M. fattua was not effective in inhibiting the growth of X. campestris.

Saluang Belum (Luvunga sarmentosa Kurz) adalah tumbuhan yang dikenal sebagai obat tradisional di Kalimantan Tengah. Batang L. sarmentosa Kurz dipercayai memiliki manfaat secara empiris sebagai antioksidan yang dimanfaatkan sebagai anti-aging (anti penuaan dini). Batang L. sarmentosa Kurz mengandung metabolit sekunder yaitu tanin, saponin, steroid dan flavonoid yang diduga memiliki aktivitas sebagai antibakteri. Berdasarkan khasiat empiris dan kandungan metabolit sekunder yang ada dalam tumbuhan L. sarmentosa Kurz, maka dilakukan penelitian tentang aktivitas daya hambat ekstrak etanol tumbuhan ini terhadap bakteri P. acnes yang merupakan salah satu bakteri penyebab jerawat. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas daya hambat ekstrak etanol batang L. sarmentosa Kurz terhadap bakteri P.acnes. Metode penelitian yang dilakukan pada penelitian ini dimulai dari pengambilan simplisia, pembuatan ekstrak etanol dengan metode perkolasi dan uji daya hambat dengan menggunakan metode Disk Difusion yang menggunakan kontrol positif antibiotik klindamicin. Analisis data dilakukan dengan membandingkan lebar zona hambat dari ekstrak etanol batang L. sarmentosa Kurz  dengan kategori penghambatan antimikroba klindamisin berdasarkan diameter zona hambat dari Clinical and Laboratory Standards Institute.  Hasil uji daya hambat didapatkan zona hambat ekstrak etanol batang Saluang Belum pada semua konsentrasi 0,5%, 1%, 5%, 10% dan 15% secara berturut-turut yaitu  Intermediate, Resistant, Intermediate, Susceptible dan Susceptible dan hasil dari zona hambat klindamicin pada semua konsentrasi 0,5%,1%,5%,10%,dan 15% adalah susceptible. Simpulan dari penelitian ini adalah ekstrak etanol batang L. sarmentosa Kurz mempunyai kemampuan dalam menghambat pertumbuhan bakteri P.acnes.           Kata Kunci: Obat Tradisional, Jerawat, Uji Daya Hambat, Propionibacterium acnes, Batang Saluang Belum Saluang belum (Luvunga sarmentosa Kurz) is one of traditional medicine in Central Kalimantan. The stem of L. sarmentosa Kurz is believed to have empirical benefits as an antioxidant which is used as anti-ageing (anti-ageing). The stem of L. sarmentosa Kurz contains secondary metabolites, namely tannins, saponins, steroids and flavonoids which are thought to have antibacterial activity. Based on the empirical properties and the content of secondary metabolites in L. sarmentosa Kurz, a study was conducted on the inhibitory activity of this plant's ethanol extract against P. acnes, which is one of the bacteria that causes acne. The purpose of this study was to determine the inhibitory activity of the ethanol extract of L. sarmentosa Kurz stem against P.acnes bacteria. The research method carried out in this study started from taking simplicia, making ethanol extract using the percolation method and the inhibition test using the disk diffusion method which used a positive control of the clindamycin antibiotic. Data analysis was performed by comparing the inhibition zone width of the ethanol extract of L. sarmentosa Kurz stem with the clindamycin antimicrobial inhibition category based on the inhibition zone diameter of the Clinical and Laboratory Standards Institute. The results of the inhibition test obtained the inhibition zone of Saluang Belum stem ethanol extract at all concentrations of 0.5%, 1%, 5%, 10% and 15% respectively, Intermediate, Resistant, Intermediate, Susceptible and Susceptible and the results of the inhibition zone. Clindamycin at all concentrations of 0.5%, 1%, 5%, 10%, and 15% are susceptible. This research concludes that the ethanol extract of L. sarmentosa Kurz stem can inhibit the growth of P.acnes bacteria.

ABSTRACT Introduction The main goal of endodontics is the prevention of apical periodontitis. This is due to the presence of persistent pathogenic microorganisms, such as Enterococcus faecalis, and its ability to directly cause acute and chronic inflammation in the periapical tissues. Lactobacillus has been shown to promote health in the intestines as well as to inhibit the growth of certain problematic oral bacteria. This study explores shifting the established paradigm of endodontic treatment, which has focused on eliminating all bacteria from the canal system and on elimination of the problematic bacteria through introducing probiotics. A preliminary work was performed to evaluate the possible effectiveness of probiotics in preventing the growth of E. faecalis. Materials and methods Two probiotic cultures Lactobacillus plantarum ATCC 8041 and Lactobacillus rhamnosus ATCC 7408 were selected to check their antimicrobial activity against E. faecalis ATCC 29212 by two methods: agar cup/well diffusion method and deferred antagonism test. Agar cup method A total of 0.5 mL of requisite test pathogen culture was inoculated into 20 mL of molten sterile Mueller and Hinton agar and cooled to 45 ± 2°C. Circular wells of diameter 10 mm were punched in each of the poured plates. Appropriately diluted test samples were added to the above-punched wells. The plates were incubated upright position at 37°C for 24 hours in aerobic conditions. Postincubation, zone of inhibition was measured. The cell-free supernatant of Lactobacillus species was also evaluated for antimicrobial activity. Deferred antagonism test The test probiotic strain was standardized to 0.1 optical density (OD) at 600 nm and inoculated in a 1 cm wide diametric streak across the surface of trypticase soy agar + yeast extract + calcium carbonate (TSYCa) agar using a sterile cotton swab. Then, the plate agar containing the test strain was incubated at 37°C for 24 hours under aerophilic condition and then standardized to 0.1 OD at 600 nm for overnight (18 hours, 37°C). A purified culture of indicator strain (pathogen) was streaked at right angles to the line of original producer growth. Postincubation plates were observed for the inhibition zone width of the indicator strain. Results Under the conditions of this study, Lactobacilli had an inhibitory effect on the growth of E. faecalis by agar cup method but not by deferred antagonism test. Conclusion This pilot study demonstrated that probiotics show a potential in root canal therapy. Clinical Significance If probioitcs are effective against endodontic pathogens, they can be potentially used as intracanal medicaments. This will be a novel concept of introducing bacteriotherapy in endodontics and replacing pathogenic bacteria by healthy bacteria, normal flora. How to cite this article Bohora AA, Kokate SR.
