This study evaluated the effect of aqueous extract of Senna occindentalis leaves on some biochemical parameters in Wistar rats. Twenty albino rats equally divided into four experimental groups were used. One group served as control and received the carrier solvent treatment. Three test groups were treated with S. occidentalis extract at 500, 1000 and 1500 mg/kg body weight respectively. The experiment lasted for 14 days after which the rats were sacrificed and blood collected for biochemical and haematological evaluation. Liver-body weight ratio was computed and liver histoarchitecture was investigated. The results showed that all haematological parameters were significantly (P<0.05) affected except the mean corpuscular haemoglobin concentration and mean corpuscular volume. There were also significant (P<0.05) alterations in the activities of gamma-glutamyl transferase, alanine aminotransferase, and aspartate aminotransferase, as well as the levels of total protein, albumin and globulin in the serum. No significant (P>0.05) alterations were observed in the computed liver-body weight ratio but marked alterations in histoarchitecture of the liver cells were present. These alterations in the haematological parameters, liver function enzymes and histological evaluation suggest a selective toxicity of the extract on the animals.

Background: Garcinia kola seed is consumed indiscriminately in Nigeria without recourse to its potential toxicity. Therefore, this study was aimed at assessing the toxicity of the aqueous extract of G. kola seeds on selected tissues of male rats. Methods: Thirty male rats (215.00 ± 18.58 g) were assigned into four groups: A, B, C and D which received 0.5 ml of distilled water, 25, 50 and 100 mg/kg body weight of the extract respectively, once daily for 7 days. Biochemical indices of organ damage and toxicity were determined using standard methods. Results: The extract significantly (P<0.05) increased the testes-body weight ratio, activities of testicular alkaline phosphatase (ALP), heart, testes and serum gamma glutamyl transferase (GGT) activity, serum concentrations of uric acid, K+, creatinine and PO43-. The liver-body weight ratio, activities of kidney and serum ALP, liver, heart and serum alanine and aspartate aminotransferases (ALT and AST), serum and testicular acid phosphatase (ACP), concentrations of serum albumin, globulin, urea, Na+ , HCO3-, conjugated and total bilirubin were reduced. The heart- and kidney-body weight ratios and liver ALP were not significantly (P>0.05) altered. Conclusion: The treatment related alterations in the present study indicates that the aqueous extract of G. kola seeds at the doses of 25, 50 and 100 mg/kg body weight caused functional toxicity to the organs of the animals and thus not safe as an oral remedy.

The toxicological effect of the aqueous extract of fresh, dried and boiled berries of Solanum aculeastrum Dunal at 1, 10 and 25 mg/kg body weight was investigated in male Wistar rats for 28 days. The parameters used were the body weight of the animals and absolute weights of the organs, haematological parameters, renal and liver functional endpoints. The animals gained appreciable weight and showed no signs of clinical toxicity. The dried (DB), boiled dried (BDB), fresh (FB) and boiled fresh berry (BFB) extracts reduced (p < .05) the heart-, liver-and spleen-body weight ratio of the animals whereas that of the lung was not altered. The kidney and testes-body weight ratios were specifically altered by the different extract. All these were not accompanied by any histomorphological changes. The extracts did not alter (p > .05) the levels of RBC, Hb, PCV and albumin of the animals. The platelets were decreased by the DB and FB whereas BFB increased this parameter. The FB and BFB at all the doses also reduced the mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC) of the animals. With the exception of the FB where the creatinine and chloride levels decreased, other extracts did not alter the level of these kidney parameters. Only FB increased the levels of uric acid and urea. All the extract decreased the serum alanine aminotransferase (ALT) of the animal. The levels of total protein, globulin, total and conjugated bilirubin were not altered by DB and BDB whereas these indices were increased by FB and BFB. The DB and BDB increased the serum alkaline phosphatase (ALP) activity whereas FB decreased the activity of the enzyme. In contrast, DB and BDB decreased the serum aspartate aminotransferase (AST) activity of the animals whereas FB and BFB increased the activity of the enzyme. The FB and BFB also increased the levels of potassium, magnesium and phosphorus of the animals. Overall, the alterations in the biochemical parameters by the various extracts of S. aculeastrum berries at these doses indicated that the normal functioning of these organs may be adversely affected. However, drying and boiling might reduce the toxic effect of the berries.

Khat is a psychoactive herbal drug of pronounced ethno-pharmacological significance often abused due to its unregulated use. It affects many brain centers including the prefrontal cortex which is the anterior most part of the frontal lobe. The prefrontal cortex modulates working memory, planning complex cognitive behaviors however; it is linked to many psychological disorders such as depression, schizophrenia and memory loss. We studied the effects exerted by khat on the PFC cytoarchitecture and functions since this part of the brain is highly interconnected with various cortical regions. This was an experimental study of 6 weeks. A total of 24 male adult wistar rats of 130g-155g were divided into four groups of 6 animals that received respective khat doses of 2000mg/kg, 1000mg/kg, 500mg/kg and 10ml/kg of distilled water for the controls. Brain to body weight ratio was determined at week 6 using an analytical balance (Fisher Science Education™, RS232C; USA). Histology of the brain was determined using H and E and Kulvers staining technique. Khat exhibited features of prefrontal cortex disorientation such as necrosis, vacuolations, chromatolysis, demyelination, cortical degeneration and hemorrhage in a dose dependent manner. Selective attention and working memory were impaired well as brain to body weight ratio was reduced significantly (P ≤ 0.05). Repeated exposure to khat distorts the prefrontal cortex cytoarchitecture and impairs selective attention and working memory accuracy due to ischemia and cell exhaustion by khat toxicity.Keywords: Khat, prefrontal cortex histology, working memory, selective attention

Non-alcoholic fatty liver disease (NAFLD) is currently estimated as the most prevalent chronic liver disease in all age groups. An increasing body of evidence obtained in experimental and clinical data indicates that oxidative stress is the most important pathogenic factor in the development of NAFLD. The study aimed to investigate the impact of α-lipoic acid (LA), widely used as an antioxidant, on the effects of a hypercaloric choline-deficient diet. Male Wistar rats were divided into three groups: control diet (C); hypercaloric choline-deficient diet (HCCD), and hypercaloric choline-deficient diet with α-lipoic acid (HCCD+LA). Supplementation of HCCD with LA for eight weeks led to a decrease in visceral adipose tissue/body weight ratio, the activity of liver glutathione peroxidase and paraoxonase-1, plasma, and liver total antioxidant activity, as well as an increase in liver/body weight ratio, liver total lipid and triglyceride content, and liver transaminase activities compared to the HCCD group without LA. In conclusion, our study shows that α-lipoic acid detains obesity development but exacerbates the severity of diet-induced oxidative stress and lipid accumulation in the liver of male Wistar rats fed a hypercaloric choline-deficient diet.

Despite the popular use and the biological effects of Anvillea radiata, there are no studies or data about its safety. The aim of the present study was to assess the acute toxicity of A. radiata aqueous extract in vivo. A single dose of 0.25, 0.5, 1, 1.5, 2.5 or 5 g/kg was administered to female rats by gavage. Body weight gain, general behavior and mortality were monitored for up to 2 weeks. Selected biochemical parameters, aspartate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen (BUN) and blood creatinine levels were determined, as well as, liver and kidney histology. Results showed no significant changes in body weight gain and organ indexes with no mortality during the experimentation period. A significant increase in AST and ALT levels were observed in 2.5 and 5 g/kg extract treated groups, and a significant decrease in BUN and creatinine levels in 1, 1.5, 2.5 or 5g/kg extract treated groups compared to control. Microscope examination of liver sections showed several anomalies in rats exposed to high concentrations (1.5, 2.5 and 5 g/kg) including fatty changes, glycogen accumulation and ballooning degeneration hepatocytes. Renal parenchyma anomalies were also observed in rats exposed to 2.5 and 5g / kg of plant extract including shrunken renal corpuscles with marked hypo-cellularity and atrophied glomeruli, large interstitial space, and renal tubules with dilated lumina which appear completely distorted. From this study, it can be concluded that Anvillea radiata aqueous extract at high concentration (higher than 1 g /kg b.w.) may be toxic and affect sensitive organs function such liver and kidney. Abstract Background The cytotoxic properties of nanoparticles have attracted a great deal of attention in the field of nanoscience and nanotechnology due to their small size and ability to penetrate cellular membranes. Methods The silver nanoparticles were synthesized using Elaeodendron croceum stem bark and characterized. The oral acute toxicity studies were carried out by administration of 500, 1000, 2000 mg/kg body weight to Wister rats in respective groups. An in vitro cytotoxicity assay was evaluated in MDA-MB-231 breast cancer cells using the WST-1 Cell Proliferation assay. The percentage of cell viability after treatment with aqueous extracts of Elaeodendron croceum (ECE) and Elaeodendron croceum silver nanoparticles (ECAgNPs) was compared with that of paclitaxel. Results The in vivo studies revealed that the LD50 was higher than 2000 mg/kg and there was no significant difference (p>0.05) between the treatment groups compared with the control group for mean organ-to-body weight ratio except in the liver and in all hematological parameters except WBC and hematocrit. Similarly, there was no significant difference (p>0.05) for serum electrolytes (Na+, Mg2+ K+, Cl−, and Ca2+), total protein, urea, ɣ-glutamyl transferase (GGT), Aspartate aminotransferase (AST), Alkaline phosphatase (ALP), Alanine aminotransferase (ALT), albumin, total and conjugated bilirubin between the treatment and the control group. However, there were changes in creatinine, urea, and cholesterol. In the in vitro assays, ECE and ECAgNPs showed IC50 values of 70.87±2.99 and 138.8±3.98 µg/mL respectively against MDA-MB-231 cells compared to paclitaxel, which showed an IC50 value of 80 ng/mL. Conclusion The results showed that the LD50 of the ECE and ECAgNPs in Wister rats was determined to be greater than 2000 mg/kg body weight. The aqueous extract also showed more cytotoxic than the ECAgNPs suggesting that the toxic compounds in aqueous extract were involved in the capping of the AgNPs.

The present study investigated the effect of leaf extract of Gangronema latifolium (G. latifolium) on acetaminophen (APAP) - induced liver injury in Wistar albino rats. In this study, sixty (60) male Wistar albino rats were divided into five (5) groups of twelve (12) rats each. Animals in group 1 served as control group and received a placebo of 0.9% saline solution. Group 2 served as APAP control group, administered with 800 mg/kg body weight of APAP only. Groups 3, 4 and 5 served as the experimental groups and received oral dosage of 800 mg/kg body weight of APAP plus 150 mg/kg, 200 mg/kg and 250 mg/kg body weight of G. latifolium respectively. The results showed that the enzymatic activities of alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and gamma-glutamyl transferase (GGT) in the serum were decreased significantly (p ≤ 0.05) in the experimental groups dosed with 150 mg/kg, 200mg/kg and 250 mg/kg of G. latifolium respectively. For 150 mg/kg G. latifolium treated group, ALT decreased from 23.3 ± 7.31 to 9.00 ± 1.52 IU/L, while AST and ALP decreased from 17.6 ± 2.66 to 15.00 ± 1.00 IU/L and 92.8 ± 2.34 to 83.8 ± 7.94 IU/L respectively. In conclusion, the results showed that aqueous extract of G. latifolium has a protective effect on rat liver induced with APAP injury.

Abstract Background Garcinia lanceifolia Roxb. has been used by many ethnic communities of Northeast India to mitigate various disorders like dyspepsia, ulcers, diabetes, etc. However, a robust scientific study on its antidiabetic and antiulcer potential is unavailable till date. The aim of this present study is to scientifically validate if the antidiabetic and antiulcer effects reported by the ethnic tribes of Assam has any scientific value or not. The effects were tested in adult Wistar albino rats using approved animal models for preclinical testing of pharmacological activities. Results The hydroalcoholic extract of the bark of Garcinia lanceifolia Roxb. was prepared and its LD50 was calculated. The LD50 was determined to be greater than 5000 mg/kg body weight. The extract at doses of 250 mg/kg body weight and 500 mg/kg body weight was found to exhibit a very potent dose-dependent antidiabetic activity. The results were backed by a battery of test including analysis of serum levels of blood glucose, lipid profiles, in vivo antioxidant enzymes, and histopathological studies. Evidence of dose-dependent antiulcer activity of the extract was backed by robust scientific data. It was found that HAEGL induced a significant dose-dependent increase in the ulcer index in both alcohol-induced and acetic acid-induced ulcer models, which was evident from the macroscopic observation of the inner lining of the gastric mucosa and the histological evaluation of the extracted stomach. Conclusion The results suggested that the bark of Garcinia lanceifolia (Roxb.) has significant antidiabetic and antiulcer potential. Further studies with respect to the development herbal dosage forms and its safety evaluation are required.
