Propionibacterium acnes are aerotolerant anaerobic, gram-positive bacilli that form part of normal flora. They produce several pro-inflammatory substances that can trigger an immune response in the host by an influx of inflammatory leukocytes into the strands, causing inflammatory lesions that leave behind scars. Repeated isolation of Propionibacterium acnes may reduce efficacy among the resistant types, clearly explaining Acne lesions' importance. The Counter acne therapies are often the first treatment choice due to the convenience of cost and time over clinical appointments. However, not all of the commercially available anti-acne formulations are supported by clinical studies. The present study was conducted to test the efficacy of selected commercial anti-acne gel formulations. The microscopic observation and biochemical studies conform to the presence of anti-acne activity. A sensitivity test was performed on all the isolates of Propionibacterium acnes by well diffusion technique. The selected over-the-counter anti-acne gel formulations failed to produce any inhibition zone.

Staphylococcus aureus is one of the normal flora in the oral cavity, but these bacteria can change to be pathogen if there are predisposing factors such as changes in quantities of bacteria and decreasing of the immune response of the body. One of the ways to prevent is to inhibit the activity of these bacteria using antibiotics. Ampicillin and tetracycline are the broad-spectrum antibiotics that can inhibit the growth of positive and negative gram bacteria. This research was a laboratory experimental with Kirby-Bauer method and using CLSI standard. Total sample are 16 discs of ampicillin and 16 discs of tetracycline then placed on MHA media contain Staphylococcus aureus to see the inhibition power. The result showed that the averages of inhibition zone of ampicillin is 36,64 mm while tetracycline is 25,58 mm. Independent sample t- test showed that p<0,01 which means there is a significantly difference between ampicillin and tetracycline. Ampicillin and tetracycline can inhibit the growth of Staphylococcus aureus with sensitive categories. Ampicillin is more sensitive and effective than tetracycline to inhibit the growth of Staphylococcus aureus

Background: Acne is caused by several factors including the active secretion of sebaceous sweat glands, hyperkeratosis in the hair infundibulum and the effects of bacteria. One of the plants that has the potential as an antibacterial is the extract of arumanis mango leaves. Method: Determine the Minimum Inhibitory Concentration (MIC) of methanol extract of mango leaves which can inhibit Propionibacterium acnes activity. The antibacterial activity tests were performed using agar diffusion. The ointment formulation, the characteristics of ointment preparations and the ointment activity against P. Acnes are discussed. Result: MIC of methanol extract of mango leaves value is 5 ppm with an inhibition zone of 1 mm. The ointment obtained is white, has distinctive smell, semisolid form, possesses a pH of 4.92 - 5.87, dispersive power of 5.05 - 6.30 cm, adhesive power of 1 - 3.67 seconds, homogeneous and protective. Ointment preparations of methanol extract of mango leaves has activity on P. acnes on the 0 and 15th day of storage. The activities of ointment preparation on day 0 with concentrations of 0, 5, 10 and 15 ppm are 0.00 mm; 10.20 mm; 19.97 mm and 23.60 mm respectively, while the inhibition zones produced by the preparation of ointment on day 15 with concentrations of 0, 5, 10 and 15 ppm are 0.00 mm; 5.71 mm; 9.58 mm and 21.88 mm respectively. Conclusion: Methanol extract of mango leaves (Mangifera indica L.) and oinment preparation are able to inhibit the growth of Propionibacterium acnes.

ABSTRACTNB-003 and NB-003 gel formulations are oil-in-water nanoemulsions designed for use in bacterial infections.In vitrosusceptibility ofPropionibacterium acnesto NB-003 formulations and comparator drugs was evaluated. Both NB-003 formulations were bactericidal against allP. acnesisolates, including those that were erythromycin, clindamycin, and/or tetracycline resistant. In the absence of sebum, the MIC90s/minimum bactericidal concentrations (MBC90s) for NB-003, NB-003 gel, salicylic acid (SA), and benzoyl peroxide (BPO) were 0.5/2.0, 1.0/2.0, 1,000/2,000, and 50/200 μg/ml, respectively. In the presence of 50% sebum, the MIC90s/MBC90s of NB003 and BPOs increased to 128/1,024 and 400/1,600 μg/ml, respectively. The MIC90s/MBC90s of SA were not significantly impacted by the presence of sebum. A reduction in the MBC90s for NB-003 and BPO was observed when 2% SA or 0.5% BPO was integrated into the formulation, resulting in MIC90s/MBC90s of 128/256 μg/ml for NB003 and 214/428 μg/ml for BPO. The addition of EDTA enhanced thein vitroefficacy of 0.5% NB-003 in the presence or absence of 25% sebum. The addition of 5 mM EDTA to each well of the microtiter plate resulted in a >16- and >256-fold decrease in MIC90and MBC90, yielding a more potent MIC90/MBC90of ≤1/<1 μg/ml. The kinetics of bactericidal activity of NB-003 againstP. acneswere compared to those of a commercially available product of BPO. Electron micrographs ofP. acnestreated with NB-003 showed complete disruption of bacteria. Assessment of spontaneous resistance ofP. acnesrevealed no stably resistant mutant strains.

Structure and biological application based analysis has been carried out for Schiff’s base ligand containing nitrogen and sulphur donor atoms and also for a series of its coordination compounds. Ligand benzil-bis(5-amino-1,3,4-thiadiazole-2-thiol) (L) has been prepared and structural features of ligand investigated by elemental analyses, IR, 1 H-NMR, mass spectra and molecular modeling studies. The chemical reaction of the prepared ligand with metal ions afforded mononuclear coordination compounds of Mn(II) and Co(II) metal ions. The characterization of the coordination compounds is being based on the techniques i.e. elemental analysis, infra-red, UV-visible as well as melting point, molar conductivity and magnetic moment measurement. A computational study has been done to understand the miscellaneous coordination modes of ligand to metal ions. Characterization result shows that coordination compounds exhibit a six coordinated geometrical arrangement i.e. octahedral geometry. The biological inhibition zone (antifungal and antibacterial activities) of synthesized compounds, i.e. ligand and its coordination compounds, has been monitored using well diffusion technique

The previously reported sodium polyanethol sulfonate disk test for the identification of Peptostreptococcus anaerobius (Graves et al., 1974) was evaluated, with modifications. Three bands of brucella agar, three inoculum sizes, and two inoculum sources were compared. Nine stock cultures of P. anaerobius (eight normal flora isolates and ATCC 27337) and 16 fresh clinical isolates were used. All cultures of P. anaerobius showed inhibition zones of 12 to 30 mm in diameter, regardless of test conditions. Out of 103 clinical isolates of other species of anaerobic gram-positive cocci tested, only two had an inhibition zone size in this range (one P. micros of 11 studied had a zone of 12 mm and one P. prevotii of 14 studied had a zone of 16). The test had an overall accuracy of 98% in the identification of P. anaerobius from clinical specimens. Since P. anaerobius accounts for one-fifth to one-third of all anaerobic gram-positive cocci encountered in clinical specimens, this simple and rapid technique can be very useful for presumptive identification.

Benchalokawichian (BLW) remedy is a Thai traditional medicine that has been notified in the List of Medicine Products of the National List of Essential Drugs A.D. 2006 and has long been used as an antipyretic. The phytochemicals of the 70% and 95% ethanolic extracts of were studied by color reaction test and HPTLC analysis. The BLW remedy showed a highest amounts of 70% ethanolic extractives (ca. 3.99 ± 0.60% of dry material weight). The several ethanolic extracts showed similar qualitative phytochemicals. The major of phytochemicals identified in these extracts were alkaloids, flavonoids, tannins, and polyphenols. The antioxidant activity was assessed using the 2,2-diphenyl-1-picrylhydrazyl assay. The analysed 70% ethanolic extract showed a highest antioxidant activity at IC50= 526.09 µg/ml and both extracts not showed tyrosinase inhibitory effect. The 95% ethanolic extract showed the greatest anti-propionibacterium acnes activity with inhibition zone of 10.10 ± 0.45 mm and minimum inhibitory concentration (MIC) was 5.05 mg/ml

The cellular and humoral immune responses of mice inoculated with rabies virus and treated with the Bacillus of Calmette-Guérin, Avridine and Propionibacterium acnes were evaluated in this paper. There was a higher percentage of surviving mice in groups submitted to P. acnes treatment. Lower levels of interferon-<FONT FACE="Symbol">g</font> (IFN-<FONT FACE="Symbol">g</font>) were found in infected mice. The intra-pad inoculation test (IPI) was not effective to detect cellular immune response, contrary to the results found in MIF reaction. The survival of mice did not present correlation with the levels of antirabies serum neutralizing (SN) antibodies titers, IFN-<FONT FACE="Symbol">g</font> concentration and MIF response.

Acne is associated with hyperkeratosis, elevated levels of skin sebum and growth of Propionibacterium acnes (P. acnes) and Staphylococcus aureus (S. aureus). Furthermore, P. acnes promotes inflammation by inducing IL-6 production and oxidative stress. The aim of this study was to assess the antioxidant, anti-inflammatory and antibacterial potential of a hop-CO2-extract with 50% humulone and lupulone. The susceptibility of P. acnes and S. aureus to the hop extract was tested by using the broth microdilution technique. The minimal inhibitory concentrations (MIC) for P. acnes and S. aureus were 3.1 and 9.4 µg/mL, respectively. In addition, the hop extract showed an antioxidative effect with a half maximal inhibitory concentration (IC50) of 29.43 µg/mL as well as additional anti-inflammatory effects by reducing the IL-6 expression (IC50: 0.8 µg/mL). In addition, a gel formulation with 0.3% hop extract (w/w) had antibacterial activity against P. acnes and S. aureus (inhibition zone value: 5.5 mm and 3 mm, respectively) which was significantly superior to the placebo gel. The positive control (a gel with the antibiotic clindamycin) showed an inhibition zone of 9 mm. Due to its antioxidant, anti-inflammatory and antibacterial effects hop extract might be a treatment option for acne-prone skin.

Objective: The objective of this research was to investigate the preliminary phytochemical screening and antibacterial activity of Bawang Dayak(Eleutherine sp.) and Hati Tanah (Angiopteris sp.) and their combination against Propionibacterium acnes.Methods: The extracts were used for phytochemical screening. Antibacterial activity was performed using disc diffusion technique, with twovariations of the concentration of 5% and 10% for each extracts, and combination of both extracts with three combinations: (1) 5%: 5%, (2) 5%: 10%, and (3) 10%: 5%.Results: Both extracts contained tannins, saponins, and steroids. The antibacterial activity against P. acnes showed that the inhibition zones ofBawang Dayak ethanol extract were 6.1±1.5 mm (5%) and 8.7±1.3 mm (10%). On the other hand, the inhibition zones of Hati Tanah ethanol extract were 4.0±1.6 mm (5%) and 9.2±2.5 mm (10%). The inhibition zones produced in combinations 1, 2, and 3 were 5.8±0.3 mm, 10.8±2.0 mm, and 15.5±2.8 mm, respectively.Conclusion: In this study showed the presence of tannins, saponins, and steroids that might be responsible for antibacterial activity in both extract. The best antibacterial activity was produced by combining the two extracts (combination 3).
