Senecavirus A (SVA), classified into the genus Senecavirus in the family Picornaviridae, causes an infectious disease in pigs. This virus can efficiently replicate in some non-pig-derived cells, such as the BHK cell line and its derivative (BSR-T7/5 cell line). We had recovered a wild-type SVA from its cDNA clone previously, and then uncovered the proteomic profile of SVA-infected BSR-T7/5 cells at 12 h post inoculation (hpi). In order to explore the cellular metabolomics further, the SVA-inoculated BSR-T7/5 cell monolayer was collected at 12 hpi for assay via liquid chromatography-tandem mass spectrometry (LC-MS/MS). The resultant data set was comprehensively analyzed using bioinformatics tools. A total of 451 metabolites were identified using in-house and public databases. Out of these metabolites, sixty-one showed significantly differential values (p value < 0.05). The Kyoto Encyclopedia of Genes and Genomes (KEGG) database was used to analyze metabolic pathways of the significantly differential metabolites. There were eighty-one identified KEGG pathways, out of which twenty-seven showed their p values < 0.05. The pyrimidine metabolism revealed the minimum p value and the maximum number of significantly differential metabolites, implying the pyrimidine played a key role in cellular metabolism after SVA infection. SVA replication must rely on the cellular metabolism. The present study on metabolomics would shed light on impacts of SVA-induced multiple interactions among metabolites on cells or even on natural hosts.

This paper examines the relationship between orange economy activity which is an activity that allow for ideas to be transformed into cultural goods, local genius, place identity, cultural policies, and sustainability intangible cultural heritage (ICH) of Balinese handwoven textiles. A questionnaire survey was administered to 145 respondents. Partial Least Squares-Structural Equation Modelling (PLS-SEM) was applied to the resultant data using SmartPLS 3.0 software. The result revealed local genius, orange economy activity, and place identity have positive and significant influence on sustainability ICH of Balinese handwoven textiles. A positive and significant direct effect between local genius and place identity to the orange economy activity was also found. The result also proved that the orange economy mediates the relationship between local genius and place identity on sustainability. Moreover, cultural policies moderates the relationship between orange economy and sustainability. Our findings might also be relevant to sustainability Bali’s ICH to strengthening the involvement of cultural industry through orange economy activity, in enhancing their place identity and local genius, in supporting and promoting the sustainability ICH of Balinese handwoven textiles. Furthermore, the role of government through cultural policies might also relevant as moderating effect the relationship between orange economy activity and sustainability of ICH.

Graphene is one of the most commonly researched materials in nanoscience and finding a cheap and efficient method to manufacture it is highly desirable because of its incredible properties. Electrochemical exfoliation involves splitting graphite into graphene by soaking the foil in an electrolyte solution and then providing an electric current. This paper evaluates the extent to which the sulphate electrolyte used in the electrochemical exfoliation process affects the electrical resistance of films created using flakes generated from the reaction. Using the method and conducting an ANOVA test with Tukey HSD Post-Hoc test on the resultant data provides significant and varied results when concerning the electrolyte variety. This implies that changing the quality and speed of the electrolyte reaction has a definitive effect on the resistance of composite films created out of graphene flakes produced from the reaction.

Objective: The aim of the study was to monitor the magnitude and trends in antimicrobial resistance through surveillance programs and bacteriological diagnostic methods. Methods: The study research has performed a retrospective observational study for period of 6 months (Dec 2019–June 2020) to determine the antimicrobial susceptibility pattern on cultures of various specimens at Vivekanandha Medical Care Hospital. The resultant data were computed in Microsoft Excel. Analyses were expressed as the total number, frequencies, and percentage. Results: A total of 110 culture and sensitivity reports were collected from microbiology departments. Among the culture report, 84% accounted for gram-negative and 16% for Gram-positive organisms. Out of eight specimens, 15 different bacteria were identified and the most common bacteria were Klebsiella pneumonia (25%), E. coli (14%), Staphylococcus aureus (13%), and Pseudomonas aeruginosa (12%). Conclusion: The interferences from the present study can help in bridging the gap in future educations, training, and adequate decision-making.

Objectives: This study aims to uncover variants of large effect size and allele frequency below 5% by sequencing all extant genes associated with rheumatoid arthritis (RA) in a homogeneous patient cohort. Patients and methods: This retrospective study was conducted between January 2001 and December 2017. We selected Chinese RA patients positive for anti-citrullinated peptide antibody (ACPA). All the 128 known candidate genes identified through genome-wide association studies were sequenced in 48 RA patients (15 males, 33 females; mean age 53.32±8.98 years; range, 32 to 75 years) and 45 controls (11 males, 34 females; mean age 32.18±9.54; range, 21 to 57 years). The exonic regions of these genes were sequenced. The resultant data were analyzed for association using single variant association and pathway-based association enrichment tests. The genetic burden due to low-frequency variants was assessed with the C-alpha test. The candidate variants that showed significant association were validated in a larger cohort of 500 RA cases (71 males, 429 females; mean age 48.6±12.2 years; range, 24 to 92 years) and 500 controls (66 males, 434 females; mean age 32.3±10.1 years; range, 21 to 73 years). Results: Thirty-nine variants in 21 genes were identified using single variant association analysis and C-alpha test, with stepwise filtering. Among these, the missense variant in interleukin-6 signal transducer (IL-6ST) 5:55260065 (p.Cys47Phe) was significantly associated with RA in Chinese patients in Singapore. Conclusion: Our results suggest that a mutation in IL-6ST (5:55260065) confers risk of RA in Chinese patients in Singapore.

Background & objectives: The 2019 novel coronavirus disease (COVID-19) pandemic is a big challenge for scientic and medical eld. Progression of severe disease is a difcult problem in treatment. Therefore, there is essential need to recognize severe forms of COVID-19 early in the disease course. Identication of effective biomarkers are able to classify patients based on severity. In this pilot study, we aimed to validate the association between immunologic biomarkers Interleukin-6 and ferritin with the severity of the COVID-19. Methods: A total of 1443 patients diagnosed with COVID-19 were enrolled including a severe group and a nonsevere group. Baseline clinical characteristics were collected. Serum interleukin-6 and ferritin were measured and the resultant data was statistically analyzed. Results: The most common initial symptoms were fever (68.81%) and cough (47.82%), followed by loss of appetite (9.28%), fatigue (17.46%) and breathlessness (15.52%). Level of IL-6 and Ferritin were signicantly higher in the severe patients (p<.05 for both) compared with nonsevere patients. Interpretation & conclusions: This pilot study conrmed that IL-6 and ferritin biomarkers are closely associated with the severity of COVID-19. Assessment of these biomarkers could be helpful to medical practitioners in starting treatment and close monitoring against COVID-19 infection which could improve prognosis and lower mortality

Task Analysis (TA) represents not one standard method, but rather a toolkit of methods that come with a large and complex range of outputs. Due to this fluidity in methods and resultant data, a standard method of analysis and visual data representation does not currently exist. Depending on the research methods used and desired outcomes, some visualization methods may be more appropriate than others. This body of work seeks to demonstrate the need for the establishment of a graphical grammar in this domain. Initial recommendations are provided for visualizing different task analysis methods. This research lays the groundwork towards the development of a full set of visualization best practices to allow practitioners to gain the most insight from their TA methods of choice.

Abstract This research employs machine learning (Mask Region-Based Convolutional Neural Networks [Mask R-CNN]) and cluster analysis (Density-based spatial clustering of applications with noise [DBSCAN]) to identify more than 20,000 relict charcoal hearths (RCHs) organized in large “fields” within and around State Game Lands (SGLs) in Pennsylvania. This research has two important threads that we hope will advance the archaeological study of landscapes. The first is the significant historical impact of charcoal production, a poorly understood industry of the late eighteenth to early twentieth century, on the historic and present landscape of the United States. Although this research focuses on charcoal production in Pennsylvania, it has broad application for both identifying and contextualizing historical charcoal production throughout the world and for better understanding modern charcoal production. The second thread is the use of open data, open source, and open access tools to conduct this analysis, as well as the open publication of the resultant data. Not only does this research demonstrate the significance of open access tools and data but the open publication of our code as well as our data allow others to replicate our work, to tweak our code and protocols for their own work, and reuse our results.

Background: Our previous studies showed the safe preventive and therapeutic effects of immunization using the nanoliposomal antiPCSK9 vaccine called “Liposomal Immunogenic Fused PCSK9-Tetanus plus Alum adjuvant” (L-IFPTA), in mouse models of atherosclerosis. Here we aimed to ascertain the immunogenicity and safety of the L-IFPTA vaccine in a pre-clinical study in healthy non-human primates. Methods: Five male rhesus macaque monkeys were subcutaneously immunized with the L-IFPTA vaccine, four times with bi-weekly intervals. To evaluate immunogenicity, the plasma antiPCSK9 antibody in immunized monkeys was detected and quantified using the ELISA method. The functionality of the induced antiPCSK9 antibodies was determined by the PCSK9/LDLR in vitro binding assay kit. The safety of the vaccine was tested using the evaluation of several major circulating indicators including plasma lipid alterations, inflammatory biomarkers and organ injury biomarkers. Results: The resultant data indicated that the L-IFPTA vaccine significantly and highly induced the generation of functional and safe antiPCSK9 antibodies in immunized monkeys. Plasma levels of specific biomarkers indicating organ performance including creatinine, urea, uric acid, bilirubin, ALP, AS, ALT and TSH were not significantly altered. After immunization in healthy monkeys, non-prespecified endpoints (plasma levels of TC, LDL-C, VLDL-C and TG) were non-significantly reduced by 11.6 ± 36%; 16 ± 28%; 22 ± 53% and 24 ± 51%, respectively, while HDL-C was slightly increased by 2 ± 64%. There were also no significant changes in plasma levels of pro- and anti-inflammatory biomarkers. Conclusion: The L-IFPTA vaccine could efficiently stimulate the host humoral immune response to produce active antibodies that inhibit plasma PCSK9 while not provoking systemic inflammation and not adversely affecting organ performance.

In this paper, a secure keyword search which provide the resultant data in a encrypted form where the end user can decrypt using the key given to them. It uses the Blowfish to encrypt the data and it also supports the data owner to delete or modify the content of their document. It also ensure accurate relevance score calculation between encrypted index and query vectors.
