Colistin is a decapeptide antibiotic with narrow spectrum activity, mainly used as treatment for Gram negative bacteria. This study aims to scientifically determine the effect of colistin administration as medicated feed on alanine aminotransferase (ALT) and creatinine level in broiler infected with Escherichia coli. KTOP group as positive control, KTON group as negative control, while I, II, and III groups were infected with Escherichia coli 1 x 108 CFU/ml 0.1 ml via intratracheal route. Group I, II, and III were given colistin treatment dosage of 0.3 g/kg food, 0.6 g/kg food, and 1.2 g/kg food. Blood samples were taken through brachial veins for ALT and creatinine examination with a Caretium NB-201 semi-auto chemistry analyzer. Data were examined statistically using IBM SPSS Statistics 24 software and graphically using Microsoft Excel 365. Conclusion of the research by statistical analysis with Kruskal-Wallis test obtained ALT test results P = 0.147 and creatinine test results P = 0.815. Based on the results of this study, the administration of colistin medicated feed did not cause a significant effect on ALT and creatinine level in broiler infected with Escherichia coli, indicating that colistin has low potential toxicity while given as medicated feed.

This study was conducted to investigate the antipyretic effect of the hydroalcoholic extract of Corchorus depressus Linn. against Escherichia coli ( E. coli)-induced pyrexia in rabbits. Hydroalcohalic extracts of C. depressus were given orally at 25, 50, and 100 mg/kg for antipyretic affect in E. coli-induced fever in rabbits. The animals were divided into five groups of five each. Among these five groups, three received various doses of experimental treatments, whereas the fourth one served as positive control and received paracetamol. The fifth group of animals served as negative control and received no treatment. The body temperature of the rabbits was measured rectally over a period of 5 h. C. depressus exhibited better effects at dose rate of 25, 50, and 100 mg/kg. The hydroalcoholic extract of C. depressus has significant antipyretic effect. These results lend support to the popular use of C. depressus in traditional medicine as a remedy for pyrexia and suggest that the characterization of the principles for such activity deserves further investigation.

The niacin in energy drinks has metabolic product that cause oxidative stress and liver damage, while the liver damage can be prevented by hepatoprotective agents. Scoparone in Artemisia vulgaris L. can act as a hepatoprotector by its antioxidant effect. This study aimed to investigate the effectivity of Artemisia vulgaris L. extract as a hepatoprotector in wistar hepatocytes induced by niacin. This study used 25 male rats which were divided into 5 groups: normal, the negative control, the positive control, the treatment group I, and II. Treatment was conducted for 28 days. The samples were terminated and the hepatocyte were prepared for histological examination. Histological appearance was catagorized as mild, moderate, and severe damage with or without inflamatory cells activity. The data analysis by Kruskal Wallis showed significant difference (p<0,001). Further analysis by Mann Whitney revealed significantly difference (p<0,05) between normal group and all groups, negative control group and positive control group, and positive control group and treatment group I, but not significantly difference between negative control group and treatment group I, negative control group and treatment group II, positive control group and treatment group II, and between treatment groups. The study concluded that the effectivity of Artemisia vulgaris L. extract has not been proven as a hepatoprotector but further study is needed to draw a definite conclusion. Keywords: energy drink, niacin, Artemisia vulgaris L., hepatoprotector

BACKGROUND: Mastitis is an inflammation of the breast tissue, usually caused by bacteria. Mastitis stimulates pro-inflammatory cytokines. The cytokine interleukin-1β (IL-1β) is a crucial mediator of the inflammatory response. This cytokine has adverse effects of hosting immunity that mediates resistance to pathogens and also exacerbates damage during chronic disease and acute tissue injury. Scaevola taccada (Gaertn.) Roxb. has been used as an ethnomedicine for healing sores in several provinces in Indonesia. AIM: This study aimed to assess the profile of pro-inflammatory cytokine IL-1β through the treatment effect of leaf extracts of S. taccada (Gaertn.) Roxb. as adjuvant for healing mastitis. METHODS: This study was a true control group experiment using the pre-test-post-test control design aimed to measure the effect of hydroalcoholic compounds in leaf extracts of S. taccada on the systemic pro-inflammatory activity of interleukin-1β (IL-1β). The treated animals were 18 mice of Sprague Dawley strain induced by Staphylococcus aureus. These treated mice were divided into three groups in which each group consisted of six mice. The mice in the Group I (negative control) were given 1 ml aquabides/250 g body weight, those in the Group II (positive control) were delivered with 9.6 mg/ml amoxicillin/250 g body weight, and those in the Group III (experimental) were given 9.6 mg amoxicillin/250 g body weight + 400 mg/ml leaf extracts of S. taccada/g body weight for 5 days, respectively. Pathological examinations were carried out from the inflamed tissues to prove the healing process of the treated mice. IL-1Β levels were determined using the enzyme-linked immunosorbent assay method. Data were analyzed using ANOVA and post hoc tests. RESULTS: There were statistically significant differences of IL-1β levels after the administration of leaf extracts of S. taccada among all the treated mice groups at p < 0.05. The Group III had the lowest IL-1β level with the mean value ± 1.45 pg/ml compared to the IL-1β level in the Group II (positive control) with the mean value ± 3.82 pg/ml and the IL-1β level in the Group I (negative control) with the with mean value ± 5.22 pg/ml. The pathological analysis of breast tissues of the treated mice proved that leaf extracts of S. taccada (Gaertn Roxb.) could reduce damaged tissues, cellular infiltration, and subcutaneous edema induced by this pathogenic microorganism. CONCLUSION: Leaf extracts of S. taccada had a significant function as adjuvant for healing mastitis by reducing pro-inflammatory cytokine IL-1β.

Noni juice can inhibit the growth of Escherichia coli bacteria. Noni juice extraction needs concentration to extract so that the preparation is more stable in the storage process. The purpose of this study was to determine the antibacterial activity and the amount of the minimum inhibitory concentration of noni juice extract from E. coli bacteria in vitro. This type of experimental research with a completely randomized one-way design. The extract was obtained by concentrating the Noni leaf extract. Extract quality control parameters include organoleptic, yield, drying shrinkage, and qualitative tests of flavonoid and anthraquinone compounds. Antibacterial activity test using the disc diffusion method with an extract concentration of 1.56%; 3.12%; 6.25%; 12.5%; and 25% with 3 replications. Positive control of amoxicillin and negative control of distilled water. Inhibition is known from the zone formed around the paper disc. The data obtained were statistically analyzed using Kruskall Wallis followed by Mann Whitney with a 95% confidence level. The results showed that the variation in the concentration of the noni juice extract had a significant effect (p <0.05). The biggest inhibitory zone was seen at 25% concentration of 10.16 mm and included in the strong category. The minimum inhibitory power was produced at a concentration of 3.12% at 2.50 mm with a weak treatment category.

Curcumin has remarkable anti-inflammatory and antioxidant properties. The aim of this study was to investigate the protective effects of curcumin on a rat model of carbapenem-resistant Escherichia coli–induced acute lung injury (ALI). Thirty-two rats were randomly allocated to 4 groups to induce an ALI: negative control group (rats not infected with E coli with no antibiotic treatment), positive control group (rats infected with E coli with no antibiotic treatment), imipenem group (rats infected with E coli that received intraperitoneal injection of imipenem), and the imipenem+curcumin group (rats infected with E coli that received intraperitoneal injection of imipenem and were fed on curcumin). The rats were killed, and lung tissues samples were harvested for biochemical analyses and histopathologic examination. Total antioxidant status (TAS), total oxidant status (TOS), tumor necrosis factor α (TNFα), and interleukin-6 (IL6) were measured. TOS increased in the positive control group (P < 0.001) and decreased in the imipenem and imipenem+curcumin groups (P < 0.001 and P < 0.001, respectively). TAS decreased in the positive control group (P = 0.005). Imipenem treatment did not increase TAS, but the imipenem+curcumin group increased TAS (P = 0.014). TNFα and IL6 increased in the positive control group compared with the negative control group (P < 0.001 and P = 0.010, respectively). Imipenem decreased TNFα (P < 0.001), but did not decrease IL6 (P = 0.418). Imipenem+curcumin decreased TNFα (P < 0.001); this decrease was more pronounced compared with the imipenem group (P = 0.008). IL6 decreased in the curcumin group compared with the positive control group (P = 0.011). Curcumin combined with imipenem can be an alternative therapeutic agent to overcome the resistance of E coli strains.

This article presents the results of a randomized clinical trial, designed to compare the efficacy and therapeutic profiles of YpozaneTM (osaterone acetate—OA) or SuprelorinTM (deslorelin acetate—DA) in male dogs with clinical signs of benign prostate hyperplasia (BPH). Forty-five intact male dogs were used in the study. The Group I (negative control) included 10 healthy dogs, the Group II (positive control) included 10 dogs with confirmed BPH and no treatment, whereas Group III and IV consisted of dogs with BPH and treated either with DA (15 dogs) or OA (10 dogs). The clinical response, testosterone and estradiol levels, hematology, biochemistry, and adverse effects incidence were evaluated. Both OA and DA proved to be effective for BPH treatment in dogs, as they allowed for the clinical remission in all treated dogs. The complete alleviation of BPH symptoms was noticed sooner with the use of OA (in 80% of dogs from day 7) compared to DA (in 40% of dogs within the first 21 days). The recurrence of clinical signs related to BPH was observed from week 24 in dogs treated with OA, whereas no relapse was noticed in dogs treated with DA at the end of the 36 weeks of the observation period. In 5 dogs (33%) treated with DA, a flare-up effect (increase in the clinical signs associated with BPH) was noticed on day 7. Despite individual differences in the clinical action, both medications were effective and safe options for the treatment of symptoms related to BPH in dogs.

A study was conducted to determine the response of Turkey poults to graded levels of Alphamune G (0.00+, 0.03, 0.04, 0.05, 0.06 and 0.00 %-) when challenged with Escherichia coli orally for 7 days. The graded levels were the treatments viz 0.00%+ (positive control), Alphamune G at 0.03, 0.04, 0.05, 0.06% and 0.00%- (negative control; infected without Alphamune G supplementation). Each treatment was allotted 3 replicates of 6 poults. The experiment which was conducted for 56 days employed a completely randomized design. E. coli was isolated from the intestinal digesta of a colisepticaemic chicken. 108 turkey poults were used in this study. Poults were infected with E.coli for 7 days through the drinking water and given the treatment. The performance parameters of Alphamune G supplementation were significantly affected. The cumulative weight, Feed intake and weight gain were highest for turkey poults fed 0.06% Alphamune G supplementation. These values were also directly proportional to the supplementation levels of Alphamune G. The birds given the negative treatment (0.00 %-) had relatively poor performance compared to the other treatments. The specific enzymes studied were significantly affected (p<0.05) by the treatments. ALT and AST were significantly highest for turkey poults fed the negative control. Enzyme values became optimum at 0.05% Alphamune G supplementation. At 0.06% of Alphamune G supplementation, cellular mitigations of the effects of E. coli was measurable. Urea and creatinine were not significantly (p>0.05) influenced by the treatments. Haematological indices such as WBC and specific differential counts (lymphocytes and neutrophils) were affected significantly (P<0.05) by supplemental levels of Alphamune G The Inclusion of Alphamune G at 0.06% in the diets improved performance of turkey poults when challenged with Escherichia coli.
