Abstract Study question Is GSTM1 polymorphism a putative biomarker of male infertility. Is there a possible correlation between GSTM1 presence, oxidative stress and male infertility? Summary answer A possible correlation may be established between GSTM1 polymorphism, and sperm mobility and morphology. Additionally oxidative potential may also be associated with fertility. What is known already Approximately 7% of men worldwide are affected by male infertility, which contributes to 40–50% of all infertility cases. Basic sperm analysis remains the main procedure of diagnosing male infertility, although there is still doubt whether it provides accurate clinical outcomes More accurate tests are essential for the diagnosis of male infertility. Oxidative stress is involved in the etiology of male infertility, with 30% to 80% of infertile men having increased levels of sperm free radicals. Recent research has shown that oxidative stress when combined with GSTM1-null genotype negatively affected the sperm quality of infertility group compared to the control group. Study design, size, duration Ninety semen samples were collected and divided into 2 groups. The study group consisted of sperm samples from 51 men identified as infertile according to WHO guidelines(case group).Sperm samples from 39 men with normal sperm count parameters (control group) were used for the control group. Participants/materials, setting, methods For all samples a sperm diagram was performed. and DNA was extracted. Polymerase chain reaction with specific for GSTM1 gene primers followed by agarose electrophoresis was applied to detect the presence of polymorphism. The MiOXSYS method was used to detect the oxidative potential. Main results and the role of chance This study shows that in the control group the presence of polymorphism was associated with a reduced number of immobile sperm cells (p = 0,035) while it appears to affect the normal morphology of the sperm(p = 0,042). In the infertility group the presence of the gene was significantly correlated with age(p = 0.046). No statistically significant difference was observed for the presence of the polymorphism between the 2 groups.In addition, we investigated the effect of oxidative potential with the MiOXSYS system and its relationship with sperm parameters. It was found that the two groups differed significantly when measuring oxidative potential, and that oxidoreduction potential affects sperm concentration/ml, total sperm count, type B motility and viscosity in the infertile male group. Limitations, reasons for caution A larger sample size could increase the accuracy of the results. Wider implications of the findings: Studying the relation of the oxidative stress with sperm parameters may lead to the establishment of a genetic profile of increased risk of infertility, which would be of major importance especially in cases of idiopathic infertility. Background: Some plants stimulate spermatogenesis and increase fertility, while some cause spermatogenesis arrest. So far, the effects of bene (Pistacia atlantica) on male fertility have not been studied. The aim of this study was to investigate the effects of bene on sperm parameters, testicular histopathology, sperm quality, and oxidative stress in busulfan-induced infertile mice. Methods: Thirty-five male BALB/c mice were randomly assigned to control, sham, busulfan, bene, and bene + busulfan groups. The busulfan group received 10 mg/kg as a single dose and intraperitoneally. The bene group received pellets containing 10% of bene. Another group received 10 mg/kg busulfan and was fed with pellet containing 10% bene. Then, sperms, sperm chromatin quality, testicular histopathology, and oxidative stress levels were studied on the 35th day of the experiment. Results: Busulfan injection resulted in a significant reduction in sperm parameters compared to the control group (p<0.001); it decreased after bene administration (p<0.001). In addition, in the group treated with bene, the sperm count with damaged DNA was reduced and the level of malondialdehyde decreased compared to the busulfan group. A significant increase was observed in the mean level of superoxide dismutase and catalase enzymes in the bene + busulfan group compared to the busulfan group (p<0.001). The histopathological improvement of the testis was observed in the bene + busulfan group. Conclusion: The administration of 10 mg/kg of bene powder for 35 days reduced the oxidative stress, improved testicular histopathology, sperm chromatin quality, and sperm parameters in the infertile mice model.

Male infertility may be the result of some identifiable conditions (varicocele, cryptorchidism, hypogonadism, and genetic abnormalities), but there is no identified cause in 30-40% of infertile men. Studies show that the use of antioxidants has a beneficial effect on fertility, especially on sperm quality, and therefore recommended as a potentially effective treatment for male infertility. However, it is argued that this treatment is not based on sufficient evidence and has no effect on the rate of healthy pregnancy. Aim: In this study, two different antioxidant combinations with different doses and contents were evaluated in terms of their effect on sperm parameters. Materials and Methods: A total of 122 patients diagnosed with idiopathic infertility were enrolled in our multicenter study. The patients were divided into two different groups: The first group used Proxeed Plus sachets 2x1 and the second group used Carniact Duo tablets 2x1 for six months. The total semen volume, total sperm count and concentration, motile sperm count, and morphological findings of the patients were compared at the end of the six months. The mean age of the patients participating in the study was 30.8 ± 6.05 years. No significant difference was found between the two groups in terms of baseline sperm count. Results: There was a significant difference between the baseline and sixth-month values of the patients using both combinations. However, no significant statistical difference was found between the groups according to the sixth-month data. Conclusion: The combinations of both antioxidants had a positive effect on sperm parameters, and the use of different doses and contents had a similar effect. Background: The aging process is accompanied by low secretion of sex hormones and testicular apoptosis. The antioxidant properties of thymoquinone (TQ) may prevent the effects of aging. Therefore, in the present study, the effects of different doses of TQ were investigated on sperm parameters, testosterone level, apoptosis, and oxidative stress in a mouse model of D-galactose-induced aging. Methods: In this experimental study, 30 adult male mice were randomly divided into 5 groups. The control group did not receive any injections, while the D-galactose group received an intraperitoneal injection of 300 mg/kg of D-galactose for 42 days. The TQ1-TQ3 groups received intraperitoneal injections of 5, 2.5, and 1.25 mg/kg of TQ plus D-galactose, respectively for 14 days (from the 1st to the 14th day of the experiment). The morphometric analysis, testicular apoptosis examination, and sperm analysis were performed, and testosterone level, total antioxidant capacity, and malondialdehyde level were evaluated on day 42 of the experiment. Data were analyzed using SPSS. Results: Administration of TQ in the TQ1 group caused a significant difference in sperm parameters, compared to the D-galactose group (P<0.05). The lowest amount of positive tunnel cells was related to 5 mg/kg of TQ and the highest to 2.5 mg/kg of TQ. There was no significant difference in the parameters of seminal vesicles, epididymis, prostate, and testis between the groups (P>0.05). The malondialdehyde level were decreased in the TQ1-TQ3 groups, compared to the D-galactose group (P<0.001). On the other hand, the total antioxidant capacity was increased significantly in the TQ1 group, compared to the D-galactose group (P<0.001). Conclusion: Administration of 5 mg of TQ for 14 days improved sperm quality and biochemical parameters, while reducing apoptotic cells of the testes in a mouse model of aging.

 Síndrome metabólica (SM) é o nome dado ao conjunto de doenças de origem metabólica, que diretamente aumenta o risco de desenvolvimento de doenças cardiovasculares e ou diabetes tipo 2. A definição mais aceita para SM inclui o sujeito com a presença de três ou mais dos seguintes critérios: 1) obesidade abdominal (circunferência abdominal ≥ 102 com para homens e ≥ 88 cm para mulheres); 2) triglicérides ≥ 150 mg/dL; 3) HDL-colesterol baixo (< 40 mg/dL para homens e < 50 para mulheres; 4) pressão arterial elevada (> 130/85 mmHg e 5) glicemia de jejum elevada (>110 mg/dL). Dentre os numerosos efeitos deletérios da SM, a infertilidade masculina pode representar uma condição patológica significativa, decorrente de alterações metabólicas ocasionadas por esta síndrome. As evidências apontam principalmente para alterações hormonais que resultam em hipogonadismo, que pode prejudicar o desenvolvimento e maturação do espermatozoide. Além disso, os vários componentes da SM podem ocasionar a instalação de um estado de estresse oxidativo, que é prejudicial ao espermatozoide. A investigação inicial da infertilidade masculina consiste de um exame clínico, avaliação hormonal (e.g., dosagem de testosterona, hormônio luteinizante e folículo estimulante) e análise do sêmen (espermograma). O espermograma é o principal e mais importante teste a ser solicitado quando a investigação da infertilidade masculina está sendo realizada. Entretanto, novas estratégias diagnósticas, tais como, quantificação dos parâmetros de estresse oxidativo e capacidade antioxidante do sêmen podem ser ferramentas úteis no diagnóstico da infertilidade masculina. Palavras chave: obesidade, espermograma, diabetes, radicais livres, estresse oxidativo.   Metabolic syndrome (MS) includes a group of metabolic diseases who are directly linked to increased risk of to develop cardiovascular diseases or type 2 diabetes. The most accepted definition for MS includes patients with three or more of the following criteria: 1) abdominal obesity (waist circumference ≥ 102 in men and ≥88 cm for women), 2) triglycerides ≥ 150 mg/dL; 3) low HDL cholesterol (<40 mg/dL for men and <50 for women, 4) high blood pressure (>130/85 mmHg and 5) high fasting glucose (>110 mg/dL). Among the deleterious effects of MS, male infertility may represent a pathological condition due to significant metabolic alterations caused by this syndrome. The evidences point out to hormonal changes that result in hypogonadism that can promote detrimental changes in development and maturation of sperm. Moreover, the various components of MS cause an oxidative stress status that is deleterious to sperm morphology and motility. The investigation of male infertility consists of the clinical examination, hormonal evaluation (quantification of plasma testosterone, luteinizing and follicle-stimulating hormone) and semen analysis (sperm count). Semen analysis is the main and most important test to be requested when the investigation of male infertility is conducted. However, new diagnostic strategies such as the quantification of oxidative stress parameters and semen antioxidant capacity can be useful tools in the male infertility diagnosis.

Background: The role of minerals in preventing the generation of oxidative stress in obese individuals has been evaluated. Magnesium is an antioxidant nutrient and a cofactor of enzymes involved in the cell membrane stabilization, attenuating the effects of oxidative stress. Objective: To evaluate the association between magnesium and concentrations of thiobarbituric acid reactive substances (TBARS) in patients with obesity and eutrophic women. Methods: A cross-sectional study was conducted with 73 women, divided into two groups: case group (patients with obesity, n=27) and control group (eutrophic women, n=46). Measurements of body mass index and waist circumference were performed. Dietary magnesium intake was assessed by the three-day food record using the NutWin software. Urinary magnesium concentration was measured by atomic absorption spectrophotometry method. Plasma concentrations of thiobarbituric acid reactive substances (TBARS) were also determined. Results: Mean values of dietary magnesium intake were 161.59 ± 60.04 and 158.73 ± 31.96 for patients with obesity and control group, respectively, with no significant difference between the groups studied (p >0.05). The value of urinary excretion of magnesium was lower than the reference values in both groups, with no significant difference between the groups studied (p >0.05). The plasma concentration of thiobarbituric acid reactive substances was significantly higher in patients with obesity compared to the control group (p <0.001). There was no correlation between levels of magnesium biomarkers and the concentration of TBARS (p >0.05). Conclusion: Patients with obesity showed a reduced dietary magnesium intake which seems to induce hypomagnesuria as a compensatory mechanism. The marker of oxidative stress evaluated in this study was not influenced by magnesium.

Background: There is a growing interest in the correlation between antioxidants and periodontal disease. In this study, we aimed to investigate the effect of oxidative stress and the impact of two antioxidants, curcumin and rutin, respectively, in the etiopathology of experimentally induced periodontitis in diabetic rats. Methods: Fifty Wistar albino rats were randomly divided into five groups and were induced with diabetes mellitus and periodontitis: (1) (CONTROL)—control group, (2) (DPP)—experimentally induced diabetes mellitus and periodontitis, (3) (DPC)—experimentally induced diabetes mellitus and periodontitis treated with curcumin (C), (4) (DPR)—experimentally induced diabetes mellitus and periodontitis treated with rutin (R) and (5) (DPCR)—experimentally induced diabetes mellitus and periodontitis treated with C and R. We evaluated malondialdehyde (MDA) as a biomarker of oxidative stress and reduced glutathione (GSH), oxidized glutathione (GSSG), GSH/GSSG and catalase (CAT) as biomarkers of the antioxidant capacity in blood harvested from the animals we tested. The MDA levels and CAT activities were also evaluated in the gingival tissue. Results: The control group effect was statistically significantly different from any other groups, regardless of whether or not the treatment was applied. There was also a significant difference between the untreated group and the three treatment groups for variables MDA, GSH, GSSG, GSH/GSSG and CAT. There was no significant difference in the mean effect for the MDA, GSH, GSSG, GSH/GSSG and CAT variables in the treated groups of rats with curcumin, rutin and the combination of curcumin and rutin. Conclusions: The oral administration of curcumin and rutin, single or combined, could reduce the oxidative stress and enhance the antioxidant status in hyperglycemic periodontitis rats.

Background: The aim of this study was to compare consequences in single and triple testicular biopsy by biopty gun in pubertal rats using histological and immunohistochemical analysis. Methods: Thirty-two Sprague-Dawley male rats were used as the experimental model. The rats were randomly divided into three study groups. The rats from the first group (n = 12) received a single-biopsy of upper pole of the left testis, while the rats from the second group (n = 10) received triple-biopsy of upper and lower poles and lateral surface of left testis. The third group (n = 10) was a control group. On the eightieth day after the biopsy in all rats bilateral orchiectomy and funiculectomy were performed to obtain testicular tissue and sperm for analysis. The consequences of the puncture were observed by pathohistology, immunohistochemistry and semen analysis. Results: The results of the study showed lower percentage of sperm count (14.5 mill/mL vs. 16 mill/mL, p = 0.130), sperm motility (24.6% vs. 32.7%, p > 0.05), abnormal sperm (30% vs. 27%, p > 0.05), atrophic tubules (21% vs. 6%, p < 0.001), volume (1.7 mL vs. 2.28 mL, p < 0.01) and apoptotic index (1.56 vs. 1.19, p = 0.650) in the testes with a triple-biopsy compared to the testes with a single-biopsy. Semen analysis showed a borderline significant difference between the group with triple-biopsy where sperm count was lower than it in the control group (14.5 mill/mL vs. 17.5 mill/mL, p = 0.05). A single-biopsy has little effect on the testis, especially on overall fertility. A triple-biopsy showed higher degree of the testicular damage but without a significant impact on overall fertility. Semen analysis showed that single- and triple-biopsies did not have a significant effect on sperm count, motility and morphology. Conclusion: Biopty gun procedure is a cheap, simple and reliable method for testicular biopsy in rats without a significant effect on sperm count, motility and morphology.

Introduction Lichen planus (LP) which is a chronic inflammatory disease can cause impaired atrial electromechanical coupling, leading to increased risk of atrial fibrillation. Purpose The present study aimed to evaluate atrial electromechanical coupling in LP patients by using electrocardiography (ECG) and echocardiography. Methods Forty-six LP patients were investigated in this cross-sectional case-control study. The control group comprised healthy individuals selected in age and gender-matched manner. Echocardiography and ECG were done for all patients to show inter and intra-atrial electromechanical delays and P wave dispersion respectively. The electromechanical delays were calculated by using the difference between the delays from the onset of the P wave on ECG to the onset of A wave on tissue Doppler recordings of the different areas. Results The baseline characteristics of the case and control group were similar and did not differ significantly. The P wave dispersion was 45.63 ± 3.48 milliseconds in the LP group in comparison to 36.56 ± 2.87 milliseconds in the control group (p &lt; 0.001). As shown in the table, the intra and inter-atrial electromechanical delays were also significantly prolonged in LP patients when compared to the control group (p &lt; 0.001). There was no significant difference between the left and right ventricular systolic function and diastolic function of the two groups. Conclusion The results of the study indicate the presence of significant impaired atrial electromechanical coupling in patients with LP confirmed by both electrocardiographic and echocardiographic tools. Abstract Sperm structural and functional defects are leading causes of male infertility. Patients with immotile sperm disorders suffer from axoneme failure and show a significant reduction in sperm count. The kinesin family member 3B (KIF3B) is one of the genes involved in the proper formation of sperm with a critical role in intraflagellar and intramanchette transport. A part of exon 2 and exons 3–5 of the KIF3B encodes a protein coiled-coil domain that interacts with IFT20 from the IFT protein complex. In the present study, the coding region of KIF3B coiled-coil domain was assessed in 88 oligoasthenoteratozoospermic patients, and the protein expression was evaluated in the mature spermatozoa of the case and control groups using immunocytochemistry and western blotting. According to the results, there was no genetic variation in the exons 3–5 of the KIF3B, but a new A &gt; T variant was identified within the exon 2 in 30 patients, where nothing was detected in the control group. In contrast to healthy individuals, significantly reduced protein expression was observable in oligoasthenoteratozoospermic (OAT) patients carrying variation where protein organization was disarranged, especially in the principal piece and midpiece of the sperm tail. Besides, the protein expression level was lower in the patients’ samples compared to that of the control group. According to the results of the present study the NM_004798.3:c.1032A &gt; T, p.Pro344 = variant; which has been recently submitted to the Clinvar database; although synonymous, causes alterations in the transcription factor binding site, exon skipping, and also exonic splicing enhancer-binding site. Therefore, KIF3B can play an important role in spermatogenesis and the related protein reduction can cause male infertility.

Objective This study aimed to evaluate the impact of the dietary supplement of Moringa oleifera leaves (MOL) on semen quality and characteristics in rabbits. Methods Eighteen (n=18) breeding bucks of New Zealand white, of similar age group, were used for the study. Three feeding regimes, (i) 100% commercial rabbit pellets (CRP)-Group I (ii) 90% CRP + 10% fresh MOL on a dry matter (DM) basis – Group II and (iii) 80% CRP + 20% fresh MOL on a DM basis – Group III, were adopted and the trial continued for 21 days. After adaptation to the diet, semen was collected from each buck and subjected to evaluation using a computer-assisted semen analyser. Results In Group III, the sperm count, normal sperm morphology, and sperm motility increased (52.0%) in comparison with the control (Group I; 50.1%). The inclusion of 20% Moringa oliefera in the diet (Group III) caused a significant increase (P<0.05) in semen concentration (Control =136.2 M/mL; Group III=297.2 M/mL). There was no significant difference (P>0.05) in sperm motility and semen volume among the groups. Conclusion The results suggest that supplementing commercial rabbit pellets with 20% fresh Moringa oliefera leaves on a DM basis can improve the quality and characteristics of semen in breeding bucks.
