The administration of diets contaminated with low to intermediate doses of deoxynivalenol and supplemented with antioxidants and binding agents slightly affects the growth, antioxidant status and vaccine response in weanling pigs

Abstract This study aimed to evaluate the impact of grading levels of deoxynivalenol (DON) in the diet of weaned pigs, as well as the effects of a supplementation with antioxidants (AOX), hydrated sodium calcium aluminosilicates (HSCAS) and their combination on the growth, antioxidant status, immune and vaccine response against the porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus 2 (PCV2). At weaning, 336 piglets were allocated to six dietary treatments according to a randomized complete block design. Treatments were as follows: basal diet (CTRL); basal diet containing DON at 1.2 mg/kg (DON1.2); basal diet containing DON at 2.4 mg/kg (DON2.4); DON2.4 diet + a mix of AOX which included vitamins A and E at 20,000 IU and 200 IU/kg feed respectively, selenized yeast at 0.3 mg/kg and a grape seed extracts at 100 mg/kg feed (DON2.4+AOX); DON2.4 diet + modified HSCAS at 1 g/kg (DON2.4+HSCAS); DON2.4+AOX+HSCAS. Pigs were vaccinated against PRRSV and PCV2 at 7 d; at 0, 14 and 35 d growth performance were recorded, and blood samples were collected in order to evaluate the oxidative status, inflammatory blood markers, lymphocyte blastogenic response and vaccine antibody response. Increasing intake of DON resulted in a quadratic effect at 35 d in the lymphocyte proliferative response to Concanavalin A and PCV2 as well as in the anti-PRRSV antibody response, whereas the catalase activity decreased in DON2.4 pigs compared to the CTRL and DON1.2 groups (P ≤ 0.05). Compared to the DON2.4 diet, the AOX supplementation slightly reduced G:F ratio (P = 0.026) and increased the ferric reducing ability of plasma as well as α-tocopherol concentration (P < 0.05), whereas the association AOX+HSCAS increased the anti-PRRSV IgG (P < 0.05). Furthermore, the HSCAS supplement reduced haptoglobin levels in serum at 14 d compared to the DON2.4 group; however, its concentration decreased in all the experimental treatments from 14 d to 35 d and particularly in the DON2.4+AOX pigs, whereas a different trend was evidenced in the DON2.4+HSCAS group, where over the same period haptoglobin concentration increased (P < 0.05). Overall, our results show that the addition of AOX and HSCAS in the diet may alleviate the negative effects due to DON contamination on the antioxidant status and immune response of vaccinated weanling pigs.

Abstract An experiment was conducted to test the hypothesis that Val from a Val-containing fermentation biomass product (Val-FB; 64.4% L-Val) has a bioavailability of 100% relative to L-Val (98% Val) when fed to weanling pigs. A Val-deficient basal diet containing 0.63% standardized ileal digestible (SID) Val was formulated. Six additional diets were prepared by supplementing the basal diet with 0.08, 0.16, or 0.24% L-Val or 0.12, 0.25, or 0.37% Val-FB to create experimental diets containing 0.71, 0.79, or 0.87% SID Val. A total of 224 weanling pigs (6.87 ± 0.64 kg) were allotted to a randomized complete block design with 7 diets and 8 replicate pens per diet. Diets were fed for 21 d and growth performance was measured on a pen basis. Orthogonal-polynomial contrasts were used to determine linear and quadratic effects of L-Val and Val-FB levels on performance and the effect of Val sources. A linear regression model based on performance was used to estimate the relative bioavailability (RBV) of Val in Val-FB relative to L-Val. The final body-weight (BW) and average-daily-gain (ADG) were greater (P < 0.01) for pigs fed diets supplemented with Val-FB than pigs fed diets supplemented with L-Val (Table 1). The average-daily-feed-intake (ADFI) decreased linearly (P < 0.01), whereas gain-to-feed ratio (G:F) increased (P < 0.01) by supplementing graded levels of both Val sources to the diets. The RBV of Val in Val-FB as determined by ADG, G:F, and final BW was 146, 135, and 143%, respectively, with 95% confidence intervals of 99 to 191%, 83 to 187%, and 70 to 217%, respectively. In conclusion, the linear regression estimated a RBV of at least 100% for Val-FB relative to L-Val, and pigs fed diets supplemented with Val-FB had greater ADG and final BW than pigs fed diets supplemented with the same amount of L-Val.

AbstractBackgroundThe mRNA vaccines for SARS-CoV2 have proven highly effective and are currently used to vaccinate all age groups against COVID-19. Despite their high efficacy in clinical trials, there is limited data on the impact of age, sex, and side effects on vaccine-induced immune responses.MethodsWe here studied the development of SARS-CoV-2 Spike protein RBD domain antibodies after two doses of the Pfizer-BioNTech Comirnaty mRNA vaccine in 118 healthy volunteers and correlated their immune response with age, sex, and side effects reported after the vaccinations.FindingsOur findings show a robust immune response to the Spike protein’s RBD region after the first and the second vaccination dose. However, we also saw a decline of antibody levels at 6 weeks versus 1 week after the second dose, suggesting a waning of the immune response over time. Regardless of this, the antibody levels at 6 weeks after the second dose remained significantly higher than before the vaccination, after the first dose, or in COVID-19 convalescent individuals. We found a decreased vaccination efficacy but fewer adverse events in older individuals, and that mRNA vaccination is less efficient in older males whereas the detrimental impact of age on vaccination outcome is abolished in females at 6 weeks after the second dose.InterpretationThe Pfizer-BioNTech Comirnaty mRNA vaccine induces a strong immune response after two doses of vaccination but older individuals develop fewer side effects and decreased antibody levels at 6 weeks. The waning of anti-viral antibodies in particular in older male individuals suggests that both age and male sex act as risk factors in the immune response to the SARS-CoV-2 mRNA vaccine.FundingThe study was supported by the Centre of Excellence in Translational Genomics (EXCEGEN), and the Estonian Research Council grant PRG377 and SYNLAB Estonia.Research in contextEvidence before this studyThe first studies addressing the immune responses in older individuals after the single-dose administration of the SARS-CoV-2 mRNA vaccines have been published. We searched PubMed and medRxiv for publications on the immune response of SARS-CoV-2-mRNA vaccines, published in English, using the search terms “SARS-CoV-2”, “COVID-19”, “vaccine response”, “mRNA vaccine”, up to April 15th, 2021. To date, most mRNA vaccine response studies have not been peer-reviewed, and data on the role of age, sex and side effects on SARS-CoV-2-mRNA vaccines in real vaccination situations is limited. Some studies have found a weaker immune response in older individuals after the first dose and these have been measured at a relatively short period (within 1-2 weeks) after the first dose but little longer-term evidence exists on the postvaccination antibody persistence. Even less information is available on sex differences or correlations with mRNA vaccine side effects.Added value of this studyIn this study, we assessed the antibody response up to 6 weeks after the second dose of Pfizer-BioNTech Comirnaty mRNA vaccine in 118 individuals. Our findings show a strong initial immune response after the first dose and an even higher Spike RBD antibody levels at 1 week after the second dose, but these significantly declined at 6 weeks after the second dose. We also found a weaker immune response and faster waning of antibodies in older vaccinated individuals, which correlated with fewer side effects at the time of vaccinations. Furthermore, although overall female and male vaccinees responded similarly, we found that age-related waning of the vaccine-related antibodies was stronger amongst older males whereas in females the impact of age was lost at 6 weeks after the second dose.Implications of all the available evidenceNew mRNA vaccines are now applied worldwide as they have shown high efficacy in clinical trials. Our results show that two doses of Pfizer-BioNTech Comirnaty mRNA vaccine induce a strong antibody response to Spike RBD region but these high levels decline 1.5 months after the second dose in most of the vaccinated individuals. Nevertheless, even at 6 weeks after the second dose, they stay significantly higher than at prevaccination, after the first dose of vaccine, or in Covid-19 postinfection. These findings also implicate that fewer adverse effects may indicate lower antibody response after the vaccination and point to the need for more individualized vaccination protocols, in particular among older people.

An experiment was conducted to evaluate the addition of sodium butyrate, plant extracts and nucleotides on weanling pig performance, digestive content pH, organ morphometry, and intestinal epithelial histology. A total of 90 piglets at 21 days of age and an average initial weight of 6.35±0.34kg were used. The piglets were distributed in a randomized complete block design with five treatments, six replicates, and three animals per experimental unit. The treatments consisted of the following: Control: basal diet without antibiotic; Antibiotic: basal diet with 40mg kg-1 colistin sulfate and Additive: 1000, 1500 and 2000mg kg-1 of a combination of sodium butyrate + plant extracts + nucleotides. The experiment lasted 35 days, at which time one animal was slaughtered to assess pH of the digestive contents, morphometry of the organs and histology of the intestinal epithelium. No differences were found (P>0.05) in the performance, pH of the digestive contents, morphometry of the organs or histology of the intestinal epithelium by the analysis of orthogonal polynomials or contrasts. The combination of sodium butyrate, plant extracts and nucleotides not improved the productive characteristics of weanling pigs.

There is limited information on the impact of dietary supplementation with separate rumen-protected (RP) amino acids (AA), or with their combination, on ewes’ oxidative status. Sixty ewes were divided into five groups; C: basal diet (control); M: basal diet + 6 g/ewe RP methionine; L: basal diet + 5 g/ewe RP lysine; LML: basal diet + 6 g methionine and 5 g lysine/ewe; and HML: basal diet + 12 g methionine + 5 g lysine/ewe. Milk’s fat content increased in RP-AA fed ewes, while that of protein in M and L only. In blood plasma, the malondialdehyde (MDA) content was reduced in the M, LML, and HML compared to C-fed ewes. An increase in glutathione transferase activity in the blood plasma of the M and LML compared to the C and HML-fed ewes were found. In milk, lower values of the ferric reducing ability of plasma (FRAP) in the LML and HML-fed ewes and of 2,2′-Azino-bis 3-ethylbenzthiazoline-6-sulfonic acid (ABTS) in LML only, were found. Lysine increased milk’s FRAP values and MDA content. Both L and HML diets increased milk’s protein carbonyls content. Methionine improves the organism’s oxidative status, without adversely affecting milk’s oxidative stability. Lysine dietary inclusion affects negatively the oxidative stability of milk.

Abstract An experiment was conducted to test the hypothesis that Val from a spray dried L-Val fermentation biomass (Val-FB; 64.4% L-Val) has a bioavailability of 100% relative to Val from L-Val (98% L-Val) when fed to weanling pigs. A Val-deficient basal diet containing 0.63% standardized ileal digestible (SID) Val was formulated. Six additional diets were prepared by supplementing the basal diet with 0.08%, 0.17%, or 0.25% L-Val or 0.12%, 0.25%, or 0.37% Val-FB to create experimental diets from both Val sources that contained 0.71%, 0.79%, or 0.87% SID Val. Two hundred twenty-four weaned pigs (6.87 ± 0.64 kg initial BW) were allotted to a randomized complete block design with 7 diets, 4 pigs per pen, and 8 replicate pens per diet. Diets were fed for 20 d. At the conclusion of the experiment, a blood sample from 1 pig per pen was analyzed for blood urea nitrogen (BUN) and plasma free AA. A linear regression model was used to estimate the relative bioavailability (RBV) of Val in Val-FB relative to Val from L-Val. Results indicated that the final BW and ADG were greater (P < 0.01) for pigs fed diets supplemented with Val-FB than pigs fed diets supplemented with L-Val. The ADFI decreased (linear, P ≤ 0.01), whereas G:F increased (linear, P < 0.01) by increasing inclusion of both Val sources in the diets. Regardless of source of dietary Val, BUN values were reduced (linear and quadratic, P < 0.01) as the concentration of Val in the diet increased. Pigs fed diets supplemented with L-Val had increased (linear and quadratic, P ≤ 0.05) concentrations of Val and Arg in plasma, and plasma concentrations of Ile, Leu, Lys, Ala, Cys, and Pro linearly increased (P < 0.05). There was also an increase (linear, P < 0.05) in plasma concentrations of Ile, Leu, Met, Ala, Asp, Cys, and Pro as Val-FB was added to the diets, and the concentration of Val in plasma increased (linear and quadratic, P < 0.05). Using L-Val as the standard, the RBV of Val in Val-FB as determined by ADG, G:F, and final BW was 146%, 135%, and 143%, respectively, with 95% confidence intervals of 99% to 191%, 83% to 187%, and 70% to 217%, respectively. In conclusion, the linear regression estimated a RBV of at least 100% for Val in Val-FB relative to Val from L-Val, and pigs fed diet supplemented with Val-FB had greater final BW, ADG, and G:F than pigs fed diets supplemented with the same amount of Val from L-Val.

Abstract. The aim of this study is to evaluate the effect of α-tocopherol supplementation at two doses (600 and 1200 mg × kg–1) on kidney antioxidant status and the histopathological changes in Wistar rats after 12 weeks of exposure at different diets. Forty rats has been divided into 4 groups of 10 rats each, the control group received basal diet with 5 % fresh sunflower oil (FSO), the second group: 5 % oxidized sunflower oil (OSO), the third group: 5 % OSO supplemented with 600 mg × kg–1 α-tocopherol and the fourth group: 5 % OSO supplemented with 1200 mg × kg–1 α-tocopherol. In OSO groups, the results showed highly significant increases of LPO (from 31.3 ± 0.9 to 53.8 ± 1.2 nmol of MDA formed/min/mg protein, p < 0.0001) with a significant decrease (p < = 0.001) of the antioxidant enzymatic activities (CAT, SOD, GPX, GR and G6PDH), body weight (339 ± 9 to 290 ± 3 g) and α-tocopherol levels (13.6 ± 0.6 to 6.5 ± 0.4 μg/mg protein). In OSO groups with 600 mg × kg–1 α-tocopherol, an antioxidant effect was found, reflected by a return of the parameters to values similar to those of the control group. However, higher doses of α-tocopherol (1200 mg × kg–1) induced a depletion of antioxidant status, α-tocopherol levels (6.0 ± 0.3 μg/mg protein, p < 0.001) and a very highly significant rise (p < 0.0001) of LPO content (54.86 ± 0.01 nmol of MDA formed/min/mg protein). The kidney tissues also showed changes in glomerular, severe inflammatory cells infiltration, and formation of novel vessels. So, we can conclude that the oxidative stress is attenuated by a moderate administration of 600 mg × kg–1 α-tocopherol, while a pro-oxidant effect occurs at 1200 mg × kg–1 α-tocopherol.

The experiment was conducted in a greenhouse during 2017 and 2018 growing seasons to evaluate the impact of the shading and various nutrition programs on mitigating heat stress, reducing the use of chemical minerals, improving the reproductive growth and yield of tomato plant. Split-plot within Randomized Complete Block Design (RCBD) with three replications was conducted in this study. Shading factor was allocated in the main plots and the nutrition programs distributed randomly in the subplots. Results indicate that shading resulted in the decrease of daytime temperature by 5.7˚C as an average for both seasons; thus a significant increasing was found in leaf contents of macro nutrients (Nitrogen, Phosphorous, and Potassium), and micro nutrients (Iron, Zinc and Boron), except the Iron content in 2018 growing season. Furthermore, shading improved significantly the reproductive growth and tomato yield. Among the plant nutrition programs, the integrated nutrient management (INM) including the application of organic substances, bio inoculum of AMF and 50% of the recommended dose of chemical fertilizers; lead to the enhancement of nutrients content, reproductive characteristics and plant yield. Generally, combination of both shading and INM showed positive effects on plants nutrient status and persisting balance on tomato flowering growth and fruits yield.

An experiment was conducted at Horticultural Research Centre, SVP University of Agriculture and Technology, Meerut (UP) during Rabi season of 2018-19 to assess the impact of different INM doses on yield and quality parameters of garlic. A total of ten treatments consisting of combinations of inorganic fertilizers, organic fertilizers and bio-fertilizers like T1- (Control), T2RDF (100:50:50 kg NPK ha-1), T3-RDF + 20 kg sulphur + FYM 20 ton ha-1, T4- RDF + 20 kg sulphur + VC 4 ton ha-1, T5- 75% RDF + 40 kg sulphur + 5 ton FYM ha-1+ PSB 5 kg ha-1, T6-75% RDF + 40 kg sulphur + 2 ton VC + Azotobacter 5 kg ha-1, T7- 75% RDF + 40 kg sulphur + FYM 3 ton + VC 1 ton+ PSB 5 kg + Azotobacter 5kg ha-1, T8- 50% RDF + 40 kg sulphur + FYM 5 ton + VC2 ton + PSB 5 kg ha-1, T9- 50% RDF + 40 kg sulphur + FYM5 ton + VC 2 ton+ Azotobacter 5 kg ha-1and T10- 50% RDF + 40 kg sulphur + FYM 5 ton + VC2 ton + PSB 5 kg + Azotobacter 5 kg ha-1 were used in Randomized Block Design and replicated thrice. Out of these an application of T7 (75% RDF + 40 kg sulphur + FYM3 ton + VC 1 ton ha-1 + PSB 5 kg + Azotobacter 5 kg ha-1) was found to be significantly superior in term of yield and attributing parameters of garlic.

Recent evidences suggest that gut microbiota plays an important role in regulating physiological and metabolic activities of pregnant sows, and β-carotene has a potentially positive effect on reproduction, but the impact of β-carotene on gut microbiota in pregnant sows remains unknown. This study aimed to explore the effect and mechanisms of β-carotene on the reproductive performance of sows from the aspect of gut microbiota. A total of 48 hybrid pregnant sows (Landrace × Yorkshire) with similar parity were randomly allocated into three groups (n = 16) and fed with a basal diet or a diet containing 30 or 90 mg/kg of β-carotene from day 90 of gestation until parturition. Dietary supplementation of 30 or 90 mg/kg β-carotene increased the number of live birth to 11.82 ± 1.54 and 12.29 ± 2.09, respectively, while the control group was 11.00 ± 1.41 (P = 0.201). Moreover, β-carotene increased significantly the serum nitric oxide (NO) level and glutathione peroxidase (GSH-Px) activity (P &lt; 0.05). Characterization of fecal microbiota revealed that 90 mg/kg β-carotene increased the diversity of the gut flora (P &lt; 0.05). In particular, β-carotene decreased the relative abundance of Firmicutes including Lachnospiraceae AC2044 group, Lachnospiraceae NK4B4 group and Ruminococcaceae UCG-008, but enriched Proteobacteria including Bilophila and Sutterella, and Actinobacteria including Corynebacterium and Corynebacterium 1 which are related to NO synthesis. These data demonstrated that dietary supplementation of β-carotene may increase antioxidant enzyme activity and NO, an important vasodilator to promote the neonatal blood circulation, through regulating gut microbiota in sows.

Abstract Immunoglobulin G antibody response, measured as sample-to-positive (S/P) ratio, to Porcine Reproductive and Respiratory Syndrome virus (PRRSV) has been proposed as an indicator trait for improved reproductive performance in PRRSV-infected purebred sows and PRRSV-vaccinated crossbred gilts. In this study, we investigated the genetic correlations (rg) of S/P ratio following a PRRSV outbreak and PRRSV-vaccination with performance in non-exposed and PRRSV-exposed sows. PRRSV outbreak phase was defined based on previously described methodologies after the detection of typical clinical signs of PRRSV infection. 541 Landrace sows had S/P ratio measured at ~54 days after the beginning of the PRRSV outbreak (S/Poutbreak), and 906 Landrace x Large White naïve F1 gilts had S/P ratio measured at ~50 days after vaccination with a commercial modified live PRRSV vaccine (S/PVx). 711 and 428 Landrace sows had reproductive performance recorded before and during the PRRSV outbreak, respectively. 811 vaccinated F1 animals had farrowing performance for up to 3 parities. All animals were genotyped for ~28K SNPs. The estimate of rg of S/Poutbreakwith S/PVx was high (rg±SE = 0.72±0.18). Estimates of rg of S/Poutbreak with reproductive performance in F1 sows were low to moderate, ranging from 0.05±0.23 (number stillborn) to 0.30±0.20 (total number born). Estimates of rg of S/PVxwith reproductive performance in non-infected purebred sows were moderate and favorable with number born alive (0.50±0.23), but low (0 to -0.11±0.23) with litter mortality traits. Estimates of rg of S/PVx were moderate and negative (-0.47±0.18) with the number of mummies in PRRSV-infected purebred sows and low with other traits (-0.29±0.18 for total number born to 0.05±0.18 for number stillborn). These results indicate that selection for antibody response following a PRRSV outbreak collected in purebred sows and to PRRSV vaccination collected in commercial crossbred gilts may increase litter size of non-infected and PRRSV-exposed purebred and commercial crossbred sows.
