Pro-oxidant effects of a high α-tocopherol dose on kidney antioxidant biomarkers and histopathological aspects

Abstract. The aim of this study is to evaluate the effect of α-tocopherol supplementation at two doses (600 and 1200 mg × kg–1) on kidney antioxidant status and the histopathological changes in Wistar rats after 12 weeks of exposure at different diets. Forty rats has been divided into 4 groups of 10 rats each, the control group received basal diet with 5 % fresh sunflower oil (FSO), the second group: 5 % oxidized sunflower oil (OSO), the third group: 5 % OSO supplemented with 600 mg × kg–1 α-tocopherol and the fourth group: 5 % OSO supplemented with 1200 mg × kg–1 α-tocopherol. In OSO groups, the results showed highly significant increases of LPO (from 31.3 ± 0.9 to 53.8 ± 1.2 nmol of MDA formed/min/mg protein, p < 0.0001) with a significant decrease (p < = 0.001) of the antioxidant enzymatic activities (CAT, SOD, GPX, GR and G6PDH), body weight (339 ± 9 to 290 ± 3 g) and α-tocopherol levels (13.6 ± 0.6 to 6.5 ± 0.4 μg/mg protein). In OSO groups with 600 mg × kg–1 α-tocopherol, an antioxidant effect was found, reflected by a return of the parameters to values similar to those of the control group. However, higher doses of α-tocopherol (1200 mg × kg–1) induced a depletion of antioxidant status, α-tocopherol levels (6.0 ± 0.3 μg/mg protein, p < 0.001) and a very highly significant rise (p < 0.0001) of LPO content (54.86 ± 0.01 nmol of MDA formed/min/mg protein). The kidney tissues also showed changes in glomerular, severe inflammatory cells infiltration, and formation of novel vessels. So, we can conclude that the oxidative stress is attenuated by a moderate administration of 600 mg × kg–1 α-tocopherol, while a pro-oxidant effect occurs at 1200 mg × kg–1 α-tocopherol.

In this study, experimentally lindane-induced histopathological changes and proliferation and/or apoptosis in germ cells in the rat testes were investigated. A total of 40 healthy fertile 3-month-old male rats were used. Animals were divided into 4 groups, each containing 10 rats. Group 1 (control) was given only pure olive oil, Groups 2, 3 and 4 were administered lindane at 10, 20 and 40 mg/kg/bw, respectively, by gastric gavage for 30 days. Microscopically, degenerative changes were observed in the lindane-treated groups. For proliferative activity PCNA immunolabelling and for germ cells apoptosis TUNEL methods were performed. Although a strong PCNA positivity in the control group was observed, a gradual decrease was noted in the lindane-treated groups especially at higher doses. Significant increases of apoptosis were seen in the lindane-treated groups compared to the control group. A decrease in testosterone concentrations was observed in lindane-treated groups compared to the control group. The study indicates that high-dose lindane intoxication contributes to the suppression of spermatogenesis through a reduction of germ cell proliferation and an increase of germ cell death in rat testes.

The aim of the study was to detect significant biochemical changes in antioxidant status during acute period of ischemic stroke in its various pathogenic subtypes in patients aged 45–74years. The most important results were blood levels of ascorbic acid, malondialdehyde and free/ oxidized glutathione ratio. Cardioembolic stroke was associated with: the reduced ascorbic acid level of 62–74% comparable to control group; 5-times increased malondialdehyde level at the end of the first week of the disease; and also 50% decreasing of cell antioxidant capacity comparable to control group and other stroke subtypes. In lacunary stroke minimal changes in antioxidant status were measured, possibly because of small size of brain damage. Positive clinical dynamics was followed by decreasing of free glutathione levels. It could be related with its previous significant expenditure in process of cell recovery in penumbra area. In case of favorable outcome all of the markers return to the levels which were comparable to control group. The differences in the dynam-ics of the cells antioxidant capacity and consumption of native antioxidants, the time from the onset of the disease, the severity of the clinical picture, depending on the stroke subtype, were revealed. Further research of this problem may help to create new methods of diagnostics and selective pharmacological correction of this pathology.

Abstract: Cayenne pepper (Capsicum frustescens L ) is commonly in daily food. This study aimed to determine the effect of cayenne pepper to histopathological changes in gaster of Wistar rats induced by aspirin. This was an experimental study. Subjects were Wistar rats consisted of 5 rats as the negative control and 20 rats as the treatment group. Group A, the negative control group, was untreated; group B was treated with aspirin 21mg/day for 10 days; group C was treated with aspirin 21mg/day for 10 days together with cayenne pepper 84mg once daily; group D was treated with aspirin 21mg/day for 10 days followed by cayenne 84mg twice daily; and group E was treated with aspirin 21mg/day for 10 days and then was untreated for 3 days. The histopathological changes in group C showed a lot of inflammatory cells and erosion of gastric mucosa epithelium compared to group B, D, and E. Group D also showed inflammatory cells but no erosion as in group C. Group E had less inflammatory cells than group B, C, and D. Conclusion: Administration of aspirin 21mg/kg BW for 10 days led to acute gastritis in Wistar rat. Administration of aspirin together with cayenne 84mg/day led to acute gastritis and erosion of gastric mucosa epithelium. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian cabe rawit terhadap gambaran histopatologik lambung tikus wistar yang diinduksi aspirin. Jenis penelitian ialah eksprimental. Subjek penelitian ialah tikus wistar, yang dibagi atas: 5 ekor untuk kontrol negatif dan 20 ekor untuk perlakuan. Kelompok A (kontrol negatif) tidak diberi perlakuan; kelompok B diberikan aspirin 21mg/hari selama 10 hari; kelompok C diberikan aspirin 21mg/hari selama 10 hari bersama cabe rawit 84mg sekali sehari; kelompok D diberikan aspirin 21mg/hari selama 10 hari kemudian dilanjutkan pemberian cabe rawit 84mg selama 3 hari 2 kali sehari; dan kelompok E diberikan aspirin 21mg/hari selama 10 hari kemudian tidak diberi perlakuan selama 3 hari. Hasil penelitian menunjukkan bahwa pada kelompok C terlihat banyak sel-sel radang disertai erosi epitel mukosa lambung dibanding dengan kelompok B, D, dan E. Pada kelompok D pemberian aspirin dilanjutkan dengan cabe rawit dosis 2 kali sehari masih terdapat sel-sel radang tetapi tidak terjadi erosi seperti pada kelompok C. Kelompok E memiliki sel-sel radang yang lebih sedikit dibandingkan kelompok B, C, dan D. Simpulan: Pemberian aspirin 21mg/hari selama 10 hari menyebabkan gastritis akut pada tikus wistar. Pemberian aspirin bersama cabe rawit 84mg/hari menyebabkan gastritis akut erosi sedangkan pemberian cabe rawit 168mg/hari setelah diberi aspirin menyebabkan gastritis akut tanpa erosi.Kata kunci: cabe rawit, lambung, aspirin

The present study was conducted on 40 females and males white mice of approximately the same ages (4-6 weeks) and body weights (23-25 gm), for the aim of observing the toxic effects and histopathological changes on bones of mice due to prolonged treatment (6 months) with anticancer chemotherapeutic agents namely methotrexate. Forty mice were divided into 4 equal groups (10 mice of each group – 5 mice per sex). The first group (low or therapeutic dose group) was received 0.15 mg/ kg, I/M once weekly. The second group (intermediate dose group) received 0.30 mg/kg, I/M once weekly. The third (toxic dose group) received 0.45 mg/kg I/M once weekly. The fourth group was a control group, it received 0.2 ml buffered normal saline, I/M once weekly.The results showed Methotrexate can cause osteoporosis and bone marrow suppression, due to cytotoxic effects of it on bone marrow.

     The study was designed to determine the infection rate of Toxoplasma gondii in the rabbits and histopathological changes of infected internal organs in mice. A total of 60 blood samples were collected from clinically healthy rabbits from different region of Baghdad city for the serological detection of T. gondii infection. Biological assay in mice was performed by intraperitoneal inoculation of 0.1 ml digested organs suspension, (lungs, heart, liver, spleen, kidney, muscles, and brain) during the period from October 2013 until July 2014. The results revealed histopathological changes in bioassay mice infected by T. gondii isolated from domestic rabbit, no histopathological changes in the brain of animals of first group1 (control group), but in the group 2 (killed at day 15) the main pathological changes were perivascular perineuronal edema with presence of some degenerated neurons characterized  by shrunken dark blue stained (basophilic) cell bodies. In the animals of the group 2 (killed at day 21) the main brain histopathological changes were Sever congestion of meningeal blood vessels with infiltration of inflammatory cells and focal aggregation of microglial cells with diffuse obvious focal gliosis, whereas, in the same group animals that killed at day 28 the main brain histopathological changes were focal encephalomalacia and edema between molecular and granular layer.

The present study assessed the effect of dietary supplementation with coconut oil (CNO), watercress oil (WCO), and their mixture as promoters of growth, antioxidant status, immunity, and intestinal microbiota in growing rabbits. A total of 120 rabbits were distributed into six groups (20 rabbits/group) receiving a basal diet without supplementation (G1) or diet supplemented with 2 g CNO (G2), 2 g WCO (G3), 0.5 g CNO plus 1.5 g WCO (G4), 1 g CNO plus 1 g WCO (G5), or 1.5 g CNO plus 0.5 g WCO/kg (G6). Live body weight and feed conversion ratio were significantly higher in the G4 and G5 groups than in the other groups. Superoxide dismutase activity and reduced glutathione concentration were significantly improved in the CNO or WCO diets. Supplemental CNO plus WCO at all tested levels produced the best lysozyme and complement 3 activities. Cecal lactobacilli, coliform, Enterobacteriaceae, and Clostridium spp. populations were lower in the group who received the 1 g CNO + 1 g WCO/kg diet than that in the control group. Dietary supplementation of 1 g CNO + 1 g WCO or 0.5 g CNO + 1.5 g WCO/kg had the potential to improve growth, feed utilization, antioxidant status, and immunity, and reduce cecal pathogenic bacteria in rabbits.

This work aimed to explore the effects of dietary supplementation of Chlorella vulgaris (CLV) on the growth performance, carcass traits, hematobiochemical variables, immunity responses, and the antioxidant status of growing rabbits. A total number of 100 rabbits were randomly distributed into four treatment groups, each of five replicates (25 rabbits/group). The experimental groups were as follows; control: a basal diet without supplementation, CLV0.5: basal diet + 0.5 g chlorella powder/kg diet; CLV1.0: basal diet + 1.0 g chlorella powder/kg diet, CLV1.5: basal diet + 1.5 g chlorella powder/kg diet. Live body weight (LBW), cumulative body weight gain (CBWG), feed intake (FI), and feed conversion ratio (FCR) were not affected by dietary CLV supplementation. Platelet count (PLT), hematocrit (HCT), means corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) values were significantly increased in the CLV0.5 group compared with the other treatment groups. Dietary supplementation of CLV (1.5 g/kg diet) significantly reduced the alanine aminotransferase (ALT) activity. The concentrations of serum triglycerides and very low-density lipoprotein (VLDL) were lower (p < 0.05) in the CLV-treated groups than those of the control. Supplemental CLV at all experimental levels gave the best values of immunoglobulins (IgG and IgM) and glutathione activities. Malondialdehyde (MDA) levels were lower in the animals that received CLV in their diet than those of the control group. Dietary supplementation of 1.0 g CLV/kg had the potential to enhance immune responses and antioxidant status, as well as reduce blood lipid accumulation. Therefore, it could be concluded that CLV supplementation to growing rabbit diets can improve the health status.

This study investigated the effects of supplementing different levels of Moringa oleifera leaf powder (MOLP) on antioxidant status and blood biochemical indices during early gestation in Beetal goats. A total of 30 goats were randomly divided into three groups (n = 10) comprising control (basal diet without MOLP), the 1.6% MOLP supplemented group (basal diet + 1.6% MOLP), and the 3.2% MOLP supplemented group (basal diet + 3.2% MOLP). The experiment started 5 days before estrus synchronization and lasted till day 60 of gestation. The MOLP significantly increased plasma flavonoids in 1.6% as well as 3.2% supplemented group on days 40 and 60 of pregnancy, while total phenolic contents were observed to be higher in the 3.2% MOLP supplemented group throughout the experiment in comparison with the control group. The supplementation improved plasma total antioxidant capacity (TAC) by decreasing malondialdehyde (MDA) and total oxidant status (TOS) in contrast to the control group. The activities of superoxide dismutase (SOD) and peroxidase (POD) were enhanced in both supplemented groups, whereas catalase (CAT) activity was noticed to be significantly high in the 3.2% MOLP supplemented group. The protein contents were significantly elevated with 1.6 and 3.2% supplementation levels from day 40 to day 60 of the experiment. Plasma sugar level, carotenoids, progesterone profile, and hydrolytic (protease and amylase) enzymes activities were improved only when supplemented with 3.2% MOLP. The findings suggest that supplementing with 3.2% MOLP provides beneficial effects on early pregnancy stress in Beetal goats.

The aim of the present work is to found out if there are histopathological and enzymatic effects on the heart and liver after oral administration of Citrullus colocynthis (CC) fruit (bitter apple) for mature male rabbits, as a model for mammals. Two experiments were performed for sixty mature male rabbits that equally and randomly divided into six groups each containing five animals .In the two experiments ,independently, three groups in each one were considered as treated groups and represented as T1, T2 and T3 groups .These rabbits groups were orally administrated with low dosage of CC (4.8mg/kg/day,experiment No.1 ) and double dosage of CC extract (9.6mg /kg /day , experiment No2 ) for three intervals 2, 4 and 8 weeks ,respectively. The other three groups in each experiment were considered control and represented as C1, C2 and C3 groups, respectively. All the rabbits in control groups were given orally distilled water (DW) at the same periods of treated groups. Histopathological changes of the heart and liver with related enzymes namely; serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT) and creatinine kinase (SCK) were studied.The histopathological study revealed no changes in rabbit’s heart tissue particularly after treatment with 4.8mg/kg/day of CC for 2 and 4 weeks. While no histopathological changes were observed in the heart of rabbit groups treated with 9.6mg/kg/day of CC extract after 2 weeks. The results of the treatment with the two dosages of the CC for 8 weeks indicated a mild degenerative changes and mild necrosis of the myocardial cells. There was swelling of the hepatocytes and perivascular cuffing of mononuclear inflammatory cells after two weeks of daily treatment with 4.8mg/kg/day of CC exposure. After four weeks with the low dosage of the CC extract, caused initiation of necrosis, more inflammation picture of liver portal tract with sinusoid All rabbit groups showed statistically a significant gradual increase (P<0.05) in the value of serum enzyme GOT, GPT and CK levels after treatment with the two dosages of CC at the end of each different period compared with before treatment and control group. Concluded from this study that the dosage 4.8mg/Kg/day of CC plant resulted in simple histopathological effects on the heart and mild histopathological changes on the liver during the entire period of the study, instead of serum enzymatic elevation of SGOT ,SGPT and SCK. Further studies are recommended to found out the possibility to use and effects of CC on animal hygiene and reproduction

This study was conducted with the objectives to determine the effects of licorice root powder (Glycyrrhriza glabra) on performance, serum parameters, egg yolk cholesterol and antioxidant capacity in laying Japanese quail. Two hundred and forty 10-wk-old Japanese quails were randomly assigned to four dietary groups, each one four times replicated with fifteen quails per dietary groups. Control group fed the basal diet; other groups were fed basal diet supplemented with 0, 0.5, 1.0 and 1.5% licorice root powder. There were no significant differences in terms of final live weight, feed intake, feed conversion ratio, egg weight, serum cholesterol, triglyceride and High-Density Lipoprotein (HDL) concentration between treatment groups. Egg production was higher in control group and 0.5% licorice root powder supplemented groups. 1.0% licorice root powder supplemented group had lower egg production rate. Licorice root powder supplementation decreased Low Density Lipoprotein (LDL) concentration and increased glucose concentration significantly. Licorice root powder supplementation increased total antioxidant status (TAS) and reduced total oxidant status (TOS) and oxidative stress index (OSI). It was concluded from this study that supplementation of licorice root powder at the level of 0.5, 1.0 and 1.5% to the feed of laying quails had no adverse effects on performance. Licorice root powder supplementation can be used to reduce cholesterol level and increase antioxidant status in quails.
