Treatment of multiple sclerosis is effective when anti-inflammatory, neuroprotective and regenerative strategies are combined. Dendropanax morbiferus ( DM ) has anti-inflammatory, anti-oxidative properties, which may be beneficial for multiple sclerosis. However, there have been no reports on the effects of DM on myelination, which is critical for regenerative processes. To know whether DM benefits myelination, we checked differentiation and myelination of oligodendrocytes (OLs) in various primary culture systems treated with DM leaf EtOH extracts or control. DM extracts increased the OL membrane size in the mixed glial and pure OL precursor cell (OPC) cultures and changed OL-lineage gene expression patterns in the OPC cultures. Western blot analysis of DM -treated OPC cultures showed upregulation of MBP and phosphorylation of ERK1/2. In myelinating cocultures, DM extracts enhanced OL differentiation, followed by increased axonal contacts and myelin gene upregulations such as Myrf, CNP and PLP. Phytochemical analysis by LC-MS/MS identified multiple components from DM extracts, containing bioactive molecules such as quercetin, cannabidiol, etc. Our results suggest DM extracts enhance OL differentiation, followed by an increase in membrane size and axonal contacts, thereby indicating enhanced myelination. In addition, we found that DM extracts contain multiple bioactive components, warranting further studies in relation to finding effective components for enhancing myelination.

Background: The study was conducted to identify the bacterial strain associated with marine sponge Hyrtiosaff. erectus collected from the Red Sea coastal water and to assess the utilization of their secondary metabolites for human benefit as antioxidant, anti-Alzheimer, anti-viral, anticancer and anti-inflammatory agent. Methods: After biochemical identification of Pesudomance sp. bacterial strain, the total polyphenol contents, cytotoxic, antioxidant, anti-Alzheimer, anti-viral, anticancer and anti-inflammatory activity of the Pesudomance sp. ethyl acetate extract were investigated by applying different biochemical assays. Polyphenol contents were investigated using spectrophotometric techniques. Antioxidant activity was determined by 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), and 2,2/-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ABTS radical scavenging activity assays. The cytotoxic effects were investigated by using the human cancerous cell lines. Results: The anti-Alzheimer, anti-viral, anticancer and anti-inflammatory activities were determined using ELISA. Qualitative phytochemical analysis of the Pesudomance sp. extract demonstrated the presence of a large and diverse group of substances such as alkaloids, carbohydrates, flavonoids, phenols, terpenoids, saponins, and tannins. The strong antioxidant activity of the Pesudomance sp. extract was mainly attributed to the protective role of polyphenols against reactive oxygen. It was also observed that Pesudomance sp. extract possessed significant anti-Alzheimer activity with 94% at 1 mg. The extract showed also high antiviral activity (90%) using reverse transcriptase enzymes inhibition assay. The examination of the anticancer activity by applying two experimental models, i.e., PTK and SHKI cleared out high significant percentages of 76.19 and 83.09 %; respectively. Conclusion: The anti-inflammatory profiling using TNF, COX1, COX2, IL6 also revealed high antiinflammatory activity with different metabolic pathway of 62.70, 75.444, 79.27 and 54.15 %; respectively. The present study concluded that ethyl acetate extract of Pesudomance sp. possessed strong antioxidant, anti-Alzheimer, and anti-viral, anticancer and anti-inflammatory activities. Further studies are required to purify the bioactive compounds.

Spirulina is used as dietary supplement in form of capsules and tablets. It is rich in Bioactive molecules and exhibits various activities. Commercially available Spirulina was collected. Extracts were prepared using various polar and non-polar solvents. The extracts were used for qualitative phytochemical analysis and to determine their Anti-Oxidant, Anti-Inflammatory and Anti-Arthritic activity. Phytochemicals like Tannin, Carbohydrates and Glycosides were present in all the extracts, while others were present only in few extracts. Biosynthesis of Nanoparticles was done by using the extract. Chloroform extract and Nanoparticles showed higher Anti-Oxidant activity, Ethanol extract showed higher Anti-Inflammatory activity and Chloroform extract showed higher Anti-Arthritic activity.

Objective: Plant-derived bioactive molecules are providing infinite opportunities for new drug development as they possess a wide range of actions against diseases with lesser side effects. The present study is made to analyze the qualitative phytochemicals and to evaluate in vitro antiproliferative and anti-inflammatory potentials of leaf extracts of Thespesia populnea. Methods: T. populnea leaf extracts were prepared sequentially according to the increasing polarity of the solvents, i.e., petroleum ether, chloroform, ethyl acetate, and methanol. Qualitative phytochemical analysis was performed to identify the chemical constituents of the extracts, and antiproliferative properties were evaluated against different cell lines using the 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide assay. Bovine serum albumin anti-denaturation assay was done to identify the anti-inflammatory activity.Results: Phytochemical analysis of the extracts revealed the presence of alkaloids, terpenoids, glycosides, tannins, flavonoids, and phenolics. The chloroform extract (CHFE) of T. populnea has a dose-dependent antiproliferative effect against acute T-cell leukemia (Jurkat E6-1), prostate Grade IV adenocarcinoma (PC-3), mouse fibroblast (L-929), and monkey kidney normal (Vero) cells. Their inhibitory concentration 50% (IC50) values were found to be 35.73±0.94 μg/ml for Jurkat E6-1, 60.79±1.84 μg/ml for PC-3, 60.88±1.45 μg/ml for L-929, and 83.482±2.05 μg/ml for Vero, respectively. CHFE also displayed the anti-inflammatory potential.Conclusion: The chloroform leaf extract of T. populnea possesses potent antiproliferative and anti-inflammatory activity. These properties present in leaf extract may be explained by the presence of biologically active constituents.

This work aims to extract and identify bioactive molecules of the plant Anethum graveolens, commonly known as Dill, which occupies an important place in Moroccan vegetation. In order to optimise their later use in the field of herbal medicine and nutrition, a focus on the phytochemical analysis of an extract from different parts of the plant were examined by using preliminary tests and staining reaction precipitation by the use of specific reagents, or by thin layer chromatographic analysis (TLC). Phytochemical tests, different organs, aerial and underground part (on different areas of the plant), revealed the richness of this plants tannins, reducing compounds, alkaloids, coumarins, terpenoids, sterols, flavonoids, irridoides, and this screening has allowed us to show that for the extraction of polyphenols, methanol is the best extraction solvent.

Background: Multiple sclerosis is an autoimmune chronic inflammatory disease of the central nervous system that can lead to some serious disabilities. Despite using various immunomodulatory and anti-inflammatory drugs that have therapeutic effects, they cannot reduce its progression completely, and have some unwanted side effects too. The immunomodulatory and anti-inflammatory effects of the β-D-Mannuronic acid [M2000] have been proven in several surveys, and the present research was designed to determine its toxicity and therapeutic effects in MS patients. Methods: This study was performed on 15 MS patients who took 25 mg/kg/day the oral form of the β-D-Mannuronic acid for six months, and 15 healthy people as a control group. Serum levels of Urea, Creatinine, GGT, Vitamin D3, Uric acid, and Anti-Phospholipids were compared to evaluate the therapeutic and possible toxic effects of this drug after this period. Results: Non- toxic effects through the study of Urea, Creatinine, GGT, and non-significant changes in Uric acid and AntiPhospholipids levels, besides a significant rise in Vitamin, D3 levels in the M2000 treated cases were found. Conclusions: Our results suggested that β-D-Mannuronic acid is a safe drug and has no toxicity when administered orally and also has some therapeutic effects in MS patients.

Background: Since its inception, people are using herbal extracts as natural remedies for the treatment of various diseases. Bryophyllum pinnatum, also known as the air plant, is a well-recognized herb used in folk medicine. It is native to Madagascar and further planted in tropical and subtropical areas around the world. It is known for the profusion of miniature plantlets which arise from the margins of phylloclade. Objective: The aim of this review is to provide the information regarding local and traditional uses, pharmacological activities and different phytochemicals reported from leaves extracts of Bryophyllum pinnatum. Methods: This review article contains a detailed survey of the literature about Bryophyllum pinnatum available in different online databases, such as; PubMed, Web of Science, Science Direct, Elsevier, and Google Scholar, etc. In this review, authors have focused on ethnopharmacological importance and phytochemicals present in Bryophyllum pinnatum and their structure. The structures of the phytochemical were prepared by the ChemDraw tool. Results: This plant is used as a traditional herbal medicine around the globe due to medicinal properties like; anthelmintic, immunosuppressive, hepatoprotective, antinociceptive, anti-inflammatory, antidiabetic, nephroprotective, antioxidant, antimicrobial, analgesic, anticonvulsant and antipyretic. Phytochemical analysis revealed the presence of many bioactive compounds like; alkaloids, flavonoids, triterpenes, steroids, glycosides, bufadienolides, lipids, and organic acids, etc. which are associated with different medicinal properties. Conclusion: Bryophyllum pinnatum possesses diverse pharmacological importance and remarkable medicinal properties. Investigators have reported a large number of phytochemicals exhibiting different medicinal properties and correlation of medicinal properties.

Abstract Euphorbia myrsinites is one of the oldest spurges described and used in folk medicine. It is characterized by blue-grey stems similar to myrtle, and it is spread in the Mediterranean region, Asia, and the USA. Chemical analysis of E. myrsinites collected in Turkey afforded the isolation of 4 diterpenes based on the so-called myrsinane skeleton being tetraesters of the tetracyclic diterpene alcohol myrsinol. In this study, the phytochemical analysis of this species collected in Italy has been undertaken to afford the isolation of a new atisane diterpene, named myrsatisane, 3 ingenol derivatives, along with the 4 tetraester derivatives previously found. A triterpene compound based on the euphane skeleton has also been isolated. Structural elucidation of the new myrsatisane was based on spectroscopic techniques, including HR-MS and 1- and 2-dimensional NMR experiments. Its relative configuration was determined by NOE correlations, while absolute stereochemistry was obtained by quantum-mechanical DFT studies. While diterpenes with the atisane skeleton are relatively common in Euphorbia species, this is the first report of an atisane diterpene from E. myrsinites. All the isolated terpenes were tested for anti-inflammatory activity on J774A.1 macrophages stimulated with lipopolysaccharide by evaluation of nitrite and pro-inflammatory cytokine Il-1β levels. Among tested compounds, the 3 ingenol diterpenes exhibited a dose-dependent (0.001 – 3 µM) significant activity, thus showing their potential as anti-inflammatory drug candidates.

Mesenchymal stromal cells derived from the fetal placenta, composed of an amnion membrane, chorion membrane, and umbilical cord, have emerged as promising sources for regenerative medicine. Here, we used next-generation sequencing technology to comprehensively compare amniotic stromal cells (ASCs) with chorionic stromal cells (CSCs) at the molecular and signaling levels. Principal component analysis showed a clear dichotomy of gene expression profiles between ASCs and CSCs. Unsupervised hierarchical clustering confirmed that the biological repeats of ASCs and CSCs were able to respectively group together. Supervised analysis identified differentially expressed genes, such as LMO3, HOXA11, and HOXA13, and differentially expressed isoforms, such as CXCL6 and HGF. Gene Ontology (GO) analysis showed that the GO terms of the extracellular matrix, angiogenesis, and cell adhesion were significantly enriched in CSCs. We further explored the factors associated with inflammation and angiogenesis using a multiplex assay. In comparison with ASCs, CSCs secreted higher levels of angiogenic factors, including angiogenin, VEGFA, HGF, and bFGF. The results of a tube formation assay proved that CSCs exhibited a strong angiogenic function. However, ASCs secreted two-fold more of an anti-inflammatory factor, TSG-6, than CSCs. In conclusion, our study demonstrated the differential gene expression patterns between ASCs and CSCs. CSCs have superior angiogenic potential, whereas ASCs exhibit increased anti-inflammatory properties.
