Novel, non-invasive, painless oral therapeutic agents are needed to replace the painful conventional treatment of arsenic-associated health hazards with metal chelators. Our aim was to examine the effect of spirulina ( Spirulina platensis ( Geitler, 1925 )) on arsenic-mediated uterine toxicity. Female Wistar rats were divided equally into four experimental treatment groups: control group, sodium arsenite group (1.0 mg/100 g body mass), spirulina placebo group (20 mg/100 g body mass), and sodium arsenite + spirulina group. In contrast with the control group, spectrophotometric and electrozymographic evaluation revealed that rats that ingested arsenic for 8 d showed significant diminution of the activities of superoxide dismutase, catalase, and peroxidase ( p < 0.001). Mutagenic uterine DNA breakage and tissue damage were prominent following arsenic consumption by the rats. Oral delivery of spirulina resulted in a significant amelioration of arsenic-induced adverse oxidative stress and genotoxic state of rats. A significant low-signaling ( p < 0.001) of gonadotropins and estradiol was also noted in the arsenic-treated rats, which was terminated by spirulina; this arsenic-primed adverse effect was significant ( p < 0.05, p < 0.01). The spirulina treatment mechanism could be associated with augmentation of the antioxidant defense system that protects the arsenic-mediated pathological state of the uterus.

This study was undertaken to determine the osteotropic effect of different doses of lipoic acid (LA) on the mineralization of bone tissue in female Wistar rats with experimental osteopenia induced by bilateral ovariectomy. Fifty-six rats were randomly selected and submitted to either a sham operation (n = 8) or an ovariectomy (n = 48). The ovariectomized rats were randomly placed into two control groups, treated subcutaneously with either physiological saline or 17β-estradiol in the dose of 4 μg/kg body mass per day, and four experimental groups that received LA subcutaneously in the doses of 12.5, 25, 50, and 100 mg/kg body mass per day (n = 8 in each group). After 28 days of experimental treatment, the rats were sacrificed, and body mass, total skeletal density, and body composition were recorded. Blood serum and isolated femora were stored for further analysis. Our results revealed that the osteoprotective effect of LA was dose-dependent and was observed in rats treated with 50 and 100 mg/kg of LA. Moreover, the LA applied to the ovariectomized rats in the dose of 50 mg/kg not only stopped the bone resorption, but stimulated its formation.

Skin aging is multitarget persistence processing that immediately involve hyperproduction of free radicals under influence of intrinsic and extrinsic factors and deterioration in intimal antioxidant defense system. The goal of the study was the evaluation of the anti-oxidant potential of artichoke standartizated extracts, 2%, as a protective strategy against skin age-associated oxidative damage caused by D-galactose (D-gal) in rats. 58 female Wistar rats included in the experimental design. D-gal-induced aging was reproduced in 36 animals of main group, and 12 rats included in control group.  All animals in main group were randomized for 3 groups: I – animals with skin aging reproduced model receive saline, II – animals with skin aging rats receive artichoke extracts (with content of chloroagenic acid 2.0%) in a dose of intradermal injection 0.13 mg/kg and main III group - animals with skin aging receive 1.3 mg/kg artichoke extract twice at weeks during 4 weeks. Influence of artichoke extracts restores skin relative weight and leads to decreasing the rate of generation of superoxide anion, hydrogen peroxide and lipid peroxidation (LPx), increasing activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and reverse ratio SOD/(catalase+GPx) to the production of H2O2 from superoxide dismutation coupling with the decrease ratio of generated O2-/H2O2. Low-dose of intradermally microinjection of artichoke extracts, 2%, activated the enzymatic link in innate antioxidant defense system in D-gal-induced skin aging model and could be recommended for applications in cosmetics as antiaging mesotherapy.


According to the Tambovagrochemcenter data, zinc deficiency in animal rations exceeds 33%. To de-termine the efficiency of ration additives 3 groups of calves were formed, one of which was control and received regular feed, first test group received additional Zinc Sulphate, and second test group received additional Zinc Bioplex. To assess the results, we take into account the following values: chemical com-position and nutritional values of the feed, body mass value, amount of immunoglobulins in blood, bio-chemical and hematological values of blood, morphological values of fur and skin. Noticeable results were achieved in 4 months, with first and second test groups receiving accordingly 3,15% and 6,13% more body mass increase over the control group. A tendency towards higher albumin concentration in both test groups was noted, and calves of the second test group had higher amounts of hemoglobin and higher concentrations of it per erythrocyte. In samples of epidermis and hairs of the second test group the papillary layer was well developed, with the amount of hair and hair follicles exceeding 7 in the field of view, with the thickness of hairs being at least 70 μm. Inner papilla of hair contained 6 to 8 layers of hair, thickness of awn hairs reaching 80-90 μm, down hairs reaching 40 μm, with the hair cortex making up more than 80% of the hair thickness. Thus, introducing zinc salts and chelated zinc compounds to the milk-fed calves’ ration, has resulted in monetary profit increases over the control group by 255,0 and 612,0 roubles accordingly. By all researched values, the highest results were reached from adding zinc bioplex with approximately 360 mg added per animal for the entire duration of the growth process.

Summary Background: It has been reported that obesity is associated with metabolic syndrome, insulin resistance, cardiovascular risk but also with nonalcoholic fatty liver disease (NAFLD). The prevalence of obesity in children and adolescents is increasing rapidly all over the world. The aim of this study was to analyze the value of liver enzymes: AST, ALT and γGT in a group of obese students in order to establish their correlation to anthropometric parameters such as: BMI (body mass index), WC (waist circumference), HC (hip circumference), and WHR (waist-to-hip ratio) compared to non-obese students who comprised the control group (CG). Methods: In this study, 238 students from the University of Novi Sad of both sexes (126 men and 112 women) with a mean age of 22.32 ± 1.85 years were included. According to the body mass index (BMI) lower and higher than 25 kg/m2 and waist circumference (WC) lower and higher than 94 cm (80 cm for females) the whole group of 238 students was divided into 2 subgroups: the obese group at increased risk for CVD (Group 1) and the group at lower risk for CVD (Group 2). AST, ALT and γGT activities were determined in fasting blood samples. Results: Statistical processing data revealed significantly higher values of AST, ALT and γGT in the group of students with BMI>25 kg/m2, WC>94 cm for males and WC>80 cm for females, HC>108 cm for males and HC>111 cm for females, and WHR>0.90 for males and WHR>0.80 for females (P<0.001). Significant association was established between anthropometric parameters and liver enzyme levels (P<0.0001). Conclusions: Obese students with higher BMI, WC, HC and WHR values have higher liver enzyme activites and a higher chance to develop NAFLD in the future.

To analyse the change on leptin, body composition, blood pressure (BP), cardiorespiratory fitness (CRF) and some biochemical parameters in physically inactive women and men with primary hypertension (HTN) and obesity, and to evaluate the potential sex differences in the change after intervention. METHODS. Participants (n = 37 women, n= 40 men, 52.9 ± 6.9 yrs) from the EXERDIET-HTA study were randomized into attention control group (physical activity recommendations) or one of three supervised aerobic exercise groups [two days/week: high-volume (HV) with 45 min of moderate-intensity continuous training, HV and high-intensity interval training (HV-HIIT), and low volume-HIIT (LV-HIIT, 20 min)]. All participants received the same hypocaloric diet. All variables were assessed pre and post intervention (16 weeks). 24 h ambulatory BP monitoring was used to analyze systolic and diastolic BP. A cardiopulmonary exercise test was performed to determine peak oxygen uptake (VO2peak). RESULTS. Following the intervention, there were increments (P &lt; 0.01) in CRF by VO2peak (Women, W = 21.1 ± 3.7 vs. 24.6 ± 4.4 mL·kg-1·min-1, Men, M = 26.3 ± 6.0 vs. 33.1 ± 10.2 mL·kg-1·min-1) and decreases (P &lt; 0.05) in leptin (W = 49.5 ± 23.0 vs. 41.8 ± 19.9 ng/mL, M = 20.5 ± 14.8 vs. 12.9 ± 18.6 ng/mL), body mass (W = 84.7 ± 12.1 vs. 80.3 ± 11.5 kg, M = 97.9 ± 14.4 vs. 91.5 ± 13.3 kg), waist perimeter (W = 97.3 ± 10.7 vs. 94.3 ± 10.9 cm, M = 107.9 ± 8.7 vs. 101.5 ± 7.9 cm), fat mass (W = 42.3 ± 5.1 vs. 38.6 ± 8.4 %, M = 31.2 ± 5.0 vs. 28.0 ± 4.4 %), systolic BP (M = 136.5 ± 12.1 vs. 129.3 ± 12.5 mmHg), diastolic BP (W = 76.2 ± 8.9 vs. 74.1 ± 8.7 mmHg, M = 79.3 ± 7.2 vs. 75.0 ± 8.2 mmHg), total cholesterol (M = 216.1 ± 44.5 vs. 196.1 ± 35.0 mg/dL), insulin (W = 13.4 ± 7.9 vs. 9.4 ± 4.2 mU/L) values. There were significant between-sex differences in body mass (W=-5.2%, M=-6.5%, effect size, ES = 0.073), waist circumference (W=-3.1%, M=-5.9%, ES = 0.114), and VO2peak (W = 14.2%, M = 20.5%, ES = 0.059). CONCLUSIONS. Aerobic exercise along with hypocaloric diet is an effective non-pharmacological tool to induce beneficial changes in women and men in BP and leptin as a mediator of obesity-induced HTN, and other regulatory mechanisms such as body composition, CRF and biochemical profile. The found sex-related differences could confirm the need of individual non-pharmacological strategies.

Background and objectives: Reactive oxygen species (ROS) overwhelm the antioxidant defense system, induce oxidative stress, and increase matrix metalloproteinase (MMP) expression, resulting in skin aging. Thus, preventing ultraviolet B (UVB)-induced skin damage can attenuate skin aging. Spirulina (a biomass of cyanobacteria, also called blue-green algae) is comprised of prokaryotes, whereas microalgae are eukaryotes and are rich in phycocyanin, a powerful antioxidant. Materials and Methods: Here, we investigated the photoprotective effects of spirulina-derived C-phycocyanin (C-PC) against UVB radiation using keratinocytes (HaCaT cells). Results: UVB radiation increased MMP-1 and MMP-9 expression but decreased involucrin, filaggrin, and loricrin expression. C-PC showed no toxicity at concentrations of 5–80 μg/mL in terms of HaCaT cell viability. UVB-irradiated HaCaT cells had a 50.8% survival rate, which increased to 80.3% with C-PC treatment. MMP expression increased with UVB treatment, whereas MMP-1 and MMP-9 concentrations decreased with C-PC treatment. UVB reduced involucrin, filaggrin, and loricrin expression in HaCaT cells, but 80 μg/mL C-PC increased their expression by >25%. In the UVB radiation group, dichlorofluorescin diacetate fluorescence intensity in HaCaT cells increased by 81.6% compared with that in the control group, whereas ROS production was reduced by 51.2% and 55.1% upon treatment with 40 and 80 μg/mL C-PC, respectively. Conclusions: C-PC might reduce or prevent skin aging by reducing UVB irradiation-induced skin wrinkles and free radicals.

Abstract Objectives This study examined the possible protective roles of exogenous glycine (Gly) and L-Arginine (l-Arg) against Diclofenac (DIC)-induced gastro-duodenal damage in rats. Methods Rats were divided into Group A (control), Group B (DIC group) and Groups C–F which were pre-treated for five days with Gly1 (250 mg/kg), Gly2 (500 mg/kg), l-Arg1 (200 mg/kg) and l-Arg2 (400 mg/kg), respectively, before co-treatment with DIC for another three days. Hematological, biochemical and histopathological analyses were then carried out. Results DIC produced significant (p<0.05) reduction in PCV (13.82%), Hb (46.58%), RBC (30.53%), serum total protein (32.72%), albumin (28.44%) and globulin (38.01%) along with significant (p<0.05) elevation of serum MPO activity (83.30%), when compared with control. In addition, DIC increased gastric H2O2 and MDA levels by 33.93 and 48.59%, respectively, while the duodenal levels of the same parameters increased by 19.43 and 85.56%, respectively. Moreover, SOD, GPx and GST activities in the DIC group were significantly (p<0.05) reduced in the stomach (21.12, 24.35 and 51.28%, respectively) and duodenum (30.59, 16.35 and 37.90%, respectively), compared to control. Treatment with Gly and l-Arg resulted in significant amelioration of the DIC-induced alterations although l-Arg produced better amelioration of RBC (29.78%), total protein (10.12%), albumin (9.93%) and MPO (65.01%), compared to the DIC group. The protective effects of both amino acids against oxidative stress parameters and histological lesions were largely similar. Conclusions The data from this study suggest that Gly or l-Arg prevented DIC-induced gastro-duodenal toxicity and might, therefore be useful in improving the therapeutic index of DIC.

The increasing prevalence of allergic asthma has become the world’s major health issue. Current treatments for allergic asthma focus on treating symptoms, while permanent cures still remain undiscovered. In this study, we investigated the effect of Korean traditional herbal remedy, Pyunkang-tang (PGT)—composed of six plants—on asthma alleviation in a mouse model. The PGT mixture was orally gavaged to mice (PM group, 20 mg/mouse/day) from 7 days before sensitization with ovalbumin (OVA) (day −7). On day 0 and day 14, mice from OVA-control (n = 9) and PM group (n = 8) were sensitized with OVA and alum through intraperitoneal injection. On days 18~20, OVA was challenged to mice through nasal injection and sacrificed next day. Cell profile in lung tissue was analyzed by flow cytometry and RT-qPCR analysis, and the number of eosinophils and expression of siglec-F were significantly reduced in the PM group. Lung tissue was examined with hematoxylin and eosin (H&E) and Alcian blue/periodic acid–Schiff (AB-PAS) staining. Noticeably reduced eosinophil infiltration around bronchioles was displayed in the PM group compared to the OVA-control group. Furthermore, PGT-treated mice showed a significant reduction in IL-13 and a mild reduction in IL-5 in lungs. A decreasing tendency of IL-5/13 (+) CD4+ T cells and IL-13(+) innate lymphoid cells (ILCs) and a significant reduction in IL5(+) ILCs were also observed. When treating PGT on murine lung epithelial cells stimulated by papain, there was a significant reduction in IL-33 mRNA expression levels. Taken together, oral delivery of PGT successfully alleviated asthmatic responses provoked by OVA in a mouse model and could lead to novel therapies for allergic asthma.

AbstractSpirulina platensis protein hydrolysates were prepared by digesting protein extracts with papain, and the hydrolysates were separated into 30, 10, and 3 kDa weights using membrane ultrafiltration. The 0–3 kDa low-molecular-weight Spirulina peptides (LMWSPs) proved the highest chemical antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, hydroxyl radical (·OH) scavenging activities and total antioxidant capacity. Cellular antioxidant ability of LMWPs fractions against 2000 μg/mL H2O2 induced oxidative damage of L02 cells were investigated. The MTT assay results displayed that LMWSPs at different concentrations (0–1000 μg/mL) had proliferation effect on the L02 cells and that treatment of the L02 cells with the 1000 μg/mL LMWSPs (0–3 kDa) significantly prevented H2O2-induced oxidative damage compared with control cells. Moreover, the 2′,7′-dichlorofluorescein diacetate (DCFH-DA) fluorescent probe assay showed that the levels of ROS and NO were significantly lower in the experimental group that was treated with the peptides for 24 h than in the control group. Furthermore, using the corresponding kits, the treatment inhibited the reduction of SOD activity and the increase of MDA contents in the L02 cells. Therefore, LMWSPs (0–3 kDa) may have potential applications in antioxidant and liver health products.

Abstract Although simultaneous supplementation with iron and folic acid is justified, the potential interactions between these micronutrients are unknown. The aim of this study was to determine the effects of oral iron and folic acid, administered together or separately, on iron concentration in tissues in rats with a deficiency of both these micronutrients. In the first stage of the experiment (28 days), 150 8-week-old female Wistar rats were randomly assigned to a control group (C; n = 30) fed the standard diet and to a study group (n = 120) fed a diet deficit in iron and folate. The study group was then randomly divided to four groups: D group fed a deficit diet, FE group fed a deficit diet with iron gluconate, the FOL group fed a deficit diet with folate acid, and the FEFOL group fed a deficit diet with iron gluconate and folate acid. After 2, 10, and 21 days of supplementation, ten animals from each group were killed. Morphological parameters were measured in whole blood. Iron concentration was assayed in serum, liver, spleen, pancreas, heart, and kidneys. Folic acid supplementation more significantly decreased iron concentrations in the pancreas and spleen than in the D group after 10 and 21 days of supplementation. Moreover, the combination of iron with folic acid markedly decreased iron levels in the liver and spleen, in comparison with iron alone, after 10 and 21 days of the experiment. In conclusion, folic acid affects iron status in female rats deficient in these micronutrients in moderate and long-term supplementation.
