At present, gold nanoparticles (GNPs) are widely used in biomedical applications such as cancer diagnostics and therapy. Accordingly, the potential toxicity hazards of these nanomaterials and human safety concerns are gaining significant attention. Here, we report the effects of prolonged peroral administration of GNPs with different sizes (2, 15 and 50[Formula: see text]nm) on morphological changes in lymphoid organs and indicators of peripheral blood of laboratory animals. The experiment was conducted on 24 white mongrel male rats weighing 180–220[Formula: see text]g, gold nanospheres sizes 2, 15 and 50[Formula: see text]nm were administered orally for 15 days at a dosage of 190[Formula: see text][Formula: see text]g/kg of animal body weight. The GNPs were conjugated with polyethylene glycol to increase their biocompatibility and bioavailability. The size-dependent decrease of the number of neutrophils and lymphocytes was noted in the study of peripheral blood, especially pronounced after administration of GNPs with size of 50[Formula: see text]nm. The stimulation of myelocytic germ of hematopoiesis was recorded at morphological study of the bone marrow. The signs of strengthening of the processes of differentiation and maturation of cellular elements were found in lymph nodes, which were showed as the increasing number of immunoblasts and large lymphocytes. The quantitative changes of cellular component morphology of lymphoid organs due to activation of migration, proliferation and differentiation of immune cells indicate the presence of immunostimulation effect of GNPs.

Recently gold nanoparticles (GNPs) have been actively studied as photothermal converters, drug carriers, and imaging agents in a wide range of applications in cancer diagnosis and therapy. The prolonged peroral administration of GNPs in a range of sizes was performed to investigate the morphological changes and their reversibility in the internal organs of laboratory animals. In this study, GNPs with average diameters of 2[Formula: see text]nm, 15[Formula: see text]nm and 50[Formula: see text]nm were administered during 30 days, and the reversibility of morphological changes was investigated 14 days after administration. After the prolonged administration of GNPs, the severity of morphological changes in the liver, kidney, spleen and lymph nodes depended on the nanoparticle size. Specifically, 50[Formula: see text]nm nanoparticles caused the most pronounced dystrophic and necrobiotic effects, whereas the smallest 2[Formula: see text]nm nanoparticles caused proliferative changes. Most importantly, the development of pathological processes was reversible, as evidenced by the gradual restoration of the organ structure at 14 days after the end of GNPs administration.

Live vaccines are used for specific brucellosis prophylaxis in the Russian Federation. These vaccines in addition to a number of  positive properties have some limitations including high  agglutinogenicity, reactogenicity, sensitizing activity. In this  connection, the development of subunit vaccines without adverse  reactions is a perspective direction in modern vaccinology. Complex  morphological research of the adrenal glands and comparative  analysis of peripheral blood (leukogram, leukocytic index and index  of allergization) of white mice immunized with thermoextracts (ТE)  of Brucella abortus I-206 in L- and S-form and inactivated vaccine B.  abortus 19 ВА were conducted. It was shown that ТE unlike B.  abortus 19 ВА caused minor alterations in peripheral blood of the  experimental animals in early periods of observation (increase of allergization index, changes in leukogram) with the subsequent  levelling to the values in control. Expositions of the adrenal gland  zoning were determined and cellular structure was estimated in  consideration of morphometry. Changes in architectonics of the mice  organ immunized with B. abortus 19 ВА were revealed. These alterations could indicate the stress-reaction development. In  case of ТE application the given changes were insignificant and were  developed in early periods. The revealed morphological changes in  adrenal glands of laboratory animals permit to prove the necessity of  realization the further experimental researches to ТE application  as the components for development of a subcellular brucellosis vaccine.

Introduction. Epoxiconazole is a highly effective triazole fungicide. It is a part of many modern compounds for the control of phytopathogens. Widely used in agriculture in all over the world and in Ukraine, in particular. Despite its hepatotoxic characteristics, this fungicide alters the hematological parameters of blood.The aim of the study – to investigate the effects of generic triazole fungicide epoxiconazole, 95% on hematological and cytochemical parameters of peripheral blood of Wistar Hannover rats in acute experiment.Research Methods. 10 healthy males of Wistar Han rats were equally divided into control (0 mg/kg/bw) and experimental groups. Dose 1580 mg/kg/bw of epoxiconazole (1/2 LD50) was administrated once orally by gavage to 5 experimental rats. Peripheral blood was studied at 0 and 1, 3, 7, 14 day after exposure (DAE). RBC, HGB, HCT, erytrocyte indices MCV, MCH, MCHC, WBC and PLT were, hemogram and morphological disturbances of cells were studied,percentage ratio of different types of leukocytes was calculated.The cytochemical status of leukocytes was assessed on the basis of determining the enzymatic activity of specific naphthol-AS-D-chloroacetateterase in neutrophils, succinate dehydrogenase and acid phosphatase in lymphocytes.Results and Discussion. Erythrocytosis in response to pesticide intoxication with a decrease of mean cellhemoglobin concentration in one erythrocyte; the reduction of hemoglobin concentration in the end of experiment against the background of active polychromasia confirmed the anemic effect of epoxiconazole. Reactive neutrophilia, stimulation of lymphocytopoiesis and monocytopoiesis, appearance of macrophages in the peripheral blood, increased activity of naphthol-AS-D-chloroacetateterase, succinate dehydrogenase and acid phosphatase were considered as compensatory mechanisms. Reverse quantitative changes in the ratio of lymphocytes subpopulations based on the activity of acid phosphatase were noted.Conclusions. Results of the study of generic fungicide epoxiconazole, 95% hematotoxic activity in the acute experiment on Wistar Hannover male rats showed the development of latent anemia, which was manifested in the late-term study; activation of monocytopoesis; increase of macrophages in the peripheral blood; changes in the immune response of the body and the appearance of atypical normocytes and lymphocytes with nuclear abnormalities.

Cytokin regulation of local immune reactions in pulmonary tissues (IL-10, IL-4, TGFb1, TNFα), and morphology of the vascular bed and alveolar epithelium of the lung air compartments were studied in a 15-day tail-suspended Wistar male rats with a body mass of 180–200 grams. Cytokin regulation of the local immunity after suspension was characterized by activation of cell proliferation and differentiation regulator TGFβ1 aimed to forestall the inflammatory and auto immune processes and to promote remodeling of the lung epithelial structure and extracellular matrix.

The effect of prolonged oral administration of gold nanoparticles of various sizes on the structure of the liver, spleen and lymph nodes of mature laboratory rats and their offspring was studied. The morphometric study was performed using the system of digital image analysis of medical Mikrovizor µVizo-103. The detection of conglomerates of gold nanoparticles in tissues was carried out using the dark field microscopy method. It was found that gold nanoparticles of sizes 2 and 50 nm cause the most pronounced morphological changes in the organs, which indicates the size-dependent character of their effects. In the liver of the group with 50 nm gold nanoparticles administration the expressed dystrophy of hepatocytes and activation of monocytic-macrophage system were observed. In the spleen and lymph nodes, the processes of proliferation and differentiation of immunocompetent cells and macrophages were activated. Oral administration of gold nanoparticles during pregnancy in females was accompanied by an increase in the number of hepatocytes with necrobiotic changes and an increase in the number of non-parenchymatous elements in the liver of the offspring, most pronounced in the groups with the administration of 2 and 50 nm gold nanoparticles.

Abstract In Poland, rabbit is a highly valued animal, due to dietetic and flavour values of its meat, but above all, rabbits tend to be commonly used laboratory animals. The aim of the study was developing standards for counts of B-cells with CD19+ receptor, T-cells with CD5+ receptor, and their subpopulations, namely T-cells with CD4+, CD8+ and CD25+ receptor in the peripheral blood of mixed-breed Polish rabbits with addition of blood of meet breeds, including the assessment of the impact of four seasons of the year and animal sex on the values of the immunological parameters determined. The results showed that the counts of B- and T-cells and their subpopulations in peripheral blood remain within the following ranges: for CD19+ B-cells: 1.05 - 3.05%, for CD5+ T-cells: 34.00 - 43.07%, CD4+ T-cells: 23.52 - 33.23%, CD8+ T-cells: 12.55 - 17.30%, whereas for CD25+ T-cells: 0.72 - 2.81%. As it comes to the season of the year, it was observed that it principally affects the values of CD25+ T-cells, while in the case of rabbit sex, more changes were found in females.

In this work, we studied the pharmacological activity of zinc and magnesium salts of 2-aminoethanesulfonic acid in white non-linear male rats with amyotrophic lateral sclerosis, which was modeled by neurotoxicantsimplication into the pelvic part of spinal cord. After the reproduction of the pathology in animals, the indices of motor activity were recorded in the Rotarod test, and morphological studies of spinal cord sections stained according to Nisl in the Belshovsky modification were carried out. It was shown that the magnesium salt of 2-aminoethanesulfonic acid (compound LHT-317) to a greater extent reduces the development of motor disorders in experimental animals compared with the control group on the 4th day of observation. The course of intravenous administration of the studied compounds of 2-aminoethanesulfonic acid did not inhibit morphological changes in the spinal cord that develop in degenerative-dystrophic pathology of the central nervous system: connections. Moreover, if, against the background of treatment with zinc salt, the total area of motor zones in animals of the experimental group exceeded that of control rats, then the number of motoneurons did not differ from the control.

Background: The liver disease problem prompts investigators to search for new methods of liver treatment. Introduction: Silymarin (Sil) protects the liver by reducing the concentration of free radicals and the extent of damage to the cell membranes. A particularly interesting method to increase the bioavailability of Sil is to use synthesized gold nanoparticles (AuNPs) as reagents. The study considered whether it was possible to use the silymarin-AuNP conjugate as a potential liver-protecting drug. Method: AuNPs were conjugated to Sil and examine the liver-protecting activity of the conjugate. Experimental hepatitis and hepatocyte cytolysis after carbon tetrachloride actionwere used as a model system, and the experiments were conducted on laboratory animals. Result: For the first time, silymarin was conjugated to colloidal gold nanoparticles (AuNPs). Electron microscopy showed that the resultant preparations were monodisperse and that the mean conjugate diameter was 18–30 nm ± 0.5 nm (mean diameter of the native nanoparticles, 15 ± 0.5 nm). In experimental hepatitis in mice, conjugate administration interfered with glutathione depletion in hepatocytes in response to carbon tetrachloride was conducive to an increase in energy metabolism, and stimulated the monocyte–macrophage function of the liver. The results were confirmed by the high respiratory activity of the hepatocytes in cell culture. Conclusion: We conclude that the silymarin-AuNP conjugate holds promise as a liver-protecting agent in acute liver disease caused by carbon tetrachloride poisoning.
