Paracetamol (PCM) is a well-known drug widely used for its analgesic and antipyretic properties. PCM is generally considered as safe but overdose of PCM can cause nephrotoxicity. Traditionally, herbs have been used for the treatment of drug or toxin-induced renal disorders and numerous medicinal plants were tested for nephroprotection effect in PCM-induced nephrotoxicity model. The aim of the present study was to evaluate the protective effect of the herbal extract Eurycoma longifolia (EL) against PCM-induced nephrotoxicity rat model. Forty Wistar rats were randomly divided into five groups of eight rats each: control (vehicle 10 ml/kg), PCM alone (200 mg/kg PCM), EL 100 (EL 100 mg/kg+200 mg/kg PCM), EL 200 (EL 200 mg/kg+200 mg/kg PCM), and EL 400 (EL 400 mg/kg+200 mg/kg PCM). All animals from control group received vehicle daily and animals from groups PCM alone, EL 100, EL 200, and EL 400 received repeated dose of PCM and the assigned treatment of EL daily for a period of 14 days. On the 15th day, serum creatinine, blood urea nitrogen, protein, and albumin were measured in blood and creatinine clearance was measured in urine collected over 24 hours. Kidney sections of all experimental groups underwent histopathological analysis. There was a significant (p<0.05) increase in serum creatinine and blood urea levels in the PCM alone group compared to the treatment groups due to nephrotoxicity. In the treatment groups, there was a dose-dependent protection against PCM-induced changes observed in serum total protein, albumin, urea, and creatinine. Significant (p<0.05) drop was seen in serum creatinine and blood urea content in EL 200 and EL 400 groups. Creatinine clearance significantly increased for EL 200 (p<0.01) and EL 400 (p < 0.001) groups. Serum total protein and serum albumin content were significantly increased (p<0.05) in EL 200 and EL 400 groups compared to PCM alone group. Histopathological examination (H&E staining) of the rat kidneys revealed severe degeneration in the PCM alone group, while there was evidence of significant dose-dependent protection in the treatment groups against PCM-induced changes. The serum and urine biochemical results and histopathology analysis of the kidney indicate the nephroprotective potential of EL extract against PCM-induced nephrotoxicity.

AbstractObjectivesThe main objective of the present study is to evaluate the mitigative effect of hydroalcoholic extract of Momordica cymbalaria fruits against sodium fluoride (NaF) induced hepatotoxicity.MethodsIn this study, Wistar male albino rats were randomly divided into five groups of six rats each. Group I and II served as normal and toxic controls. Group III as plant control received extract at a dose of 400 mg/kg b. wt, p.o and Groups IV and V as treatment groups received extract at a dose 200 and 400 mg/kg b. wt, p.o for 30 days. All groups except Groups I and III received 100 ppm of NaF through drinking water. After completion of the study, blood collected for the estimation of liver blood serum biomarkers such as aspartate aminotransferases (AST), alanine aminotransferases (ALT), alkaline Phosphatase (ALP), direct and total bilirubin, total protein and albumin. The liver tissue homogenate was for estimation of lipid peroxidation, catalase, and reduced glutathione levels.ResultsThe results showed that NaF intoxication caused elevation of liver blood serum levels and lipid peroxidation; decreased levels of serum total protein, albumin and liver reduced glutathione, and catalase observed. The treatment groups showed decreased elevated serum biomarkers (ALT, AST, and ALP), liver lipid peroxidation and increased serum total protein and albumin, liver reduced glutathione and catalase levels in a dose-dependent manner. Histopathological studies also further strongly supported for mitigative effects of the plant.ConclusionsIn conclusion, our findings of the study indicated that M. cymbalaria fruits were a potential drug candidate in the treatment of NaF induced hepatotoxicity.

Abstract Objectives This study examined the possible protective roles of exogenous glycine (Gly) and L-Arginine (l-Arg) against Diclofenac (DIC)-induced gastro-duodenal damage in rats. Methods Rats were divided into Group A (control), Group B (DIC group) and Groups C–F which were pre-treated for five days with Gly1 (250 mg/kg), Gly2 (500 mg/kg), l-Arg1 (200 mg/kg) and l-Arg2 (400 mg/kg), respectively, before co-treatment with DIC for another three days. Hematological, biochemical and histopathological analyses were then carried out. Results DIC produced significant (p<0.05) reduction in PCV (13.82%), Hb (46.58%), RBC (30.53%), serum total protein (32.72%), albumin (28.44%) and globulin (38.01%) along with significant (p<0.05) elevation of serum MPO activity (83.30%), when compared with control. In addition, DIC increased gastric H2O2 and MDA levels by 33.93 and 48.59%, respectively, while the duodenal levels of the same parameters increased by 19.43 and 85.56%, respectively. Moreover, SOD, GPx and GST activities in the DIC group were significantly (p<0.05) reduced in the stomach (21.12, 24.35 and 51.28%, respectively) and duodenum (30.59, 16.35 and 37.90%, respectively), compared to control. Treatment with Gly and l-Arg resulted in significant amelioration of the DIC-induced alterations although l-Arg produced better amelioration of RBC (29.78%), total protein (10.12%), albumin (9.93%) and MPO (65.01%), compared to the DIC group. The protective effects of both amino acids against oxidative stress parameters and histological lesions were largely similar. Conclusions The data from this study suggest that Gly or l-Arg prevented DIC-induced gastro-duodenal toxicity and might, therefore be useful in improving the therapeutic index of DIC.

<title>Abstract</title><sec><title>Objective</title><p>Evaluate the biological performance of titanium alloys grade IV under different surface treatments: sandblasting and double etching (Experimental surface 1; Exp1, NEODENT); surface with wettability increase (Experimental surface 2; Exp2, NEODENT) on response of preliminary differentiation and cell maturation.</p></sec><sec><title>Material and method</title><p>Immortalized osteoblast cells were plated on Exp1 and Exp2 titanium discs. The polystyrene plate surface without disc was used as control group (C). Cell viability was assessed by measuring mitochondrial activity (MTT) at 4 and 24 h (n = 5), cell attachment was performed using trypan blue exclusion within 4 hours (n = 5), serum total protein and alkaline phosphatase normalization was performed at 4, 7 and 14 days (n = 5). Data were analyzed using one-way ANOVA and Tukey test.</p></sec><sec><title>Result</title><p>The values of cell viability were: 4h: C– 0.32±0.01<sup>A</sup>; Exp1– 0.34±0.08<sup>A</sup>; Exp2– 0.29±0.03<sup>A</sup>. 24h: C– 0.43±0.02<sup>A</sup>; Exp1– 0.39±0.01<sup>A</sup>; Exp2– 0.37±0.03<sup>A</sup>. Lipoprotein phospholipid (PL) plays a major role in neutralization of endotoxin. This study tested the hypothesis that prophylactic administration of a PL-enriched emulsion (PRE), which augments PL content of serum lipoproteins and neutralizes endotoxin in vitro, would preserve cardiovascular function and improve survival in porcine septic peritonitis. A control group was compared with low-, mid-, and high-dose treatment groups that received PRE by primed continuous infusion for 48 h. A fibrin clot containing live Escherichia coli 0111.B4 was implanted intraperitoneally 30 min after the priming dose. Survival increased in a dose-dependent manner and was correlated with serum PL. Infused PL was associated with high-density lipoprotein in the low-dose group and all serum lipoproteins at higher doses. Treatment significantly lowered serum endotoxin and tumor necrosis factor (TNF)-α, preserved cardiac output and ejection fraction, and attenuated increases in systemic and pulmonary vascular resistances. This study demonstrated that augmentation of lipoprotein PL via administration of PRE improved survival and offered a novel therapeutic approach to sepsis.

AbstractIntroductionXylazine, a type of α2-adrenoceptors, is a commonly used drug in veterinary medicine. Xylazine-induced changes in the content of amino acid neurotransmitters – glycine (Gly) and aspartic acid (Asp), in different brain regions and neurons were studied.Material and MethodsWistar rats were administered 50 mg/kg or 70 mg/kg of xylazine by intraperitoneal injection. In addition, in vitro experiments were conducted, in which neurons were treated with 15 μg/mL, 25 μg/mL, 35μg/mL, and 45 μg/mL of xylazine. Test methods were based on the enzyme-linked immunosorbent assays (ELISA).ResultsDuring anaesthesia, Asp levels in each brain area were significantly lower compared to the control group. Except for the cerebrum, levels of Gly in other brain areas were significantly increased during the anaesthesia period. In vitro, xylazine-related neuron secretion of Gly increased significantly compared to the control group at 60 min and 90 min. Moreover, xylazine caused a significant decrease in the levels of Asp secreted by neurons at 20 min, but gradually returned to the level of the control group.ConclusionThe data showed that during anaesthesia the overall levels of Asp decreased and overall levels of Gly increased. In addition, the inhibitory effect of xylazine on Asp and the promotion of Gly were dose-dependent. Our data showed that different effects of xylazine on excitatory and inhibitory neurotransmitters provided a theoretical basis for the mechanism of xylazine activity in clinical anaesthesia.

Background and Purpose: Shigella is a human shigellosis and its lipopolysaccharide is identified by 4TLR. The 4TLR is a family of pseudo-TOLL receptors and many immune routes are triggered by stimulating these receptors. Many studies show increasing of 4TLR expression in Mesenchyme stem cells under the influence of lipopolysaccharide. The main objective of this study was to identify the appropriate lipopolysaccharide of Shigella strains by stimulating the immune system for vaccine studies. Materials and Methods: In this experimental study, the stem cell of human Mesenchymal derived from bone marrow was treated by three dilution of 0.1, 0.01, and 0.001 extract of Shigella strains (Flexneri, Dysentery and Sonnei) containing lipopolysaccharide. Then, the expression of 4TLR at RNA level was evaluated by RT-PCR and Q-PCR techniques. Cells treated with phosphate buffer saline were considered as control group. Findings: The expression of 4TLR was observed in all treatments groups except for treatment groups with relative concentration of 0.001 sonnei and dysentery as well as control group. Changes in 4TLR expression were dose-dependent on all treatment groups. The highest expression was related to the treatment with Shigella Flexneri extract and the smallest was related to Shigella sonnei. The use of pure lipopolysaccharide of Escherichia coli as a positive control showed that the lipopolysaccharide in Shigella extract is responsible for increasing the expression of 4TLR. Conclusion: given the increased expression of 4TLR by Shigella extract, this extract is recommended to increase the efficacy of the vaccine.

Terminalia Chebula extract is used for regeneration of hepatic cells and protection of liver against damage due to its active component. This study aims to observe the protective effect of Terminalia Chebula against Paracetamol induced change of serum total protein level in Wister Albino rats. The study was carried out in the Department of Physiology, Dhaka Medical College (DMC) during January 2013 to December 2013. A total number of 44 rats, age ranging from 90 to 120 days, weight between 150 to 200 gm (initial body weight) were selected for the study. After acclimatization for 14 days, they were divided into control groups and experimental groups. Before sacrifice, final body weights of all the rats were measured. then all the rats were sacrificed on 22nd day and then blood samples were collected. For assessment of liver function, serum total protein level was done by using standard laboratory kits. The mean serum total protein level was significantly (p<0.001) lower in paracetamol treated control group in comparison to those of baseline control group. Serum total protein level of all experimental groups were significantly (P<0.001) higher than Paracetamol treated control group. From the results of this study, it may be concluded that Terminalia Chebula may have some protective effect against Paracetamol induced liver damage in rats.Bangladesh Med J. 2017 Jan; 46 (1): 11-14

<p class="Abstract">The diabetic patients (n=109) with mild or moderate renal insufficiency underwent coronary intervention were randomly divided into control group and observation group. The patients in observation group were treated with injection Shenkang and alprostadil before operation. The results showed that the level of serum creatinine, creatinine clearance and blood urea nitrogen had no significant difference (p&gt;0.05) before and postoperative day 1 and day 2 in both groups. However, the value of serum creatinine, creatinine clearance and blood urea nitrogen were significantly different on post-operative day 3 (p&lt;0.05). The incidence of contrast-induced nephropathy was 14.8% in control group, and markedly higher than observation group (3.6%) (p&lt;0.05). Shenkang injection combined with alprostadil injection decreased the incidence of contrast-induced nephropathy in elderly patients with diabetes mellitus complicated with mild to moderate renal insufficiency.</p>

There is accumulating evidence suggesting that depression is a risk factor for cardiovascular diseases. This study aimed to examine the hypothesis that the proinflammatory cytokine TNF-α would partially explain the link between depression and atherosclerotic endothelial changes. Rats were distributed among 6 groups: (i) control group; (ii) group subjected to chronic mild stress (CMS); (iii) group fed a cholesterol–cholic acid–thiouracil (CCT diet); and (iv) CMS group fed the CCT diet and treated with the vehicle for 8 weeks. The last 2 groups were subjected to CMS–CCT and received thalidomide (THAL) or imipramine (IMIP). Rats were assessed behaviorally (sucrose preference, open field, and forced-swimming tests). TNF-α protein was assessed from the serum, aorta, and liver. Aortic TNF-α gene expression (assessed using RT–PCR), serum lipid profile, and insulin levels were measured. Endothelial function was assessed in isolated aortic rings. The THAL and IMIP groups showed ameliorated CMS–CCT-related behavioral changes. CMS–CCT-induced metabolic and endothelial dysfunctions were improved in the THAL group but were worsened in the IMIP group. RT–PCR showed a significant reduction of aortic TNF-α mRNA expression in the THAL and IMIP treatment groups. These data paralleled the findings for aortic immunohistochemistry. The THAL group, but not the IMIP group, showed improved CMS–CCT-induced changes in the vascular reactivity of the aortic rings. Thus, TNF-α provides a target link between depression, metabolic syndrome, and endothelial dysfunction. This could open a new therapeutic approach to address the comorbidities of depression.

AbstractIntroduction: The aim of this study was to determine the effect of deoxynivalenol (DON), given alone or with bentonite (which eliminates mycotoxicity) in the diet of mink dams throughout mating, pregnancy, and lactation period to pelt harvesting, on the mechanical properties and geometry of their long bones.Material and Methods: The minks were randomly assigned into two groups: a control group (not supplemented with DON, n = 15) and a group fed naturally DON-contaminated wheat and divided into three sub-groups (each sub-group n = 15), depending on bentonite dose: 0 M – sub-group fed naturally DON-contaminated wheat at a concentration of 3.7 mg kg−1 alone; 2 M – sub-group fed naturally DON-contaminated wheat at a concentration of 3.7 mg kg−1 and bentonite at a concentration of 2 kg 1000 kg−1; 0.5 M – sub-group fed naturally DON-contaminated wheat at a concentration of 3.7 mg kg−1 and bentonite at a concentration of 0.5 kg 1000 kg−1.Results: The DON treatment reduced the length of the femur compared to the control group and reduced the bone weight dependently on the amount of bentonite supplementation. However, DON treatment reduced the MRWT and CI of the femur, irrespective of the bentonite supplementation, compared to the control. The total BTD and BMC decreased in all DON-treated groups (irrespective of the bentonite supplementation). Furthermore, the densitometric analysis showed that the main changes in BMD and BMC indicated bone loss in the proximal and distal parts of bone covering the trabecular bone; whereas when bentonite was given at the dose of 2 kg 1000 kg−1 an increase in the whole BMD and BMC was observed in the femoral midshaft.Conclusion: Analysis of the geometrical parameters seems to indicate that endosteal resorption was delayed after bentonite supplementation. The addition of bentonite diminished the DON action on bone homeostasis in the mink dams. Thus bentonite could prevent DON-induced bone loss in a dose-dependent manner.
