Subacute administration of tributyltin chloride modulates neurotransmitters and their metabolites in discrete brain regions of maternal mice and their F1 offspring

Tributyltin (TBT) compounds have been used as anti-fouling agents and the central nervous system is one of its target organs. TBT-induced modulations of neurotransmitters in the brains of adult mice have been reported. However, little is known about the developmental neurotoxicity of TBT. In this study, we evaluated the effects of TBT on neurotransmitters and their metabolites in discrete brain regions of female ICR mice and their offspring. Pregnant ICR mice were exposed to TBT chloride at concentrations of 0, 15 or 50 ppm in water or 125 ppm in food. Male offspring were sacrificed at one, two and three weeks after birth. The concentrations of norepinephrine, dopamine (DA), dihydoxyphenylacetic acid, homovanillic acid (HVA), serotonin (5-HT), and 5-hydroxyindoleacetic acid (5-HIAA) were determined in different brain regions by HPLC. All offspring from the 125 ppm group died immediately after birth. A significant decrease in the body weight of the TBT-treated F1 groups compared to the control group was observed in the first week. Significant increases compared to the controls were observed for the DA concentration in the striatum of the 50 ppm F1 group, and for the HVA concentration in the cerebrum and the 5-HT concentration in the medulla oblongata of the 15 and 50 ppm F1 groups in the third week. At three weeks of age, the neurotransmitters and their metabolites may be useful indexes for developmental neurotoxicity. For the dams, a significant decrease in the 5-HT concentration was observed in the cerebellum, medulla, midbrain and striatum of the 125 ppm group compared to the control group. A significant decrease in the 5-HIAA concentration was also observed in the cerebellum, midbrain and striatum of the dams in the 125 ppm group compared to the control. TBT may induce a decrease in the synthesis of 5-HT in the dams. The discrepancy between dams and offspring may be due to several factors such as age, dose, route, sex and pregnancy.

Patients with fibromyalgia (FM) show widespread pain associated with other symptoms such as cognitive problems, depression, and anxiety among others associated with alterations in the central nervous system. The hippocampal subfields had differences in function, histology, and connectivity with other brain regions, and are altered in different diseases. This study evaluates the volumetric differences between patients with FM compared with a healthy control group. A total of 49 women with, and 43 healthy women completed this study. T1-weighted MRI was used to assess brain volume, and FreeSurfer software was used to segment the hippocampal subfields. Women with FM had a significant reduction in most of the hippocampal subfields. The regression equation models were obtained to predict the volume of specific subfields of the right and left hippocampus. These findings provide that women with FM have lower hippocampal subfields volumes compared with healthy women. Besides, regression models show that different covariates, such as age, cognitive impairment, or depression, are related to specific subfields.

Objective: Potash known as potassium carbonate (K2CO3) is a mixture of salt with other components, including impurities which coexist in mineral and salt is highly consumed in various forms by pregnant women. The aim of this research is to determine the effect of potash on the weight index of pregnant Wistar rats. Methods: A total of 25 albino Wistar rat with weights ranging from 180 to 300 g were used and allocated into five groups of five animals each (four females and one male) designated as Groups A, B, C, D, and E. The experimental Groups B, C, D, and E were administered through oral route different doses of potash of 300 mg/kg, 600 mg/kg, 900 mg/kg, and 1200 mg/kg, respectively, after pregnancy was detected by checking for mucus plug in the vagina. Group A served as the control group and was administered distilled water only. The animals were allowed for 1 week for acclimatization under normal temperature (270–300°C), which they were being fed with normal feed (grower’s mash) and water ad libitum for 1 week. Results: The result showed a significant (p˂0.05) reduction in weight with the highest level seen with the 1200 mg/kg group when compared to the control. Conclusion: Therefore, the effect of potash alters the physical activity and decreases weight, by implication may induce growth retardation of the Wistar rats which is not healthy for a pregnant animal.

Abstract Background Precocious puberty is frequently associated with obesity, which will lead to long-term effects, especially on growth and reproduction. However, the effect of precocious puberty on children's neurodevelopment is still unknown. Objectives Here we evaluated the effect of High fat diet (HFD)-induced precocious puberty on neurodevelopment and behaviors of animals. Methods Ovaries sections were stained with hematoxylin–eosin (H&E) using standard techniques. Behavioral tests included elevated plus maze (EPM), open field exploration, Y-Maze, marble burying test, and novelty- suppressed feeding. The expression of genes related to puberty and neural development was detected by immunohistochemistry and Western blot. Results Our results showed HFD-induced precocious puberty increased the risk-taking behavior and decreased memory of mice. The content of Tyrosine hydroxylase (TH) and Arginine vasopressin (AVP) in hypothalamus were higher in HFD group than control group. Although the recovery of normal diet will gradually restore the body fat and other physiological index of mice, the anxiety increases in adult mice, and the memory is also damaged. Conclusions These findings describe the sensitivity of mice brain to HFD-induced precocious puberty and the irrecoverability of neural damage caused by precocious puberty. Therefore, avoiding HFD in childhood is important to prevent precocious puberty and neurodevelopmental impairment in mice. Graphic abstract

AbstractIntroduction:Tiletamine-xylazine-tramadol (XFM) has few side effects and can provide good sedation and analgesia. Adenosine 5’-monophosphate-activated protein kinase (AMPK) can attenuate trigeminal neuralgia. The study aimed to investigate the effects of XFM and its specific antagonist on AMPK in different regions of the brain.Material and Methods:A model of XFM in the rat was established. A total of 72 Sprague Dawley (SD) rats were randomly divided into three equally sized groups: XFM anaesthesia (M group), antagonist (W group), and XFM with antagonist interactive groups (MW group). Eighteen SD rats were in the control group and were injected intraperitoneally with saline (C group). The rats were sacrificed and the cerebral cortex, cerebellum, hippocampus, thalamus, and brain stem were immediately separated, in order to detect AMPKα mRNA expression by quantitative PCR.Results:XFM was able to increase the mRNA expression of AMPKα1 and AMPKα2 in all brain regions, and the antagonist caused the opposite effect, although the effects of XFM could not be completely reversed in some areas.Conclusion:XFM can influence the expression of AMPK in the central nervous system of the rat, which can provide a reference for the future development of anaesthetics for animals.

Background: Sthoulya is Medovaha Strotodushtijanya vyadhi, which includes abnormal and excessive accumulation of Medodhatu in the body. This is caused by lack of physical and mental activity, daytime sleep, excessive intake of madhur (sweet), snigdha ahar (oily diet) results in  increase Kaphadosha and meda which results in Sthoulya (overweight) having symptoms of mild dysponea, thirst, drowsiness, excess sleep, appetite, offensive smell from the body, incapability to work and incapability to participate in sexual intercourse. Aim: Comparative clinical efficacy of Tryushanadi Guggul and Navaka Guggul in Sthoulya (overweight). Materials and methods: Total 60 patients of Sthoulya will enrolled and will divided into two groups (each group contains 30). Patients in group A (experimental group) will be given 1 gram Tryushanadi Guggul two times a day after meal with honey and in group B (control group)1 gram Navaka Guggul will administered two times a day after meal with honey for 30 days. Dietary changes and walking (30 minutes) will be advised to patients of both groups. Follow up will be taken on 15th day and 30th day. Assessment of subjective parameters like kshudrashwasa (exertional dyspnoea), swedadhikya (perspiration), atikshudha (increased appetite), nidradhikya (increased sleep) and objective parameters like body weight, B.M.I., mid arm circumference, waist-hip ratio and lipid profile will be done before and after treatment. Results: Subjective and objectives outcomes will be assessed by statistical analysis. Conclusion: It will be drawn from the result obtained.

The Timed Up and Go (TUG) test quantifies physical mobility by measuring the total performance time. In this study, we quantified the single TUG subcomponents and, for the first time, explored the effects of gait cycle and pelvis asymmetries on them. Transfemoral (TF) and transtibial (TT) amputees were compared with a control group. A single wearable inertial sensor, applied to the back, captured kinematic data from the body and pelvis during the 10-m walk test and the TUG test. From these data, two categories of symmetry indexes (SI) were computed: One SI captured the differences between the antero-posterior accelerations of the two sides during the gait cycle, while another set of SI quantified the symmetry over the three-dimensional pelvis motions. Moreover, the total time of the TUG test, the time of each subcomponent, and the velocity of the turning subcomponents were measured. Only the TF amputees showed significant reductions in each SI category when compared to the controls. During the TUG test, the TF group showed a longer duration and velocity reduction mainly over the turning subtasks. However, for all the amputees there were significant correlations between the level of asymmetries and the velocity during the turning tasks. Overall, gait cycle and pelvis asymmetries had a specific detrimental effect on the turning performance instead of on linear walking.

Obesity is one of the most common complex health problem. The pathway of serotonin synthesis takes part in neuroendocrine regulation, as well as in the regulation of a number of behavioral functions of the body and fat deposition. Serotonin is a mediator of the amine nature, which functions as a neurotransmitter and tissue hormone. The greatest amount of serotonin is synthesized in the brain and 12 duodenum. As a neurotransmitter, serotonin affects both directly and indirectly on the function of most brain cells. Female hormone progesterone influence on serotonin functions. One of the effect of progesterone is increasing of amount of fat tissue during the pregnancy. Long-term using of progesterone in hormone substitution therapy or as part of contraception also lead to fat accumulation effect. The levels of activity of serotonergic system enzymes, tryptophan hydroxylase, tryptophan decarboxylase and monoamine oxidase (MAO), and tryptophan, 5-hydroxytryptophan, serotonin and 5-hydroxyindoleacetic acid concentrations in the rat brain under obesity conditions caused by prolonged administration of progesterone were determined in this study. Studies have shown that the content of tryptophan, 5-hydroxytryptophan, serotonin and 5-hydroxyindoleacetic acid in the brain of rats under obesity caused by prolonged administration of progesterone increased in comparison with the rats of the control group. The levels of tryptophan hydroxylase and MAO activity decreased, and tryptophan decarboxylase activity levels increased in the rat brain under obesity conditions caused by prolonged administration of progesterone. Thus, as a result of our studies, we found an imbalance in the system of serotonin metabolism in the brain of rats with the development of hormonal obesity induced by prolonged administration of progesterone, which may indicate the involvement of the serotonergic neurotransmitter system in the mechanisms of the development of obesity and concomitant diseases.

Effects of neem, betel leaf, devil’s tree, jute and turmeric against natural gastrointestinal nematodes in sheep and on some hematological parameters (TEC, Hb and PCV) and body weight were studied. Thirty (30) naturally parasitized sheep were randomly divided into six groups(A, B, C, D, E and F), each consisting of five sheep. Ten percent water extract of leaves of neem, betel leaf, devil’s tree and jute were administered orally to the sheep of group A, B, C and D, respectively. Sheep of group E was treated orally with 10% water extract of rhizome of turmeric. Sheep of group F was kept as infected control group. Fecal samples, hematological parameters and body weight were examined before treatment and on 7th, 14th, 21st and 28th day. A significant (p<0.01) reduction of EPG count was found following administration of neem (37.60-47.03%), betel leaf (6.43-14.00%), devil’s tree (3.04-11.04%), jute (0.50-5.26%) and turmeric (0.46-8.30) in sheep. The EPG count of the control group (F) were significantly (p<0.01) increased up to the last day of experimental period. After treatment with neem, betel leaf, devil’s tree, jute and turmeric total erythrocyte count (TEC), hemoglobin (Hb) content and packed cell volume (PCV) were increased significantly (p<0.01 and p<0.05) in sheep. The body weight was increased significantly (p<0.01 and p<0.05) in neem, betel leaf, devil’s tree, jute and turmeric treated sheep. On the other hand, body weight was decreased in untreated control group. The present study reveal that 10% water extract of neem was moderately effective and betel leaf, devil’s tree, jute and turmeric were relatively less effective against gastrointestinal nematodes in sheep. Background: Stroke is a condition in which Spasticity in the body musculature greatly affect the functional independence of the patients. Hold- Relax and Static Stretching is one of the useful treatment to reduce Spasticity. Aim: To find out the effect of Hold – Relax V/S Static Stretching on Elbow flexors muscle Spasticity in Stroke Patient Materials and Methods: 20 subjects were randomly allocated into two groups (Group A and Group B). For 10 subjects in Group A Control group was given Static Stretching with conventional rehab and Group B Experimental group was given Hold- Relax with conventional rehab. Treatment were given to 3 weeks 3 sets per sessions. The outcome measure used to assess the Spasticity before and after the intervention was Modified Ashworth Scale (MAS). Results and Discussion: The Hold-Relax group showed a significant reduction in Spasticity of Elbow flexors muscle, compared to the control group (p<0.05). Conclusions: Hold- Relax is more effective than Static Stretching. Key words: Stroke, Spasticity, Hold- Relax, Static Stretching, MAS.

Цель исследования - оценка возможности использования интегральных гематологических индексов для выбора оптимальной модели ушивания послеоперационных ран в эксперименте. Методика. Исследование проводили на 297 белых беспородных крысах-самцах массой 190-210 г, которые были разделены на 3 группы, по 99 крыс в каждой. Одна группа являлась контрольной, у второй группы крыс был смоделирован раневой процесс с натяжением брюшной стенки; у третьей группы животных - модель раневого процесса с применением трансплантата нанокомпозитного материала. Определяли следующие интегральные гематологические индексы: индекс Кребса; коэффициент Бредекка; лейкоцитарный индекс; индекс соотношения нейтрофилов и моноцитов; индекс соотношения лимфоцитов и моноцитов; индекс соотношения лимфоцитов и эозинофилов; коэффициент отношения альбуминов к глобулинам; оксипролиновый коэффициент. Результаты. Установлены статистически значимые колебания показателей интегральных гематологических индексов в динамике заживления послеоперационной раны у крыс. Заключение. На основе полученных результатов был сделан вывод, что особенности динамики интегральных гематологических индексов, выявленные на различных моделях раневого процесса, отражают неспецифические и специфические защитные реакции организма в целом, позволяют делать прогноз дальнейшего течения и исхода раневого процесса, а также обеспечивают возможность выбора оптимальной модели ушивания послеоперационной раны. The article presents results of predicting the wound healing process using integral hematological indexes. The aim of the study was to describe the dynamics of integral hematological indexes of wound healing process in rats to predict its outcome. The study included 297 white mongrel male rats weighing 190-210 g, which were divided into three groups, 99 rats in each. One group was a control group; in the second group, a wound healing process with abdominal wall tension was modeled; in the third group, a wound healing process with a graft was modeled. The following integrated hematological indexes were determined: Krebs index; Bredeck index; leukocyte index; neutrophil/monocyte ratio; lymphocyte/monocyte ratio; lymphocyte/eosinophil ratio; albumin/globulin ratio; and oxyproline coefficient. The study showed that changes in values of integral hematological indexes were mostly statistically and clinically significant. We concluded that changes in integral hematological indexes in different models of wound healing reflect nonspecific and specific protective responses of the body as a whole and allow to predict further course and outcome of the wound healing process.
