BACKGROUND Numerous studies have revealed that aerobic exercise (AE) or environmental enrichment (EE) exert positive effects on the molecular, cellular, and structural changes responsible for functional plasticity. Therefore, this study was aimed to investigate whether the combination of AE and EE yields a greater effect on spatial memory and increases the expression of the adhesion molecule neuroligin 1 compared with either of the therapies alone. METHODS Twenty 6-month-old male Wistar rats were randomly divided into four groups: (1) the control group (C), (2) the AE group, (3) the EE group, and (4) the AE-EE group. Escape latency in a Water-E-maze (WEM) task was used as a parameter to assess spatial memory function. Neuroligin 1 protein expression was examined via the immunohistochemistry (IHC) technique followed by IHC optical density (OD) score analysis of the CA1 hippocampal region. RESULTS Data acquired from the WEM task show that escape latency progressively decreased over time in all groups. However, animals in the AE-EE group required less time to complete the task compared with those in the control, AE, and EE groups. IHC OD scores revealed that the combination of AE and EE caused the highest expression of neuroligin 1 in the CA1 hippocampal region among the therapies studied. Statistical analysis indicated significant differences in OD score between the AE-EE and control, AE, and EE groups (p ≤ 0.001). CONCLUSIONS The combination of AE and EE rats increased neuroligin 1 expression and improved the spatial memory of male Wistar rats compared with application of either therapy alone.

Memory is an important cognitive function in humans. Exercise and environmental enrichment (EE) exposure have positive effects on memory function via improved neurogenesis through expression of growth factors such as brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF). Exercise and environmental enrichment have independently been shown to increase BDNF and NGF, but the effect of the combination of these treatments has not been widely studied. This experimental study aims to analyse the effect of aerobic exercise, EE exposure, and combination of aerobic exercise and EE exposure on memory function. This study used twenty 7-month old male Wistar rats that were given treatment for 8 weeks. Memory function was tested using forced alternation Y-maze. Hippocampal expression of BDNF and NGF were also assessed. The results showed the combination group has highest performance in memory function test and also the highest level of hippocampal BDNF and NGF (P<0.05). It can be concluded that the combination of aerobic exercise and continuous EE exposure produces the best results for memory function through higher levels of hippocampal BDNF and NGF in adult rats.

Abstract Purpose Information on monosodium glutamate (MSG) safety in the eye-brain axis remains scarce. The objective of the study was to establish changes in calcium (Ca2+) concentrations, catalase activity, Malondialdehyde (MDA) levels and identify any major histological changes associated with parenteral and feed supplementation of MSG in male Wistar rats. This is important to guide policy on human consumption of food additives.Methods Thirty adult male Wistar rats were divided into 5 groups (n = 6) for a period of one month. Group I and IV were treated with MSG 6g/kg and 60 mg/kg subcutaneous, while groups II and III at 5% and 2% MSG feed supplement. Group V was the control group treated with normal saline (no MSG). The eye and brain were analyzed for Ca2+, catalase, MDA levels and histological analysis was done using Hematoxylin and Eosin staining.Results Calcium levels in the eye was reduced at high (6g/kg/day and 5% s.c) MSG treatments while in the brain these remained constant. Since Ca2+ plays a crucial role in catalase activity, levels of catalase were not affected in eye-brain axis. MDA levels were elevated at high MSG treatments and no structural changes in the retina of the eye and no damage were found in the brain.Conclusion MSG maintained catalase levels, Ca2+ and no structural changes were observed in the eye and brain.

BACKGROUND: Complex aerobic exercise is believed to induce positive effects on neuron structure and cognitive function. Long-term and continual cognitive stimulation increases neuroplasticity by stimulating the synthesis of neuronal growth proteins and the formation of new synapses. Exercise also increases the ability of neurons to survive and improves brain vascularization. Further investigations should be conducted to explore what types of aerobic exercise are beneficial for cognitive function. AIM: This study investigated the effects of hurdle aerobic exercise on developmentally regulated brain protein-A (Drebrin-A) as a neuroplasticity indicator, and on vascular endothelial growth factor (VEGF) as an angiogenesis marker in the hippocampus. METHODS: This study was an experimental study with post-test only control group design. Thirty-three adult mice were divided into control, hurdle aerobic runner (HAR), and plain aerobic runner (PAR) groups (n = 11 for each group). Fiberglass running wheels were originally designed and modified to assemble hurdles inside with adjustable speed. Speed adaptation was intended to achieve aerobic intensity. The experiment was performed 5 times a week for 8 weeks. The Morris water maze test (MWMT) was used to assess spatial memory ability. One day after the last running exercise and final MWMT, the mice were sacrificed and the right side of the hippocampus was obtained for Drebrin-A analysis by enzyme-linked immunosorbent assay (ELISA). The entire right side brain tissue after hippocampus was removed then used for the neuroglobin ELISA assay. To analyze VEGF expression and calculation of blood vessel, the left side of the brain was prepared for hematoxylin eosin and immunohistochemistry staining. To assess the effect of exercise on vascular widening, the analysis of the slides was performed by calculating the percentage of blood vessels with diameters more than 15 μm. One-way ANOVA and Fisher’s least significant difference test was used for statistical analysis. RESULTS: There was a significant difference in the levels of Drebrin-A between the HAR and PAR groups. Both exercise groups had higher levels of Drebrin-A than the control group. HAR and PAR groups exhibited significantly higher percentages of blood vessels expressing VEGF in hippocampus compared to control. HAR and PAR groups had the higher percentages of larger vessels compare to control. There was no significant difference of neuroglobin levels among the three groups. Both the HAR and PAR groups exhibited better spatial memory than the control group. CONCLUSION: Both aerobic exercises induced positive effects on brain angiogenesis, while the intensity of aerobic exercises did not result in high hypoxic stress in the brain.

BACKGROUND: Exposure to environmental enrichment has a positive effect on brain function, including improved cognition. Environmental enrichment has many aspects, including social interactions, object stimulations, and physical activities. Exercise and environmental enrichment can be considered to improve cognitive function with different underlying mechanisms. This study aims to compare the effects of environmental enrichment and aerobic exercise at both synaptic and whole-organism levels using synapyophysin as a measure of synaptic physiology and spatial memory as a measure of cognitive function.METHODS: A six-week in vivo experimental study on 15, 6-month old male Wistar rats randomly divided into three groups (n=5): aerobic group (A), enriched environment group (EE), and enriched with an aerobic or combined group (EEA). All rats were tested four times in the Water-E maze (WEM) task at weeks 0, 2, 4, and 6 of the study. We used immunohistochemistry to determine the synaptophysin expression in hippocampal CA1 region.RESULTS: Based on synaptophysin immunostaining, there were higher optical density scores for synaptophysin in hippocampal CA1 region following EEA, but there were no statistically significant differences between groups (ANOVA test, p>0.05). The spatial memory test showed there were significantly reduced travel time and total errors from the 2nd and 4th weeks in the EEA group, respectively (p<0.05).CONCLUSION: The combination of enriched environment and aerobic exercise seems to rapidly improve spatial memory and enhances the presynaptic protein, synaptophysin in hippocampal CA1 region.KEYWORDS: aerobic exercise, environmental enrichment, spatial memory, synaptophysin, Water-E maze

Background and aims: Irisin myokine whose secretion is induced by exercise is associated with an increase in thermogenesis. However, the effect of aerobic exercise with different intensities on the production and release of irisin in overweight individuals is controversial. This study aimed to investigate the effect of eight weeks of aerobic training with different intensities on PGC-1α, FNDC5, UCP1, and irisin in obese male Wistar rats. Methods: In this experimental study, 24 adult obese male Wistar rats (weight: 250-300 g; body mass index [BMI] >30 g/cm2) divided into three groups, including aerobic training with 28 m/min (moderate intensity [MI]), aerobic training with 34 m/min (high intensity [HI]), and control. All training groups exercised for eight weeks walking on a treadmill (five 60-min sessions per week). The paired sample t test and one-way ANOVA were used to determine the intra- and inter-group differences. Furthermore, Tukey’s post hoc test was used. Results: The levels of PGC-1α in muscle tissue in aerobic training groups with MI and HI increased (P=0.001 and F=11.81) compared to control group. The levels of irisin (P=0.006 and F=6.10) and UCP1 (P=0.04) were significantly increased in MI group. On the other hand, FNDC5 increased significantly in both MI and HI groups (P=0.001 and F=12.49). While irisin and UCP1 levels increased in HI group compared to the control group, the change was not statistically significant (P>0.05). The levels of irisin, UCP1, and FNDC5 increased more significantly in MI group than in HI group (P<0.05). Conclusion: Both types of aerobic training (MI and HI) had a beneficial effect on changes in irisin, UCP1, FNDC5, and PGC-1α levels. Accordingly, moderate exercise possibly changes the phenotype of white to brown adipose tissue leading to an increase in thermogenesis, body weight loss, and sensitivity to insulin.

BACKGROUND Memory is a vital function of the brain. Aerobic exercise has a positive effect on memory’s function, but the appropriate combination of intensity and duration of aerobic exercise is still unknown. This study was aimed to investigate the effect of optimum combinations of intensity and duration of aerobic exercise on spatial memory function.  METHODS In this study, the authors performed in vivo experiment using 20 male Wistar rats (6-month-old). They were randomly divided into four groups: (1) low-intensity and short duration aerobic exercise group (L–S); (2) low-intensity and long duration aerobic exercise group (L–L); (3) high-intensity and short duration aerobic exercise group (H– S); and (4) high-intensity and long duration aerobic exercise group (H–L). The aerobic exercise treatment of each group was conducted for three weeks with a frequency of five days a week. The memory function was assessed with the help of water-E-maze on week 0, 1, 2, and 3 (a total of four times).  RESULTS This study indicates that the central nervous system responds to aerobic exercise as an external stimulus differently depending on the combinations of intensity and duration. Moreover, this study demonstrates that changes in memory functions are best observed in the group with low-intensity and long duration aerobic exercise.  CONCLUSIONS The combination of low-intensity and long duration of aerobic exercise for animal study can improve spatial memory functions better than any other combinations of intensity and duration of aerobic exercises in male Wistar rats.

<p>Liver is an organ with complex metabolism. When the liver is inflamed, cellular immunity will defend against inflammatory agents by stimulating immune cells to produce reactive oxygen species (ROS). Excessive ROS accumulation cause oxydative stress with increased  liver malondialdehyde (MDA) level. Some researches showed that purple sweet potato contain flavonoids (anthocyanins) that functioned as antioxydants. This study aimed to show the prophylactic effect of purple sweet potato extract to the liver MDA level of male Wistar rats induced by carrageenan.</p><p>This study used post-only control group method using 18 male Wistar rats divided into 3 groups: group of rats without treatment, group of rats induced by 0,1 ml of 1% carrageenan by intraplantar injection on day-8, and group of rats given with 872 mg/kgBW of purple sweet potato extract for 7 days and induced by 0,1 ml of 1% carrageenan. In the end of the study, the liver MDA levels were measured by Thio-Barbituric Acid method on each groups.</p><p>The results of One-Way ANOVA test showed there was no significant difference (p = 0,290) between group of rats without treatment (<em>x̅</em>= 207,50) and group of rats induced by carrageenan (<em>x̅</em>=233,17). Then, there is no significant difference (p = 0.978) between group of rats induced by carrageenan and group of rats given with prophylactic purple sweet potato extract and induced by carrageenan (<em>x̅</em>= 232,50).</p><p>The conclusion of this study is giving intraplantar injection of carrageenan can increase liver MDA level insignificantly and giving prophylactic purple sweet potato extract has an effect to decrease the liver MDA level of rats induced by carragenan insignificantly because it contains anthocyanins as antioxidants.</p><p> </p><strong>Keywords: </strong>Liver, <em>Ipomoea batatas</em> L., Malondialdehyde, Anthocyanins

The aim of this study was to investigate the clinical efficacy of vanadium complexes on triglycerides (TG), total cholesterol (Chol), uric acid (UA), urea (U), and antioxidant parameters: nonenzymatic (FRAP—ferric reducing ability of plasma, and reduced glutathione—GSH) and enzymatic (glutathione peroxidase—GPx, catalase—CAT, and GPx/CAT ratio) activity in the plasma of healthy male Wistar rats. Three vanadium complexes: [VO(bpy)2]SO4⋅2H2O, [VO(4,4′Me2bpy)2]SO4⋅2H2O, and Na[VO(O2)2(bpy)]⋅8H2O are administered by gavage during 5 weeks in two different diets such as control (C) and high fatty (F) diets. Changes of biochemical and antioxidants parameters are measured in plasma. All three vanadium complexes statistically decrease the body mass growth in comparison to the control and fatty diet. In plasma GSH was statistically increased in all vanadium complexes-treated rats from control and fatty group in comparison to only control group. Calculated GPX/CAT ratio was the highest in the control group in comparison to others.

The present investigation was carried out to evaluate the hepatoprotective effect of probiotic fermented milk (FM) containing Lactobacillus rhamnosus GG and Lactobacillus casei strain Shirota, alone as well as in combination with chlorophyllin (CHL) as an antioxidant agent in male Wistar rats administered aflatoxin-B1 (AFB1). AFB1 was injected intraperitoneally at the rate of 450 μg/kg body weight per animal twice a week for 6 weeks, maintaining an equal time interval between the two consecutive AFB1 administrations. A total of 125 male Wistar rats were randomly allocated to five groups, each group having twenty-five animals. Group I was offered FM containing L. rhamnosus GG and L. casei strain Shirota. Group II was administered AFB1 and served as the control group; group III was administered FM-AFB1, in which besides administering AFB1, FM was also offered. Group IV was offered CHL and AFB1, and group V was offered both FM and CHL along with AFB1. The rats were euthanised at the 15th and 25th week of the experiment and examined for the biochemical and hepatopathological profile. A significant reduction in thiobarbituric acid-reactive substances (TBARS) was observed in the FM–CHL–AFB1 group compared with the AFB1 control group. FM alone or in combination with CHL was found to show a significant (P < 0·05) hepatoprotective effect by lowering the levels of TBARS and by enhancing the activities of antioxidant enzymes such as glutathione peroxidase, superoxide dismutase, catalase and glutathione-S-transferase, indicating that probiotic FM alone or in combination with CHL possesses a potent protective effect against AFB1-induced hepatic damage.

Background: The process of wound healing is influenced by various factors such as age, hormones, and wound care. Wound care is done to accelerate wound healing which can be done by various methods, one of them is traditional care. Traditional wound care can use medicinal plants. Rhodomyrtus tomentosa is a medicinal plant that has an antioxidant, anti-inflammatory, antitumor and antibacterial content. Thus this study aims to evaluate the effectiveness of the antiseptic solution of the Rodhomyrtus tomentosa leaf extract on wound healing in male Wistar rats. Method: this research is pure experimental research with post test only control group design. Thirty male white rats were divided into five groups, namely negative control, positive control, Rhodomyrtus tomentosa leaf extract 15%, 30%, and 60%. Rhodomyrtus tomentosa leaf extraction was carried out by maceration method with 70% ethano solvent. The extraction results are divided into 3 concentrations (15%, 30% and 60%). The wound healing process was evaluated by measuring the length of the wound manually from 0 to 10 days in each group. Meanwhile, the number of fibroblast cells was calculated through hematoxylin eosin (HE) staining and observed using an Olympus CX41 microscope with a 10x magnification and objective lens magnification in 3 fields. Result: There was a significant difference in the reduction in wound length (p =< 0,000) between the five experimental groups (Rhodomyrtus tomentosa leaf extract solution 15%, 30% and 60%, negative control and positive control. Solution of rhodomyrtus tomentosa leaf extract accelerated the increase in the number of fibroblasts compared to the negative control group (p = 0.003), but did not make a difference (p = 0.403) with the positive control group. Rhodomyrtus tomentosa leaf extraction solution had the same microscopic effect on the number of fibroblasts with a positive control group given 0.9% NaCl solution. Conclusion: There was a significant difference in the number of fibroblasts between all groups, but no difference in wound healing length.
