Background and Aim: Chlorpyrifos (CPF) is a widely used organophosphate insecticide. Nanoparticles of zinc oxide (ZnO NPs) physically showed effective adsorbing property for some insecticides. The study was conducted to estimate the potential effect of ZnO NPs against CPF toxicity. Materials and Methods: Four groups of male rats were used; control group and three groups received drinking water contained 75 mg/L CPF, combined 75 mg/L CPF and 200 mg/L ZnO NPs, and 200 mg/L ZnO NPs, respectively. Results: CPF significantly decreased macrophage activity, serum lysozyme activity, and levels of interleukin-2 (IL-2) and IL-6; increased the percentage of DNA degeneration on comet assay of lymphocytes and significantly elevated hepatic and splenic malondialdehyde contents; and decreased their glutathione contents. The liver and spleen showed marked histological alterations after exposure to CPF with decreased expression of acetylcholinesterase. The coadministration of ZnO NPs ameliorated most of the undesirable effects of CPF, through elevation of macrophage and serum lysozyme activities, increased the levels of IL-2 and IL-6, corrected the oxidative stress markers, and alleviated most of the adverse effect exerted by CPF in liver and spleen tissues. Conclusion: The addition of ZnO NPs to CPF-contaminated drinking water may be useful as a powerful antioxidant agent against toxic damage induced by CPF particularly in individuals who are on daily occupational exposure to low doses of CPF.

Background: Zinc oxide Nanoparticles (NPs) present irreversible effects on the nervous system, memory, and learning. Objective: The current study aimed to investigate the effects of pentoxifylline on memory impairments, CA1 hippocampal pyramidal cells, and blood serum antioxidant enzymes in male rats treated with zinc oxide NPs. Methods: Male Wistar rats were divided into the control, zinc oxide NPs (1.25 mg/kg), pentoxifylline (50 mg/kg), and pentoxifylline with zinc oxide NPs groups. In all study groups, saline, zinc oxide NPs, and pentoxifylline were intraperitoneally injected 30 minutes before training. In the co-treatment group, pentoxifylline was injected one hour before injecting Zno NPs. After performing the behavioral test, the tested animals’ brains were fixed and the number of healthy neurons in the CA1 region of the hippocampus was counted. In all research groups, malondialdehyde levels, total antioxidant power, superoxide dismutase levels, and glutathione peroxidase in blood serum were measured. Results: Zinc oxide nanoparticles decreased memory and the number of healthy neurons in the CA1 region of the hippocampus and increased oxidative stress in blood serum, compared to the controls. In the co-treatment group, using pentoxifylline improved the above-mentioned factors and reached the level of the control group. Pentoxifylline alone presented no significant effect on the aforementioned characteristics, compared to the control group. Conclusion: ZnO NPs may decrease memory retrieval and cause cell death in the pyramidal neurons of the CA1 region of the hippocampus by increasing oxidative stress. Pentoxifylline, as a potent antioxidant, can prevent the harmful effects of ZnO NPs.

The present study aimed to investigate the effect of turmeric and ginger on lipid profile of male rats exposed to oxidative stress induced by hydrogen peroxide H2O2 at a concentration of 1% given with consumed drinking water to male rats. Methods: 200 mg/kg from turmeric and ginger were used, and the animals were treatment for 30 days. Results: the results showed a significant increase in cholesterol, triglycerides, low density lipoprotein (LDL), very low density lipoprotein (VLDL), whereas it explained a significant decrease in high density lipoprotein (HDL) of male rats exposed to oxidative stress when compared with control group. the results showed a significant decrease in cholesterol, triglycerides, (LDL), (VLDL), whereas it explained a significant increase in (HDL) of rats treated with turmeric and ginger at dose 200 mg/kg when compared with male rats exposed to oxidative stress.

Various studies in rodents have shown that nanoparticles are transferred to the breast milk. Under the present study, lactating Wistar rats were repetitively gavaged 5, 25, and 50 mg/kg bw of zinc oxide nanoparticles (ZnO-NPs) and 50 mg kg−1 bw of bulk zinc oxide (bZnO) for 19 days after parturition. The results showed that ZnO-NPs were absorbed in the small intestine of dams and distributed to the liver. Furthermore, ZnO-NPs were distributed to the intestine and liver of rat pups through dam’s milk. No significant change in body weight was observed in the dams treated with ZnO-NPs or bZnO and their offsprings as compared to the control group. The spleen weight significantly increased in the rat dams treated with 50 mg kg−1 of ZnO-NPs. ZnO-NPs were mostly excreted through feces. The levels of liver cytochrome P450 reductase and serum total antioxidant capacity significantly decreased in the rat dams treated with ZnO-NPs (50 mg kg−1) and their offsprings. The levels of serum cytokines (tumor necrosis factor-alpha and interleukin-1 beta) and liver injury marker enzymes (alanine aminotransferase and aspartate aminotransferase) significantly increased in the rat dams treated with ZnO-NPs (25 and 50 mg kg−1) and their offsprings. The level of immunoglobulin A secretion in the intestinal fluid of rat dams and their offsprings is significantly increased by increasing the dose of ZnO-NPs. Histopathology of intestine and liver of offsprings whose rat dams were treated with ZnO-NPs (50 mg kg−1) showed gross pathological changes. These results provide information for the safety evaluation of ZnO-NPs use during lactation. In conclusion, a dose-dependent postnatal transfer of ZnO-NPs is hazardous to the breastfed offsprings.

Cadmium (Cd) is considered to be one of the major environmental pollutants which have potential threat to human health. Reports of declining male fertility have renewed interest in the role of environmental and occupational exposures in the etiology of human infertility. Cd exposure led to obvious degenerative changes in testicular tissue. This study was performed to investigate the Cd-induced structural effects on the testes and to evaluate the possible protective effect of omega-3 oil in adult albino rats. Thirty adult male rats were used in the present work, divided randomly into five groups, six rats in each group; the first group was considered as a control group and left without treatment except the standard rat chow and tap water. The second group was given 40 mg/L of CdCl2 in drinking water while the third group was given 60 mg/L of CdCl2 in drinking water. The fourth group was given 40 mg/L of CdCl2 in drinking water plus omega-3 oil (4 g/kg diet), while the fifth group was given 60 mg/L of CdCl2 in drinking water plus omega-3 oil (4 g/kg diet), the Cd-treated rats showed dose-dependent histological and ultrastructural alterations which have been ameliorated after exposure to omega-3 oil. The present investigation concluded that omega-3 played a protective role against Cd-induced histopathological changes in rat testis.

Chromium galvanic production have leaded to biosphere pollution. Therefore advisable to study of role in the neurodegenerative development in retinal diseases under experimental conditions. The aim is to study the Enterosgel effect on morphological changes in rats retina with Cr(VI) – induced retinopathy. Materials and methods. An experimental study had carried out on 72 outbred white male rats. The rats had divided into 3 groups: I – control group of intact rats (n = 24). Control rats were received drinking water, II group – rats (n = 24), were received drinking water with Cr (VI) (K2Cr2O7) – 0.02 mol/L, III group – animals (n = 24) were received drinking water with K2Cr2O7– 0.02 mol/L and hydrogel methylsilicic acid (Enterosgel) at a dose of 0.8 mg/kg per day as a corrector. The animals had been decapitated under ether anesthesia. The retina had been studied on days 20, 40 and 60 of the experiment. Morphologically and morphometrically they had analyzed. Results. According to histological studies, it has proved that Cr (VI) causes dystrophic and degenerative changes in all rats retina layers. They increase as the duration of the experiment. The use of Enterosgel as a corrective therapy showed positive results in restoring the morphological structure of rats retina. After Enterosgel 20 days using as a corrector of Cr (VI) exposure, there is a barely noticeable swelling of the outer and inner nuclear layers. Other layers of the retina, morphologically, look undamaged. Forty days Enterosgel treatment have outer and inner nuclear layer edema of retina of animals persists but does not increase. It is easy noticeable swelling of the outer and inner layers of mesh, but no signs of damage processes of cell populations nuclear layers. State ganglionic layer and nerve fiber layer entirely satisfactory. These pathological changes are not critical. After 60 days from the beginning of loading of Cr (VI) and application of Enterosgel in the retina of rats there are initial degenerative changes in the photosensory layer. Cystic dilated outer segments of rods and cones were visible throughout, and areas of their fragmentation were observed. Ganglion neurons are not damaged, but their axons appear somewhat thickened and fluffy. But in general, the typical structure of the retina is preserved. Conclusions. Chromium-induced toxicity in rats is characterized by pronounced histological and morphometric changes and retinal thickness, which appear after 20 days, increase by 40 days and acquire maximum transformations after 60 days of the experiment. The use of Enterosgel improves picture morphological structures of the retina in rats under the influence of Cr (VI). The changes were expressed on days 20 and 40, which indicates the presence of protective properties for the retina

Abstract. Resveratrol (RES) is a well-known phytocompound and food component which has antioxidative and multifunctional bioactivities. The present study aims to examine how resveratrol administration affects plasma leptin and liver glycogen levels in rats subjected to an acute swimming exercise bout. The study was carried out on Wistar-Albino type adult male rats, each group include 7 rats. Group 1, Control Group. Group 2, Control Swimming Group: The group fed on a standard diet and subjected to an acute swimming exercise bout for 30 minutes at the end of the study. Group 3, Resveratrol Group: The group fed on a standard diet and given (10 mg/kg) resveratrol in drinking water for four weeks. Group 4, Resveratrol + Swimming Group: The group fed on a standard diet, given (10 mg/kg) resveratrol in drinking water for four weeks and subjected to a 30-minute acute swimming exercise at the end of the study. Plasma leptin levels using ELISA method (ng/l) and liver glycogen levels were determined by using histochemical method (number/0.1 mm2). Four weeks resveratrol administration to exercised and not-exercised rats did not cause a change in plasma leptin levels. Liver glycogen levels were 17.00 ± 3.16; 14.12 ± 2.98; 20.82 ± 1.97; 16.38 ± 1.27 (mean ± sd); respectively in groups 1, 2, 3, 4. Resveratrol administration to rats subjected to a bout of acute swimming exercise produced an effect that prevented the decrease in liver glycogen (p < 0.05). The study highlights that resveratrol supplementation may have regulatory effects on liver glycogen levels in exercised and non-exercised rats.

The evaluation of the effect of ginger on the modulation of toxic effects induced by lead is the objective of our study. Forty male rats were randomly divided into four groups and treated daily for 3 consecutive months. Group I (0-0) was kept as control; group II (0-G) received an experimental diet with 2% of ginger; group III (Pb-0) received 2% lead acetate dissolved in drinking water with a normal diet; and group IV (Pb-G) received 2% lead acetate in drinking water and an experimental diet containing 2% ginger. Lead acetate exposure caused a significant increase of organosomatic indexes, hepatic, lipid, and urine profiles. In addition, lead acetate has a pro-oxidative effect expressed by a significant decrease in tissue GSH levels and the enzymatic activity of GPx and CAT. This pro-oxidative action was also marked by an increase in MDA level and GST activity in lead-treated group. Feeding ginger-supplemented diet to lead acetate-treated rats restored all the parameters studied as compared to control. These results suggest that ginger treatment exerts a protective effect on metabolic disorders by decreasing the oxidative stress.

Background: Zinc oxide nanoparticles (ZnO NPs) are increasingly utilized in both industrial and medical applications. Therefore, the study was aimed to investigate the effect of green nanoparticle complex (green tea extract/zinc oxide nanoparticles complex, GTE/ZnO NPs) on oxidative stress induced by monosodium glutamate (MSG) on the liver of rats. Methods: Wistar male rats (n=64) weighing between 200-250 g were divided randomly into eight groups: control group was given physiological saline (1 mg/kg), two groups were treated with two different doses of MSG (MSG-LD, MSG-HD; 6 and 17.5 mg/Kg, respectively), GTE was given 1 mg/mL, 5th group was treated with ZnO NPs and 6th group was treated with GTE/ZnO NPs complex while, 7th and 8th groups were treated with MSG-LD + GTE/ZnO NPs complex and MSG-HD + GTE/ZnO NPs complex, respectively. All substances were given orally for 30 consecutive days. At the end of the study, the liver was homogenized for measurement of the oxidative stress status and anti-inflammatory biomarkers as well as histological and transmission alternations. Results: Results showed that the antioxidant enzymes activity and glutathione level were significantly decreased in MSG groups than control in a dose-dependent manner. Conversely, the malondialdehyde and inflammatory cytokines levels were significantly increased in MSG groups than the control group. The liver indicated no evidence of alteration in oxidative status, anti-inflammatory and morphological parameters in GTE, ZnO NPs and GTE/ZnO NPs complex groups. Conclusion: In conclusion, MSG at both doses caused oxidative stress and inflammation on liver after 28 days of exposure that supported histological analysis and transmission view of hepatic parenchyma. GTE/ZnO NPs act as partial hepato-protective against MSG.

Abstract Background A-to-I RNA editing represents a new player in the pathogenesis of cancer. However, the knowledge of RNA editing process in cancer is still limited and represents only the tip of the iceberg. The ADAR gene family regulate the dynamic landscape of RNA editing. Aberrant RNA editing status played a vital role in the pathogenesis of hepatocellular carcinoma (HCC). The nutri-epigenomic agent- pterostilbene- exhibits anti-inflammatory, antioxidative and antiproliferative activities. However, the effect of pterostilbene on ADAR(s) expression in HCC was not studied before. Aim of the work to evaluate the potential effect of pterostilbene administration on Adar(s) expression in HCC rats. Materials and methods Twenty four adult male rats were randomly divided into 4 groups: the control group, untreated HCC group received diethylnitrosamine (DENA) for 14 weeks, HCC group take received pterostilbene and DENA for 14 weeks, and non-HCC rats were given pterostilbene for 14 weeks. These groups were subjected to histological examination of liver tissues, laboratory measures (serum albumin, ALT, AST, and α fetoprotein), and Adar(s) expression by real time-PCR. Results liver enzymes (ALT, AST) and α fetoprotein levels in treated HCC group were significantly lower than untreated HCC group (p&lt;0.05). Serum albumin levels were significantly higher in treated HCC rats than untreated HCC group (P&lt;0.05). Adar1 was highly expressed in untreated HCC rats in comparison to the control group (p&lt;0.05). Meanwhile, treated HCC group had lower expression levels of Adar1 in comparison to untreated HCC rats. Conclusions pterostilbene had a beneficial effect on HCC and it may alleviate the aberrant expression of Adar1 in HCC rats. Key words HCC, ADARs, pterostilbene, RNA editing enzymes. Preliminary protection of zinc oxide nanoparticles (ZnO NPs) with terpenoids such as betulin, its derivatives, and essential oils components has been proposed to produce gel-like oleophilic and hydrophilic formulations. We studied the properties of gel-like dispersions of ZnO NPs with immobilized terpenoids and their effects on the activity of LDH, GR, G6PDH, restoration of redox balance of co-enzyme pairs NAD+/NADH and NADP+/NADPH, as well as the activity of SOD, catalase, AlDH in erythrocytes in the treatment of burns in rats. Hysteresis loops on the rheograms of studied dispersions characterize their thixotropic properties. ZnO NPs with betulin diphosphate in the water–ethanol medium lead to a 20-fold increase in the hydrodynamic radius at pH 7.3 compared to the initial ZnO NPs, and facilitate the formation of Zn2+ ions and their penetration into the viable epidermis, unlike oleophilic dispersions. All dispersions reduce the healing time by one and a half times compared with the untreated control group, increase the activity of LDH, GR, G6PDH, SOD, catalase, AlDH, and contribute to the normalization of coenzyme balance. Normalization of the redox balance and wound state was more effective using hydrophilic dispersions due to Zn2 + penetration.
