Background: ocimum sanctum (Tulsi) “Queen of herbs” is considered as sacred and medicinal plant in a lot of countries around the world especially India. Therefore this study was carried out to investigate the heptorenal protective effect of ocimum sanctum aqueous extract against gentamicin induced hepatorenal toxicity in chickens.Materials and Methods: A total 100, one-day old unsexed broiler chicks of a commercial breed (Cobb breed) were used in this study, the chickens were subjected to different treatments blood and tissue samples were collected at 5th week of age post administration, biochemical and histopathological examinations were utilized to investigate protective effect of ocimum sanctum aqueous extract on liver and kidney.Results: gentamicin recorded significant (P<0.05)increase in serum level of aspartate aminotransferase (AST), alanine aminotransferase (ALT), uric acid and creatinine levels when compared with control group, the administration of ocimum sanctum aqueous extract with gentamicin ameliorate their toxic effect, the group treated with ocimum sanctum aqueous extract only similar to control, while gentamicin induce significant (P<0.05)decrease in serum albumin, total protein and globulin levels when compared with control group, ocimum sanctum aqueous extract with gentamicin restore their toxic effect. The group treated with ocimum sanctum aqueous extract only as control. Gentamicin induce histopathological alterations in liver and kidney tissues by comparison with control group and ocimum sanctum aqueous extract treated group as control, while the administration of ocimum sanctum aqueous extract with gentamicin showing improvement in histopathological lesion compared with toxic effect induced by gentamicin.Conclusions: The hepatotoxicity and nephrotoxicity induced by gentamicin were ameliorated by aqueous extract of ocimum sanctum especially at dose of 2ml/liter in drinking water for 12 days.

Background: Liquorice (Glycyrrhiza glabra) is a high valued plant and used in a lot of countries around the world. The liquorice (LQ) has a remarkable medicinal, nutritional and socio-economic value. Therefore this study was designed to clarify the protective effect of liquorice hydroethanolic extract against Thioacetamide (TAA) induced hepatotoxicity and hematotoxicity in rats.Materials and Methods: Seventy white Albino male rats were used in this study and after acclimatization rats were subjected to different treatments blood and tissue samples were collected after day 30 post administration, biochemical, antioxidant, hematological and histopathological examinations were utilized to investigate hepatoprotective activity of liquorice hydroethanolic extract.Results: TAA significant (P<0.05) increase the activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) and hepatic malondialdehyde (MDA) compared to control group and silymarin(SL)treated group as stander, admin-istration of LQ extract restore the toxic effect of TAA , while TAA significant (P<0.05) decrease the levels of catalase(CAT) ,hepatic glutathione(GSH) hepatic superoxide dismutase (SOD) , total protein and albumin level compared to control group and silymarin(SL)treated group as stander, LQ extract ameliorate toxic effect of TAA. TAA induce hematotoxicity in rats by significant (P<0.05) decrease Hb content and RBCs, whereas WBCs count significantly (P<0.05) increased throughout the period of administration when compared to the rats in control group and silymarin(SL)treated group as stander,LQ administration protect against the hematotoxicity effect of TAA, TAA induce histopathological alteration in liver tissue by comparison with control group and silymarin(SL)treated group as stander, while LQ extract showing improvement in the histopathological lesion compared with toxic effect induced by TAA.Conclusions: The hepatotoxicity and hematotoxicity induced by TAA were ameliorated by hydroethanolic extract of LQ especially in double dose (200 mg/kg b.wt). This effect was attributed to free radical scavenging activity and potent antioxidant activity of its components (flavonoid, tannin and saponin).

Abstract Background Hepatorenal injuries and dyslipidemia are common global health challenges but medicinal plant extracts may have potential to prevent them. Thus, this study evaluated the protective effect of aqueous extract of Lophira lanceolata leaf (LLE) against cisplatin-induced hepatorenal injuries and dyslipidemia in albino Wistar rats. Methods Thirty rats were randomly divided into 6 groups of 5 rats each. Group I rats received distilled water and served as control, group II rats were given 5 mg/kg cisplatin (CIS) intraperitoneally, groups III and IV rats were treated with 200 and 400 mg/kg LLE respectively for 26 days by oral gavages while groups V and VI rats were treated with 200 and 400 mg/kg LLE respectively, followed by CIS on the 21st day as in group II. About 24 h after treatment, blood was collected from the rats; then serum was separated and used for estimations of biochemical parameters. The kidney and liver of rats were removed, rinsed in normal saline, stored in 10% formalin and used for histological analyses. Results The biomarkers of hepatic (Aminotransferases, Alkaline phosphatase and Bilirubin) and renal (urea and creatinine) injuries, and dyslipidemia (Total cholesterol, triglycerides and LDL-cholesterol) significantly (p < 0.05) increased in the rats exclusively exposed to cisplatin when compared with normal control. However, treatment of cisplatin-exposed rats with 200 and 400 mg/kg LLE significantly (p < 0.05) reduced the levels of these biomarkers of hepatorenal injuries and dyslipidemia when compared with cisplatin control. Photomicrographs showed pathological signs in the liver and kidney of rats exclusively exposed to cisplatin, but there was moderate protection of these tissues in the rats treated with LLE and cisplatin. Conclusion The current findings have shown that Lophira lanceolata leaf extract may provide moderate protection against cisplatin-induced hepatorenal injuries and dyslipidemia in albino Wistar rats.

In agriculture, excessive use of various pesticides had increased in developing countries. These pesticides may be highly toxic, not only to aquatic organisms like fishes but also to humans. In recent years, synthetic pyrethroids were developed for major uses in various agriculture practices and public health purposes. We aim to find the toxic Effect of Cypermethrin on Freshwater Fish Oreochromis mossambicus (Tilapia). The fish Oreochromis mossambicus were exposed for 30 days to various sub lethal concentrations (1/10, 1/20, and 1/30) of cypermethrin. After completion of a 30 days exposure, the fish Oreochromis mossambicus was sacrificed and tissue samples of muscle, liver, and kidney were analyzed. Decreased value of ALP, ACP, and Increased value of AST, ALT was observed in all the sub lethal exposure of cypermethrin on treated freshwater fish Oreochromis mossambicus at 30 days on comparing with the control group. High variation of AST and ALT were observed in the liver at 1/10th concentration of cypermethrin and elevated variation of ALP and ACP was observed in the liver at 1/10th concentration of sub lethal level for 30 days exposure. While comparing with the control group, the effects of cypermethrin on experimental fish showed increased levels of AST, ALT in the tissues of muscle, liver, and kidney of O. mossambicus, and the level of ACP and ALP were found in a decreased manner. Increased levels of ALT and AST activity observed in freshwater fishes may be due to the reduction of metabolic activity and concentration of ACP and ALP in various tissues due to cellular necrosis. In the present study, we found that the effect of cypermethrin alters the activity of various enzymes in freshwater fish Oreochromis mossambicus. It is concluded from this study that exposure to cypermethrin affects the enzyme activities of fish.

The colonization of Lactobacillus spp., enterobacteria and facultatively anaerobic gram-positive cocci was monitored in intestinal samples of growing broiler chicks from 24 h to 28 days of age. Rapid bacterial growth occurred within the first week, followed by stabilization and decline of colony forming units (CFU). Xylanase supplementation led to significantly lower CFU per gram of wet weight for total presumptive enterobacteria and total gram-positive cocci in luminal and tissue samples in the first 3 weeks. Lactobacillus spp. colony counts from tissue samples were higher for animals with the xylanase-supplemented diet, but luminal CFU were not. The composition of dominant Lactobacillus spp. strains was different in duodenal and jejunal tissues, but distribution of Lactobacillus spp. colony forms was unaffected by xylanase treatment. Mucosa-associated Enterococcus spp. displaced the dominant gram-positive cocci in the jejunal samples. d- and l-lactic acid and acetic acid concentrations were significantly higher in ileal samples from the control group on days 7 and 14, while butyric acid concentrations were higher in the xylanase-treated group. It is concluded that the less viscous intestinal environment caused by the xylanase slowed proliferation of gram-positive cocci and presumptive enterobacteria in enzyme-supplemented animals in the first 3 weeks of life.

Methotrexate (MTX) is a drug used in the treatment of various types of cancer and inflammatory diseases, but its clinical use has been restricted due to its toxicity. Apigenin (API) is an effective flavonoid with antioxidant and anti-inflammatory properties. The aim of this study was to determine the protective effect of API against MTX-induced liver and kidney toxicity. Four groups with 12 male mice each were used. The control and API groups were received 0.9% saline (ip) and API (3 mg/kg ip) for 4 days, respectively. The MTX group were given a single dose of MTX (20 mg/kg ip) on the fourth day. The MTX + API group were administered API for 7 days and then MTX on fourth day. Blood, liver and kidney were collected to evaluate tissue injury markers, oxidative stress biomarkers, and histopathological and immunohistochemical assessments. In MTX-treated group, significant increases in aminotransferases activities, creatinine and malondialdehyde (MDA) levels and significant decreases in catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase1 (SOD1) activities and glutathione (GSH) levels were determined compared to the control group. Furthermore, histopathological changes and significant increases in caspase-3, C-reactive protein (CRP), granulocyte colony-stimulating factor (G-CSF), and inducible nitric oxide synthase (iNOS) expressions were detected in both liver and kidney tissues of MTX-treated mice. Pretreatment with API alleviates liver and kidney toxicity by attenuating oxidative stress and tissue injury markers, histopathological alterations, and apoptosis and inflammation. These results suggest that API has a protective effect against oxidative stress and liver-kidney toxicity induced by MTX.

Introduction Increasing the immune system's function of fighting infectious diseases is very important in the poultry industry. Ginger, scientifically known as Zingiber officinale, belongs to the Zingiberaceae family. The use of ginger in the diet of poultry increases serum levels of superoxide dismutase enzymes and glutathione peroxidase, which are considered to be important antioxidant enzymes. The main objective of the present study is to evaluate the effect of ginger on the spleen tissue of broiler chickens. Material and Methods The specimens comprised 2 groups of 20 Ross breed broiler chicks, for 42 days and were then, examined and tested. The diet was supplemented with 1 g/kg of ginger powder from the beginning of the rearing period. Blood samples of the chicks were randomly collected to measure the levels of hemagglutination (HI). The removed spleens were fixed with 10% formalin buffer. The specimens were cut in 5-micron diameters and stained with hematoxylin and eosin. Results and Conclusion There was a statistically significant difference in the mean of HI blood titers between the chicks in the growth period and final period groups (p < 0.05). The white-pulp tissue samples were more clearly seen in the treatment group than in the control group, and also, it was observed that the wall of the central artery of the white pulp was thicker in the ginger-treated group as compared with the control group. The nutritional value of ginger may vary. Thus, it is necessary to investigate the effect of this plant final on weight gain; the serum factors associated with the metabolic chart, and the response of the immune system to this plant.

PURPOSE: Aloe vera is a tropical plant popularly known in Brazil as babosa. We have investigated the effect of aqueous extract of Aloe vera on the biodistribution of Na99mTcO4 and laboratorial parameters in Wistar rats. METHODS: Twelve animals were divided into treated and control groups. In the treated group, Aloe vera was given by gavage (5mg/mL/day) during 10 days. The control group received sorbitol by the same way and period. One hour after the last dose, we injected 0.1mL of Na99mTcO4 by orbital plexus. After 60 min, all the animals were killed. Samples were harvested from the brain, liver, heart, muscle, pancreas, stomach, femur, kidneys, blood, testis and thyroid and the percentage of radioactivity (%ATI/g) was determined. Biochemical dosages were performed. RESULTS: There was a significant increase of %ATI/g in blood, femur, kidneys, liver, stomach, testis and thyroid and also in blood levels of AST and ALT. A significant decrease in levels of glucose, cholesterol, triglycerides, creatinine and urea occurred. The statistical analyses were performed by Mann-Whitney test and T-Student test (p<0.05). CONCLUSION: The aqueous extract of Aloe vera facilitated the uptake of Na99mTcO4 in organs of rats and it was responsible to a high increase of levels of AST and ALT.

The aim of this study was to evaluate the effect of Vernonia amygdalina leaf extract (VALE) on the carbon tetrachloride-induced hepatotoxicity (CCl4) in broiler chickens. A total of 360-day-old broilers were divided into 4 treatments of 90 birds each consisting of 6 replicates of 15 birds each. The treatments were birds offered 1 mL/kg BW saline (control group), 100 mg/kg BW VALE, 1 mL/kg BW CCl4 (CCl4-treated group), and 100 mg/kg BW VALE + 1 mL/kg BW CCl4 (VALE + CCl4 group). Blood samples were collected at 42 days of age and analyzed for the liver enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and selected biochemical parameters. The experiment was laid out in a completely randomized design. The results obtained showed that VALE had the potential to mitigate the adverse effects of CCl4 on protein and lipid metabolism as reflected in the low serum malondialdehyde (MDA) levels, which is a marker of lipid peroxidation. The aqueous extract of Vernonia amygdalina (VA) at a dose of 100 mg/kg body weight showed a moderate hepatoprotective effect by reducing serum AST levels (p < 0.05). The levels of serum AST, ALP, ALT, and GGT were significantly increased in CCl4-treated birds compared to the control group, reflecting carbon tetrachloride-induced liver damage. The VALE + CCl4 group showed a significantly higher amount of ALP compared to birds treated with carbon tetrachloride, suggesting a hepatoprotective effect. To conclude, Vernonia amygdalina aqueous extract can be used to confer protection against hepatotoxicity, which can induce severe hepatocellular damage in birds.

This study aimed to evaluate the toxic effect of silver nanoparticles (AgNPs) on the parotid glands (PGs) of albino rats histologically and ultrastructurally and assess the possible protective effect of ascorbic acid as an antioxidant. Thirty male albino rats weighing between 150 mg and 200 mg were divided into three groups: the control group (C1) contained 10 rats that received 2 mg/kg (body weight (bw)) of aqueous nitrate buffer by intraperitoneal (IP) injection daily for 28 days; the AgNPs group contained 10 rats that received 2 mg/kg (bw) IP AgNPs daily for 28 days; and the AgNPs-vitamin C group contained 10 albino rats that received 2 mg/kg (bw) AgNPs IP daily for 28 days with oral administration of 100 mg/kg (bw) vitamin C in drinking water daily for 28 days. The PG acinar and ductal cells of the AgNPs group showed signs of toxicity and degeneration characterized as pleomorphic nuclei, binucleation, cytoplasmic vacuolations, and stagnated secretion in the ductal lumen. In addition to degenerated mitochondria, dilated rough endoplasmic reticulum and lysosomes were filled with AgNPs ( p < 0.001). The AgNPs-vitamin C group showed significantly less degenerative changes histologically and ultrastructurally compared to the AgNPs group ( p = 0.002). AgNPs produced significant toxic effects on the PG of albino rats, presumably through the generation of reactive oxygen species and toxic ion release, and administration of vitamin C was shown effective in decreasing these toxic effects.

Melatonin (MEL) and coenzyme Q10 (CoQ10) both display antioxidant and free radical scavenger properties. In the present study, the effect of MEL and CoQ10 on the oxidative stress and fibrosis induced by ochratoxin A (OTA) administration in rats was investigated. Rats were divided into five equal groups, each consisting of seven rats: (1) controls; (2) OTA-treated rats (289 μg/kg/day); (3) OTA+MEL–treated rats (289 μg/kg/day OTA + 10 mg/kg/day MEL); and (4) OTA+CoQ10–treated rats (289 μg/kg/day OTA +1 mg/100 g/day body weight (bw) CoQ10). After 4 weeks of treatment, the level of malondialdehyde (MDA), glutathione peroxidase (GPx), and hydroxyproline (Hyp) were measured in the homogenates of liver and kidney. In the OTA-treated group, the levels of MDA and Hyp in both liver and kidney were significantly increased when compared with the levels of control, whereas GPx activities decreased. In OTA+MEL–treated rats, the levels of MDA and Hyp in both liver and kidney were significantly decreased when compared with the levels of OTA-treated rats; however; GPX activities increased. In the OTA+CoQ10–treated group, the levels of MDA and Hyp were decreased when compared with the levels of OTA-treated rats, whereas GPx activities increased. In the OTA+CoQ 10–treated group, the levels of MDA, Hyp, and GPx were not significantly changed in kidney when compared with OTA-treated group. MEL has a protective effect against OTA toxicity through an inhibition of the oxidative damage and fibrosis both liver and kidney. Although CoQ10 has protective effect against OTA toxicity in liver tissue, it has no effect in kidney tissue.
