The aim of this study was to evaluate the effect of Vernonia amygdalina leaf extract (VALE) on the carbon tetrachloride-induced hepatotoxicity (CCl4) in broiler chickens. A total of 360-day-old broilers were divided into 4 treatments of 90 birds each consisting of 6 replicates of 15 birds each. The treatments were birds offered 1 mL/kg BW saline (control group), 100 mg/kg BW VALE, 1 mL/kg BW CCl4 (CCl4-treated group), and 100 mg/kg BW VALE + 1 mL/kg BW CCl4 (VALE + CCl4 group). Blood samples were collected at 42 days of age and analyzed for the liver enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), and selected biochemical parameters. The experiment was laid out in a completely randomized design. The results obtained showed that VALE had the potential to mitigate the adverse effects of CCl4 on protein and lipid metabolism as reflected in the low serum malondialdehyde (MDA) levels, which is a marker of lipid peroxidation. The aqueous extract of Vernonia amygdalina (VA) at a dose of 100 mg/kg body weight showed a moderate hepatoprotective effect by reducing serum AST levels (p < 0.05). The levels of serum AST, ALP, ALT, and GGT were significantly increased in CCl4-treated birds compared to the control group, reflecting carbon tetrachloride-induced liver damage. The VALE + CCl4 group showed a significantly higher amount of ALP compared to birds treated with carbon tetrachloride, suggesting a hepatoprotective effect. To conclude, Vernonia amygdalina aqueous extract can be used to confer protection against hepatotoxicity, which can induce severe hepatocellular damage in birds.

Carbon tetrachloride (CCl₄) is a chemical that can cause damage to liver cells. One of the natural ingredients developed to reduce liver damage due to chemicals and infections is Chlorella sp. The research aimed to explore the hepatoprotective effect of the antioxidant superoxide dismutase (SOD) by administering Sun Chlorella in Rattus norvegicus CCl₄ induced rats. The study design used a post-test with a control group design with a completely randomized design trial on 30 male rats of Wistar strains, aged 2–3 months, 200–250 grams in weight. The research was conducted in November 2016–January 2017 in Purwokerto. Rats were divided into five groups and treated for four weeks as follows: K1 group was given aquades; K2 was given CCl₄; K3, K4, K5 were given CCl₄ and Sun Chlorella 3.6 mg/200 gBW, 7.2 mg/200 gBW, and 14.4 mg/200 gBW respectively, by the gastric probe. Statistical analysis with correlation test and one way ANOVA multivariate test showed that Sun Chlorella 7.2 and 14.4 mg/200 gBW significantly increased SOD levels in rats induced CCl₄ (p=0.004, p=0.009). SOD rates were significantly strong associated with aspartate aminotransferase/AST (r=−0.685, p=0.000) and alanine aminotransferase/ALT (r=−0.659, p=0.000). The conclusion is Sun Chlorella increases SOD levels in CCl₄-induced rats. Increased SOD levels may decrease AST and ALT levels. EFEK HEPATOPROTEKTIF SUN CHLORELLA SEBAGAI ANTIOKSIDAN PADA RATTUS NORVEGICUS YANG DIINDUKSI KARBON TETRAKLORIDAKarbon tetraklorida (CCl₄) merupakan bahan kimia yang dapat menyebabkan kerusakan sel hati. Berbagai bahan alami telah dikembangkan untuk mengurangi kerusakan hati baik akibat bahan kimia maupun infeksi, salah satunya adalah Chlorella sp. Penelitian ini bertujuan melihat efek hepatoprotektif  dengan pemberian Sun Chlorella pada tikus Rattus norvegicus yang diinduksi CCl₄. Desain penelitian menggunakan post-test with a control group dengan rancangan percobaan rancangan acak lengkap pada 30 tikus jantan galur Wistar, usia 2–3 bulan, dan berat 200–250 gram. Penelitian ini dilakukan di Purwokerto pada periode November 2016–Januari 2017. Tikus dibagi menjadi lima kelompok dan mendapatkan perlakuan per oral dengan sonde lambung selama 4 minggu sebagai berikut: kelompok K1 sebagi kontrol negatif diberikan aquades; kelompok control positif K2 diberikan CCl₄; kelompok perlakuan K3, K4, dan K5 diberikan CCl4 dan Sun Chlorella 3,6 mg/200 gBB; 7,2 mg/200 gBB; 14,4 mg/200 gBB tikus berurutan. Uji statistik dengan menggunakan uji korelasi dan ANOVA satu arah menunjukkan bahwa pemberian Sun Chlorella 7,2 mg dan 14,4 mg meningkatkan kadar SOD pada tikus yang diinduksi CCl₄ secara bermakna (p=0,004; p=0,009). Kadar SOD berhubungan kuat dengan kadar aspartat aminotransferase/AST (r=−0,685; p=0,000) dan alanin aminotransferase/ALT (r=−0,659; p=0,000). Kesimpulan penelitian ini adalah pemberian Sun Chlorella meningkatkan kadar SOD pada tikus yang diinduksi CCl₄. Peningkatan kadar SOD menyebabkan penurunan kadar AST dan ALT.

To investigate the protective effects of galangin on liver toxicity induced by carbon tetrachloride (CCl4) in mice. Mouse hepatotoxicity model was established by intraperitoneal injection (i.p.) of 10 ml/kg body weight CCl4 that diluted with corn oil to a proportion of 1:500 on Kunming mice. The mice were randomly divided into five groups named control group, model group, and 1, 5, and 10 mg/kg galangin group. The levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were analyzed by ELISA. Liver histopathological examination was observed via optical microscopy. The levels of superoxide dismutase (SOD), malondialdehyde (MDA), glutathione (GSH), and glutathion (GSSG) were analyzed to assess oxidative stress. Finally, western blot assay was carried out to analyse the expression levels of total AMP-activated protein kinase (AMPK), phospho-AMPK (p-AMPK), total liver kinase B1 (LKB1), and phospho-LKB1 (p-LKB1). Compared with the control group, in the model group, the levels of AST, ALT, MDA, and GSSG increased significantly ( p < 0.01); the activity of SOD and GSH decreased significantly ( p < 0.01); and the histopathological examination revealed liver necrosis. However, treatment with galangin (5 and 10 mg/kg) significantly reversed these CCl4-induced liver damage indicators. Furthermore, treatment with galangin (10 mg/kg) significantly increased the p-AMPK and p-LKB1 expression levels ( p < 0.01). This study supports the hepatoprotective effect of galangin against hepatotoxicity, perhaps occurring mainly through the LKB1/AMPK-mediated pathway.

PURPOSE: Aloe vera is a tropical plant popularly known in Brazil as babosa. We have investigated the effect of aqueous extract of Aloe vera on the biodistribution of Na99mTcO4 and laboratorial parameters in Wistar rats. METHODS: Twelve animals were divided into treated and control groups. In the treated group, Aloe vera was given by gavage (5mg/mL/day) during 10 days. The control group received sorbitol by the same way and period. One hour after the last dose, we injected 0.1mL of Na99mTcO4 by orbital plexus. After 60 min, all the animals were killed. Samples were harvested from the brain, liver, heart, muscle, pancreas, stomach, femur, kidneys, blood, testis and thyroid and the percentage of radioactivity (%ATI/g) was determined. Biochemical dosages were performed. RESULTS: There was a significant increase of %ATI/g in blood, femur, kidneys, liver, stomach, testis and thyroid and also in blood levels of AST and ALT. A significant decrease in levels of glucose, cholesterol, triglycerides, creatinine and urea occurred. The statistical analyses were performed by Mann-Whitney test and T-Student test (p<0.05). CONCLUSION: The aqueous extract of Aloe vera facilitated the uptake of Na99mTcO4 in organs of rats and it was responsible to a high increase of levels of AST and ALT.

Purpose : Aortic valve stenosis is the most common valvular heart disease, and standard curative therapy remains open-heart surgical valve replacement. The aim of our experimental study was to determine if ApoA-I mimetic peptide infusions could induce regression of aortic valve stenosis. Methods : Twenty-seven New-Zealand White male rabbits received a cholesterol-enriched diet and vitamin D 2 until significant aortic valve stenosis was detected by echocardiography. The enriched diet was then stopped to mimic cholesterol-lowering therapy and animals were randomized to receive saline (control group, n =14) or an ApoA-I mimetic peptide (treated group, n =13), 3 times per week for 2 weeks. Serial echocardiograms and post mortem valve histology were performed. Results : Aortic valve area improved significantly in the treated group compared to controls after 7 days (20.9±0.9 mm 2 vs. 18.2±0.6 mm 2 , P <0.0001) (corresponding to increases of 15.9% and 1.9%), 10 days (21.5±1.0 mm 2 vs. 19.5±0.6 mm 2 , P =0.0032) (increases of 19.2% vs. 9.1%), and 14 days of treatment (22.4±0.9 mm 2 vs. 20.4±0.6 mm 2 , P =0.0028) (increases of 24.4% vs. 14.2%). Likewise, aortic valve thickness decreased by 19% after 14 days of treatment in the treated group (0.951±0.070 mm at baseline vs. 0.768±0.074 mm at follow-up) whereas it was unchanged in controls ( P <0.0001). Histological analysis revealed that lesion extent at the base of valve leaflets and sinuses of Valsalva was smaller in the treated compared to control group (52.8±12.5% vs. 66.7±9.9%, P =0.032). The ApoA-I mimetic peptide treatment also leads to a reduction in aortic valve calcifications as revealed by the loss of the positive relationship observed in the control group ( r =0.87, P =0.004) between calcifications area and aortic valve thickness. Conclusions : Infusions of an ApoA-I mimetic peptide lead to regression of experimental aortic valve stenosis. These positive results justify the further testing of HDL-based therapies in patients with valvular aortic stenosis. Regression of aortic stenosis, if achieved safely, could transform our clinical approach of this disease.

  ABSTRAK Tujuan penelitian ini adalah untuk mengetahui pengaruh dari penggunaan ramuan herbal terhadap income over feed cost  ayam broiler. Materi  yang digunakan pada  penelitian ini adalah: ayam broiler DOC. Penelitian ini dirancang dengan menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari 4 perlakuan dan masing-masing perlakuan terdiri dari 4 ulangan. Perlakuan yang digunakan dalam penelitian ini adalah penggunaan ramuan herbal dalam air minum dengan level penggunaan sebagai berikut : P0: tanpa penambahan ramuan herbal, P1:air + 10 % ramuan herbal, P2: air  + 15 % ramuan herbal, P3: air + 20% ramuan herbal. Variabel yang di amati  dalam penelitian ini adalah konsumsi pakan, biaya pakan, dan income over feed cost ayam broiler. Data yang diperoleh (income over feed cost) di analisis dengan menggunakan analisis ragam dengan rancangan Acak Lengkap (RAL). Hasil penelitian menunjukan bahwa biaya pakan tertinggi terdapat pada kelompok perlakuan P3 diikuti berturut-turut oleh P1, P2, P0 dan yang terendah adalah P0. Sedangkan pada P0 biaya pakan paling rendah karena tidak ada penggunaan ramuan herbal. Berdasarkan data penerimaan, penerimaan tertinggi terdapat pada kelompok kontrol P1 Rp 31.430,6  sedangkan penerimaan terandah terdapat pada kelompok P0 sebesar Rp 24.814,8.100 Rendahnya penerimaan dalam penelitian ini disebabkan oleh rendahnya bobot badan ayam broiler selama penelitian. Income Over Feed Cost selama penelitian yang tertinggi terdapat pada kelompok P3 (3.568), dan terendah terdapat pada perlakuan  P1 (979) P0 (1.964), P2 (1.937) karena biaya pakan lebih tinggi dari pada penerimaan.Ini berarti bahwa penggunaan ramuan herbal  dalam air minum berpengaruh sangat nyata (P< 0,01) terhadap Income Over Feed Cost. Berdasarkan hasil penelitian dapat disimpulkan bahwa penggunaan ramuan herbal sebagai feed additive  memberikan pengaruh sangat nyata terhadap IOFC ayam broiler.   Abstract The purpose of this study was to determine the effect of using herbal ingredients on income over feed costs of broiler chickens. The material used in this study was: broiler DOC. This study was designed using a Completely Randomized Design (CRD) consisting of 4 treatments and each treatment consisted of 4 replications. The treatment used in this study is the use of herbal ingredients in drinking water with the use level as follows: P0: without the addition of herbal ingredients, P1: water + 10% herbal ingredients, P2: water + 15% herbal ingredients, P3: water + 20% herbal concoctions. The variables observed in this study were feed consumption, feed costs, and income over feed costs for broiler chickens. The data obtained (income over feed cost) is analyzed using a variety of analysis with a completely randomized design (CRD). The results showed that the highest feed costs were found in the P3 treatment group followed by P1, P2, P0 and the lowest was P0. Whereas at P0 the cost of feed is lowest because there is no use of herbal ingredients. Based on acceptance data, the highest revenue was in the P1 control group of Rp. 31,430.6 while the lowest acceptance was in the P0 group of Rp. 24,814.8,100. The low acceptance in this study was caused by the low body weight of broiler chickens during the study. Income Over Feed Cost during the highest research was found in the P3 group (3,568), and the lowest was found in the treatment of P1 (979) P0 (1,964), P2 (1,937) because the cost of feed was higher than acceptance. drinking water has a very significant effect (P <0.01) on Income Over Feed Cost. Based on the results of the study it can be concluded that the use of herbal ingredients as additive feeds has a very significant effect on IOFC broiler chickens.

Background and objectives: toxic liver injury results in nitrooxidative stress. Melatonin is a potent free radical scavenger, an inducible nitric oxide synthase (iNOS) inhibitor and an activator of antioxidant enzymes. The aim of this study was to investigate the hepatoprotective effect of exogenous melatonin on animals with acute toxic hepatitis. Material and methods: 36 healthy Sprague-Dawley male rats were split into three equal groups and given carbon tetrachloride (CCl4), 2 g/kg (CCl4 group) or the same dose of CCl4 and melatonin, 10 mg/kg (CCl4/melatonin group) or saline (control group). The effect of melatonin on prooxidant and antioxidant system indexes, NO and NOS levels in serum and liver, data of mitochondrial chain functions and cytolysis in liver were evaluated in all three groups. Results: melatonin significantly decreased activities of AST, ALT, ceruloplasmine and thiobarbituric acid reactive substance (TBARS) in serum. Catalase activity was lowered in serum but not in the liver. Hepatic TBARS, lipid hydroperoxides and glutathione concentrations were decreased, while superoxide dismutase, mitochondrial cytochrome oxidase and succinate dehydrogenase activities increased. Melatonin inhibited synthesis of stable NO metabolites in serum: NO2-by 37.9%; NO3-by 29.2%. There was no significant difference in content NO2-in the liver, but concentration of NO3-increased by 32.6%. Melatonin significantly reduced iNOS concentrations both in serum (59.7%) and liver (57.8%) but did not affect endothelial isoform enzyme activities neither in serum, nor in liver. The histopathological liver lesions observed in the CCl4/melatonin group were less severe than those seen in the CCl4 group. Conclusions: we demonstrated an ameliorating effect of melatonin on prooxidants and antioxidants, NO-NOS systems balance, mitochondrial function and histopathological lesions in the liver in rats with CCl4-induced hepatitis.

The effect of the in ovo application of selected prebiotics and synbiotics on the humoral immune response against T-dependent (SRBC) and T-independent (dextran) antigens and delayed-type hypersensitivity (DTH) to phytohemagglutinin was studied. On the 12th day of incubation, 800 eggs (Ross 308) were divided into five groups and injected into the egg air chamber with prebiotic inulin (Pre1), Bi2tos (Pre2), a synbiotic composed of inulin and Lactococcus lactis subsp. lactis IBB SL1 (Syn1), a synbiotic composed of Bi2tos and L. lactis subsp. cremoris IBB SC1 (Syn2), and physiological saline (control group; C). The chickens were immunized twice at the 7th and 21st day of life with SRBC and dextran. A DTH test was performed on the 7th, 21st, and 35th day. The application of prebiotics and synbiotics had no significant effect on the humoral immune response. SRBC-immunized in ovo Pre1- and Pre2-treated chickens showed significantly higher serum IgG levels than the control. A significant effect on the DTH reaction was detected on the 7th (Pre1 < C) and 21st (Pre2 > Syn2) day. However; Bi2tos may transiently stimulate the cellular immune response on the 21st day. It may be concluded that the application of inulin in an egg air chamber on the 12th day of incubation may stimulate the secondary immune response. The inulin-treated group exhibited a lower mortality rate than the control group.

The anti-diabetic effect of ethanolic and aqueous extracts of Imperata cylindrical  rhizomes was investigated in alloxan-induced diabeties in rats. Diabetes was induced by a single 150 mg/kg intraperitoneal dose of alloxan. Rats were divided into five groups with six rats in each group i.e. the normal control group, diabetic control group, standard group (glibenclamide, 10mg/kg, p.o.), Test-I group (200 mg/kg ethanolic extract) and Test-II group (200 mg/kg aqueous extract). The above concerned groups were inoculated on 21st day. On the last day of the experiment, fasted rats were killed by cervical dislocation. The body weight was measured at the initial day and final day. The blood samples were collected for estimation of glucose. The loss of body weight in control group, but recovery was observed in drug treated group. The serum glucose level was significant increased in diabetic rats. However, significant improvement was observed in treated group. The biochemical parameters such as HDL and proteins level were decreased in the control group but maintained in drug treated group. LDL, cholesterol, triglyceride creatinine and urea were significant increase in control group however, reduced level in drug treated group. The present study concluded that ethanolic and aqueous extracts of I. cylindrical  rhizome showed an appreciable effect in reducing the hyperglycemia and the complications associated with diabetes. However, aqueous extract is found more significant in decreasing blood glucose level in comparison to the ethanolic extract. The study results justify the traditional use of the plant as anti-diabetic.

Aim of the Study: This study was undertaken to ascertain if Hibiscus sabdariffa extract can affect the lipid profile (Total cholesterol (TC), triglycerides (TG), high density lipoprotein (HDL), very low density lipoprotein (VLDL), and low density lipoprotein (LDL)) levels in a paracetamol- induced hepatotoxicity using albino Wistar rat as a model. Materials and Methods: Thirty (30) rats used for this study were divided into three groups. Group A (n=10) served as control. Group B (n=10) was administered paracetamol only at a dose of 750 mg/kg body weight. Group C (n=10) was administered paracetamol (dose 750 mg/kg body weight) and aqueous extract of H. sabdariffa (dose 10 ml/kg body weight) of the animal for 3 weeks. All animals were allowed free access to clean drinking water and normal rat chow. Results: Results of the study revealed that TC was significantly lower (p<0.05) in the paracetamol + H. sabdariffa-treated group as compared to paracetamol-treated group and control respectively. Similar trend was observed with TG, VLDL-c, LDL-c and HDL-c. However, the decrease in HDL-c was not statistically significant when compared to control. Conclusion: The presence of bioactive constituents vis; anthocyanins, flavonoids, polyvenols and free radical scavenging properties in H. sabdariffa enabled a hypolipidemic effect on the animals by lowering the levels of serum TG, VLDL-c, LDL-c despite challenge on the liver. However, it was unable to produce significant effect on HDL concentration -very important cholesterol required in high level to maintain homeostasis inside the body. This may be due to the challenge on the liver as a result of the paracetamol abuse.
