Cholesterol is a natural substance with physical characteristic similar to fat but has a steroidal group. The body requires cholesterol in normal amount; however, it will harm the body in excess amount. High cholesterol levels in the blood are dangerous because of the precipitation of cholesterol and other fatty substances resulting in atherosclerosis. Suji leaf (Dracaena angustifolia Roxb.) used as a natural dye has a high flavonoid content that is inferred to have cholesterol-lowering activity. This study aims to test the in vitro activity of suji leaf (Dracaena angustifolia Roxb.) extract in decreasing cholesterol level with various concentrations and to find the effective concentration (EC50). The method of extraction used was remaceration method with 70% ethanol solvent. Analysis of cholesterol-lowering activity was done by Lieberman-Burchard method by making variation of ethanol extract 400 ppm, 500 ppm, 600 ppm, 700 ppm, and 800 ppm. The results showed the percentage of cholesterol-lowering activity by 33.62%, 36.15%, 46.61%, 56.39% and 64.05% respectively. Value of EC50 activity of suji leaf extract is 632.50 ppm.

Objective: Inflammation is a localized reaction in the tissue in response to injury wherein mediators such as leukotrienes are produced as protectiveresponse leukotrienes are synthesized during the metabolism of arachidonic acid by lipoxygenase and they increase capillary permeability andleukocyte adhesion. Artocarpus heterophyllus is used in many medicines due to its natural anti-inflammatory activity. The aim of this study was to usethe bark and leaf extracts of A. heterophyllus to obtain the most active fraction that could inhibit lipoxygenase and to investigate the total phenol andflavonoid levels in these extracts.Methods: Barks and leaves were extracted by a multistage reflux method. An in vitro lipoxygenase inhibition assay was performed by measuring thetotal phenol and flavonoid contents for each fraction.Results: Ethanol extract was found to be the most active extract in each fraction. IC50 values obtained in the two most active fractions were 31.82μg/mL in the ethyl acetate of bark extract and 46.61 μg/mL in the ethyl acetate of leaf extract. This value was higher than that of apigenin standardwhich has an IC50 value of 2.08 μg/mL. The highest phenol content was present in the ethanol leaf extract with 404.903 mg gallic acid equivalent/gextract. Meanwhile, the highest flavonoid content was found in the ethyl acetate of bark extract with 372.362 mg QE/g extract.Conclusion: The bark and leaf extract tests showed that the increase in the total phenol or flavonoid content was proportional to the increase in theenzyme inhibitory activity.

A total of 85 strains of lactic acid bacteria were isolated from corn silage in this study and analyzed in vitro for their cholesterol removal, NPC1L1 protein down-regulation and bile salt deconjugation ability, respectively. Nineteen strains were selected for further analysis for their probiotic potential. Finally, 3 strains showing better probiotic potential were evaluated for their cholesterol-lowering activity in hamsters. The strains showing the greater cholesterol removal and NPC1L1 protein down-regulation activity had no significant effects on serum and hepatic cholesterol levels in hamsters (p > 0.05). However, Lactobacillus plantarum CAAS 18008 (1 × 109 CFU/d) showing the greater bile salt deconjugation ability significantly reduced serum low-density lipoprotein cholesterol, total cholesterol, and hepatic total cholesterol levels by 28.8%, 21.7%, and 30.9%, respectively (p < 0.05). The cholesterol-lowering mechanism was attributed to its bile salt hydrolase activity, which enhanced daily fecal bile acid excretion levels and thereby accelerated new bile acid synthesis from cholesterol in liver. This study demonstrated that the strains showing greater cholesterol removal and NPC1L1 protein down-regulation activity in vitro hardly reveal cholesterol-lowering activity in vivo, whereas the strains showing greater bile salt deconjugation ability in vitro has large potential to decrease serum cholesterol levels in vivo.

AbstractCholesterol is an important sterol in human body tissue which belongs to the lipid group but cannot be hydrolyzed. Cholesterol has various uses in the body such as forming steroid hormones in the hormones estrogen and progesterone. However, if cholesterol levels in the blood are too high, it can cause blockage of blood flow which can lead to atherosclerosis. Lime contains secondary metabolites that function to reduce the increase in cholesterol levels in the blood. The purpose of this study was to determine the anticholesterol activity and to determine the EC₅₀ value of partition n-hexane, methanol, ethanol extract of lime peel (Citrus aurantiifolia) in vitro. Analysis of cholesterol activity is known by measuring cholesterol levels in vitro using Lieberman Burchard reagent. The analytical method used UV-Vis spectrophotometry at a wavelength of 665.0 nm with a series of test sample concentrations of 150 µg/ml; 300 µg/ml; 450 µg/ml; 600 µg/ml and 750 µg/ml. The research data shows that the decrease in cholesterol levels is directly proportional to the increase in the concentration in the sample. The EC₅₀ value of the n-hexane partition was 448.76 µg/ml; methanol partition as much as 448.98 µg/ml and ethanol extract as much as 450.18 µg/ml.Keywords: Anticholesterol; in vitro; lime peel; partition. Jeruk nipis memiliki kandungan metabolit sekunder yang berfungsi untuk mengurangi kenaikan kadar kolesterol dalam darah. Tujuan pada penelitian ini yaitu mengetahui aktivitas antikolesterol dan mengetahui nilai EC₅₀ dari partisi n-heksana, metanol, ekstrak etanol kulit jeruk nipis (Citrus aurantiifolia) secara in vitro. Analisis aktivitas kolesterol diketahui dengan mengukur kadar kolesterol secara in vitro menggunakan pereaksi Lieberman Burchard. Metode analisis menggunakan spektrofotometri UV-Vis pada panjang gelombang 665,0 nm dengan seri konsentrasi sampel uji 150 µg/ml; 300 µg/ml; 450 µg/ml; 600 µg/ml dan 750 µg/ml. Data penelitian menunjukkan penurunan kadar kolesterol berbanding lurus dengan peningkatan konsentrasi pada sampel. The Rambusa plant (Passiflora foetida L) is known to contain compounds for various treatments, empirically the people in Indonesia, especially Central Kalimantan, use the leaves of rambusa known as the cemot plant to reduce cholesterol levels in the blood. Chemically, cholesterol is a complex compound produced by the body with various functions, including producing sex hormones, adrenal cortex hormones, vitamin D, and the production of bile salts that help the intestines to absorb fat. So, if the dose is right or reasonable, the role of cholesterol in the body will be optimal. The purpose of this study was to determine the effect of anticholesterol in rambusa leaf extract. This study is an experimental study. The anticolesterol effect was tested by the modified enzymatic endpoint cholesterol method. The leaf extract of Rambusa was made with a concentration of 0.5 mg/ml, 1 mg/ml and 1.5 mg/ml as a positive control (multiplier) using simvastatin which was then reacted with reagent R1 and read the results using a photometer and UV-spectrophotometer. Vis with a wavelength of 546 nm. The measurement results using a photometer showed standard cholesterol levels (control negative) 200 mg/dl, positive control 195 mg/dl, 144 mg/dl (extract 0.5 mg/ml), 108 mg/dl (extract 1 mg/ml) and 103 mg/dl (extract 1.5 mg/ml). While the measurement of cholesterol absorbance and concentration using a UV-Vis spectrophotometer at a wavelength of 546 nm 0.179�159.8 mg/dl (Control positive), 0.123�109.8 mg/dl (extract 0.5 mg/ml), 0.110�98.2 mg/dl (extract 1 mg/ml), and 0.109�97.3 mg/dl (extract 1.5 mg/ml). As a conclusion, Rambusa leaf extract can reduce cholesterol levels in vitro.

Cholesterol belongs to the non-hydrolyzed lipid group and is the main sterol in body tissues. When cholesterol levels increase, it can cause blockages in blood vessels. Several studies have explained that one of the compounds that can reduce cholesterol is flavonoid compounds, one of the plants containing flavonoid compounds is guava leaf (Syzygium aqueum (Burm.f.) Alston). The purpose of this study was to determine the activity and percent reduction in cholesterol levels and the EC50 value of the ethanol extract of guava leaves (Syzygium aqueum (Burm.f.) Alston) in vitro using UV-Vis spectrophotometry with a wavelength of 665 nm. Anticholesterol activity analysis was carried out using Lieberman-Burchard reagent with a test solution series of 150; 300; 450; 600; and 750 ppm. The results showed that the greater the concentration, the lower the absorbance produced and the higher the percent reduction in cholesterol levels. At a concentration of 750 ppm cholesterol decreased by 58.74%. And the obtained EC50 value of 462 ppm, which means at that concentration the ethanol extract of guava leaves (Syzygium aqueum (Burm.f.) Tujuan dari penelitian ini adalah untuk mengetahui aktivitas dan persen penurunan kadar kolesterol serta nilai EC50 dari ekstrak etanol daun jambu air (Syzygium aqueum (Burm.f.)Alston) secara in vitro menggunakan spektrofotometri UV-Vis dengan panjang gelombang 665 nm. Analisis aktivitas antikolesterol dilakukan dengan menggunakan pereaksi Lieberman-Burchard dengan seri larutan sampeli 150; 300; 450; 600; dan 750 ppm. Hasil penelitian menunjukkan semakin besar konsentrasi, absorbansi yang dihasilkan adalah semakin rendah dan persen penurunan kadar kolesterolnya semakin tinggi. Pada konsentrasi 750 ppm terjadi penurunan kolesterol sebesar 58,74%. Kegunaan labu kuning di Indonesia masih sebatas daging buah yang dapat diolah menjadi panganan seperti kue basah, kolak dan sayur berkuah. Secara empiris, biji labu kuning telah digunakan untuk mengatasi cacingan. Penelitian ini dilakukan untuk mengetahui mortalitas cacing gelang (Ascaridia galli) dalam ekstrak etanol biji labu kuning (Cucurbita moschata Duchesne). Penelitian ini menggunakan 25 ekor Ascaridia galli yang dibagi menjadi 5 kelompok, kelompok I kontrol negatif menggunakan larutan NaCl fisiologis, kelompok II kontrol positif menggunakan larutan pirantel pamoat 0,5 %, kelompok III, IV dan V berturut-turut menggunakan 25 mg/ml, 50 mg/ml dan 100 mg/ml ekstrak etanol biji labu kuning. Parameter penelitian ini ditentukan dengan melihat persentase nilai skor pasca inkubasi 12 jam, 24 jam, dan 36 jam. Skor 3 diberikan apabila seluruh tubuh Ascaridia galli bergerak, skor 2 diberikan jika hanya sebagian tubuh Ascaridia galli bergerak, skor 1 jika Ascaridia galli diam tetapi masih hidup, dan skor 0 apabila Ascaridia galli mati. Hasil uji in vitro dengan perlakuan 25 mg/ml ekstrak etanol biji labu kuning menyebabkan kematian 3 ekor Ascaridia galli  atau 60% pasca inkubasi 36 jam, sedangkan ekstrak etanol biji labu kuning dengan perlakuan 50 mg/ml, 100 mg/ml dan kelompok kontrol positif mengakibatkan kematian 4 ekor Ascaridia galli atau 80% pasca inkubasi 36 jam. Dari hasil penelitian disimpulkan bahwa ekstrak etanol biji labu kuning (Cucurbita moschata Duchesne) dosis 25 mg/ml, 50 mg/ml, dan 100 mg/ml secara in vitro dalam waktu 36 jam mampu mengakibatkan mortalitas Ascaridia galli. The use of yellow pumpkin in Indonesia is still limited to fruit meat that can be processed into snacks such as soggy cakes, porridge and vegetable soup. This research was conducted to determine the mortality of Ascaridia galli in ethanol extract of yellow pumpkin seeds (Cucurbita moschata Duchesne). This study used 25 Ascaridia galli which were divided into 5 groups, group I was negative control using physiological NaCl solution, group II was positive control using 0.5% pirantel pamoate solution, group III, IV and V respectively used 25 mg / ml, 50 mg/ml and 100 mg/ml ethanol extract of yellow pumpkin seeds. The parameters of this study were determined by looking at the percentage of post-incubation scores 12 hours, 24 hours, and 36 hours. A score of 3 is given if the whole body of Ascaridia galli moves, a score of 2 is given if only part of the body of Ascaridia galli moves, a score of 1 if Ascaridia galli is still but still alive, and a score of 0 if Ascaridia galli dies. In vitro test results with 25 mg/ml ethanol extract of pumpkin seeds caused 3 deaths of Ascaridia galli or 60% after incubation for 36 hours, while ethanol extract of yellow pumpkin seeds treated with 50 mg / ml, 100 mg/ml and positive control group resulting in the death of 4 Ascaridia galli or 80% after 36 hours incubation. From the results of the study concluded that the ethanol extract of yellow pumpkin seeds (Cucurbita moschata Duchesne) doses of 25 mg / ml, 50 mg / ml, and 100 mg / ml in vitro within 36 hours can lead to Ascaridia galli mortality.

Various strategies have been employed in the treatment and management of Salmonella infection however, Salmonella strains have gained resistance to antibiotics. This study was to determine in vitro anti-Salmonella activity of Boerhaavia diffusa leaf extract against clinical isolate of Salmonella typhi and Salmonella typhi ATCC 14028. The aqueous and ethanol extracts of B. diffusa were studied for their antibacterial activity against pathogenic Salmonella typhi. This study was carried out between April and September 2018. The in vitro antibacterial activity was performed by agar well diffusion method and broth dilution using spectrophotometric method and the results were expressed as the average diameter of zone of inhibition of bacterial growth around the well and optical density respectively. It was observed that aqueous extract exerted slightly higher activity than ethanolic extract as revealed by the mean diameter of zone of inhibitions at a concentration of 200 mg/ml, the aqueous extract had 35.21±0.47 mm (Salmonella typhi ATCC 14028) compared with ethanol extract 26.41±0.32 mm (clinical). However, in the broth dilution method, ethanol extract significantly (p=0.05) reduced the cell, at 48 hours, the optical density of clinical isolate of S. typhi treated at concentration of 200 mg/ml of extract was 0.47±0.02 nm while at the same concentration of extract, aqueous extract had an optical density of 0.52±0.11 nm respectively. The phytochemical assay revealed that tannin (5.18±0.02 mg/g) and quinone (8.45±0.13 mg/g) in ethanol extract was significantly (p=0.05) higher than aqueous extract while saponin (14.18±0.06 mg/g) was higher in the aqueous extract. The ethanol and aqueous extracts of leaves of B. diffusa whole plant exhibited significant antibacterial activity against both clinical and typed Salmonella typhi. Therefore, the plant extract could be used for the treatment of Salmonellosis, however, the in vivo studies is needed to ascertain the safety of the extract.

Malacca is one of traditional medicine that possesses a potent antimicrobial activity. This study aims to determine the inhibitory activity of Malacca leaf extract on the growth of Salmonella sp in vitro. The bacteria was obtained from Microbiology Laboratory of the Faculty of Veterinary Medicine, Universitas Syiah Kuala. The study was conducted using n-hexane extract, ethyl acetate extract and ethanol of malacca leaves with dilution concentrations of 5%, 25%, and 50%.The inhibitory property of malacca leaf was tested using Kirby-Bauer method. Data were analyzed descriptively. The results of this study indicate that n-hexane extract, ethyl acetate extract and ethanol extract of malacca leaves can inhibit the growth of Salmonella sp. The n-hexane extract of malacca leaves showed a greater inhibition than the ethyl acetate and ethanol extract of malacca leaves. n-hexane extract with a concentration of 5%, 25%, and 50% showed average diameter inhibition of 1.35 mm (weak), 4.97 mm (moderate), and 12.87 mm (strong), respectively ethyl acetate extract with a concentration of 5%, 25%, and 50% showed average diameter inhibition of 2.00 mm (weak), 5.72 mm (moderate), and 7.58 mm (moderate), whereas in ethanol extract were 0.47 mm (weak), 2.58 mm (weak), and 4.35 mm (weak), repectively. The clear zone areas in negative and positive control were 0.00 mm 20.00 mm, respectively. Malacca leaf extract possess inhibitory property against the growth of the Salmonella sp.

Belimbing wuluh merupakan tanaman jenis buah dan obat tradisional. Tanaman belimbing wuluh sudah sering dimanfaatkan masyarakat salah satunya untuk mengobati penyakit seperti batuk dan radang rektum. Tujuan penelitian ini adalah menguji senyawa fitokimia, menganalisis kandungan total fenolik dan flavonoid, serta aktivitas antioksidan dan antiinflamasi secara in vitro pada ekstrak etanol daun belimbing wuluh. Metode yang dilakukan pada penelitian ini adalah ekstraksi daun belimbing wuluh, skrining fitokimia, perhitungan total fenol dan flavonoid, uji aktivitas antioksidan, dan uji aktivitas antiinflamasi. Senyawa fitokimia yang terkandung pada ekstrak etanol daun belimbing wuluh adalah saponin, tanin, steroid, flavonoid, dan alkaloid. Kandungan total fenolik dan flavonoid ekstrak etanol daun belimbing wuluh secara berturut-turut sebesar 39,03 dan 97,28 µg QE/mg. Ekstrak etanol daun belimbing wuluh memiliki aktivitas antioksidan yang tergolong sangat kuat, sementara aktivitas antiinflamasinya terutama ditunjukkan pada konsentrasi ekstrak 200 µg/ ml, memiliki nilai persen inhibisi hemolisis yang paling tinggi. Kesimpulannya, ekstrak etanol daun belimbing wuluh dapat menjadi salah satu sumber antioksidan dan antiinflamasi alami.Ethanol Extracts of Averrhoa Bilimbi Leaf Demonstrated Antioxidative and Anti-inflammatory ActivityAbstractAverrhoa bilimbi, a fruit-bearing plant, has been traditionally used for medicinal purposes such as treatment of cough and rectal inflammation. In this current work, ethanol extract of Averrhoa bilimbi leaf was analyzed, with regard to phytochemical composition, i.e. total phenolic, and flavonoid, antioxidant activity, and in vitro antiinflammation activity. Extraction of carambola leaves, phytochemical screening, total phenolic and flavonoid contens, assay of antioxidant activity, and assay of anti-inflammatory activity were done in this research. As a result, the leaf extract positively contained some phytochemical compounds, i.e. saponin, tannin, steroid, flavonoid, and alkaloid. Furthermore, total phenolic and flavonoid of the leaf extract was found at 39.03 and 97.28 µg QE/mg extract, respectively. Additionally, antioxidant activity of the leaf extract was classified as very strong, while its anti-inflammatory feature at extract concentration of 200 µg/ml exhibited the highest inhibition of hemolysis. In summary, the ability of Averrhoa bilimbi leaf ethanol extracts to act as antioxidative and anti-inflammatory agents was determined and this may open the use for natural antioxidant and anti-inflammatory agents.
