The effects of phosphate on physiological responses and carbohydrate production in Ulva fasciata (Chlorophyta) from upwelling and non-upwelling sites

Abstract Phosphorus is a key macronutrient in macroalgal physiology, including carbohydrate anabolism. To determine the effects of phosphorus on different physiological parameters, we cultivated Ulva fasciata specimens from distinct localities (upwelling and non-upwelling sites) in the presence of different phosphate concentrations (0, 2, and 4 µM PO 4 3 − ${\text{PO}}_{4}^{3-}$ ). After 15 days, growth rates were similar (approx. 12% day−1) and carbohydrate contents had increased in individuals from both sites. In individuals from the upwelling site, carbohydrate contents were high in all treatments (71% dry mass), whereas non-upwelling site individuals cultivated under the highest phosphate concentration showed the lowest carbohydrate content (46% DM). Nevertheless, we observed higher rates of phosphorus uptake in individuals from the non-upwelling site cultivated under the highest phosphate concentration, indicating a stress response to elevated nutrient concentrations. Individuals from both sites cultivated with phosphate maintained healthy photosystems over the experimental period (F v/F m = 0.788), whereas those cultivated in the absence of phosphate showed evidence of photosystem impairment, as indicated by a progressive decline in maximum quantum yield. Altogether, our results indicate that site origin and phosphate concentration influence the carbohydrate content in U. fasciata and that individuals from sites with broad environmental variation, such as upwelling events, can show higher productivity.

Seaweeds are a rich source of health beneficial bioactive nutraceuticals and currently they are under-utilized in Sri Lanka. In the present study, proximate analysis of seaweed varieties Chnoospora minima and Porphyra sp. obtained from Mirissa, Matara, Sri Lanka and Ulva fasciata was taken from Point Dondra Matara, Sri Lanka on June, 2018 were investigated. The moisture content, total fat content, protein content and ash content were determined according to the Official methods of Analysis by Association of Official Analytical Chemists after drying for 8h at 600 C. The results revealed that the moisture contents (%) of Chnoospora minima, Porphyra sp. and Ulva fasciata were 13.24 ± 0.20, 14.30 ± 0.14 and 18.11 ± 0.01 respectively. Total fat contents (%) of Chnoospora minima, Porphyra sp. and Ulva fasciata were 0.21 ± 0.11, 0.19 ± 0.03 and 0.28 ± 0.05 respectively. Protein contents (%) of Chnoospora minima, Porphyra sp. and Ulva fasciata were 13.70 ± 0.2, 21.14 ± 0.04 and 11.84 ± 0.1. Total ash contents (%) of Chnoospora minima, Porphyra sp. and Ulva fasciata were 17.20 ± 0.24, 5.40 ± 0.7 and 18.05 ± 0.21 respectively. Total carbohydrate content (%) was analyzed according to the Dubois method. Chnoospora minima, Porphyra sp. and Ulva fasciata showed total carbohydrate content (%) as 3.87 + 0.66, 20.59 ± 0.24 and 7.68 ± 1.16 respectively. Moreover, the sulphate content was analyzed according to the precipitate method. Chnoospora minima, Porphyra sp. and Ulva fasciata showed 1.45 ± 0.35, 2.75 ± 0.07 and 4.54 ± 0.06, sulfate contents (%) respectively. In conclusion, highest ash content which indicates a good mineral content was observed in Ulva fasciata and Chnoospora minima. Fibre, protein and carbohydrate contents are significantly different among the 3 samples. Highest fibre content was observed in Chnoospora minima. Highest protein and carbohydrate contents were observed in Porphyra sp. But there is no significant difference in fat contents among the three samples.

Choosing the best season collection of cuttings is essential to have success with propagation, due mainly to carbohydrate content, since is a source of energy for roots and shoots development. In literature, there are no papers that associate the collection season of cuttings with carbohydrate contents in figs. Therefore, the current study aimed to evaluate carbohydrate contents and season collection of cuttings to propagate ‘Roxo de Valinhos’ fig tree. The experiment was conducted in a greenhouse; cuttings were collected in the first week of the following months: June, July, August and September. The design was in completely randomized blocks, with four replicates of 30 cuttings collected per season. The following traits were evaluated: carbohydrate content; percentage of root cuttings, bud cuttings, cuttings without bud and root; number of buds, leaves and roots per cutting; length of the largest root and bud; bud diameter; bud and root dry mass; and root volume. Results indicated high carbohydrate content in cuttings that were collected in August; therefore, the best month to collect cuttings of ‘Roxo de Valinhos’ fig tree in the city of São Manuel, state of São Paulo. This result is due to high percentage of rooted cuttings and better development. Furthermore, carbohydrate content mainly affects root growth; besides that, reducing sugars are the most important feature in the development of cuttings.

Pollution abatement through phosphorus and nitrogen retention is a key ecosystem service provided by streams. Human activities have been changing in-stream nutrient concentrations, thereby altering lotic ecosystem functioning, especially in developing countries. We estimated nutrient uptake metrics (ambient uptake length, areal uptake rate, and uptake velocity) for nitrate (NO3–N), ammonium (NH4–N), and soluble reactive phosphorus (SRP) in four tropical Cerrado headwater streams during 2017, through whole-stream nutrient addition experiments. According to multiple regression models, ambient SRP concentration was an important explanatory variable of nutrient uptake. Further, best models included ambient NO3–N and water velocity (for NO3–N uptake metrics), dissolved oxygen (DO) and canopy cover (for NH4–N); and DO, discharge, water velocity, and temperature (for SRP). The best kinetic models describing nutrient uptake were efficiency-loss (R2 from 0.47–0.88) and first-order models (R2 from 0.60–0.85). NO3–N, NH4–N, and SRP uptake in these streams seemed coupled as a result of complex interactions of biotic P limitation, abiotic P cycling processes, and the preferential uptake of NH4–N among N-forms. Global change effects on these tropical streams, such as temperature increase and nutrient enrichment due to urban and agricultural expansion, may have adverse and partially unpredictable impacts on whole-stream nutrient processing.

A trial was carried out in Campinas, Brazil, from August 2005 to August 2006, in order to analyze the effect of two propagation methods (seeds and cuttings) on the development of Pfaffia glomerata (Spreng.) Pedersen. The experimental design was a completely randomized block, with five replications, and two methods of plant propagation. Plant height, aerial part dry mass and root dry mass were evaluated. Plants propagated from cuttings showed greater accumulation of dry mass on the aerial part. Root dry mass accumulation curves showed an exponential pattern, and at the end of the experimental period, the average of roots dry mass for both treatments were similar, up to 349,65 g plant-1.

ABSTRACT: The objective of the following research was to investigate the physiological responses of two bahiagrass genotypes (Paspalum notatum Flügge) submitted to water restriction, induced by polyethylene glycol (PEG6000), during seed germination. Seeds of cv. Pensacola (diploid) and Bagual ecotype (tetraploid) were placed on paper moistened with osmotic solutions at potentials of 0.0; -0.1; -0.2 and -0.3 MPa. Composed of two subsamples of 100 seeds and four experimental units per treatment, the germination test was conducted in a germination chamber under alternating temperatures of 30 °C (8 hours) and 20 °C (16 hours), with presence of light during the higher temperature. Final germination percentage and germination speed index of the seeds, as well as early growth, activity and expression of antioxidant enzymes of seedlings were evaluated. The water restriction induced by PEG6000 reduced the percentage of germination in Pensacola (from 74% to 59%) and Bagual (34% to 10%). For the latter, the evaluation of seedlings showed higher dry mass in relation to cv. Pensacola (1.7 times higher in root and 1.6 times in shoot). In response to moderate water restriction, the genotypes showed increases in root growth (1.7 times higher in -0.1 compared to 0.0 MPa), both in expression and enzymatic activity (superoxide dismutase and catalase).

One of the bottlenecks in relation to the production of forest seedlings is irrigation, especially in less-tech nurseries. The small volumes of substrate, where the seedlings develop, lead to low water storage. This fact can lead to water deficit and significant losses in the production of seedlings, generating even mortality. The objective of this study was to determine the best irrigation frequencies for Eucalyptus grandis seedling production in less-tech nurseries. The experiment was conducted between 2013/08/04 and 2013/23/07 (106 days) and conducted in Chapadão do Sul county, Brazil. The experiment was set up in a randomized complete block design, with five replications. Four irrigation frequencies were tested: IF1 (one daily irrigation - 11:00 a.m.), IF2 (two daily irrigations - 11:00 a.m. and 7:00 p.m.), IF3 (three daily irrigations - 07:00 a.m., 11:00 a.m. and 7:00 p.m.) and IF4 (four daily irrigations - 07:00 a.m., 11:00 a.m., 3:00 p.m. and 7:00 p.m.). The sample units were composed of eight seedlings in 50 cm3 conical tubes filled with soil and vermiculite in a volume ratio of 1:1. The irrigation depth was estimated by reference evapotranspiration (Penman-Monteith) multiplied by a crop coefficient equal to two. Plant height, number of definitive leaves, shoot dry mass (root and total), seed quality index, survival and efficiency of water use by eucalyptus seedlings were evaluated. The average daily irrigation depth in the experimental period was 5.2 mm. Based on the results, it is recommended for eucalyptus seedling producers, in less-tech nursery, irrigation management twice per days. (11:00 a.m. and 7:00 p.m.).

This study was carried out during 2012-2014 to determine the effect of locally available casing materials with association of bio-inoculant Pseudomonas putida. Six different combinations of casing mixtures were amended for evaluating its effect on yield, biological efficiency, protein and carbohydrate contents of Agaricus bisporus. A mixture of FYM + GLS + Vermi-compost + P. putida gave higher mushroom yield, biological efficiency, protein and carbohydrate content. It gave better yield (1306 g), biological efficiency (28.7%), protein (34.07%) and carbohydrate content (5.07%) respectively when compared with other treatments. In addition, waste tea leaves took minimum period (33.00 days) for initiation of pin head when compared with others. Locally available casing materials along with P. putida incorporated in the casing soil can be an important factor to obtain maximum and assured yield in mushroom cultivation.

Phosphorus is generally considered the limiting nutrient for algae growth in the aquatic environment. The phosphorus uptake for algae can give information for bioavailability-P. The contribution of phosphorous to bioavailability was evaluated using uptake-P in water solution under anaerobic conditions. The more is the incubation time, the more is the phosphate bioavailability. The maximum bioavailability occurred at 14 days with a phosphate concentration of 50 ppm resulting in P-available of 18.41 ppm. There was a linear correlation between P-available with P-medium with R2 = 0.97. The phosphorus bioavailability can be resulted from the equation: [P-Available] = 0.3789 x [P-medium]. The phosphate absorbed by the algae Oscillatoria sp in anaerobic conditions with initial concentrations of 10, 20, 30, 40 and 50 ppm was 5.46, 8.65, 11.63, 17.99 and 18.41 ppm, respectively. The maximum efficiency adsorption occurred in the concentration of 30 ppm with 49.18% adsorption.

This study aimed to evaluate the effect of cattle manure on young plants of Khaya senegalensis submitted to different irrigation intervals. A greenhouse experiment was carried out during 2017 cold season, based in a 5x3 factorial arrangement – cattle manure fertilizing with 0, 10, 20, 30 and 40% doses, and 3-day, 6-day and 12-day irrigation intervals. Water potential, relative water content (RWC), water consumption (WC), water use efficiency (WUE), chlorophyll content estimation (SPAD index), plant height, stem diameter, plant total dry mass and free amino acid and proline content were measured. Irrigation intervals had no significant effect on physiological traits and plant water relationships, except for WUE, which was higher under 12-day irrigation interval. This was assumed as an indication of non-occurrence of water stress, even under the longest irrigation interval. Cattle manure, in turn, played a significant effect on these traits, except for water potential, irrespective of irrigation intervals. Increasing cattle manure doses improved RWC (just doses up to 20%), reduced WC and increased WUE, SPAD index, and both free amino acid and proline contents. On the other hand, growth-related parameters were not affected by cattle manure, but a significant effect of irrigation intervals was observed. Both plant height and total dry mass were lower under 3-day and 6-day irrigation intervals. We suspect a possibly low evapotranspiration, given the relatively low temperatures of that season (14-16 ºC), which may have reduced the demand for irrigation in the experimental period. Thus, soil moisture under the shorter irrigation intervals may have increased until reach to a close to saturation level, limiting the amount of soil oxygen and hampers nutrient uptake.

Variations in the photosynthetic pigments and total carbohydrate contents of needles of different age classes (current-year, 1-year-old, 2-year-old and 3-year-old) of Pinus nigra subsp. pallasiana (Lambert) Holmboe trees in a young natural stand were investigated during the growing season. In current-year needles, total carbohydrate content was lower during June and July when the needle growth continued than in older age classes, but it was similar to other age classes in the months of August to October. Seasonal patterns of variations in total carbohydrate content were almost similar in 1-, 2-, and 3-year-old needles. Chlorophyll and carotenoid contents increased from May to June, remained relatively constant or declined slightly during summer and autumn in 1-, 2-, and 3-year-old needles. In October, the pigment content was highest in 1-year-old needles, and lowest in 3-year-old needles. Our study indicated that total carbohydrate and pigment contents were affected by needle age classes and seasons.
