The study compares the effect of one-time administration of nonsteroidal and/or steroidal anti-inflammatory combinations by topical or intramuscular (IM) routes to pigeons with monosodium urate (MSU)-induced arthritis. Forty-five adult domestic pigeons were assigned into nine equal groups: NC, negative control; PC, positive control with arthritis; sham, sham control; T1, meloxicam + hydrocortisone; T2, dexamethasone + piroxicam; T3, meloxicam + dexamethasone; T4, hydrocortisone + piroxicam; T5, dexamethasone + hydrocortisone; T6, meloxicam + piroxicam. Arthritis was also induced in T1 to T6 birds. Meloxicam and dexamethasone were administered by IM injection and the other drugs topically right after the induction of arthritis. Different drug combinations significantly decreased one-leg standing time. Induction of arthritis significantly increased TNF-α and IL-6 levels in synovial fluid and serum corticosterone and epinephrine in the PC group. Administration of drugs to birds of Groups T1 and T5 did not significantly change corticosterone concentration, while all different drug combinations decreased epinephrine level. Drug combinations that demonstrated better analgesic effect more strongly reduced serum epinephrine concentration. Meloxicam + hydrocortisone was the most effective combination in reducing inflammatory cytokines. In conclusion, one-time combination therapy with anti-inflammatory agents was effective in the acute management of inflammatory pain due to MSU-induced arthritis in pigeons, even by the topical route.

Background. Root canal treatment is a main role in decreasing infection from root canal and pulp. The main cause of periapical damage mostly are bacteries. E.faecalis is a bactery that is found as an etiology of endodontic treatment failure. Cell wall of this bacteria is containing Lipoteichoic acid (LTA). LTA can penetrate into the periradicular tissue, act as endotoxin in host and cause periradicular inflammation then lead to bone destruction. LTA stimulates immunology reaction that produce Tumor Necrosis Factor alpha (TNF-α) and Transforming growth factors beta (TGF-ß). TNF-α is a main mediator and also have an important role in inflamation response otherwise TGF-ß is working as a multifunction  regulator of cell growth and differentiation during reforming and remodelling.  Purpose. The aim of this study is to know about the expression of TNF-α and TGF-ß during the periapical tissue damage due to induction of E.faecalis. Method. This study used laboratory experimental with the post test only control group design. A total of 30 male rats were randomly divided into 3 main groups, Group A (control negative) : normal tooth. Group B (control positive) : every tooth was induced only by sterile BHI-b. Group C (treated group) : every tooth  was induced by 10 μl BHI-b E.faecalis ATCC212(106 CFU). The animals were sacrificed 21 days later and prepared for histological examination of tissue damage, then we did the immunohistochemistry  followed by calculation on the light microscope. Result. The analysis revealed that the expression of TNF-α at treated group are higher than negative control and positive control but the expression of  TGF-ß at treated group are higher than the negative control group but lower than positive control. Conclusion. From this study we know that the expression of TNF-α and TGF-ß are changing during the periapical tissue damage that induced by E.faecalis.

 Objective: The objective of this study was to evaluate the anti-inflammatory activity in acute and subacute models of inflammation from ethanolic fruit extract of Asam kandis (Garcinia xanthochymus Hook. f. ex T. Anderson) in animal (rats) models.Methods: Pleliminary phytochemical screening was carried out by using standard procedures.. Assessment of acute and subacute models of inflammation was using carrageenan-induced paw edema method and cotton pellet granuloma method using three dosage treatments; 200 mg/kg BW, 400 mg/kg BW, and 800 mg/kg BW along with a negative control group (0.5% Na CMC) and positive control (Na diclofenac 2.25 mg/kg BW). The inhibition period was observed at 30, 60, 90, 120, 150, and 180 min time intervals.Result: The phytochemical screening showed that the ethanolic fruit extract from Asam kandis contain contains flavonoids, glycosides, steroids, and triterpenoids. The anti-inflammatory result showed that the strongest inhibition produced by ethanolic fruit extract of Asam kandis occurred on the dosage of 800 mg/kg BW compared to the other doses (200 and 400 mg/kg BW) throughout the observation period.Conclusion: This finding indicated that ethanolic fruit extract of Asam kandis (G. xanthochymus Hook. f. ex T. Anderson) might become an interesting candidate for treatment of inflammation.

Trigona honey (TH) is well known for its therapeutic characteristics. To date, the study of Trigona honey as a prophylactic or immune booster prior to the bacterial infection of the invivo model is not well covered. This study aims to investigate anti-inflammatory and immune activities in Wistar rats infected with respiratory infection following with Trigona honey. 25 Wistar rats were assigned to possitive groups, negative control group, positive control group was fed TH (5 g / kg body weight) orally, the untreated group was infected with Staphylococcus aureus to induce respiratory infection, the treated group has been infected with S. aureus followed by treatment with TH at a dose of 1.5 ×108 CFU / mL and the preventive group ingested TH one week before S. aureus infection. Blood was obtained for biochemical analysis. Lung tissues have been collected for molecular examination. The results showed a significant decrease in serum levels of ALT, AST, urea and creatinine in the preventive and treated groups, serum IgG increased significantly (P<0.05) in the preventive and treated groups, IFN-y increased in the preventive group while decreased in the treated group, and IL-8 increased in the treated group while decreased in the preventive group. The mRNA expression of AGP is up-regulated in the positive control, preventive and treated groups. The α2-MG, TNF-α , and mRNA expressions showed lower regulation after administration of TH in preventive and treated groups. The results show the ability of TH to counteract immune and inflammatory changes in serum levels and gene expressions.

Abstract Background: Consumption of herbal flavonoids instead of chemical drugs has increased in recent years due to fewer side effects and affordability. In this study, the effect of apigenin was investigated on inflammation induced by lipopolysaccharide in male rat's serum by measuring the pro-inflammatory cytokines, i.e., IL-1β, IL-6, and TNF-α.Methods: 90 male Wistar rats weighing 200 ±2 grams were used and divided into control, sham (solvent), and positive control (dexamethasone 15 mg/kg. ip), and 3 experimental groups which received 5, 15 or 30 mg/kg of apigenin, intraperitoneally. In 30 minutes after interventions, lipopolysaccharide (LPS) [30 μg/kg. ip] was injected. Then, at 4, 12- and 24-hour intervals, rats were anesthetized, and blood samples were prepared intracardially. Samples were centrifuged, and serums were separated and stored at -80 ° C. Measurement of IL-1β, IL-6, and TNF-α were conducted by the enzyme-linked immunosorbent assay (ELISA) method. Data were analyzed using the SPSS software version 19.Results: Pre-injection of apigenin at 5 mg/kg dosage were reduced TNF-α and IL-1β levels at 24-hours after LPS injection, compared to control (for both P <0.05). Pre-injection of 15 mg/kg of apigenin was reduced IL-6 level at 24-hours after LPS injection (P <0.05). Pre-injection of 30 mg/kg of apigenin were reduced TNF-α level at 4- (P <0.05), 12- (P <0.01) and 24- (P <0.01) hours, IL-1β level at 24-hours (P <0.01), and IL-6 level at 4- (P <0.05) and 24- (P <0.01) hours after LPS injection.Conclusions: Apigenin reduces proinflammatory cytokines in serum in acute inflammation induction. This impact is close to the dexamethasone effect as an anti-inflammatory steroid drug.

Ipomoea pes-caprae is one of the plants in Indonesia that has the potential as a traditional herbal medicine to treat inflammation. Hence, the objective of this study was to evaluate the effect of I. pes-capraeleaf extract as anti-inflammatory in edema rats induced by 5% egg white solution. The experimental design used was a Completely Randomized Design (CRD) consisted of 5 treatments, each consisted of 5 rats.The treatment consisted of KN = negative control, KP = positive control, giving  of 25 mg diclofenac sodium, K 100 = giving 100 mg kg-1body weight (BW) of I. pes-capraeleaf extract , K300 = 300 mg kg-1BW of I. pes-capraeleaf extract, and K500 = 500 mg kg-1BW of I. pes-capraeleaf extract. The anti-inflammatory data obtained were tested statistically with a one-way ANOVA test at  99% confidence level and followed by Duncan's test. The ANOVA test results showed that the giving ofI. pes-capraeleaf extract had a very significant effect (p 0.01) on the percentage of inflammatory, obtained Fcount Ftable, 21.46 4.79 at the level of α = 0.01. The treatment of I. pes-capraeleaf extract of 500 mg kg-1BW had the effect of decreasing the volume of edema with the positive control variable using diclofenac sodium 25 mg. It is concluded that I. pes-capraeleaf extract can be used as an anti-inflammatory by decreasing rat feet edema volume. I. pes-capraeleaf extract has a prospect for non-immunological inflammatory natural drug candidates.

In this study, 34 Traditional Chinese Medicine (TCM) compounds were screened for potential anabolic and anti-inflammatory properties on human osteoarthritic (OA) chondrocytes. The anabolic effects were assessed by measuring the glycosaminoglycan (GAG) relative to the DNA content using a 3D pellet culture model. The most chondrogenic compounds were tested in an inflammatory model consisting of 3 days of treatment with cytokines (IL-1β/TNF-α) with or without supplementation of TCM compounds. The anti-inflammatory effects were assessed transcriptionally, biochemically and histologically. From the 34 compounds, Vanilic acid (VA), Epimedin A (Epi A) and C (Epi C), 2′′-O-rhamnosylicariside II (2-O-rhs II), Icariin, Psoralidin (PS), Protocatechuicaldehyde (PCA), 4-Hydroxybenzoic acid (4-HBA) and 5-Hydroxymethylfurfural (5-HMF) showed the most profound anabolic effects. After induction of inflammation, pro-inflammatory and catabolic genes were upregulated, and GAG/DNA was decreased. VA, Epi C, PS, PCA, 4-HBA and 5-HMF exhibited anti-catabolic and anti-inflammatory effects and prevented the up-regulation of pro-inflammatory markers including metalloproteinases and cyclooxygenase 2. After two weeks of treatment with TCM compounds, the GAG/DNA ratio was restored compared with the negative control group. Immunohistochemistry and Safranin-O staining confirmed superior amounts of cartilaginous matrix in treated pellets. In conclusion, VA, Epi C, PS, PCA, 4-HBA and 5-HMF showed promising anabolic and anti-inflammatory effects.

The inflammatory response is one of natural process in the body to protect itself following tissue injury, but it can cause discomfort. Noni (Morinda citrifolia L.) leaf known as a traditional medicament to help reduce the inflammatory effect. The leaves empirically applied as a wrapping on fever or wound. The purpose of this study was to identify the anti-inflammatory effect of topical noni leaf extract paste in 5% and 10% concentration by examination of Wistar rat paw edema induced by λ-carrageenan. Twenty-four Wistar rats divided into four groups, which were negative control, positive control with diclofenac sodium 1% gel, 5% noni leaf, and 10% noni leaf paste groups. Paw edema was induced by intraplantar injection of 1% λ-carrageenan to every rat. Every treatment subsequently applied in the plantar area before injection, and the changed paw volume measured with plethysmometer at minutes 0, 30, 60, and 90. This study was at the Animal Laboratory, Faculty of Medicine, Universitas Jenderal Achmad Yani, Cimahi city in October–December 2017. The result displayed that the minimum volume after 90 minutes was on 5% and 10% noni leaf paste group, which is 1.00 mL. Kruskal-Wallis test result of inflammatory percentage was significantly different among every group in each examination time (p<0.05). Post-hoc test showed that inflammatory reduction on paw edema with noni leaf paste application on both concentrations were significantly different compared to the negative control. However, it was not different from the positive control group. This study showed that application of noni leaf paste in 5% and 10% concentration could help reduce inflammatory response on skin possibly by the active anti-inflammatory ingredients of noni leaf. EFEK PASTA EKSTRAK DAUN MENGKUDU (MORINDA CITRIFOLIA L.) TOPIKAL TERHADAP EDEMA KAKI TIKUS GALUR WISTAR YANG DIINDUKSI KARAGENANInflamasi adalah proses alami tubuh untuk melindunginya setelah cedera, namun hal tersebut dapat menyebabkan ketidaknyamanan. Daun mengkudu (Morinda citrifolia L.) dikenal sebagai obat tradisional untuk menurunkan efek inflamasi yang secara empiris digunakan untuk mengobati demam dan luka. Tujuan penelitian ini adalah mengetahui efek anti-inflamasi pasta ekstrak daun mengkudu melalui pemeriksaan edema kaki tikus yang diinduksi karagenan-λ. Dua puluh empat tikus galur Wistar dibagi menjadi 4 kelompok, yaitu kontrol negatif dengan akuades, kontrol positif (aplikasi gel Na diklofenak 1%), serta perlakuan pasta daun mengkudu 5% dan 10%. Edema dibuat dengan menginjeksi intraplantar tikus dengan 1% karagenan-λ pada tiap kelompok. Tiap-tiap perlakuan diaplikasikan sebelum tikus diinjeksi dan perubahan volume kaki tikus diukur dengan pletismometer di menit ke-0, 30, 60, dan 90. Penelitian ini dilakukan di Laboratorium Hewan, Fakultas Kedokteran, Universitas Jenderal Achmad Yani, Kota Cimahi pada Oktober–Desember 2017. Hasil pengukuran memperlihatkan penurunan volume edema kaki terkecil setelah 90 menit pada kelompok pasta daun mengkudu 10%, yaitu 1,00 mL. Hasil Uji Kruskal-Wallis terhadap persentase inflamasi berbeda nyata pada tiap kelompok dan tiap waktu pengamatan (p<0,05). Hasil uji beda menunjukkan bahwa penurunan inflamasi kaki tikus pada perlakuan pasta daun mengkudu kedua konsentrasi berbeda nyata dibanding dengan kontrol negatif, tetapi tidak berbeda dengan kontrol positif. Penelitian ini menunjukkan bahwa pasta daun mengkudu 5% dan 10% dapat membantu menurunkan reaksi inflamasi kulit dan efeknya sejalan dengan Na diklofenak karena zat aktif yang bersifat anti-inflamasi dalam daun mengkudu.

AbstractBackgroundAlcoholic liver disease (ALD) is regarded as a global health problem with limited therapeutic options. Previous studies highlighted some anticancer, antiviral, and hepatoprotective activities of limonoids, but the effects of these compounds on ALD remain unknown. The present study aimed to evaluate the effect of some natural limonoids on ethanol-induced liver injury.MethodsThirty-five albino mice (Mus musculus) were administered with 40% ethanol in the presence or absence of the different limonoids [including three havanensin-type limonoids, TS1, TS3, Rubescin D isolated from an African medicinal plant, Trichilia rubescens Oliv. (Meliaceae), and one limonin], or silymarin at 50 mg/kg for 3 days. Thereafter, the effect of the most active compound was evaluated in a chronic model of ALD. For this purpose, 24 mice with each group consisting of six mice were administered orally with 40% ethanol and limonoid at different doses (50, 75, and 100 mg/kg) for 28 days. Finally, biochemical parameters such as alanine aminotransferase (ALT), superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), triglyceride (TG), and tumor necrosis factor α (TNF-α) levels were quantified in liver homogenates.ResultsAll tested limonoids significantly (p < 0.01) reduced ALT levels relative to the negative control in the acute model. However, in comparison to other limonoids, limonin at 50 and 75 mg/kg significantly reduced TG, MDA, and TNF-α levels (1.8-fold); alleviated leukocyte infiltration in liver tissue; significantly increased the activity of SOD; and decreased those of CAT better than silymarin used as a positive control at 50 mg/kg.ConclusionsThese data suggest that limonin possesses protective effects on long-term alcohol poisoning partially due to antioxidant and anti-inflammatory mechanisms.

Objective: Colorectal cancer (CRC) contributes to 9.7% of all cancer, and its pathogenesis is related to chronic inflammation. However, current cancer therapy options are lacking, and peptide in food has become popular among researchers because it is cheap, easy to get, has a low toxicity, and is a promising cancer-preventing agent. This research aimed to investigate whether lunasin from soybeans can reduce the expression of pro-inflammatory cytokine TNF-α in colonic epithelial cells. Methods: Thirty Swiss Webster mice were randomly allocated to six groups. One group was normal, and in five groups, carcinogenesis was induced using azoxymethane (AOM) and dextran sodium sulfate (DSS). The mice were then given nothing (negative control), aspirin (positive control), and lunasin-rich soybean extract (LSE) in three different doses (250, 300, and 350 mg/kgBW) for four weeks. Distal colon tissue was immunohistochemically stained and then observed under a light microscope with 400X magnification to count epithelial cells, based on their color. The index was calculated using optical density scores. Results: LSE was shown to decrease the expression of tumor necrosis factor (TNF)-α. This decrease was statistically significant between the negative control and a dose of 300 mg/kgBW (p=0.016) and 350 mg/kgBW (p=0.009), yet it was not significant with a dose of 250 mg/kgBW (p=0.754). Conclusion: a dose of 300 mg/kgBW or higher of LSE can reduce the expression of TNF-α.

Abstract The study aimed to investigate the possible anti-inflammatory activity of larch sawdust as feed supplement in lactating sows’ diet and its possible effect on the prevalence of Postpartum Dysgalactia Syndrome under field conditions. In a Greek farrow-to-finish pig farm, fifteen sows were randomly and equally allocated to a negative control group (NC group), a positive control group (PC group), and a treatment group (LT group). The animals of the first two groups received 99% basic diet and 1% corn starch, while LT group animals received 99% basic diet and 1% larch sawdust. The whole trial period lasted 35 days (7 days prior to farrow – day of weaning). At parturition day, animals of the PC group received 2 ml of an anti-inflammatory drug intramuscularly (meloxicam, Metacam®, Boehringer Ingelheim Vetmedica), while the animals of both other groups, received 2 ml of normal saline. Results showed insignificant differences among experimental groups for parameters such as post-partum rectal temperature and piglets performance. On the contrary, a significant increase of mean milk lactation index was observed in LT and PC groups on the 4th day of lactation period, when compared with NC group (p=0.014). Additionally, mean IL-6 concentrations in blood in the LT group showed a tendency for reduction when compared with those found in NC, and insignificant difference (p>0.05) when compared with those observed in PC group 24 hours postpartum. Moreover, the respective TNFα mean level in the LT group at 24 and 72 hours after parturition was similar to that found in PC group, respectively) and significantly lower than that determined in the NC group (p=0.003, p=0.024. The results suggest a possible anti-inflammatory effect of larch sawdust in sows.
