In this paper the author considers the possibility of using an automated method of diagnostics of the causative agent of trichinosis Trichinella spp. at wild trade animals. In the course of trichinosis monitoring in the Central region of Russia naturally infested carcasses of animals were selected. The infected animals were stored at a low (-5C) temperature in the climatic camera before carrying out researches. With the help of AVT devices diagnostic tests on trichinosis of different types of trade animals muscular tissue samples (diaphragm, masseter, extremities muscles, tongue) were conducted. During the research refrigerated and later cooled muscles samples were used. Samples weighing 500,5 g. were used for the research. To get qualitative mincemeat fat and connective tissue were removed from muscular tissue. Pepsin produced by Shako (Rostov region) was used for an artificial peptoliz. Diagnostic samples of boars, foxes, martens increased the weighing mass to 40 minutes, of wolves to 50 minutes and of raccoon dogs to 60 minutes. Further tissue was fermented and weighing mass of the researched samples was decreased. The optimal period of time was found for trichinellascopy test of wild animals muscular tissue by the automated method on AVT devices.

AbstractBladder cancer (BCa) patients are diagnosed by cytology and cystoscopy. However, these diagnostic tests bear some limitations. We sought for reliable biomarkers to better determine BCa extension. Prostate-specific membrane antigen (PSMA) appears to fulfill this requirement in prostate cancer but its role in BCa has not been established yet. We then analyzed 87 bladder tissue samples from 74 patients assessing PSMA expression by immunohistochemistry. The median PSMA expression, exclusively found in tumor neovasculature, in terms of H-score significantly differed between non-tumor samples and tumor samples (p = 0.00288) showing a higher neovasculature-related PSMA expression. No differences were observed in relation to tumor type, grade and stage. BCa neovasculature-related PSMA overexpression may be useful in defining the degree of extension of the neoplasm. In addition, testing PSMA expression by immunohistochemistry may hold theranostic implications both considering anti-angiogenesis agents and radio-labelled PSMA ligands for intracavitary radionuclide therapy. In our opinion, BCa neovasculature-related PSMA overexpression may be considered an apt target for anti-angiogenesis and radionuclide treatment in BCa, once the evaluation of tumor-retention time for the appropriateness of long half-life therapeutic PSMA ligands as radionuclide treatment will be performed.

AbstractCoronavirus disease 2019 (COVID-19) has brought a huge impact on global health and the economy. Early and accurate diagnosis of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections is essential for clinical intervention and pandemic control. This book chapter addresses the evolving approach to the laboratory diagnosis of COVID-19 covering preanalytical, analytical, and postanalytical steps. The rapidly changing dynamics of the COVID-19 pandemic serve as an example which will be important for laboratories to plan for future pandemics. With the quick identification of the causative pathogen and availability of the genome sequence, it will be possible to develop and implement diagnostic tests within weeks of an outbreak. Laboratories will need to be flexible to continuously adapt to changing testing needs and burdens on the healthcare system, plan mitigation strategies for bottlenecks in testing and workflow due to limitations on resources and supplies, and prepare back-up plans now in order to be better prepared for future pandemics.

Abstract This paper reports on the development of an automated method to perform sedimentation tests on a suspension, to study the settling and sedimentation behaviour of particle suspension. The standard method for measuring sedimentation rate is the jar test, but this is burdened with some errors: the possibility of misinterpretation of the interface or subjective readings by technicians. In order to overcome these problems, there are many different methods that exclude subjective mistakes. The proposed solution automatically detects the phase separation boundary and, by use of a moving camera, plots the real-time sedimentation curve. The good agreement of settling curves between the manual method, another CCD image processing method and the current technique demonstrates the reliability of the system. This system can be used for testing different types of suspensions. The article presents a comparison of the commonly used method of image analysis and the proposed solution with a tracking camera, based on the example of a coal suspension.

Abstract Our objective was to identify the direct and indirect presence of Neospora caninum in dairy cattle and their aborted fetuses from Lima, Peru. A total 219 blood samples obtained from dairy cattle with records of spontaneous abortion were collected to detect antibodies against N. caninum in serum with indirect ELISA and search for risk-factor associations. 68 fetal aborted tissue samples of these cows were analyzed by PCR, indirect ELISA and histopathology assay to detect N. caninum presence. The prevalence ratio (PR) and 95% confidence intervals (CI) were estimated. Univariate analysis was performed using the chi-squared test. Among the 68 aborted fetuses collected, 10 (15%) were positive in at least two diagnostic tests. Among 219 serum samples, 46.6% (95% CI: 40.0%-53.3%) were positive. Cows with 4 years or older (PR: 7.10; 95% CI: 4.89-10.67) and multiparous (PR: 1.76; 95% CI: 1.11-2.80) were found to be more likely to possess N. caninum antibodies. This study detects presence of N. caninum in dairy cattle and their aborted fetus from Lima valley, suggesting biosecurity management improve to neosporosis control.

Abstract Background : It is defined that immature platelet fraction (IPF) is the percentage of reticulated platelet (RP) of total platelet count. We have measured an IPF reference range using XE-2100 blood cell counter with upgraded software (Sysmex, Kobe, Japan) and evaluated the clinical utility of this parameter for the laboratory diagnosis of thrombocytopenia due to increased peripheral platelet destruction. Materials and methods : Peripheral blood samples collected into K2EDTA (Beckton Dickinson, Franklin Lakes, NJ, USA) were analysed at Chonbuk National University Hospital. 142 samples from apparently healthy adults (all routine full blood count parameters including platelets within the healthy reference range) were used to establish a normal reference range for the IPF%. The patients are classified 3 group including hypoplastic (consisted of 22 patients undergoing chemotherapy with falling platelet counts and 14 with aplastic anemia), destructive (14 with ITP and 7 with infection), and cirrhotic (40 with cirrhosis of liver). Results : An IPF reference range in healthy individuals was established as 0.4~5.4%, with a mean of 1.7%. The significant increase in IPF values was found in destructive patient group. The cut-off value of IPF is 6.0%, its sensitivity and specificity are 95.2%, 82.9% respectively. Reproducibility was good. Conclusion : A rapid, inexpensive automated method for measuring the IPF% is feasible and should become a standard parameter in evaluating the thrombocytopenic patient.

Abstract Physical activity practiced on a constant, regular basis contributes directly to the improvement of the structure and functions of different organs and body systems. It can also counteract the appearance of some anatomic and functional deterioration and contribute to the correction of certain deficiencies caused by improper conditions of life/work in people’s life. This study analyses two forms of physical exercise - Aerobic Step and Pilates. The goal of the study is to identify the effects of practicing Aerobic Step and Pilates exercises on the bodies of those who practice these forms of physical exercise. The hypothesis of the research is that practicing on constant/regular basis different types of aerobic exercises combined with efficient nutrition ensures optimum physical condition for everyday activities. The activity was carried out at the “Beauty Centre” aerobic hall from Timisoara, Romania. The study was carried out between February and May 2013 on a sample of 25 people subjected to anthropometric and functional measurements (size, weight, hip perimeter and body weight index, percentage of muscular tissue) both before (initial test - It) and after (final test - Ft) Aerobic Step and Pilates programmes. Results show the efficiency of these programmes as pointed out by the diminution of hip perimeter, by normal IMC, and by the increase of the active muscular tissues.

Оптимальный срок посева свеклы столовой для получения корнеплодов с высокой товарностью и закладки их на зимнее хранение в условиях Ростовской области 5 15 июля. При посеве в этот срок улучшается качество корнеплодов, а выход товарной продукции составляет 84 90.In the conditions of field experiments the technological methods of seed growing of table beet in the conditions of the South of Russia are specified. The optimal period of red beet sowing for root crops with high marketability producing for winter storage in the Rostov region is 5 15 of July. When sowing in this period, the quality of root crops improves, and the yield of marketable products is 84 90.

The paper presents data on the problems of monitoring and diagnosing the technical conditions of critical production facilities, such as torpedo ladle cars, steel ladles. The accidents with critical production facilities, such as torpedo ladle cars, lead to losses and different types of damages in the metallurgical industry. The paper substantiates the need for a mathematical study of the operation process of the noted critical production facilities. A Markovian graph has been built that describes the states of torpedo ladle cars during their operation. A mathematical model is presented that allows determining the optimal frequency of diagnostics of torpedo ladle cars, which, in contrast to the existing approaches, take into account the procedures for preventive diagnostics of torpedo ladle cars, without taking them out of service. Dependence of the utilization coefficient on the period of diagnostics of PM350t torpedo ladle cars was developed. The results (of determining the optimal period of diagnostics for PM350t torpedo ladle cars) are demonstrated. The system for automated monitoring and diagnosing the technical conditions of torpedo ladle cars, without taking them out of service, has been developed and described.

Ultrasound-guided sampling methods are usually minimally invasive techniques applied to obtain cytological specimens or tissue samples, mainly used for the diagnosis of different types of tumors. The main benefits of ultrasound guidance is its availability. It offers high flexibility in the choice of sampling approach (transabdominal, transvaginal, and transrectal) and short duration of procedure. Ultrasound guided sampling of pelvic masses represents the diagnostic method of choice in selected patients. We carried out a narrative review of literatures regarding the ultrasound-guided methods of cytological and histological evaluation of pelvic masses as well as the positive and negative predictors for the achievement of an adequate sample.

This article examines how to increase pork meat productivity and quality indicators through the use of crossbreeding. The aim is to increase the meat productivity of animals; reduce losses during production and processing; and improve the criteria for assessing the quality of meat and the organization of its rational use. The purpose of our research is to study the productive quality of the steppe type of pigs, used in the Rostov region in breeding and crossing systems, and the complex of biological and internal indicators in connection with the defects in the quality of pork. Four groups with 16 animals in each were formed: the first group – with the intensive cultivation technology, included pigs of the DM-1 breed, the second – SM-1, the third – the SM-1 × DM-1 crossbreed, the fourth – the DM-1 × SM-1 crossbreed. Analysis of the raw meat showed that the pork had a good ability to emulsify and gel, and had a high nutritional value. The toxicity index in the muscle and fat tissue samples corresponded to the first permissible toxicity group. The study shows the advantages of crossbred individuals over purebred ones, which can be explained by the consequences of heterosis. According to the research results, the use of pig breeding in the industrial production of pork can be recommended in order to improve feedstock and meat productivity, biological and interior indicators, as well as the quality of raw meat.
