<em>During the storage period, cocoa seeds undergo various processes that will affect the content of their food reserves and will have an impact on germination capacity and seeds vigor. The objective of this study is to investigate the changes of biochemical and vigor of cocoa seeds after storage for a maximum of 4 weeks with an interval of 1 week. The experiment was conducted at the Integrated Laboratory and Greenhouse of Balittri, Sukabumi, West Java, from October to December 2019. The completely randomized design with five treatments of seeds storage duration: (a) seeds unstored, (b) seeds stored for 1 week, (c) seeds stored for 2 weeks, (d) seeds stored for 3 weeks, and (e) seeds stored for 4 weeks, and five replications were used in this study. Parameters observed during seed storage were the biochemistry change of seeds and the percentage of germinated seeds, while during seed germination is an indicator of seed vigor. The results showed that storing cocoa seeds for 1-4 weeks decreased the fat content of the seeds, while the free fatty acids, protein, and percentage of germinated seeds increased. Until the 21<sup>st</sup> day after sowing, seed storage increases electrical conductivity but decreases growth rate, maximum growth potential, viability, and hypocotyl length. Seeds stored for one week still good in their vigor (viability of 80%), while seeds stored for 2-4 weeks have decreased their vigor (maximum viability of 54.4%).</em>

ABSTRACT<br /><br />Physiological and biochemical changes in seeds indicates levels of viability and vigor during deterioration. The aims of the research were to study the patterns of biochemical changes in soybean seeds and its relationship to seed vigor during storage under controlled conditions. The experiment was arranged in a completely randomized design using two varieties as a factor which was nested into storage period and replicated four times. Two varieties were Gepak Kuning and Mallika, stored in controlled condition at temperature of 19-22 °C and 64-67% of relative humidity for six months. The seeds were evaluated for protein, peroxide value, electrical conductivity and seed viability. The exponential regression analysis was used to determine the patterns of biochemical changes of each soybean varieties during the storage period. The results showed that the biochemical behaviour of soybean seeds stored under controlled conditions for 24 weeks following the exponential equation of y = a + b expcx, which a, b, and c were constants that has different value for each variety. Electrical conductivity and peroxide value could be used as indicators of seed vigor during storage under controlled conditions for 24 weeks. Seed storage under controlled conditions are to maintain seed vigor &gt;80% up to 12 weeks for Mallika and 24 weeks for Gepak Kuning with low oxidative process and less membrane damages.<br /><br />Keywords:deterioration, storability, vigor<br /><br />

<p>ABSTRACT<br />Information of seed quality during storage can be determined through the actual storage and storability vigor estimation. This study aimed at comparing effective accelerated aging method<br />between physical and chemical, and studying the seed deterioration during storage in ambient (T =28-30 0C, RH=75-78%) and AC (T =18-20 0C, RH =51-60%) condition with three levels of initial moisture content (8-10%, 10-12%, and 12-14%) for 20 weeks. The final objective of this research<br />was to develop model for storability vigor of papaya seed. Two experiments, accelerated aging and seed storage were conducted at Seed Laboratory, Department of Agronomy and Horticulture, Bogor Agricultural University from October 2015 to May 2016. A completely randomized design with nested factors and four replications was applied to both experiments. The results showed that physical accelerated aging using IPB 77-1 MMM machine was more effective than chemical accelerated aging using IPB 77-1 MM machine for papaya seed. The viability of seed stored in AC condition remained high until the end of the storage period, whereas it declined at 16 week storage period in the ambient condition. The viability of seed with initial moisture content of 12-14% declined faster than that of initial moisture content of 8-10% after 18 week storage periode. The model used to estimate the storability vigor of papaya seed accurately was the equation y = a + b expcx where y : storability vigor estimation, x : aging time and a,b,c : constant value. Penelitian ini bertujuan untuk membandingkan metode<br />pengusangan cepat yang efektif antara fisik dengan kimia serta mempelajari pola penurunan viabilitas benih selama penyimpanan aktual pada kondisi simpan kamar (suhu =28-30 0C, RH =75-78%) dan AC (suhu =18-20 0C, RH =51-60%) dengan tiga tingkat kadar air awal (8-10%, 10-12%, dan 12-14%) selama 20 minggu. Tujuan akhirnya adalah membangun model vigor daya simpan benih pepaya. Penelitian pengusangan cepat dan penyimpanan dilakukan pada bulan Oktober 2015 sampai Mei 2016 di Laboratorium Benih, Departemen Agronomi dan Hortikultura, Institut Pertanian<br />Bogor. Kedua penelitian menggunakan rancangan acak lengkap tersarang dengan empat ulangan. Hasil penelitian menunjukkan bahwa pengusangan cepat secara fisik dengan alat IPB 77-1 MMM lebih efektif daripada pengusangan kimia dengan alat IPB 77-1 MM untuk benih pepaya. Viabilitas benih yang disimpan pada kondisi AC tetap tinggi hingga akhir periode simpan, sedangkan pada kondisi kamar penurunan viabilitas dimulai pada periode simpan 16 minggu. Benih yang disimpan dengan tingkat KA awal sebesar 12-14% lebih cepat mengalami penurunan viabilitas mulai periode simpan 18 minggu dibandingkan dengan benih dengan KA awal 8-10%. Hasil penelitian juga menunjukkan terdapat korelasi yang erat antara pola kemunduran benih pada pengusangan cepat dan penyimpanan aktual, sehingga model pendugaan vigor daya simpan (y) berdasarkan waktu pengusangan (x) dapat disusun dengan persamaan y = a + b expcx. Kandungan aluminium yang tinggi pada tanah Ultisol merupakan salah satu masalah utama dalam budidaya tanaman kedelai mulai dari fase perkecambahan hingga pertumbuhan tanaman. Perlakuan priming pada benih diyakini mampu memperbaiki perkecambahan pada lingkungan yang kurang menguntungkan. Penelitian dilakukan untuk mengetahui pengaruh priming pada perkecambahan benih kedelai yang dikecambahkan dalam media masam dan mengandung aluminium. Penelitian disusun dalam Rancangan Acak Lengkap (RAL) pola faktorial. Faktor pertama adalah jenis priming : tanpa priming, H2O (air), KNO3 1%, KNO3 2%, GA3 50 ppm, GA3 100 ppm, dan PEG 6000 7,5%. Faktor kedua adalah varietas kedelai yaitu: Anjasmoro, Burangrang, dan Grobogan. Data dianalisis sidik ragamnya dan dilanjutkan dengan uji Beda Nyata Jujur (BNJ) 5% menggunakan program statistika R Studio. Hasil penelitian menunjukkan priming PEG 7,5% dan varietas Burangrang secara mandiri merupakan perlakuan terbaik untuk meningkatkan vigor benih kedelai berdasarkan variabel waktu muncul kecambah, kecepatan perkecambahan, dan panjang hipokotil. Implikasi penelitian ini adalah PEG 7,5% dapat digunakan untuk perbaikan perkecambahan pada tanah masam dengan kandungan aluminium yang relatif tinggi. High aluminium content in Ultisol is the main problem in soybean cultivation from germination to planting growth phases.Priming treatment is believed to improve seed germination in a poor-growing environment. This study aimed to determine the effect of priming of soybean seeds germinated under acid medium and aluminium stress. The experimental design used was Completely Randomized Design (CRD) with factorial pattern. The first factor was priming type : no priming, H2O (water), 1% KNO3, 2% KNO3, 50 ppm GA3, 100 ppm GA3, and 7.5% PEG 6000. The second factor was soybean variety : Anjasmoro, Burangrang, and Grobogan.  The data obtained were analyzed using the R Studio statistic program with a 5% HSD test. The study resulted that each PEG 7.5% priming and Burangrang  variety independently was the best in increasing soybean seed vigor based on sprout time emergence, germination speed and hypocotyl length. This research implicates that PEG 7.5% can be used to improve seed germination on acid soils with high aluminium content.

ABSTRACT<br /><br />The use of probiotic bacteria as biocontrol agents is one of the methods of controlling anthracnose disease caused by Colletotrichum acutatum. The objective of this research was to suppress the infection of C. acutatum and increase chilli pepper seedling vigour. The research involved factorial experiments arranged in a completely randomized design with two factors. The first factor was seed coating involving six treatments, i.e., control, seed coating without bacteria, seed coating with Bacillus sp., Pseudomonas sp., Actinomycetes sp, and fungicide. The second factor was the seed storage period where six storage periods were experimented, i.e., 0, 1, 2, 3, 4, 5, and 6 months. The results showed that the coating treatment of Bacillus sp., Pseudomonas sp., and Actinomycetes sp. improved germination, growth rate and number of leaves and reduced the incidence of attacks and infection of hypocotyls at 5 month storage period.<br /><br />Keywords: Actinomycetes sp., Bacillus sp., Pseudomonas sp., seed coating, storage period

This study aims to determine the effect of the location of fruit on trees and treatment on seeds to germination cocoa. This study was conducted in Tumpok Teungoh Village, Banda Sakti Sub-district, Lhokseumawe City, August to September 2016. This study used Completely Randomized Design with 3 replications. Two factors studied were (a) the location of the fruit on the tree where L1 = The location of the fruit on the stem and L2 = The location of the fruit on the branch. (b) treatment factor in seeds where P1 = Without pulp clearance, P2 = Pulp cleansing. The parameters observed were potential growth, germination, vigor index, growth rate and synchronization of cocoa seed growth. The result of variance analysis showed that the fruit difference treatment did not affect the observation of maximum growth potential and cocoa seed germination, but it influenced the observation of vigor index, growth rate and synchronization of cocoa seed growth. Treatment of pulp cleansing did not affect the observation of growth potential, germination, growth rate and synchronization of seed growth, but pulp cleansing treatment had an effect on observation of vigor index of cocoa seed. There was no interaction between fruit difference treatment and treatment of cocoa bean pulp cleansing of all observed parameters

<p>This study was conducted to determine seed germination on the mechanisms of salinity stress in <em>B. rapa </em>var. <em>parachinensis </em>(Hong kong choy sum). The type of experiment was a completely randomized design with three salinity treatment NaCl, MgCl2, and KCl and minimum four replications was used. Salinity treatment consisted of 0 (control), 50, 100, 150, 200, mM concentrations. The experiment was carried out under 70% relative humidity at 23°C with a photoperiod of 8:16 h light/dark. The germinated seeds were counted every day and germination percentage, seed vigor, seed tolerance, length of seedling and biomass seedling was measured. The data were analyzed by one-way ANOVA multiple comparison test followed by Duncan’s multiple range test.</p>

Cempaka wasian is a type of woody tree which has a substantial historical value associated with the local culture of the Minahasan people. The improvement of cempaka wasian plantation is essential due to their enormous importance and usage. This study aims to discover the impacts of duration, storage room, and containers used for the appropriate storage of cempaka wasian seeds. The design used in this study is entirely randomized design organized with a factorial pattern which consists of 3 factors; 1) storage duration, 2) storage room and 3) storage containers. Duration of storage was 2, 3, 4, and 5 months; meanwhile, storage rooms comprised of refrigerators, chambers, and air-conditioned room. Moreover, storing containers consisted of calico cloth, aluminium foils, and plastic bags. The study finding indicates that the seeds of cempaka wasian could still germinate if stored for five months by using plastics bags inside refrigerators. A significant provision of seeds and suitable timing are closely related to the proper seed storage techniques.

The aim of this study were, 1) to find out and study the effect of KNO3 immersion treatment on oil palm seed germination; 2) to find out and study the effect of coconut water immersion treatment on oil palm seed germination; 3) to find out and study the interactions that occur between KNO3 immersion treatment and coconut water to oil palm seed germination. This research was conducted in July 2019 until August 2019 in the Laboratory of the Department of Agriculture Cultivation, Faculty of Agriculture, Palangka Raya University using a Completely Randomized Design with 2 factors. The first factor is the concentration of KNO3: K0 (0%); K1 (0.2%); K2 (0.4%); K3 (0.6%) and the second factor is coconut water concentration, B0 (0%); B1 (60%); B2 (80%); B3 (100%). The parameters observed were first count germination, maximum growth potential, germination power, growth speed, radicle length, plumular length and dormancy intensity. The results showed that the treatment of KNO3 concentration significantly affected first count germination, maximum growth potential, germination rate, growth speed and dormancy intensity. Treatment of coconut water concentration significantly affected first count germination, maximum growth potential, germination rate, growth speed and dormancy intensity. The interaction between the treatment of KNO3 concentration and coconut water concentration had no significant effect on all observed parameters

High-altitude fields have been subjected to an intense anthropization process. Due to this, studies on germination, seed storage and propagation of species that compose this plant formation become fundamental. Among such species is the Chresta sphaerocephala DC., Asteraceae. The purpose of this work was to study different temperatures for germination and storage of this species. Four germination temperatures (15°C, 20°C, 25°C and 30°C) and two storage temperatures (-5°C and +5°C) were tested. In order to assess seed vigor, the germination speed index (GSI) and average seed germination time were calculated. ANOVA and Tukey's test were performed at a 5% significance level. In addition, a morphological difference was observed in the analyzed Chresta sphaerocephala seeds, with these being classified as C1 (small seeds), C2 (long thin seeds), C3 (thicker seeds) and C4 (dark colored seeds). As for the storage temperature, 5°C was shown to be the best. When correlated with the germination temperature, 20°C resulted in a statistically higher number of germinated seeds.

Hard bark seeds are one of the causes of long seed dormancy. The solution of salacca seed dormancy is done by soaking the seeds using gibberellin. The aim of this research to examine the effect of different concentration and soaking time of gibberellin on seed viability and growth of salak seedling. The research was conducted on May – July 2019 in Green House of Food Plant and Horticultural Protection Center, Central Java. The study used a Completely Randomized Design (CRD) 4 x 4 with 3 replications, each replications using 5 seeds. The first factor was concentration of gibberellin (G0: 0 ppm, G1: 20 ppm, G2: 40 ppm, G3: 60 ppm). The second factor was soaking time of gibberellin (S0: 10 minutes, S1: 25 minutes, S2: 40 minutes, S3: 55 minutes). Data were analyzed by variance and proceeded with Duncan Multiple Range Test (DMRT) at level 5% and orthogonal polynomial test. The observed variables were percentage of germination, maximum growth potential, seedling height, number of leaves, and root length. The results showed that the highest of percentage germination, maximum growth potential, and seedling height were observed in the 60 ppm of gibberellin concentration. The highest of percentage germination, maximum growth potential, and seedling height were researched in the soaking time 55 minutes. The highest root length was observed in the treatment of 60 ppm of GA3 and soaking time 55 minutes.
