Abstract In this study, the self-extracted constipation treatment of traditional Chinese medicine extracts was applied to constipated rats. To explore the mechanism and role of the Chinese medicine for the treatment of constipation, the 16S rRNA sequencing and qRT-PCR technology were used to analyze the abundance and diversity of intestinal flora in rats. We found that the relative abundance of Firmicutes in the intestinal flora of rats with constipation was significantly higher than that in the control group, and the Bacteroides was significantly lower than that in the control group, while the relative abundance of Firmicutes was significantly changed after taking a certain dose of Chinese medicine, and reduced the relative abundance of Lachnospiraceae, and greatly increased the relative abundance of Lactobacillus, enhanced the symbiotic relationships of Lactobacillus with other intestinal flora. There was no significant difference in the total copies of intestinal bacteria between the constipated rats and the control group. The total copies of intestinal bacteria in the constipated rats decreased after taking the traditional Chinese medicine. Finally, this study results provides a theoretical basis for the treatment and understand the mechanism and effect of traditional Chinese medicine on rate constipation.

Finally, this study results provides a theoretical basis for the treatment and understand the mechanism and effect of traditional Chinese medicine on rate constipation.

This study aimed to determine the efficacy of Integrated Rehabilitation Techniques of Traditional Chinese Medicine (IRT-TCM) on patients with ischemic stroke as an alternative therapy to conventional rehabilitation techniques. Sixty-nine patients with ischemic stroke were randomly assigned to receive either IRT-TCM (intervention group, n = 46) or conventional rehabilitation techniques (control group, n = 23). The IRT-TCM consisted of a sequential combination of acupuncture and massage techniques. The Fugl-Meyer Assessment (FMA), National Institutes of Health Stroke Scale (NIHSS), Barthel index (BI) and modified Rankin Scale (mRS) were measured on day 0 (baseline, before treatment), day 21, and day 90. We observed that the scores in FMA and BI were increased, and NIHSS were decreased in both groups on day 21 and 90, compared with the baseline (day 0). Furthermore, significantly better scores in FMA of lower limbs and NIHSS were found in patients treated with IRT-TCM on day 21 and 90. For mRS, the percentage of patients ranking 0 and 1 in the intervention group presented a striking contrast to the control group on day 90 but with no significant difference. The results indicated that, as a feasible alternative therapy, IRT-TCM is beneficial for patients with ischemic stroke. Further research with larger sample size, long-term observation, and strict blinding are still in need to confirm the efficacy of IRT-TCM.

Aim. To study the effect and mechanism of traditional Chinese medicine Qinbai Qingfei concentrated pellet (QQCP) against Mycoplasma pneumoniae (MP). Methods. Rat airway smooth muscle (ASM) cells were used to examine the antimycoplasmal activity of QQCP via four drug-adding modes: pre- and postadding drugs, simultaneous-adding after drug and MP mixed, and simultaneous-adding drug and MP; taking roxithromycin dispersive tablets (RDT) as positive control, the cellular A570 values were determined by MTT method. Results. All of A570 values in QQCP group were significantly higher than those of the corresponding MP control group (P<0.01) in four drug-adding modes; there was no significant difference in A570 values between the QQCP group and that of the positive control group (P>0.05), confirming that QQCP could significantly inhibit the infectivity of MP to ASM cells. Conclusion. QQCP had significant activity in preventing and treating MP infection, killing MP, and antiabsorption.

Abstract Background Aberration of birth canal microbiota is one of the most important factors in the etiology of sow endomentritis. Nevertheless, reports about the structure and composition of birth canal microbiota in endomentritis sow and their relationship with intestinal microbes is limited. Therefore, understanding the relationship between birth canal microbiota and intestinal microbiota of the host has become exceedingly crucial. Results In this study, 4 healthy and 4 endomentritis sows were selected basing on whether the sow had endometritis or not in a farm of China. The microflora of their birth canal secretions and fresh feces were analyzed via sequencing the V3 + V4 region of bacterial 16S rDNA gene. The results showed that the significant difference between endometritis and healthy sows birth canal flora in the composition and abundance. Sow endomentritis was associated with increasing in the relative abundance of Porphyromonas, Clostridium_sensu_stricto_1, Streptococcus, Ezakiella, Fusobacterium, Actinobacillus, Bacteroides, and Prevotella as well as Anaerococcus. On the contrary, the majority of beneficial bacteria that belonging to Firmicutes phylum (e.g., Lactobacillus and Enterococcus ) declined in endomentritis sow. The increased relative abundance of Porphyromonas in the vaginal secretions might correlate with the decrease of Lactobacillusin the feces of endometritis sows. Moreover, the experimental result also found that some intestinal bacteria (such as Escherichia-Shigella and Bacteroides) may be bound up with the onset of sow endometritis. Conclusion Sow endometritis is closely related to the microbiota of birth canal, and that some intestinal bacteria may promote the onset of endometritis. The above results can supply a theoretical basis to research the pathogenesis of endometritis and the microbiota of sow's birth canal and gut.

Abstract BackgroundChalazion is a chronic inflammatory granuloma of the meibomian gland formed on the basis of the obstruction of the meibomian gland drainage duct and the retention of secretions. It is one of the most common clinically eye diseases in children. Chronic inflammation of the meibomian glands is responsible for this disease, and the gut flora is thought to be involved in the inflammatory process. In this study, we investigated the relationship between intestinal microbial composition and children's chalazion.MethodsFecal samples were collected from 21 children with chalazion and 26 healthy children. DNA was extracted from fecal stool samples and 16S rRNA sequences in the gut flora were detected by using second second-generation sequencing technology. The results were used to compare the composition of the microbiome between patients and healthy controls.ResultsAccording to Alpha Diversity and Beta Diversity analysis, we found that there was no significant difference in bacteria diversity and relative abundance between the two groups. We compared the flora of the control group and the diseased group through Lefse analysis, and screened out 11 different species. Based on the absolute abundance of species, 43 different species were selected. Anosim analysis and metastats analysis were used to compare the flora of the control group and the diseased group again. At the species level, we found that gut_metagenome and human_gut_metagenome are the common differences in species levels obtained from the above analysis. Finally, corrplot correlation analysis was performed, suggesting that gut_metagenome has a great correlation with the number, ulceration, and recurrence of chalazion in children.ConclusionsThere was no significant difference in the diversity index and relative abundance of flora in children with chalazion compared with healthy children, but there were significant differences in some bacterial species. The gut_metagenome strains identified in this study were significantly related to the growth, ulceration, and relapse of children with chalazion. It is suggested that gut_metagenome may be a microbiological indicator which is independent of clinicopathologic factors but associated with chalazion disease .* These authors have contributed equally to this work.

The purpose of this study was to investigate the effect of the HXBM408 bacteria on the diversity of rat intestinal bacteria and the metabolism of soybean isoflavones. The control group was administered sterilized water and daidzein by gavage for 7 days. Conversely, the experimental group was administered HXBM408 solution and daidzein by gavage for 7 days. The content of the daidzein metabolite equol in rat feces in the experimental group was significantly higher than that in the control group (P < 0.05) on the 7th and 14th days. However, the content of daidzein and its metabolites in feces was not significantly different (P > 0.05). On the 7th day, the relative abundance of Streptococcus in the feces of the experimental group was significantly higher than that of the control group (P < 0.05), but the difference disappeared over time (P > 0.05). In the intestinal digesta of rats, the proteobacteria of the experimental group was significantly lower than those of the control group (P < 0.05). HXBM408 can increase the degradation ability of soybean isoflavones in a short period after ingestion, increase the number of beneficial intestinal flora, and improve the structure of the flora.

CoTOL is a traditional Chinese medicine (TCM) formula in clinics for treating gout and hyperuricemia, especially in obese patients with recurrent attacks. However, fewer studies have investigated how CoTOL impacts the intestinal flora in reducing uric acid. In the present, we analyze the bacteria targeted by ingredients of CoTOL and evaluate the effects of CoTOL on uric acid and intestinal flora in a mice model of obese hyperuricemia inoculated with xanthine dehydrogenase- (XOD-) producing bacteria, Streptococcus faecalis. Firstly, ingredients of herbs in CoTOL and gene target by these ingredients were retrieved from TCMID 2.0, and these genes were screened by DAVID Bioinformatics Resources 6.8, deciphered to retrieve the bacteria. Then, 3-4-week-old male C57bl/6j mice were randomly divided into 6 groups and fed with high fat diet for 8 weeks up to obesity standard. The mice were inoculated intragastrically with 5 × 109 CFU Streptococcus faecalis 3 times at the 5th, 6th, and 7th week and intragastrically administrated with uricase inhibitor, potassium-oxonate (PO, 250 mg/kg), to induce hyperuricemia at the 8th week, once a day for 7 consecutive days, respectively (IB model). IB model plus CoTOL (0.4 ml/20g) and allopurinol (40 mg/kg) were administrated by gavage at the 5th week, once a day for 4 weeks. The feces and blood in each group were sampled at the 4th and 8th week. With no bacteria inoculation, CoTOL, allopurinol, and blank group were treated with CoTOL and allopurinol or water, respectively. 44 species of bacteria (i.e., Enterococcus faecalis, Streptococcus, etc.) genes were targeted by 6 ingredients of 6 herbs in CoTOL. Inoculation with Streptococcus faecalis significantly caused the elevation of uric acid and the change of intestinal flora structure, whereas treatment with CoTOL significantly increased the abundance of Akkermansia and those of Bacteroides and Alloprevotella decreased. Furthermore, CoTOL exhibited a unique effect on reducing weight unobserved in allopurinol intervention. The present study, for the first time, demonstrated that CoTOL has beneficial effects on hyperuricemia and overweight, which may be attributed to regulating material metabolism and improving the structure or function of intestinal flora. Thus, CoTOL may be a promising therapy for hyperuricemia and overweight in chronic gout management and can be integrated with conventional treatments.

Objective: To observe the clinical effect of Jieyu decoction in treating generalized anxiety disorder with stagnation-heat in liver meridian type. Methods: There were 72 cases of generalized anxiety disorders outpatient and inpatient patients, and 36 in the control and treatment group. The control group was given oral Deanxit; the treatment group was given Jieyu decoction granules, and necessary psychological counseling was given to patients in both groups. Changes in traditional Chinese medicine syndrome scoring scale, Hamilton scale (HAMA), dynamic electrocardiogram heart rate variability and other indicators were recorded and compared between the two groups. Results: The total effective rate of the control group was 76.5%, and the total effective rate of the treatment group was 80.0%. The curative effect of the treatment group and the control group was significantly higher than that before treatment (P < 0.05), and there was no significant difference in the overall curative effect between the two groups (P > 0.05). The HAMA scale score of the two groups was significantly lower than that before treatment (P < 0.05), and there was no significant difference in the reduction rate of HAMA score between the two groups (P > 0.05). The traditional Chinese medicine syndrome scores of the two groups were significantly decreased compared with that before treatment (P < 0.05). The traditional Chinese medicine syndrome scores of the treatment group were significantly different from that of the control group after 8 weeks of treatment (P < 0.05). Two groups of heart rate variability in 24 h period of standard deviation between the average normal R - R (SDNN), 24 h per 5 min normal R - R period between the standard deviation (SDANN), root mean square of phase difference between adjacent R - R (RMSSD) and the number of cardiac 50 ms accounted for the percentage of the total number of cardiac (PNN50) than before treatment significantly increased (P < 0.05), 8 weeks after treatment the SDNN, SDANN than the control group no significant difference (P > 0.05), RMSSD,PNN50 significantly difference compared with control group (P < 0.05). Conclusion: Jieyu decoction has good efficacy and high safety in the treatment of generalized anxiety disorder of the type of liver meridian and heat stagnation, which is suitable for clinical promotion and application.

Abstract Background Spleen-deficiency syndrome, an important pathological change in traditional Chinese medicine, has been proven to attribute to intestinal dysbacteriosis. Shengmai Yin (SMY), a classic formula for replenishing qi and restoring pulse, is a common medicine for critical emergencies in traditional Chinese Medicine. Interestingly, our previous study established a spleen-deficiency rat model and verified the potency of SMY formula in curing spleen-deficiency rats. Our goal herein was to explore whether SMY can modulate the composition of intestinal flora and alleviate spleen-deficiency in rats. Methods This experiment was randomly divided into three groups, namely the normal control group (NC), model control group (MC), and the Shengmai Yin group (SMY). After the treatment, the weight and symptom indexes of the rats were recorded, histological changes in the colon were observed, levels of serum D-xylose, gastrin (GAS), and vasoactive intestinal peptide (VIP) were measured, and gut microbiota profiling was conducted by 16S rRNA sequencing. Results The body mass of the spleen-deficiency model rats significantly decreased compared with that of the NC group, and SMY treatment significantly increased body mass compared with the MC group (P < 0.01). Colon histopathology revealed that SMY treatment alleviated colonic mucosal damage in spleen-deficiency rats. The serum levels of D-xylose and gastrin (GAS) were significant increased by SMY (P < 0.05, P < 0.01), and vasoactive intestinal peptide (VIP) was reduced by SMY (P < 0.01) compared with MC. Furthermore, alpha diversity was significantly decreased in the model rats compared to the normal rats (P < 0.05) and increased with SMY treatment (P < 0.01). The most abundant phyla were Firmicutes and Bacteroidetes, followed by Proteobacteria, Verrucomicrobia, and Actinobacteria. At the genus level, there was a lower relative abundance of Lactobacillus, Bacteroides, Akkermasia, and Allobaculum, and a higher relative abundance of Lachnospiraceae NK4A 136 group, Ruminococcaceae UCG-014, and Sphingomonas in the MC group. The relative abundance of Actinobacteria, Alistipes, Bifidobacterium, Bifidobacterium, Bifidobacteriaceae, Lachnospiraceae NK4A136group, Lactobacillus, Lactobacillaceae, Bacilli, Verrucomicrobiae, and Akkermansia were significantly abundant in the treatment groups, and thus may be singled out as potential biomarkers for SMY in the treatment of spleen deficiency. In addition, analysis on the correlation between species and physicochemical indexes showed that the abundance of Parasutterella was negatively correlated with the change in GAS, and positively correlated with the change in VIP (P < 0.01). Conclusion Our findings have provided preliminary evidence that modulating the gut microbiota may play a role in the treatment of spleen deficiency with SMY. However, further studies are needed to clarify the mechanism by which SMY regulation of related gut microbiota occurs.

Abstract Nigella A, also named Sieboldianoside A, has been extracted from many kinds of Traditional Chinese Medicine (TCM), such as Nigella glandulifera, Stauntonia chinensis DC., and the leaves of Acanthopanax sieboldianus. Nigella A exhibited potential analgesic, anti-inflammatory, anti-tumor, and antioxidant activities. However, whether Nigella A could treat ulcerative colitis (UC) is still unknown. As saponins always be regarded as the kinds of ingredients that could regulate immunity and intestinal flora. This research aimed to investigate the therapeutic effect of Nigella A on UC and explore its effect on intestinal flora. We noted that Nigella A and Sulfasalazine (SASP) could significantly improve the signs and symptoms, alleviate colonic pathological injury in DSS-induced mice. The changing of many specific bacterial genus such as Lactobacillus, Porphyromonadaceae, Bacteroides and Escherichia might closely related to the recovery of intestinal inflammatory response. This study initially confirmed the therapeutic effect of Nigella A and SASP on DSS-induced colitis by improving the diversity of intestinal microbial composition. Nigella A has the potential to be developed for the treatment of UC and other disorders related to the imbalance of intestinal flora.
