Obesity is one of the most prevalent health concerns among all age groups & populations worldwide, resulting into a significant increase in mortality and morbidity related to metabolic disorders. Targeting one or more enzymes involved in lipid metabolism can be selective for evaluation of anti obesity action of drug. The present study was aimed to evaluate in vitro anti obesity action by inhibiting pancreatic lipase & ᾳ amylase enzyme by various fractions of methanolic extract of aerial parts Fagonia cretica L. along with their phytochemical analysis. The n- hexane (HFFC), Chloroform (CFFC), Ethyl acetate (EAFFC), n-butanol (BFFC) & aqueous fractions (AQFFC) were prepared from methanolic extracts of F. cretica L & were analyzed for qualitative as well as quantitative phytochemical study using reported methods. The qualitative phytochemical studies of prepared extract & fractions showed presence of flavonoids, saponins, phenolics, alkaloids & carbohydrates. All the fractions were then examined for their in-vitro lipase inhibitory & ᾳ amylase inhibitory activities at a concentration level of 50, 100, 150 & 200µg/ml and their percentage inhibitory effects were reported. Among the analyzed samples, BFFC showed highest lipase inhibitory action i.e. 83.02 ± 2.47% as compared to other fractions. EAFFC showed significantly higher ᾳ amylase inhibitory action i.e. 80.22 ± 1.18% as compared to other fractions.

Objective: The aim of this research was to assess the in vitro anti-inflammatory activity and cytotoxicity of the methanolic extract and total alkaloid extract obtained from leaves and bulbs of Crinum x amabile (CA) growing in Ecuador.Methods: Methanolic extracts of dry powered leaves and bulbs of CA obtained by cold maceration method were subjected to preliminary phytochemical screening. Total alkaloid extracts of leaves and bulbs of CA were obtained by conventional extraction of alkaloids base with an organic solvent. Furthermore, the anti-inflammatory activity and cytotoxicity of the four extracts were investigated by in vitro isolated neutrophils model using stable tetrazolium salt (WST-1).Results: Phytochemical analysis of methanolic extracts revealed the major classes of phytochemicals such as alkaloids, flavonoids, tannins, triterpenes, and steroids, but no proteins or saponins could be detected on leaves extract. Extracts obtained from bulbs both methanolic and total alkaloids of CA show an interesting anti-inflammatory activity, although it was not significant compared to the standard anti-inflammatory drug, aspirin. Cytotoxicity of bulb alkaloid extract was lower compared with all of the other extracts.Conclusion: Based on the results of this research, it could be concluded that CA is a very interesting source of natural anti-inflammatory compounds (especially alkaloids) which could be used to prevent many chronic disorders. Further, phytochemical studies are necessary to identify the chemical compounds responsible for the significant anti-inflammatory activity showed.

AbstractPlant-derived phytochemicals consisting of phenols and flavonoids possess antioxidant properties, eventually rendering a lucrative tool to scavenge reactive oxygen species. This study was carried out to evaluate in vitro antioxidant and cytotoxic potential of methanolic extract and petroleum ether extracts ofPhytochemical screening was done following standard procedures. Antioxidant activity was tested using several in vitro assays, viz., 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, NO assay, HPreliminary phytochemical study revealed the presence of flavonoids and glycosides in both extracts. Methanolic extract was found to possess stronger antioxidant potential than petroleum ether extracts in all assays. The ICThese findings demonstrate that methanolic extracts could be considered as potential sources of natural antioxidant, whereas petroleum ether extracts could be explored for promising anticancer molecules.

Three new 5-deoxyflavonoid and dihydroflavonoids 2, 3 and 4 have been isolated from the methanolic extract of Abutioln pakistanicum aerial parts, for which structures were elucidated explicitly by extensive MS- and NMR-experiments. In addition to these, 3,7,4′-trihydroxy-3′-methoxy flavonol (1) is reported for the first time from Abutioln pakistanicum. Compound 2 and 4 are p-coumaric acid esters while compounds 2–4 exhibited α-glucosidase inhibitory activity. Docking studies indicated that the ability of flavonoids 2, 3 and 4 to form multiple hydrogen bonds with catalytically important residues is decisive hence is responsible for the inhibition activity. The docking results signified the observed in-vitro activity quite well which is in accordance with previously obtained conclusion that phenol moiety and hydroxyl group are critical for the inhibition of α-glucosidase enzyme.

AbstractObjectivesRauvolfia vomitoria is a medicinal plant used traditionally in Africa in the management of several human diseases including psychosis. However, there is inadequate scientific information on the potency of the phenolic constituents of R. vomitoria leaf in the management of neurodegeneration. Therefore, this study characterized the phenolic constituents and investigated the effects of aqueous and methanolic extracts of R. vomitoria leaf on free radicals, Fe2+-induced lipid peroxidation, and critical enzymes linked to neurodegeneration in rat’s brain in vitro.MethodsThe polyphenols were evaluated by characterizing phenolic constituents using high-performance liquid chromatography coupled with diode array detector (HPLC-DAD). The antioxidant properties were assessed through the extracts ability to reduce Fe3+ to Fe2+; inhibit ABTS, DPPH, and OH radicals and Fe2+-induced lipid peroxidation. The effects of the extracts on AChE and MAO were also evaluated.ResultsThe phenolic characterization of R. vomitoria leaf revealed that there were more flavonoids present. Both aqueous and methanolic extracts of R. vomitoria leaf had inhibitory effects with the methanolic extract having higher significant (p≤0.05) free radicals scavenging ability coupled with inhibition of monoamine oxidases. However, there was no significant (p≤0.05) difference obtained in the inhibition of lipid peroxidation and cholinesterases.ConclusionThis study suggests that the rich phenolic constituents of R. vomitoria leaf might contribute to the observed antioxidative and neuroprotective effects. The methanolic extract was more potent than the aqueous extract; therefore, extraction of R. vomitoria leaf with methanol could offer better health-promoting effects in neurodegenerative condition.

Polyphenolic enriched extracts from two species of Cyperus, Cyperus glomeratus and Cyperus thunbergii, possess mammalian arginase inhibitory capacities, with the percentage inhibition ranging from 80% to 95% at 100 µg/mL and 40% to 64% at 10 µg/mL. Phytochemical investigation of these species led to the isolation and identification of two new natural stilbene oligomers named thunbergin A-B (1–2), together with three other stilbenes, trans-resveratrol (3), trans-scirpusin A (4), trans-cyperusphenol A (6), and two flavonoids, aureusidin (5) and luteolin (7), which were isolated for the first time from C.thunbergii and C. glomeratus. Structures were established on the basis of the spectroscopic data from MS and NMR experiments. The arginase inhibitory activity of compounds 1–7 was evaluated through an in vitro arginase inhibitory assay using purified liver bovine arginase. As a result, five compounds (1, 4–7) showed significant inhibition of arginase, with IC50 values between 17.6 and 60.6 µM, in the range of those of the natural arginase inhibitor piceatannol (12.6 µM). In addition, methanolic extract from Cyperus thunbergii exhibited an endothelium and NO-dependent vasorelaxant effect on thoracic aortic rings from rats and improved endothelial dysfunction in an adjuvant-induced arthritis rat model.

Histopathological studies of the effects of chloroform and methanolic leaf extracts of Ilex kudingcha in Trypanosoma brucei infected albino wistar rats were investigated. The toxicity and phytochemical study were also carried out using standard protocol. T. brucei infected animals were administered orally with 200 and 400 mg/kg b.w. of the extracts and 3.5 mg/kg b.w. of the standard drug (diminazene aceturate). Results on acute toxicity studies (LD50) revealed no sign of lethality up to the dose of 5,000 mg/kg body weight but the liver and kidney histology of infected animals treated with 5,000 mg/kg b.w. of I. kudingcha extracts were observed to be hepatotoxic and nephrotoxic. The methanol extracts showed appreciably high in vivo anti-trypanosomal activities compared to the reference drug. Histological examination of the organs revealed serious pathological lesions in the liver of the infected animals without treatment (negative control). In the positive control animals (infected animals administered standard drug), mild multifocal aggregate of inflammatory leucocytes was observed. In the other experimental animals, no pathological lesion was observed in the liver, kidney, brain, and heart of infected animals treated with the methanolic extract and combined methanol and chloroform extracts. The effectiveness of the methanolic extract at reducing the lesions caused by the parasite is the same compared with the standard drug. Phytochemical analysis of the plant extracts showed that methanol extract contained appreciable high levels of alkaloids, saponin, tannins, phenol, and glycoside while flavonoid was not detected. Hence, the curative properties of methanolic extract of I. kudingcha as observed in the organs indicate its anti-trypanosomal properties but it should be consumed at minimal doses.

Excoecaria agallocha plant (Leaves) was analyzed to explore In Vitro antimicrobial and membrane stabilizing activities as a crude methanolic extract. Antimicrobial activity was performed against a wide range of Gram positive(+Ve) and Gram negative(-Ve) bacteria by using disc diffusion method and various methanolic extracts of leaves of E.agallocha was tested for determining membrane stabilizing activity at hypotonic solution and heat induce condition and standard acetyl salicylic acid (0.10 mg/mL) was employed as standard. the zones of inhibition created was found to be 1.3 cm at a concentration of 100 µL/ disc in case of Gram negative(-Ve) bacteria Salmonella typhi. The results obtained were compared with that of a standard Ampicillin (10 µL) and imipenem (10µL), penicillin (10µl), cefoxitine(30µl). Crude methanolic extract of 10mg/ml concentration showed maximum value of 17.67±0.0102 and 18.92±0.086% respectively under hypotonic solution and heat induced condition. Thus the result suggest that, crude methanolic extracts of E.agallocha possessed slight to moderate antimicrobial and membrane stabilizing properties.

The main aim of study is to evaluate the anti-arthritic effect of different extracts of Boerhaavia diffusa in arthritic rats. Different extracts were prepared by successive solvent extraction methods by using the various polar and non polar solvents and their % yields were calculated. Arthritis was induced by FCA induced arthritis model in rats and paw volume was measured on different days. Body weights of all animals were also measured simultaneously and at the end of experiment some haematological parameters were measured. On preliminary phytochemical studies extracts showed the presence of alkaloids, fatty acids, terpenoids, flavonoids and phenolic compounds. Among all extracts, methanolic extract significantly decreased the paw volume in all treated groups. Methanolic extracts also restored the body weight significantly. The results of our study revealed that all the extracts treated group’s causes significant alterations in the hematological parameters and maximal effects were observed at 400 mg/kg. Since methanolic extract showed best activity in arthritic model and its phytochemical study showed presence of flavonoids and phenolic compounds, so it may be possible that anti-arthritic activity of root extracts may be due to presence flavonoids.  Keywords: Arthritis, FCA induced arthritis, Boerhaavia diffusa, haematological parameters, and Body weight

Objective: Eupatorium glandulosum is a useful medicinal plant belongs to the family Asteraceae. It is traditionally used to treat various diseases such as wound healing, antioxidant, and antiproliferative. The present study was aimed to investigate the physicochemical and phytochemical properties of various extracts of aerial parts of E. glandulosum. Methods: The E. glandulosum plant materials were extracted using the solvents alcohol, ethyl acetate, and chloroform by Soxhlet method. The extracts were screened for physiochemical constants, preliminary phytochemical analysis for carbohydrates, glycosides, alkaloids, flavonoids, phenols, tannins, and saponins. The quantitative phytochemical analysis was carried out for total flavonoid and total phenols using standard procedures. Results: The physiochemical constituents such as total ash, acid-insoluble ash, and water-soluble ash were found to be 14.25% (w/w), 5% (w/w), and 7.30% (w/w), respectively. The preliminary phytochemical screening confirmed the presence of carbohydrates, glycosides, alkaloids, flavonoids, phenols, tannins, and saponins. The flavonoid content of the plant extracts was found to be in the descending order ethyl acetate ˃alcohol ˃chloroform and the phenolic content was found to be alcohol ˃ethyl acetate ˃chloroform. Conclusion: The result showed the presence of phytochemical constituents and higher values of phenolic and flavonoid content make the plant useful for the formulation of the different drugs for human uses for treating various diseases.

The in vitro antiprotozoal activities of crude methanolic extracts from the aerial parts of five Lamiaceae plants ( Salvia tomentosa, S. sclarea, S. dichroantha, Nepeta nuda subsp. nuda and Marrubium astracanicum subsp. macrodon) were evaluated against four parasitic protozoa, i.e. Trypanosoma brucei rhodesiense, T. cruzi, Leishmania donovani and Plasmodium falciparum. The cytotoxic potentials of the extracts on L6 cells were also evaluated. Melarsoprol, benznidazole, miltefosine, chloroquine and podophyllotoxin were used as reference drugs. All crude MeOH extracts showed antiprotozoal potential against at least three parasites, so they were dispersed in water and partitioned against n-hexane and chloroform to yield three subextracts that were screened in the same test systems. The n-hexane extract of N. nuda was the most active against T. brucei rhodesiense while the CHCl3 extracts of S. tomentosa and S. dichroantha showed significant activity against L. donovani. All organic extracts displayed in vitro antimalarial and moderate trypanocidal activities against T. cruzi with the n-hexane extract of S. sclarea being the most active against the latter. The extracts displayed low or no cytotoxicity towards mammalian L6 cells.
