Phytocomponents and antioxidant activity in the methanol extract of seed lotus (Nelumbo Nucifera Gaernt.) from Viet Nam

In this study, we determined the phytocomponents using GC-MS and evaluated antioxidant activity by a 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay of the methanolic extract of six lotus varieties seed in Viet Nam (N. Nucifera). GC-MS chromatogram of the methanolic extract of six lotus varieties seed in Viet Nam showed 27 peaks which indicate the presence of 27 phytochemical constituents. The major phytochemical constituent is methyl-alpha-dgalactopyranoside (66,86-78,69%) in all lotus varieties. Antioxidant activity of liquid extract and condensed extract was found as 8.20-15.67 mg/mL and 0.850-1.286 mg/mL in comparison with IC50 of ascorbic acid (3.2 μg/mL). Moreover, 50 % of the DPPH scavenging ability of liquid and condensed extract differed from each other. From obtained results, the seeds of six lotus varieties in Viet Nam were found to be potential for bioactive compounds and antioxidant activity.

Brassicaspecies are very rich in health-promoting phytochemicals, including phenolic compounds, vitamin C, and minerals. The objective of this study was to investigate the effect of different blanching (i.e., water and steam) and cooking (i.e., water boiling, steam boiling, microwaving, and stir-frying) methods on the nutrient components, phytochemical contents (i.e., polyphenols, carotenoids, flavonoid, and ascorbic acid), antioxidant activity measured by DPPH assay, and phenolic profiles of white cauliflower. Results showed that water boiling and water blanching processes had a great effect on the nutrient components and caused significant losses of dry matter, protein, and mineral and phytochemical contents. However, steam treatments (blanching and cooking), stir-frying, and microwaving presented the lowest reductions. Methanolic extract of fresh cauliflower had significantly the highest antioxidant activity (68.91%) followed by the extracts of steam-blanched, steam-boiled, stir-fried, and microwaved cauliflower 61.83%, 59.15%, 58.93%, and 58.24%, respectively. HPLC analysis revealed that the predominant phenolics of raw cauliflower were protocatechuic acid (192.45), quercetin (202.4), pyrogallol (18.9), vanillic acid (11.90), coumaric acid (6.94), and kaempferol (25.91) mg/100 g DW, respectively.

Objective: Prasman (Ayapana triplinervis [Vahl]) is a plant that can eliminate free radicals through its antioxidant effects. However, little research hasbeen conducted to explore the antioxidant activity of the plant.Methods: A. triplinervis (Vahl) leaves, which were determined by the Indonesian Institute of Sciences (LIPI) Bogor, were used in this study. Meanwhile,extraction was performed using n-hexane, ethyl acetate, and methanol as the solvents. Based on the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay usinga UV–visible spectrophotometer and based on ferric reducing antioxidant power (FRAP) assay using a microplate reader.Results: Based on the DPPH assay using a UV–visible spectrophotometer, n-hexane, ethyl acetate, and methanol extracts with a final concentrationof 25 μg/mL inhibited DPPH radical production by 38.91, 51.03, and 54.06%, respectively. Using the percent inhibition, the IC50 for the ethyl acetateand methanol extracts were 28.71 and 23.472 μg/mL, respectively. Based on FRAP assay using a microplate reader, the n-hexane, ethyl acetate, andmethanol extracts had ferrous equivalent antioxidant capacity values of 460, 828.99, and 940.22 μmol/g, respectively. The methanol extract had thegreatest antioxidant activity. The ethyl acetate and methanol extracts at the initial concentrations contained total phenol levels of 12.06 and 42.11 mgGAE/g extract, respectively, as well as total flavonoid levels of 3.24 and 3.41 mg QE/g extract, respectively.Conclusion: Based on the determination of phenol and flavonoid levels, the methanol extract had the greatest antioxidant effects.

Free radicals are molecules that lose electrons in their outer orbitals so that the number of electrons becomes odd and unstable. Free radicals can damage various macromolecule cells including proteins, carbohydrates, fats. Antioxidants are compounds that can donate one or more electrons to free radicals. Avocado leaves contain high bioactive components that can be used as a source of natural antioxidants. Avocado leaves that have been dried, mashed and then shifted with methanol. The extracts were tested by phytochemical screening, including flavonpoid, phenolic, tannin and antioxidant activity using the DPPH (2,2-diphenyl-1-picrylhidrazyl) method. This method is based on the reduction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) against free radicals which causes a change from purple to yellow 2,2-diphenyl-1-picrylhydrazyl) (DPPH). From the research, the IC50 values ​​in avocado leaves were 118.8056 µg / mL and 7,276 µg / mL on ascorbic acid as a positive control. From the IC50 value obtained, it is known that the methanol extract of avocado leaves has strong antioxidant activity to fight free radicals.

Flemingia macrophylla locally known as ‘Charchara’ in Bangladesh is a medicinal herb. Being a member of the Fabaceae family it claims a number of medicinal uses including hypoglycemic activity, neuroprotective effect and gynecological remedies. Methanolic extract of F. macrophylla was investigated to evaluate its antioxidant and general toxic properties in this study. Antioxidant potential was evaluated using total antioxidant capacity, total phenol contents, total flavonoid contents and DPPH (1,1-diphenyl-2-picrylhydrazyl) assays. Both leaf and stem extracts of F. macrophylla were found to possess significant amount of phenolics and flavonoids, expressed as gallic acid equivalents (GAE) and quercetin equivalent (QE), respectively. Interestingly total phenol content was equal for both leaf and stem extract and it was 43.8 mg GAE/g. Total flavonoid content was found to be 64.4 and 39.1 mg QE/g for leaf and stem extract, respectively. The total antioxidant capacity was expressed as ascorbic acid equivalents (AAE) and for leaf it was 5.067 mg/gm AAE and for stem 0.8167 mg/gm AAE. DPPH scavenging activity was determined by comparing with ascorbic acid. The IC50 values were 19.95, 125.89 and 707.94 ?g/ml for ascorbic acid, leaf and stem extract, respectively. The results of the present study on methanolic extracts of F. macrophylla revealed the presence of moderate antioxidant activity and extract of leaves produced better results than stem of the plant. In brine shrimp lethality bioassay, methanolic extract of F. macrophylla exhibited insignificant cytotoxicity. This study was designed to evaluate the antioxidant activity of phenolic compounds for six cultivars of date from In Saleh region (Phoenix dactylifera L.), which were extracted by reflux with solvents, 80% methanol and 70% aceton after the phenolic and flavonoid content were estimated. The total phenolic content (TPC) and total flavonoid content (TFC) of the extracts were measured using Folin–Ciocalteau and aluminum chloride colorimetric methods, respectively. Antioxidant activity acetone/H2O and methanol/H2O extracts using ABTS, DPPH, FRAP, TAC and CUPRAC assays. The results showed that methanolic solvent reflux gives better yields. The highest level of phenolic compounds for all dates varieties was found in the 80% methanol extract. This level ranged from 8.04 ± 0.14 to 46.81 ± 1.22 mg GAE/100 g fresh weight and flavonoid content was observed relatively low for both two solvents. However, extraction into 70% acetone gave the highest antioxidant activity with all assay. These results showed that dates had strongly scavenging activity on DPPH reaching 3.12 µg/mL in methanol/H2O from Cul1 (6.9 µg/mL with ascorbic acid), also on ABTS reaching 6.51 µg/mL in acetone/H2O from Cul2 (104.98 µg/mL with ascorbic acid).

This paper describes results of phytochemical analyses and antioxidant activity of the seeds ofCucumeropsis edulis. For phytochemical analyses, extraction was performed with the same powder using different solvents: Acetone the first, ethyl acetate the second and methanol the last. Aqueous extract was used for evaluation of antioxidant activity. The results obtained show the abundance of polyphenols, gallic tannins and alkaloids in all extracts. Reducing compounds are present in ethyl acetate and acetonic extracts and leucoanthocyans are present in methanolic extract only. The results (IC50 = 2249± 87,03 µg/mL AAI = 0,022 ± 0,001) of evaluation of antioxidant activity are very low.

This study was designed to evaluate the antioxidant activity of phenolic compounds of for Lamiaceae: Salvia officinalis, Satureja calamintha, Mentha pulegium and Marubium vulgare using ferric reducing power (FRAP) and radical scavenging method (DPPH). Preliminary phytochemical analysis shows that the major constituents of crude extracts (Hydromethanolic, hydroacetonic) are tannins, saponins, quinones, flavonoids, terpenoids and reducing compounds in the four plants. In quantitative analysis, variable polyphenols, flavonoids and condensed tannins content are recorded. The highest polyphenols content is that of the aqueous/methanol crude extract of Salvia officinalis (188.92 ± 1.19 mg GAE/ gE), followed by aqueous/acetone crude extract of Mentha pulegium (176.55 ± 2.49 mg GAE/gE). Flavonoids and condensed tannins content of crude extract showing a high content in aqueous/methanol extract of Mentha pulegium and Satureja calamintha (93.64 ± 0.26mg EQC/gE and 25.44 ± 0.71 mg EQC/gE), respectively. Scavenging activity of the free radical DPPH, revealed that the aqueous/methanol extract of Salvia officinalis shown a relatively high antioxidant activity (IC50 = 1.07 μg/ml) compared with ascorbic acid (IC50 = 0.91 μg/ml). FRAP method revealed that the extracts of Mentha pulegium showed the highest power to reduce iron (EC50 = 0.147 mg/ml), but lower than that of ascorbic acid (EC50 = 0.057 mg/mL).

<strong>Extraction and Oral Suspension Formulation of Sea Cucumber Holothuria scabra as Source of Antioxidants.</strong> Sea cucumbers have long been utilized in the food and medicine for Asia and Middle East communities. Indonesia is one of the main suppliers in the trade of sea cucumbers in Asia. Sea cucumber utilization in Indonesia is generally done only in the form of raw materials. Holothuria scabra is one of the sea cucumber that can be explored as a potential source of antioxidants. This study aims to determine the antioxidant activity of sea cucumber H.scabra in extract and suspension. Sea cucumber was extracted with methanol 96% and ethanol 96%, then both antioxidant activity was tested. Extract with higher antioxidant activity were made into oral suspension preparation for further evaluation of the stability (organoleptic, pH, microorganisms) and antioxidant testing. The results showed that ethanol extracts had higher antioxidant activity of methanol extract, but still lower than ascorbic acid. Based on the calculation, IC<sub>50 </sub>of methanol extract was 316.85 ppm and ethanol extract was 130.54 ppm, while ascorbic acid showed IC<sub>50</sub> 34.06 ppm. The most stable formula (F1a) shows that there is still an antioxidant activity that is equal to 42.11 %.

Objective: To investigate the photochemical constituents present in methanol extract of martynia annua seeds using Gas Chromatography-Mass Spectroscopy(GC-MS), High-Performance Thin–Layer Chromatography(HPTLC) analysis and study antioxidant activity. Methods: Methanol extract of Martynia annua seeds were subjected to GC-MS and HPTLC analysis. HPTLC analysis was carried out using GAMAG system with a linomate5 applicator, system mobile phase (Toluene: Chloroform: Ethanol (4:4:1 V/V/V)), two different volume of extract was applied 2 µl and 5 µl. GC-MS analysis was carried out on JEOL GC MATE ΙΙ, column HP 5 MS and Quadruple double focusing mass analyzer. Antioxidant activity was determined by DPPH assay. Results: GC-MS analysis provided 17 peaks indicating the presence of seventeen different phytochemicals in methanol extract of martynia annua seeds. HPTLC fingerprint showed 6 peaks at both size 2 µl and 5 µl at 254 nm whereas 4 peaks, 9 peaks were detected at 366 nm for 2 µl and 5 µl respectively. After derivatization with 10 % methanolic sulphuric acid, 8 peaks, 11 peaks were detected for 2 µl and 5 µl respectively when the derivatized plate was scanned at 540 nm. DPPH free radical scavenging result showed EC50 value of 44.1±1.1 µg/ml. Conclusion: The GC-MS analysis showed the presence of fatty acids, ester, aldehydes and ketones whereas in HPTLC different peaks at different UV-lights before and after derivatization were observed. Maximum percentage inhibition using DPPH assay was found 74 at concentration of 50 µg/ml.

Objective: To investigate the content and antioxidant activities of polyphenol, the correlation between polyphenol content and their antioxidant activities, and phytochemistry compositions of different extracts from marine sponge Aaptos suberitoides commonly found growing in Nhatrang bay, Vietnam orienting application into functional food and pharmacy. Methods: Evaluating the toxicity of antioxidant polyphenol powder preparing from the initial concentrated extract was by the adjusted Behrens Karber method and a correlation between polyphenol content and antioxidant activities basing on the Pearson coefficient in Excel. Separating antioxidant polyphenol content was base on solvents polarization of n-hexane, chloroform, ethanol, ethyl acetate and n-butanol which the quantification of polyphenol content and antioxidant activities, and preliminary phytochemical compositions qualitative. Results: Antioxidant polyphenol powder did not affect mice weight during the assay time of 28 d. Polyphenol content and antioxidant activities got the highest value at chloroform extract in comparison to other extracts, a significant difference (p<0.05) and strong correlation (R2>0.9). Polyphenol content (122.682 mg gallic acid equivalent ml-1), total antioxidant activity (368.183 mg ascorbic acid equivalent ml-1), reducing power activity (24.08 mg FeSO4 equivalent ml-1) and DPPH scavenging (72.48±1.54 %) were the highest values. Alkaloids, flavonoids, steroids, tannins and triterpenoids existed in initial methanol extract. Weakly polarized polyphenol content was 70.27% in comparison to initial methanol extract. Conclusion: Antioxidant polyphenol of sponge Aaptos suberitoides has the potential for application into the field of functional food and pharmaceuticals.
