With this study, ascorbic acid and erythorbic acid were used for the first time to prevent melanosis in shrimp. Three shrimp species (Aristaeomorpha foliacea, Plesionika edwardsi and Melicertus hathor) were used. It was determined that melanosis scores were higher in the control groups. Combination of ascorbic acid and erythorbic acid with metabisulphite was found to be more effective in inhibiting of melanosis than the application alone. No significant differences were found between the groups in terms of TVB-N values. The TMA-N value of the control group was significantly higher than those of application groups. Ascorbic acid, Erythorbic acid and their combinations with sodium metabisulphite were found effective on inhibition of melanosis and quality changes in three shrimp species.Keywords: reducing agents; ascorbic acid; erythorbic acid; melanosis; quality; shrimp

Background Antioxidants show nephroprotective effect against vancomycin associated nephrotoxicity (VAN) in animals. This study aimed to assess the ascorbic acid nephro-protective role against VAN clinically. Methods Forty-one critically ill patients were randomly assigned to one of two groups: intervention group (vancomycin IV plus ascorbic acid, n=21) or control group (vancomycin IV only, n=20). Primary outcomes were the incidence of VAN and the absolute change in creatinine parameters, while mortality rate was the secondary outcome. Nephrotoxicity was defined as an increase in serum creatinine (S.cr) by at least 0.5 mg/dL or 50% of baseline for at least two successive measurements. This study is registered at Clinicaltrials.gov (NCT03921099), April 2019. Results Mean absolute S.cr increase was significant when compared between both groups, P-value = 0.036, where S.cr increased by 0.05(0.12) and 0.34(0.55) mg/dL in the intervention and control groups, respectively. Mean absolute Cr.cl decline was significant when compared between both groups, P-value = 0.04, where Cr.cl was decreased by 5.9(17.8) and 22.3(30.4) ml/min in the intervention and control groups, respectively. Incidence of VAN was 1/21(4.7%) versus 5/20(25%) in the intervention and control groups, respectively (RR: 0.19; CI: 0.024–1.49; P-value = 0.093). Mortality was higher in the control group; however, it was not statistically significant, P-value = 0.141. Conclusion Co-administration of ascorbic acid with vancomycin preserved renal function and reduced the absolute risk of VAN by 20.3%, however, the reduction in VAN incidence didn’t reach statistical significance level. Further large multicenter prospective trials are recommended.

The aims of this study were to determine the modulating role of ascorbic acid (AA) on rectal temperature (RT), heterophil to lymphocyte (H to L) ratio and aberrant behaviours of ostrich chicks transported by road for 4 h during hot-dry conditions. Twenty ostrich chicks aged 2.5 months, of both sexes and belonging to the Red Neck breed, served as subjects of the study. The chicks were assigned randomly to AA-treated and control groups, consisting of 10 chicks each. The AA-treated group was administered orally with 100 mg/kg body weight of AA dissolved in 5 mL of sterile water 30 min before transportation, whilst the control group was given the equivalent of sterile water only. The thermal load (TL) experienced in the vehicle during transportation fluctuated between 31 °C and 89 °C, as calculated from the ambient temperature and relative humidity. Transportation induced hyperthermia, lymphopenia, heterophilia and aberrant behaviours of pecking, wing fluffing and panting, which were ameliorated by AA administration. The relationships between the TL, journey duration and physiological variables of RT, H to L ratio and aberrant behaviours recorded during transportation were significantly and positively correlated in the control group. In AA-treated group the relationships were not significantly correlated. In conclusion, the results showed for the first time that AA ameliorated the adverse effects of stress caused by road transportation on the aberrant behaviours, RT and H to L ratio of ostrich chicks during the hot-dry season.

Moxidectin administered in January or February at a single dose was tested for efficacy in horses on two farms for 12 and 11 months, respectively. Horses were infected with cyathostomes naturally in the previous grazing period. Forty horses of farm 1 and 20 horses of farm 2 were used in controlled tests to evaluate the efficacy of moxidectin 2% gel formulation at the dosage 0.4 mg moxidectin per kg of live weight, ivermectin commercial paste formulation at the dosage 0.2 mg ivermectin per kg of live weight, mebendazole and fenbendazole commercial paste formulation at the dosage both 7.5 mg mebendazole and fenbendazole per kg of live weight, all applied orally. Three control groups of 10 horses each (farm 1) were treated twice a year with ivermectin and benzimidazoles, respectively. Individual faecal egg counts, faecal cultures and larval differentiation were performed. Moxidectin had more prolonged and greater suppressive effects on the post-treatment reappearance and magnitude of strongyle egg counts than did ivermectin or benzimidazoles. In the moxidectin treated group (M1) strongyle eggs were seen for the first time in April and a slight increase in the mean count of eggs per gram of faeces (EPG) was observed during the rest of the season. Litter larval counts significantly reflected levels of exposure during the tested season. Twenty animals of farm 2 were allocated into two groups of ten horses each based on pre-treatment eggs per gram (EPG) counts (moxidectin treated group and control group). In the moxidectin treated group mean egg counts remained very low throughout the study. A plateau was reached by autumn, with egg counts ranging from 74 to 145 EPG. The faecal egg counts of moxidectin treated group (M2) were significantly higher in March, April, May and June.

Abstract: Although some studies in the southeast part of Guizhou Province have suggested that Miaoyao Fanggan Sachets (MFS) prevent influenza, little is known about its influence on immune systems. Influenza virus mainly infects immune-compromised individuals. The effects of MFS have mainly been recognized in clinical practice. However, there have been relatively few studies on its biological mechanism. Here we investigated whether MFS was able to affect the mucosal immunization and the activation of alveolar macrophages (AM), CD4+and CD8+ T-cells: Eighty Kunming male mice were treated with MFS continuously or intermittently with Yu-Ping-Feng powder (YPF-P) (positive control group) or with normal saline (NS) (control group) for 4 weeks, respectively. Mice treated with MFS were further divided into the continuous inhalation group (12 h daily/4 weeks) and the discontinuous inhalation group (1 h, three times a day for 4 weeks). Mice in both groups were placed under 0.5 m: Mice continuously and intermittently inhaling MFS showed a moderate increase in IgA and IgG1 protein levels compared with mice in the control groups. There was also a slightly significant increase in the number of AM in the continuous inhalation group compared with mice in the control groups (p<0.05). Furthermore, compared with controls, there was also a slightly significant increase in the number and percentage of CD4+and CD8+T-cells in both the continuous inhalation group and the discontinuous inhalation group (p<0.05).: MFS was able to up-regulate the protein levels of sIgA and IgG1. Meanwhile, MFS could activate AM, CD4+and CD8+T-cells in mice. Our data have, for the first time, demonstrated that the protection against influenza by MFS is partly through activation of the innate and adaptive cell-mediated immune responses, indicating MFS as a potential new immune-modulatory agent for respiratory tract infectious disease.

The aim of this study was to determine the post-parturitional changes in the proportion of the splenic CD4+ and CD8+ subpopulation of T lymphocytes after the birth of Toxocara canis infected C57Bl/6 mice and their offspring in relation to the intensity of infection. In mothers infected on the day of mating the proportion of CD4+ T lymphocytes showed an increase for the period of 21 days after the birth in comparison with the control group of the mothers. Conversely, the proportion of CD8+ T lymphocytes showed a significant decline throughout the whole course of the observation. In the offspring of infected mothers the initial increase of CD4+ after the birth turned into a significant decrease in comparison with the offspring of the healthy mothers. The reduction of CD8+ T lymphocytes was detected in the offspring of the infected mothers almost throughout the whole period of observation. The CD4+/CD8+ ratio showed a considerable increase in infected mothers and their offspring in comparison with the control groups. T. canis larvae in the muscle of offspring were found for the first time on the fifth day after the birth and the number of larvae showed a moderate increase. The results refer to the changes in the immune regulatory mechanisms in the T. canis infected mothers and the high level of miscarriages after infection.

Background: The aim of this study was to investigate the mechanisms underlying possible toxic effects of sulphite on neurodegeneration. Methods: Male Wistar rats were assigned to each of the four groups: Control (Control), Sulphite-treated (Sulphite), 6-hydroxydopamine (6-OHDA)-injected (6-OHDA), and sulphite-treated and 6-OHDA-injected (6-OHDA+Sulphite). Sodium metabisulphite was administered orally by gavage at a dose of 100 mg/kg/day for 45 days. Experimental PD was created stereotactically via the unilateral infusion of 6-OHDA into the medial forebrain bundle (MFB). Rotarod performances, plasma S-sulfonate levels, caspase-3 activities, Bax and Bcl-2 levels, tyrosine hydroxylase (TH) and cleaved caspase-3 double staining were investigated. Results: The rotarod test showed that the 6-OHDA-injected animals exhibited shorter time on the rod mile compared to the control group; however, there was no difference between 6-OHDA and 6-OHDA+Sulphite groups. Plasma levels of S-sulfonate in Sulphite and 6-OHDA+ Sulphite groups increased in contrast to their corresponding control groups. Caspase-3 enzyme activity increased in the 6-OHDA group whereas it did not in control. However, sulphite treatment did not affect these activity levels. Anti-apoptotic protein Bcl-2 concentration decreased, but the concentration of pro-apoptotic protein Bax increased in the 6-OHDA group compared to the control group. The expression of caspase-3 increased, while the number of tyrosine hydroxylase (TH)-positive neurons decreased in 6-OHDA group as compared to the control groups. However, sulphite treatment had no effect on these parameters. Conclusion: Sulphite is not a potentially aggravating factor for the activity of caspase-3 in a 6- OHDA-induced experimental model of Parkinson’s disease.

Numerous scientific studies have been carried out on the predisposition of the Pro47Ser polymorphism of the TR53 gene to breast cancer in women living in most countries of the world. However, no studies have been conducted on the susceptibility of this Pro47Ser gene polymorphism to breast cancer in Uzbek women. In this article, genotyping of the Pro47Ser polymorphism of the TR53 gene was performed for the first time in 207 Uzbek women. According to the results of our study, the functionally dangerous T allele of the Pro47Ser polymorphism of the TR53 gene was statistically significantly higher than that in healthy donors in patients with breast cancer; (ch2 = 8.2; p = 0.004; OR = 11.2; 95% CI: 1.422-88.38; RR = 10.7; 95% CI: 1.382-82.82). The safe C allele, on the other hand, was more common in the control group than in the main group. The natural S / C genotype was found in the control group with a higher frequency than in the control group. However, the limit of the statistical difference reached a significant level: (ch2 = 8.4; p = 0.004; OR = 0.08; 95% CI: 0.01066-0.676; ch2 = 8.2; p = 0.004; RR = 0.1 ; 95% CI: 0.8489- 0.9723). The S / T genotype of this polymorphism significantly differs in the frequency of occurrence in the main and control groups. Relative risk according to statistical analysis: RR = 10.7; (95% CI: 1.395–82.07) and odds ratio: OR = 11.8; (95% CI: 1.479-93.77). It is known that the minor T allele and the S / T genotype of this polymorphism increase the risk of developing breast cancer by 10.7 times. It should be noted that the unique homozygous genotype of Pro47Ser polymorphism was not identified in patients and control groups studied by T / T. Thus, the T allele and the C / T genotype of the Pro47Ser polymorphism of the TR53 gene are important factors that increase the risk of breast cancer (r <0.05). The C allele and S / S genotype of this polymorphism are reliable protective markers against the development of this pathology. Therefore, we believe that the Pro47Ser polymorphism of the TR53 gene can be used as important genetic markers in determining the likelihood of developing breast cancer.

Conjugated linoleic acid (CLA) is a collective term for the positional and geometric isomers of a conjugated diene of linoleic acid (C18:2, n-6). The aims of the present study were to evaluate whether levels of hepatic α-tocopherol, α-tocopherol transfer protein (α-TTP), and antioxidant enzymes in mice were affected by a CLA-supplemented diet. C57BL/6 J mice were divided into the CLA and control groups, which were fed, respectively, a 5 % fat diet with or without 1 g/100 g of CLA (1:1 mixture of cis-9, trans-11 and trans-10, cis-12) for four weeks. α-Tocopherol levels in plasma and liver were significantly higher in the CLA group than in the control group. Liver α-TTP levels were also significantly increased in the CLA group, the α-TTP/β-actin ratio being 2.5-fold higher than that in control mice (p<0.01). Thiobarbituric acid-reactive substances were significantly decreased in the CLA group (p<0.01). There were no significant differences between the two groups in levels of three antioxidant enzymes (superoxide dismutase, glutathione peroxidase, and catalase). The accumulation of liver α-tocopherol seen with the CLA diet can be attributed to the antioxidant potential of CLA and the ability of α-TTP induction. The lack of changes in antioxidant enzyme protein levels and the reduced lipid peroxidation in the liver of CLA mice are due to α-tocopherol accumulation.

Previously, we have found that the addition of L-ascorbic acid to chitosan enhanced the reduction in body weight gain in guinea pigs fed a high-fat diet. We hypothesized that the addition of L-ascorbic acid to chitosan would accelerate the reduction of body weight in humans, similar to the animal model. Overweight subjects administered chitosan with or without L-ascorbic acid for 8 weeks, were assigned to three groups: Control group (N = 26, placebo, vehicle only), Chito group (N = 27, 3 g/day chitosan), and Chito-vita group (N = 27, 3 g/day chitosan plus 2 g/day L-ascorbic acid). The body weights and body mass index (BMI) of the Chito and Chito-vita groups decreased significantly (p < 0.05) compared to the Control group. The BMI of the Chito-vita group decreased significantly compared to the Chito group (Chito: -1.0 kg/m2 vs. Chito-vita: -1.6 kg/m2, p < 0.05). The results showed that the chitosan enhanced reduction of body weight and BMI was accentuated by the addition of L-ascorbic acid. The fat mass, percentage body fat, body circumference, and skinfold thickness in the Chito and Chito-vita groups decreased more than the Control group; however, these parameters were not significantly different between the three groups. Chitosan combined with L-ascorbic acid may be useful for controlling body weight.

Abstract. There is a growing interest in embitterment as psychological concept. However, little systematic research has been conducted to characterize this emotional reaction. Still, there is an ongoing debate about the distinctiveness of embitterment and its dimensions. Additionally, a categorical and a dimensional perspective on embitterment have been developed independently over the last decade. The present study investigates the dimensions of embitterment by bringing these two different approaches together, for the first time. The Bern Embitterment Inventory (BEI) was given to 49 patients diagnosed with “Posttraumatic Embitterment Disorder (PTED)” and a matched control group of 49 patients with psychological disorders with other dominant emotional dysregulations. The ability to discriminate between the two groups was assessed by t-tests and Receiver Operating Characteristic Curves (ROC curve analysis). PTED patients scored significantly higher on the BEI than the patients of the control group. ROC analyses indicated diagnostic accuracy of the inventory. Further, we conducted Confirmatory Factor Analyses (CFA) to examine the different dimensions of embitterment and their relations. As a result, we found four characteristic dimensions of embitterment, namely disappointment, lack of acknowledge, pessimism, and misanthropy. In general, our findings showed a common understanding of embitterment as a unique but multidimensional emotional reaction to distressful life-events.
