The Pecut kuda plant is one of the plants of the genus Stachytarpheta which is found in tropical regions, including Indonesia. In Indonesia this plant is found in East Kalimantan which grows wild. This study aims to determine the type of secondary metabolites and the level of toxicity of the leaves of horse whip (Stachytarpheta jamaicenesis (L.) Vahl). The research stages were sample drying, sample maceration, filtered, concentrated with a rotary evaporator, phytochemical and toxicity tests using linear regression values to determine the LC50 value with SAS probit analysis. Phytochemical test results showed that the pecut horse leaf extract in the n-hexane fraction contained flavonoids, quinones and steroids, the ethyl acetate fraction contained alkaloids, steroids, saponins, flavonoids and quinones and the methanol fraction contained phenolic flavonoids, quinones and saponins. The results of the toxicity test on Artemia salina L. shrimp larvae in the fraction of n-hexane, ethyl acetate and methanol obtained LC50 values of 0.036 ppm, 5.24 ppm and 29.51 ppm, respectively. Based on the LC50 value, it can be concluded that the three fractions are very toxic, but the most toxic is the n-hexane fraction (LC50 = 0.036 ppm).

ABSTRAKAedes aegypti merupakan serangga vektor dari virus Dengue penyebab Demam Berdarah Dengue (DBD). Kandungan senyawa dalam tumbuhan lakum berpotensi sebagai larvasida. Buah lakum dimaserasi dengan pelarut methanol kemudian difraksinasi menggunakan etil asetat. Fraksi diteliti kandungan senyawanya menggunakan analisis fitokimia. Uji larvasida diamati selama 48 jam terdiri dari tujuh konsentrasi berseri yaitu kontrol; 0,04; 0,12; 0,2; 0,6; 1; 3% (b/v). Setiap konsentrasi diulang 3 kali dan setiap ulangan terdiri dari 10 ekor larva nyamuk Ae. aegypti. Data mortalitas dianalisis menggunakan analisis probit untuk menentukan nilai LC50. Fraksi metanol buah lakum  berpotensi sebagai larvasida dengan nilai LC50 6 jam sebesar 1,125%. Fraksi etil asetat  buah lakum  berpotensi sebagai larvasida dengan nilai LC50 6 jam sebesar 1,102%. Buah lakum mengandung senyawa: polifenol, kuinon, flavonoid, alkaloid dan terpenoid. Kata kunci: Aedes aegypti; Cayratia trifolia (L.) Domin; larvasida; LC50  ABSTRACTLarvacidal activity of methanol and ethyl acetate fraction of lakum fruits (Cayratia trifolia (L.) Domin.) against Aedes aegypti larvae. Aedes aegypti is a vector from the Dengue virus which causes Dengue Hemorrhagic Fever (DHF). The compounds of lakum plants has the potential larvicide. The fruit of C. trifolia was macerated with methanol then fractionated using ethyl acetate as a solvent. The fraction was examined for its compound using phytochemical analysis. Larvacide test was observed for 48 hours consisting of seven concentration of 0 (control); 0.04; 0.12; 0.2; 0.6; 1; 3% (w/v). Each concentration was repeated 3 times and each repetition consisted of 10 larvae of Ae. aegypti. Mortality data were analyzed using probit analysis to determine the LC50 of the fraction of methanol and ethyl acetate from lakum fruit. The methanol fraction has the potential larvicide with a six hour LC50 value 1.125%. The ethyl acetate fraction has the potential larvicide with a six hour LC50 value 1.102%. Telah dilakukan penelitian mengenai Kajian Toksisitas dari Fraksi Heksana, Etil Asetat dan Etanol Daun Soyogik (Saurauia bracteosa DC). Daun Soyogik dikeringanginkan sampai kering dan dimaserasi berturut-turut menggunakan pelarut heksana, etil asetat dan etanol. Uji toksisitas dilakukan  menggunakan metode Brine Shrimp Lethality Test (BSLT), kemudian diidentifikasi presentasi kematian larva udang menggunakan analisis probit (LC50). Hasil pengujian toksisitas menunjukkan nilai LC50 yaitu : fraksi  heksana (181,97), fraksi etil asetat (12,97) dan fraksi etanol (2,82).A research has been about Toxicity  of the Fraction Soyogik Leaves (Saurauia bracteosa DC) from Hexane, Ethyl Acetate and Ethanol. Soyogik leaves drying till dunes and maceratied successively using hexane, ethyl acetate and ethanol solvent. Toxicity tests performed using Brine Shrimp Lethality Test (BSLT) methode. Toxicity testing showed the LC50 value is hexane fraction (181,97), ethyl acetate fraction (12,97) and fraction of ethanol (2,82).

Surian (Toona sinensis) is one of the herbs that contain many bioactive compounds. Part of surian plants that can be utilized as therapy is leaf and bark. This study aims to determine the toxicity of leaf and bark extract of Surian through Bhrine Shrimp Lethality Test (BSLT), then compare toxic concentration of water extract, and ethanol 70 %, and n-hexane, diethyl ether and ethyl acetate fractions to shrimp larvae (Artemia salina L). The results showed that water content of leaves simplicia was 8.08 % and bark was 6.26 %. The highest extract rendement value by ethanol solvent was 23 %. The result of BSLT test probit analysis showed that the extract of ethyl acetate bark fraction had the highest toxicity with LC50 value 405.641 ppm, while leaf water extract had the lowest toxicity with LC50 value 707.787 ppm.

Phytochemical screening and Brine Shrimp lethality test of pecut kuda leaf extract had been done against Artemia salina Leach. The aims of this study is to screen potentially bioactive extract of pecut kuda leaf as an effort ti find out the chemical substances responsible for the toxic effect. The Pecut kuda leaves (Stachytarpheta jamaicensis, L. Vahl) was extracted by methanol 80 percent, then partitioned successively with hexane, chloroform, and ethyl acetate. Phytochemical screening was done according to Ciulei method (1984). The toxicity effect was tested against A. salina, L (Brine Shrimp Test). The result of phytochemical screening known that compounds in the extract of pecut kuda leaves (S. jamaicensis, L. Vahl), hexane fraction contained of sterol and triterpenes; where the chloroform fraction contained of saponin, sterol, and triterpenes; further more the ethyl acetate fraction contained flavonoid, tannin, saponin, sterol, dan triterpenes. The result of the study shows that several fractions of extract of pecut kuda leaves (S. jamaicensis) hexane fraction, chloroform, and ethyl acetate was toxic because it was able to kill more than 50 percent larva of A. salina, L at the concentration less than 1000 ppm. The LC50 of all fractions of pecut kuda leaves extract (S. jamaicensis, L. Vahl) were shown from the most toxic to the lowest were as followed: LC50 of hexane fraction = 98.33 ppm, of chloroform raction LC50 = 204.17 ppm, and of ethyl acetate fraction LC50 = 249.80 ppm. Finally results of the TLC profile showed that all fraction (hexane, chloroform, and ethyl acetate) indicated that the pecut kuda leaves extract (S. jamaicensis) contained terpenoid compound.

Objective: This study aimed to compare the bronchorelaxation effect of the methanol extract, hexane fraction, ethyl acetate fraction, and methanol fraction of Coptosapelta flavescens Korth. roots (CFRs) on the isolated bronchial rings of guinea pig. Methods: Research design was experimental in which a 4-mm long bronchus of a male guinea pig was kept in an isolated organ bath containing Krebs-Henseleit solution at 37°C, pH 7.4, and flooded with carbogen gas. The bronchial ring was precontracted with methacholine and then given six doses of C. flavescens Korth. root’s methanol extract (MECFR) solution cumulatively. The extract’s solvent 10% dimethyl sulfoxide ethanol was used as negative control. This procedure was then repeated using CFRs hexane fraction (HFCFR) solution, CFRs ethyl acetate fraction (EAFCFR), and CFRs methanol fraction (MFCFR). Results: The bronchorelaxation effect of MECFR, MFCFR, EAFCFR, and HFCFR is shown by their dose-response curves (DRCs) which are significantly different compared with the extracts solvent’s DRC. The maximal efficacy (Emax) of MFCFR was the same (p>0.05) as the MECFRs Emax, but the EAFCFRs and HFCFRs Emax were smaller (p<0.05) than the MECFRs Emax. Conclusion: The bronchorelaxation effect of the MECFR on the guinea pig’s bronchial ring is similar to the CFRs methanol fraction, and is stronger than the CFRs ethyl acetate fraction and CFRs hexane fraction.

Purpose: To standardize Zakham-e-Hayat (Bryophyllum pinnatum Lam.) as well as determine its cytotoxic and insecticidal activity. Methods: Different parts of the plant (flowers, young leaves, old leaves and stem) and different solvents, viz, methanol, ethyl acetate and n-hexane were used for the extraction. Morphological characterization (macroscopic and microscopic), phytochemical screening for primary and secondary metabolites, and thin layer chromatography (TLC) using various solvent systems, namely, methanol/chloroform (MeOH/CHCl3), n-hexane/methanol/chloroform (n-hexane/MeOH/CHCl3), methanol/chloroform/water (MeOH/CHCl3/H2O), n-hexane/chloroform (n-hexane/CHCl3), n-hexane/ethyl acetate (n-hexane/EtOAc) were carried out. Fourier transform infrared spectroscopy (FTIR) was also performed. In addition, the extracts were evaluated for their cytotoxic and insecticidal activities. Results: The methanol fraction of the young leaves had 50% lethal dose (LD50) of 28.46 µg/mL while the ethyl acetate fraction of young and old leaves showed significant cytotoxicity at LD50 of 0.13 µg/mL and 142.68 µg/mL respectively. On the other hand, n-hexane fraction was inactive. The methanol fraction of the flower revealed the presence of carbohydrates, the ethyl acetate fraction showed the presence of carbohydrates and amino acids, while the n-hexane fraction showed the presence of carbohydrates as primary metabolites. Furthermore, alkaloids, flavonoid, saponins and resins were present in the methanol fraction, flavonoid and alkaloids in the ethyl acetate fraction, and alkaloids and resins in the n-hexane fraction as secondary metabolites. Conclusion: Methanol and n-hexane fractions of Bryophyllum pinnatum have some safe bioactive compounds which may exhibit potential health benefits.

Background: The prevalence of diabetes mellitus is increasing on a global trend. The aim of the present study is to identify the most effective antioxidants and hypoglycemic fraction of Azanza garckeana. Methods: The fractions (nhexane or ethyl-acetate or aqueous) of A. garckeana were administered to the alloxan-induced diabetic rats at doses of 100, 200, and 400 mg/kg for 15 days. Antioxidants activities were evaluated at concentrations of 62.5, 125, 250, and 500 µg/mL using the DPPH scavenging assay. Results: Results revealed that both the hexane, ethyl-acetate, and aqueous fractions exhibited hypoglycemic and antioxidants activities in a dose-dependent manner. The n-hexane fraction demonstrated highest percentage DPPH scavenging effect of 26.34±3.43, 38.44±4.35, 59.34±3.45, and 74.83±5.35 at 62.5, 125, 250, and 500 µg/mL respectively. The ethyl-acetate fraction demonstrated 19.33±2.98, 28.94±3.24, 47.34±2.90, and 57.82±4.54 respectively while the aqueous fraction exhibited the least activities of 12.45±23.45, 18.64±2.94, 27.94±3.89, and 39.43±3.89 at concentrations of 62.5, 125, 250, and 500 µg/mL respectively. In addition, the n-hexane fraction demonstrated the most significant hypoglycemic effect with the highest glucose reduction of 58.97 ±3.45 %, 63.86±5.35 %, and 66.51±4.35 %, ethyl acetate fraction demonstrated glucose reduction of 7.55±0.54%, 21.77±2.35 %, and 29.56±3.46 % while the aqueous fraction demonstrated the least hypoglycemic effect of 9.89±2.67 %, 18.09±3.45 %, and 18.87±3.24 at 100, 200 and 400 mg/kg bw respectively. Conclusion: The n-hexane fraction of Azanza garckeana extract could serve as a reservoir of bioactive agents that could be useful for the development of a new anti-diabetic agent

Antioxidants can prevent free radical formation. Natural antioxidants found in many plants, such as <em>Ficus benjamina </em>and <em>Annona reticulata</em>. The study aimed to compare the antioxidant activity of extracts and fractions of <em>Ficus benjamina </em>and <em>Annona reticulata</em> leaves against 1,1-diphenyl-2-picrilhydrazyl. The steps of this study consist of extraction, fractionation with n-hexane, ethyl acetate and water, phytochemical screening, antioxidant activity determination, and comparing the IC<sub>50</sub> values. Percentage scavenging activity of the extracts and fractions against DPPH was calculated to determine the antioxidant activity. The IC<sub>50</sub> value of <em>Ficus benjamina </em>was 127.86 ppm for ethanolic extract, 94.01 ppm for water fraction, 115.48 ppm for ethyl acetate fraction, and 335.50 ppm for n-hexane fraction. The IC<sub>50</sub> value of <em>Annona reticulata</em> was 274.31 ppm for ethanolic extract, 211.42 ppm for water fraction, 367.91 ppm for ethyl acetate fraction, and 741.08 ppm for n-hexane fraction. The results showed that the <em>Ficus benjamina</em> water fraction was the best antioxidant compared to other extract and fraction.

Mangosteen leaves have flavonoid compounds, tannins, and saponins that can be efficacious as antibacterial. The aim of this study was to determine the fraction of mangosteen leaves having an antibacterial effect and knowing the class of compounds identified as having antibacterial activity after TLC- Bioautography testing was carried out. The samples were extracted using 96% maceratarion method and fractioned using liquid-liquid fractionation method with methanol, n-hexane and ethyl acetate solvents, antibacterial activity using agar diffusion method (Kirby and Bauer) with 3 concetrations namely 10%, 20% and 30%. Thin Layer Chromatography (TLC) uses n-hexane and chloroform solvens. TLC-Bioautography uses contact bioautography methods. The resultd showed that mangosteen leaves in methanol fraction with a concentration of 30% had a very large inhibitory activity again Staphylococcus aureus and ethyl acetate fraction with a concentration of 30% had the gratest antibacterial activity against  Escherichia coli. The results of the TLC- Bioautography study showed that the flavonoids compounds after spraying with AlCl3 and the mangosteen leaf Biosutography test had inhibitory zone activity against the bacteria Staphylococcus aureus dan Escherichia coli. Keywords: Mangosteen Leaves. Antibacterial, TLC Bioautography.  ABSTRAK Daun manggis mempunyai senyawa flavonoid, tanin, dan saponin yang dapat berkhasiat sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui fraksi daun manggis memiliki efek antibakteri dan mengetahui golongan senyawa yang teridentifikasi memiliki aktivitas antibakteri setelah dilakukan pengujian KLT Bioautografi. Sampel diektrak dengan metode maserasi dengan pelarut 96% dan difraksinasi dengan metode  fraksinasi cair-cair dengan pelarut metanol, n-heksan dan etil asetat, aktivitas antibakteri menggunakan metode difusi agar (Kirby and Bauer) dengan 3 kosentrasi yaitu 10%, 20% dan 30%. Kromatografi Lapis Tipis (KLT) menggunakan pelarut n-heksan dan klorofom. KLT-Bioautografi menggunakan metode bioautografi kontak. Hasil penelitian menunjukan daun manggis pada fraksi metanol  dengan kosentrasi 30% memiliki aktivitas zona hambat ppaling besar terhadapat bakteri Staphylococcus aureus dan fraksi etil asetat dengan kosentrasi 30% memiliki aktivitas antibakteri paling besar terhadap  Escherichia coli. Hasil penelitian KLT-Bioautografi menunjukan golongan senyawa flavonoid setelah disemprotkan dengan AlCl3 dan uji Bioautografi daun manggis memiliki aktivitas zona hambat terhadap bakteri Staphylococcus aureus dan Escherichia coli. Kata kunci : Daun Manggis, Antibakteri, KLT- Bioautografi.

Objective: Arginase inhibition could be a potential therapeutic approach for endothelial dysfunction. Syzygium cumini (L.) Skeels leaves containphenolic acids and flavonoids, which have been predicted to exhibit arginase inhibitory activity. Moreover, these leaves contain tannins, which canform complexes with enzymes and lead to false-positive results during biological testing. Therefore, this study was conducted to evaluate the arginaseinhibitory activity of S. cumini leaf extract and fractions as well as to elucidate the effects of tannins on this activity.Methods: S. cumini leaves were fractionated using n-hexane, ethyl acetate, and methanol. A colorimetric method was employed to evaluate arginaseinhibitory activity. Tannin elimination was performed through the gelatin precipitation method. Total phenolic and flavonoid contents of the fractionswere calculated using the Folin–Ciocalteu and aluminum chloride methods, respectively.Results: Ethyl acetate and methanol fractions showed arginase inhibitory activity with half-maximal inhibitory concentrations (IC50) of 46.96 and15.35 μg/mL, respectively. The methanol fraction was positive for tannins. After tannin elimination, this fraction exhibited less potent arginaseinhibitory activity, with an IC50 value of 53.03 μg/mL. The ethyl acetate fraction showed higher total phenolic and flavonoid contents than the methanolfraction.Conclusion: Tannins affected the arginase inhibitory activity of the methanol fraction of S. cumini leaves; however, the ethyl acetate fraction did notcontain tannins and could inhibit arginase activity.
