Uji Aktivitas Antimikroba dan Antioksidan dari Ekstrak Etanol Daun Keji Beling (Strobilanthes crispus BI)

Uji aktivitas antimikroba dari ekstrak etanol daun keji beling (Strobilanthes crispus BI) telah dilakukan terhadap mikroba Candida albicans, Microsporum gypseum, Bacillus cereus, dan Shigella dysentriae dengan metode difusi cakram. Uji aktivitas antioksidan ekstrak etanol daun keji beling dilakukan dengan metode DPPH. Daun keji beling yang telah halus, diekstraksi dengan metode maserasi menggunakan pelarut etanol 96% selama 2 hari, dan dipekatkan dengan rotary vacum evaporator. Uji aktivitas antimikroba dilakukan dengan variasi konsentrasi 100; 200; 300; 400; 500 mg/ml. Hasil skrining fitokimia ekstrak etanol daun keji beling menunjukkan hasil positif terhadap senyawa golongan alkaloid dan fenolik. Hasil penelitian menunjukkan ekstrak etanol daun keji beling efektif terhadap mikroba Candida albicans dan tidak efektif terhadap mikroba Microsporum gypseum, Bacillus cereus, dan Shigella dysentriae. Ekstrak etanol daun kejibeling mampu menghambat pertumbuhan Candida albicans dengan diameter zona bening untuk konsentrasi 100; 200; 300; 400; 500 mg/ml masing-masing adalah 11,53; 13,96; 15,46; 16,80; dan 18,20 mm. Uji aktivitas antioksidan ekstrak etanol daun keji beling dengan metode DPPH menunjukkan nilai IC50 sebesar 155,2 ppm.   The antimicrobial and antioxidant activity evaluation of leaves ethanol extract of KejiBeling (Strobilanthescrispus BI) has been carried out. The leaves of KejiBeling was grounded into powder and extracted using ethanol 96% (v/v) for 2x24 h and concentrated with rotary vacum evaporator. The antimicrobial activity was performed against several microbes, i.e. Candida albicans, Microsporumgypseum, Bacillus cereus, andShigella dysentriaeusing disk diffusion method. The antimicrobial activity was performed using several concentrations of leaves extract from 100-500mg/mL.The phytochemical screening showed the presence of alkaloid and phenolic group intheleaves ethanol extract of KejiBeling. The leaves ethanol extract of KejiBeling was antimicrobial active against Candida albicans with inhibition zone diameter of 11,53; 13,96; 15,46; 16,80; and 18,20 mm for the concentration of 100; 200; 300; 400; 500 mg/ml, respectively. The antioxidant activity of leaves ethanol extract showed IC50 value of 155,2 ppm

ABSTRACTTunicate is an invertebrate that lives in a coral reef ecosystem and produces many compounds such as, antibacterial, antitumor and anticancer. This study aimed to determine the antimicrobial activity of extracts and fraction of tunicate (Polycarpa aurata) collected in the Lembeh Strait, Bitung against Escherichia coli, Staphylococcus aureus and Candida albicans. Tunicate (Polycarpa aurata) was extracted by maceration method using 96% ethanol solvent, fractination using liquid-liquid partition method with n-hexane, chloroform and methanol solvent, and antimicrobial testing using Kirby Bauer’s disk diffusion method. The results showed that ethanol extract of tunicate (Polycarpa aurata) had antimicrobial activity againts Escherichia coli with an inhibition of 15.12 mm, and againts Candida albicans with an inhibition of 15 mm. While the methanol fraction showed antimicrobial with a strong category and inhibition of 16.17 mm againts Staphylococcus aureus. Keyword: Tunicate (Polycarpa aurata), Extraction, Fractination, Antimicrobials  ABSTRAKTunikata merupakan invertebrata di ekosistem terumbu karang yang banyak menghasilkan senyawa seperti, antibakteri, antitumor dan antikanker.  Penelitian ini bertujuan untuk  mengetahui aktivitas antimikroba ekstrak dan fraksi tunikata (Polycarpa aurata) yang dikoleksi di Selat lembeh, Bitung terhadap Escherichia coli, Staphylococcus aureus dan Candida albicans.  Tunikata (Polycarpa aurata) diekstraksi menggunakan metode maserasi dengan pelarut etanol 96%, fraksinasi menggunakan metode partisi dengan pelarut n-heksan, kloroform dan metanol, dan pengujian antimikroba menggunakan metode difusi agar Kirby Bauer. Hasil penelitian menunjukkan bahwa ekstrak etanol tunikata (Polycarpa aurata) memiliki aktivitas antimikroba kategori kuat pada fraksi metanol dengan daya hambat sebesar 16, 17 mm terhadap Escherichia coli, pada ekstrak etanol dengan daya hambat sebesar 15, 12 mm terhadap Staphylococcus aureus sedangkan pada Candida albicans aktivitas yang sangat baik terjadi pada ektraksi etanol sebesar 15 mm. Kata Kunci: Tunikata (Polycarpa aurata), Ekstraksi dan Fraksinasi, Antimikroba

Startfruit (Averrhoa bilimbi L.) is one the kind of plant that is widely used as a traditional herbal medicineto overcome various diseases i.e. diabetes mellitus, cough, rheumatism, thrush, diabetes, toothache, acne, etc. The secondary metabolites of the starfruit leaves contain flavonoids and tannins, whereby these activecompounds can be used as an antibacterial. This study aimed to determine the inhibiting power of starfruit leaf ethanol extract against Staphylococcus epidermidis at concentrations of 25%, 50%, 75%, and 100%. The research was conducted at the  AKAFARMA Laboratory and the Chemistry Laboratory , Faculty of Teacher Training and Education, Syiah Kuala University from June to July 2020. The research method was laboratory experimental using the disk diffusion method. The population of starfruit leaves was obtained from Lhokseumawe  using a purposive sampling technique. Ethanol extract of starfruit leaves with concentrations of 25%, 50%, 75%, and 100%. The diameter of the inhibition zone of the ethanol extract  from the starfruit leaves at  concentrations of 100%, 75%, 50%, and 25% were 15 mm, 12 mm, 11 mm and 10 mm, respectively. It can be concluded that  ethanol extract of starfruit leaves can inhibit the growth  of  Staphylococcus  epidermidis  with  a  maximum inhibitory concentration  of  100%,  which is classified  as a strong category.

Genus Mentha presents group of plants which are the most studied in family Lamiaceae. Aboveground parts are used for different purposes in pharmacy, food industry or confectionery. Most important is natural product extracted from leaves - essential oil (EO). The aim of presented experiment was to demonstrate different chemotype and compare antibacterial activity of two Mentha species EO. Plant samples were obtained from various environments – from Slovakia and from Italy. Dominant compounds were determined by GC/MS. The results showed high amount of menthol and menthone in tested Slovak peppermint EO. On the other hand, carvone and 1,8-cineole were determinate as dominant compounds in Italian spearmint EO. The antimicrobial activity of the EO was investigated by disc diffusion and broth micro dilution methods. EO was evaluated for their antibacterial activity against 7 microorganisms: Enterobacter cloacae, Salmonella spp., Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus, Streptococcus pyogenes. The determination results of antibacterial activity by agar disk diffusion method ranged from 7 to 14 mm of the growth inhibition zone. MIC of tested mint EO varied from 0.625 to 2.5 μg/mL. In addition, both EO showed relatively the same antibacterial activity against the selected Gram-negative bacteria. However, there is a variation in the antibacterial activity against Gram-positive bacteria.

The ethanol extracts of selected Acinos Miller species were investigated in terms of chemical composition, antimicrobial and antioxidative activities. Qualitative and quantitative analyses of the extracts were performed using GC and GC-MS. Eighty-six constituents, accounting for 93.70-99.99% of the total composition of the extracts, were identified. The ethanol extracts of A. majoranifolius, A. suaveolens and A. alpinus were characterized by domination of monoterpenes, representing 85.03%, 57.39% and 28.02% of the total extracts, respectively. Fatty acids and their esters in the A. arvensis and A. graveolens extracts reached 28.97% and 30.75%. Also, n-alkanes were the major compounds found in A. hungaricus ethanol extract (30.98%). The extracts were characterized by determination of total polyphenols, flavonoids and tannins. Besides, the antioxidant activity of the investigated extracts was estimated by two assays: DPPH and FRAP test. The highest antioxidant activity was observed in the extract of A. alpinus which had high levels of all examined polyphenol classes. A disk diffusion method was employed for the determination of the antimicrobial activities of the ethanol extracts. Gram-positive: Bacillus subtilis, Sarcina lutea, Staphylococcus aureus, Clostridium pyogenes, Enterococcus sp., Micrococcus flavus; Gram-negative: Klebsiella pneumoniae, Pseudomonas aerugionsa, Salmonella enteritidis, Proteus vulgaris, Escherichia coli and fungal organisms: Aspergillus niger, Candida albicans and Saccharomyces cerevisiae were used as test microorganisms. The results of preliminary bioassay demonstrated that the A. alpinus ethanol extract could be a possible source of compounds with antioxidant and antimicrobial activity.

Saluang Belum (Luvunga sarmentosa Kurz) adalah tumbuhan yang dikenal sebagai obat tradisional di Kalimantan Tengah. Batang L. sarmentosa Kurz dipercayai memiliki manfaat secara empiris sebagai antioksidan yang dimanfaatkan sebagai anti-aging (anti penuaan dini). Batang L. sarmentosa Kurz mengandung metabolit sekunder yaitu tanin, saponin, steroid dan flavonoid yang diduga memiliki aktivitas sebagai antibakteri. Berdasarkan khasiat empiris dan kandungan metabolit sekunder yang ada dalam tumbuhan L. sarmentosa Kurz, maka dilakukan penelitian tentang aktivitas daya hambat ekstrak etanol tumbuhan ini terhadap bakteri P. acnes yang merupakan salah satu bakteri penyebab jerawat. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas daya hambat ekstrak etanol batang L. sarmentosa Kurz terhadap bakteri P.acnes. Metode penelitian yang dilakukan pada penelitian ini dimulai dari pengambilan simplisia, pembuatan ekstrak etanol dengan metode perkolasi dan uji daya hambat dengan menggunakan metode Disk Difusion yang menggunakan kontrol positif antibiotik klindamicin. Analisis data dilakukan dengan membandingkan lebar zona hambat dari ekstrak etanol batang L. sarmentosa Kurz  dengan kategori penghambatan antimikroba klindamisin berdasarkan diameter zona hambat dari Clinical and Laboratory Standards Institute.  Hasil uji daya hambat didapatkan zona hambat ekstrak etanol batang Saluang Belum pada semua konsentrasi 0,5%, 1%, 5%, 10% dan 15% secara berturut-turut yaitu  Intermediate, Resistant, Intermediate, Susceptible dan Susceptible dan hasil dari zona hambat klindamicin pada semua konsentrasi 0,5%,1%,5%,10%,dan 15% adalah susceptible. Simpulan dari penelitian ini adalah ekstrak etanol batang L. sarmentosa Kurz mempunyai kemampuan dalam menghambat pertumbuhan bakteri P.acnes.           Kata Kunci: Obat Tradisional, Jerawat, Uji Daya Hambat, Propionibacterium acnes, Batang Saluang Belum Saluang belum (Luvunga sarmentosa Kurz) is one of traditional medicine in Central Kalimantan. The stem of L. sarmentosa Kurz is believed to have empirical benefits as an antioxidant which is used as anti-ageing (anti-ageing). The stem of L. sarmentosa Kurz contains secondary metabolites, namely tannins, saponins, steroids and flavonoids which are thought to have antibacterial activity. Based on the empirical properties and the content of secondary metabolites in L. sarmentosa Kurz, a study was conducted on the inhibitory activity of this plant's ethanol extract against P. acnes, which is one of the bacteria that causes acne. The purpose of this study was to determine the inhibitory activity of the ethanol extract of L. sarmentosa Kurz stem against P.acnes bacteria. The research method carried out in this study started from taking simplicia, making ethanol extract using the percolation method and the inhibition test using the disk diffusion method which used a positive control of the clindamycin antibiotic. Data analysis was performed by comparing the inhibition zone width of the ethanol extract of L. sarmentosa Kurz stem with the clindamycin antimicrobial inhibition category based on the inhibition zone diameter of the Clinical and Laboratory Standards Institute. The results of the inhibition test obtained the inhibition zone of Saluang Belum stem ethanol extract at all concentrations of 0.5%, 1%, 5%, 10% and 15% respectively, Intermediate, Resistant, Intermediate, Susceptible and Susceptible and the results of the inhibition zone. Clindamycin at all concentrations of 0.5%, 1%, 5%, 10%, and 15% are susceptible. This research concludes that the ethanol extract of L. sarmentosa Kurz stem can inhibit the growth of P.acnes bacteria.

The use of natural preservatives called bacteriocin derived from lactic acid bacteria (LAB) is one way of preventing food from being contaminated by pathogenic microorganisms such as L. monocytogenes (LM). The aims of this study were to evaluate the ability of LAB isolated from dadih to inhibit the growth of LM and to obtain the antimicrobial components that play a role in inhibiting the growth of LM. The antimicrobial activity of the supernatant obtained from 12 strains of dadih LAB was determined using the paper disk diffusion method. The results showed that the supernatant from the 12 LAB strains was able to inhibit the growth of LM with various inhibition zones. However, out of the 12 LABs, only 9 strains were found to have an inhibition zone of more than 3.5 mm.  The antimicrobial compounds of 9 strains were tested and it was found that the antimicrobial compounds of strains R-8, R-14 and R-49 were derived from lactic acid. In addition, 6 strains namely R-43, R-32, R-19, R-55, R-45 and R-41 were derived from bacteriocin based on their sensitivity to pH, heat and enzyme treatments. Crude bacteriocin derived from 6 LAB strains inhibited the growth of LM, and the highest antimicrobial activity was obtained in Streptococcus faecalis subsp. liquefaciens R-55 with an average inhibition zone of 13.87 mm. Bacteriocin produced by strain R-55 can be used as natural preservatives for the prevention of food-borne disease caused by LM.

The use of antibiotics is one of the most important ways to deal with the spread and treatment of pathogenic microbial infections. The search for new antibiotic sources continues to be carried out to anticipate the emergence of microbial resistance. One of the natural resources that has the potential as an antimicrobial source is a member of the macrofungi of the Genus Ganoderma. The purpose of this study was to evaluate the antimicrobial performance of the ethanol extracts of Ganoderma  lucidum, G. applanatum dan Ganorderma sp. against fungi (Candida albicans dan Cryptococcus neoformans),  gram positive bacteria (Bacillus cereus dan Staphylococcus aureus) and gram negative bacteria (Escherichia coli dan Shigella sp.). Macrofungi samples were taken from the forest area of Nature Tourism Park (TWA) Gunung Tunak, TWA Kerandangan, TWA Suranadi, TWA Nuraksa Sesaot, TWA Lemor and Pusuk forest. The stages of the method performed are sample collection, sample preparation, extraction, and testing of antimicrobial activity using the well difusion method. The ethanol extract concentrations for testing were 20%, 40%, 60% and 80%. The results showed that the three Ganoderma species had anti-fungal and antibacterial activity and that different levels of concentration had an effect on inhibition. The size of the inhibition zone is directly proportional to the higher the extract concentration. The antimicrobial activity of the ethanol extract of G. lucidum was higher when compared to G. applanatum and Ganoderma sp. both against fungi (Candida albicans and Cryptococcus neoformans) as well as against gram-positive and gram-negative test bacteria. In addition, G. applanatum showed very weak inhibition against both groups of tested bacteria.  Key words: candidiasis; cryptococcosis; comorbid infections; macerations; pathogens

ABSTRAK Kunyit (Curcuma longa L.) merupakan salah satu jenis tanaman obat yang termasuk dalam keluarga Zingiberaceae. Senyawa aktif yang terkandung dalam rimpang kunyit (Curcuma longa L.) mampu bekerja sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari ekstrak etanol rimpang kunyit (Curcuma longa L.) terhadap Staphylococcus aureus dan Pseudomonas aeruginosa. Ekstraksi dilakukan dengan cara refluks menggunakan pelarut etanol 96%. Pengujian aktivitas antibakteri dilakukan menggunakan metode difusi paper disk dengan masing-masing konsentrasi ekstrak 10%, 20%, 30%, 40% b/v. Kontrol positif yang digunakan adalah Ciprofloxacin sedangkan kontrol negatif yang digunakan adalah DMSO. Hasil skrining fitokimia ekstrak etanol rimpang kunyit mengandung alkaloid, flavonoid, fenol, tanin dan terpenoid. Berdasarkan hasil penelitian, ekstrak etanol rimpang kunyit dapat menghambat bakteri Staphylococcus aureus dan Pseudomonas aeruginosa pada konsentrasi 10%, 20%, 30%, 40% dan konsentrasi 40% merupakan konsentrasi yang memberikan diameter zona hambat terbesar terhadap kedua bakteri uji yaitu 8,63 mm dan 7,8 mm. The active compounds contained in the Curcuma longa L. rhizome can work as antibacterial. This study aims to determine the antibacterial activity of the ethanol extract of Curcuma longa L. Antibacterial activity testing was carried out using the paper disk diffusion method with each extract concentration of 10%, 20%, 30%, 40%. The positive control used was Ciprofloxacin while the negative control used was DMSO. The results of phytochemical screening of the ethanol extract of Curcuma longa L. rhizome contain alkaloids, flavonoids, phenols, tannins and terpenoids. Based on the results of the study, the ethanol extract of turmeric rhizome can inhibit Staphylococcus aureus and Pseudomonas aeruginosa bacteria at concentrations of 10%, 20%, 30%, 40% and 40% concentrations which give the largest inhibition zone diameter of the two test bacteria which is 8.63 mm and 7.8 mm. Different concentrations of ethanolic extracts of thyme (Zataria multiflora) and rosemary (Rosmarinus officinalis) were evaluated to determine their antimicrobial activity using the agar-well diffusion method. The values of inhibition zone diameter (IZD) for Candida albicans fungus and Staphylococcus aureus Gram-positive bacteria were determined. The bioactivities of two various extracts were studied, and the chemical composition of the extracts were identified using gas chromatography-mass spectrometry (GC-MS) technique. The results of the test showed that at concentrations of 10% and 40% thyme extract, the values of IZD were 12.5 mm and 23.3 mm, respectively, against the growth of S. aureus, which were higher than C. albicans (7.0 mm and 22.5 mm, respectively). The rosemary extract at concentrations of 20% and 60% showed lower antibacterial activity against S. aureus (4.7 mm and 8.7 mm IZD, respectively) and lower antifungal activity against C. albicans (12.2 mm and 1.7 mm IZD, respectively). At a concentration of 40% thyme extract, the highest antibacterial (23.3 mm IZD) and antifungal (22.5 mm IZD) activities were observed. The GC/MS analysis showed that carvacrol (52.3%), linalool L (16%), and thymol (9.6%) were the main components of thyme extract, while in the rosemary extract β-amyrone (18.0%), verbenone (8.0%), and 1,8-cineole (7.26%) were the major constituents.

The main chemical composition, i.e. the total content of bioactive compounds (phenolics 2209.86 ? 70.32 mg GAE/100g FDR, flavonoids 831.87 ? 12.61 mg R/100g FDR and anthocyanins 144.55 ? 0.39 mg CGE/100g FDR), in freeze-dried raspberry (FDR) was evaluated spectrophotometrically. Vitamin C content was determined by HPLC analysis (88.81 ? 4.38 mg vit C/100g FDR). Antioxidant activities of FDR extract were evaluated spectrophotometrically on stable 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals and by electron spin resonance spectroscopy (ESR) method on hydroxyl radicals (?OH). EC50 values were evaluated. EC50 DPPH? was 0.127 ? 0.013 mg/ml, while EC50 ?OH was 1.366 ? 0.026 mg/ml. Antiproliferative activity of the FDR extract was evaluated in vitro in three human cell lines by colorimetric sulphorhodamine B (SRB) assay. The most pronounced effects were obtained in the breast adenocarcinoma cell line (MCF7). EC50 value was 395.07 ? 96.38 ?g/ml. Antimicrobial activity was determined by disk diffusion method. The FDR extract produced a clear inhibition zone (without visible colonies) only toward Staphylococcus aureus. The minimal inhibitory (MIC) and minimal bactericidal (MBC) concentrations of FDR extract were evaluated. The values MIC were in the range of 4.7 - 100 mg/ml, and of MBC in the range of 6.3 - > 100 mg/ml.
