The White-Headed Ukrainian breed of cattle belongs to breeds of a dairy direction of productivity and belongs to small disappearing breeds. According to calculations, in accordance with the FAO recommendations, the White-Headed Ukrainian breed of cattle is at risk. The aim of the work was to establish the karyotype variability of White-Headed Ukrainian cows, whose animals belong to closed gene pool herds. Cytogenetic monitoring showed that the quantitative chromosome abnormalities, in particular aneuploidy, in White-Headed Ukrainian cows of two groups according to the purpose of the cow (selection nucleus and production group) did not exceed the spontaneous level of this variability (1.5-8.3%) and was 6.0% and 7.0% respectively. A multiple increase in the number of chromosomes (polyploidy) was not detected. The average value of metaphase plates with asynchronous differences in the centromeric regions of chromosomes was 1.0% and corresponded to the level of this variability characteristic of the species Bos taurus. The frequency of structural chromosome abnormalities (chromosomal breaks) in animals of the two groups was 2.3% and 2.0%, respectively, which corresponds to the norm characteristic of cattle with spontaneous mutagenesis. The results of the micronucleus test showed that the proportion of lymphocytes with micronuclei in animals of the two groups was in the range of 4.8 - 5.0%, dinuclear lymphocytes 5.6-6.4 %, and the proportion of dividing cells (mitotic index) was 2.8-3.8 %. The frequency of cells with aneuploidy in cows of the Ukrainian Black-Spotted dairy breed of this farm was established higher than the spontaneous level. The difference between the mean values on this basis in cows of White-Headed Ukrainian and Ukrainian Black-Spotted dairy breeds was statistically significant at P> 0.99. In cows of the Ukrainian Black-Spotted dairy breed under conditions of intensive use, an increase in the frequency of metaphases with aneuploidy was observed, which was not manifested in cows of the White-Headed Ukrainian breed.

Abstract The development of spontaneous bacterial peritonitis (SBP) is a serious and life-threatening condition in patients with cirrhosis and ascites. The aim of this study was to determine the diagnostic potential of calprotectin in ascites, for SBP in patients with liver cirrhosis and ascites before and after antibiotic treatment and to compare the mean values of calprotectin in ascites in patients with and without SBP. This prospective-observational study was comprised of 70 patients with cirrhosis and ascites, divided into two groups, the SBP and the non-SBP group. Quantitative measurements of calprotectin in ascites was completed with the Quantum Blue Calprotectin Ascites test (LF-ASC25), using the Quantum Blue Reader. The average value of calprotectin in the SBP group was 1.5 ± 0.40 μg / mL, and in the non-SBP group it was lower (0.4 ± 0.30). The difference between the mean values was statistically significant with p <0.05. The mean value of calprotectin in ascites before therapy among the SBP group was 1.5 ± 0.4, and after antibiotic therapy, the value decreased significantly to 1.0 ± 0.6; the difference between the mean values was statistically significant with p <0.05. ROC analysis indicated that calprotectin contributed to the diagnosis of SBP with a 94.3% sensitivity rating (to correctly identify positives), and the specificity was 62.5%, which corresponded to the value of 0.275. Our research confirmed that ascitic calprotectin was a good predictor, and is significantly associated with the occurrence of SBP in patients with liver cirrhosis. By monitoring the value of calprotectin in ascites on the 7th day of antibiotic treatment, the effectiveness of antibiotic treatment in patients with SBP can be determined.

Aim: To evaluate the influence of three different working lengths on the amount of apically extruded debris. Methodology: Thirty lower premolars with single roots and straight root canals were standardized at 17 mm. Then, they were inserted into Eppendorf tubes, and the 1.5% agar gel was introduced into the tubes surrounding the roots. The coronal section of the roots was kept visible. The set of tubes and agar gel was weighed 3 times and the average value was recorded. Then, the specimens were randomly distributed in 3 different groups according to the working length (CT) used for the instrumentation: Group (CT -1) - the working length 1 mm below the major foramen (MF); Group (CT 00) - the length was determined at the MF, and Group (CT +1) - the CT was determined 1 mm beyond the MF. Instrumentation was performed with Reciproc Blue R25 (VDW, Munich, Germany) under irrigation with 0.9% saline. After preparation, samples were removed from Eppendorf tubes and weighed 3 times again. The difference between the mean values of the initial and final weight was recorded. The one-way ANOVA test (post-hoc Bonferroni) was used with P> 0.05. Results: The average weight of the extruded waste was 0.0134 ± 0.0157 for CT -1, 0.0075 ± 0.0062 for CT 00 and 0.0075 ± 0.0068 for CT +1, with no statistically significant differences between groups. Conclusion: There was no impact of the different CTs on the amount of extruded debris beyond the apex.

It is very important for patients with spontaneous bacterial peritonitis (SBP) to assess the length of survival and the risk of death, primarily because of the wide range of potential complications that can lead to multisystem organ failure and fatal outcome. The aim of this study was to determine the predictive potential of MELD and Child-Turcotte-Pugh II score for SBP in patients with cirrhosis and ascites. Material and methods: The study was designed as a prospective-analytical-observational and was conducted at the University Clinic for Gastroenterohepatology in Skopje for a period of one year. The study population included 70 hospitalized patients with established liver cirrhosis, regardless of etiology, divided into two groups, 35 patients with SBP and 35 non-SBP. Prognostic scores in patients with liver cirrhosis and ascites: MELD score, according to the formula: MELD = [(0.957 x Ln Creatinin) + (0.378 x Ln Bilirubin) + (1.12 x Ln INR) + (0.643) x 10]. The Child-Turcotte-Pugh II score includes 6 parameters: serum albumin and bilirubin, amount of ascites, degree of encephalopathy ( HE), prothrombin time (PT) and serum creatinine, and assessment of the degree of hepatic encephalopathy according to the West Haven criteria. Results: The average value of the MELD score in patients with SBP was 22.6 ± 8.27 and in non-SBP the average value was lower - 17.83±5.87. According to the Mann-Whitney U test, the difference between the mean values was statistically significant for p <0.05 (z = 2.41; p = 0.015). A score of 30 to 39 was registered in 25.7% of patients with SBP, and only in 2.9% in non-SBP; the percentage difference was statistically significant for p <0.05 (Difference test, p = 0.0064 ). Patients with SBP had an average Child-Pugh score of 13.09 ± 2.48 or 100.0% C-class points. In patients with non-SBP, an average child-Pugh score of 9.63 ± 1.62 was recorded, or class B in 65.7% and class C in 34.3%. The percentage difference was statistically significant for p <0.05 (Difference test, p = 0.000000). According to the Mann-Whitney U test, the difference between the mean values was statistically significant for p <0.05 (z = -5.44; p = 0.00001). ROC analysis indicated that the Child-Turcotte-Pugh II score contributed to the diagnosis of SBP - 90.7% (p = 0.000) (excellent predictor), closer to the ideal value of 1.0 and above the worst value of 0.5. ROC analysis indicated that the MELD score did not contribute to the diagnosis of SBP - 66.7% (p = 0.017) (weak predictor), closer to the worst value of 0.5. Conclusion:Our research confirmed that SBP occurs in patients with severe hepatic dysfunction calculated according to the CTP II score and MELD score. Mean value of the MELD score in patients with SBP was higher then in patients with non-SBP. On the other hand all patients with SBP had an average CTP II score, C-class points,  while the largest percentage of patients with non-SBP were class B-class points. MELD score is a weak predictor of SBP.  The best predictor for predicting SBP is the CTP II score (rank C).

This article presents the results of studying the dimensional accuracy of the bases of complete removable prostheses made using a 3D printer and the traditional method. Bases of complete removable prostheses were made using an intraoral laser scanner iTero Cadent (USA) and a 3D printer Asiga Max UV (Australia). To study the dimensional accuracy of the bases of complete removable prostheses, we used the DentalCAD 2.2 Valletta software. The Nonparametric Wilcoxon W-test was used for statistical analysis of the obtained data. We found that the average value of the difference with the standard for bases made using digital technologies is 0.08744±0.0484 mm. The average value of the difference with the standard for bases made by the traditional method is 0.5654±0.1611 mm. Based on these data, we concluded that the bases of complete removable prostheses made using modern digital technologies (intraoral laser scanning and 3D printer) have a higher dimensional accuracy compared to the bases of complete removable prostheses made using the traditional method with a significance level of p<0.05 (Wilcoxon's W-test=0, p=0.031). Keywords: digital technologies in dentistry, digital impressions, intraoral scanner, 3D printing, ExoCAD, complete removable dentures.

Seasonal variations in the stable isotopic composition of snow and meltwater were investigated in a sub-arctic, mountainous, but non-glacial, catchment at Okstindan in northern Norway based on analyses of δ18O and δD. Samples were collected during four field periods (August 1998; April 1999; June 1999 and August 1999) at three sites lying on an altitudinal transect (740–970 m a.s.l.). Snowpack data display an increase in the mean values of δ18O (increasing from a mean value of −13.51 to −11.49‰ between April and August), as well as a decrease in variability through the melt period. Comparison with a regional meteoric water line indicates that the slope of the δ18O–δD line for the snowpacks decreases over the same period, dropping from 7.49 to approximately 6.2.This change points to the role of evaporation in snowpack ablation and is confirmed by the vertical profile of deuterium excess. Snowpack seepage data, although limited, also suggest reduced values of δD, as might be associated with local evaporation during meltwater generation. In general, meltwaters were depleted in δ18O relative to the source snowpack at the peak of the melt (June), but later in the year (August) the difference between the two was not statistically significant. The diurnal pattern of isotopic composition indicates that the most depleted meltwaters coincide with the peak in temperature and, hence, meltwater production.

Abstract Background The homologous recombination (HR) pathway is largely inactive in early embryos prior to the first cell division, making it difficult to achieve targeted gene knock-ins. The homology-mediated end joining (HMEJ)-based strategy has been shown to increase knock-in efficiency relative to HR, non-homologous end joining (NHEJ), and microhomology-mediated end joining (MMEJ) strategies in non-dividing cells. Results By introducing gRNA/Cas9 ribonucleoprotein complex and a HMEJ-based donor template with 1 kb homology arms flanked by the H11 safe harbor locus gRNA target site, knock-in rates of 40% of a 5.1 kb bovine sex-determining region Y (SRY)-green fluorescent protein (GFP) template were achieved in Bos taurus zygotes. Embryos that developed to the blastocyst stage were screened for GFP, and nine were transferred to recipient cows resulting in a live phenotypically normal bull calf. Genomic analyses revealed no wildtype sequence at the H11 target site, but rather a 26 bp insertion allele, and a complex 38 kb knock-in allele with seven copies of the SRY-GFP template and a single copy of the donor plasmid backbone. An additional minor 18 kb allele was detected that looks to be a derivative of the 38 kb allele resulting from the deletion of an inverted repeat of four copies of the SRY-GFP template. Conclusion The allelic heterogeneity in this biallelic knock-in calf appears to have resulted from a combination of homology directed repair, homology independent targeted insertion by blunt-end ligation, NHEJ, and rearrangement following editing of the gRNA target site in the donor template. This study illustrates the potential to produce targeted gene knock-in animals by direct cytoplasmic injection of bovine embryos with gRNA/Cas9, although further optimization is required to ensure a precise single-copy gene integration event.
