 AbstractIntroduction: Rifampicin is one of anti-tuberculosis drugs are reported to have hepatotoxic effects through increasing ROS (Reactive Oxygen Species) so thattriggering necrosis of liver cells and stimulates the formation of TNF-α. Essential oil of Annona muricata Linn. contains linalool, β-caryophyllen, trans-caryophyllen, germacrene A, germacrene B, germacrene D, sesquisterpene, β-canidine, γ-canidine. The objective of this study is to investigate the effect of essential oil of Annona muricata Linn. on levels of Tumor Necrosis Alpha(TNF-α) liver in male wistar rats which induced by rifampicin.Methods: This was an invivo experimental study with control group post test only design and use 25 male wistar rats and divided into 5 groups, they are KN (corn oil), KP (Rifampicin), P1 (Rifampicin + Essential oil 1,5%), P2 (Rifampisin + Essential oil 3%), P3 (Rifampisin + Essential oil 6%) for 6 weeks and then levels of TNF-α liver will be checked. Analysis of the data using One Way (ANOVA) test and then tested by post hoc test using Least Significant Difference (LSD). The result significant if p<0,05.Result: Induced rifampicin can increased levels of TNF-α liver than group KN (p<0,05). Treatment of essential oil of Annona muricata Linn. on P1, P2, and P3 can reduced levels of TNF-α liver than group KP (p<0,05). Allegedly this is due to the active ingredient in the essential oils act as antioxidants and antiinflammatory.Conclusion: Essential oil of Annona muricata Linn. at all doses can reduced levels of TNF-α liver in male wistar rats which induced by rifampicin.Keywords:rifampicin,essential oil of Annona muricata Linn., TNF-α liver

Backgound: Non Alcoholic Fatty Liver Disease (NAFLD) occurs when the intake and free fatty acid synthesis occurs more frequently than its oxidation and resecretion in the blood. Melatonin is a powerful antioxidant that can boost the synthesis of endogenous antioxidants in the body, suppress the inflammatory response and inhibit the formation of steatosis.Objective: To analyze the effect of melatonin supplementation in reducing body weight andTNF-α levels in male Wistar rats were fed by waste cooking oil.Methods: True experimental studyusing post-test only control group design. This study was done on 18 male wistar rats were divided into 3 groups : the positive control group (P0) was administrated waste cooking oil, the treated group 1 (P1) was administrated waste cooking oil and 5mg/kgBW melatonin, and the treated group 2 (P2) was administrated waste cooking oil and 10mg/kgBW melatonin for 28 days.Data analysis using One Way ANOVA test and followed by Tukey test to determine the most effective dose of melatonin.Results: There was significant difference in body weightbetween P2group and K0 group (p=0,019) with the mean body weight difference was 19,167g lower than K0 group. There was no difference in TNF-α levels between the three groups (p=0,155). Conclusion: Melatonin dose of 10mg/kgBWloses body weight male Wistar rats have given by waste cooking oil for 28 days, but does not reduce TNF-α levels.

<p>Liver is an organ with complex metabolism. When the liver is inflamed, cellular immunity will defend against inflammatory agents by stimulating immune cells to produce reactive oxygen species (ROS). Excessive ROS accumulation cause oxydative stress with increased  liver malondialdehyde (MDA) level. Some researches showed that purple sweet potato contain flavonoids (anthocyanins) that functioned as antioxydants. This study aimed to show the prophylactic effect of purple sweet potato extract to the liver MDA level of male Wistar rats induced by carrageenan.</p><p>This study used post-only control group method using 18 male Wistar rats divided into 3 groups: group of rats without treatment, group of rats induced by 0,1 ml of 1% carrageenan by intraplantar injection on day-8, and group of rats given with 872 mg/kgBW of purple sweet potato extract for 7 days and induced by 0,1 ml of 1% carrageenan. In the end of the study, the liver MDA levels were measured by Thio-Barbituric Acid method on each groups.</p><p>The results of One-Way ANOVA test showed there was no significant difference (p = 0,290) between group of rats without treatment (<em>x̅</em>= 207,50) and group of rats induced by carrageenan (<em>x̅</em>=233,17). Then, there is no significant difference (p = 0.978) between group of rats induced by carrageenan and group of rats given with prophylactic purple sweet potato extract and induced by carrageenan (<em>x̅</em>= 232,50).</p><p>The conclusion of this study is giving intraplantar injection of carrageenan can increase liver MDA level insignificantly and giving prophylactic purple sweet potato extract has an effect to decrease the liver MDA level of rats induced by carragenan insignificantly because it contains anthocyanins as antioxidants.</p><p> </p><strong>Keywords: </strong>Liver, <em>Ipomoea batatas</em> L., Malondialdehyde, Anthocyanins

ABSTRACT The polysaccharide fucoidin is a selectin blocker that inhibits leukocyte recruitment into the cerebrospinal fluid (CSF) during experimental pneumococcal meningitis. In the present study, the effect of fucoidin treatment on the release of the proinflammatory cytokines tumor necrosis factor alpha (TNF-α), interleukin-1 (IL-1), and IL-8 into the CSF was investigated. Rabbits (n = 7) were treated intravenously with 10 mg of fucoidin/kg of body weight every second hour starting 4 h after intracisternal inoculation of ∼106 CFU of Streptococcus pneumoniae type 3 (untreated control group, n = 7). CSF samples were obtained every second hour during a 16-h study period. Treatment with fucoidin caused a consistent and significant decrease in CSF IL-1 levels (in picograms per milliliter) between 12 and 16 h (0 versus 170, 0 versus 526, and 60 versus 1,467, respectively;P < 0.02). A less consistent decrease in CSF TNF-α levels was observed in the fucoidin-treated group, but with no significant difference between the two groups (P > 0.05). In contrast, there was no attenuation in CSF IL-8 levels. Indeed, there was a significant increase in CSF IL-8 levels (in picograms per milliliter) in the fucoidin-treated group at 10 and 12 h (921 versus 574 and 1,397 versus 569, respectively;P < 0.09). In conclusion, our results suggest that blood-derived leukocytes mainly are responsible for the release of IL-1 and to some degree TNF-α into the CSF during pneumococcal meningitis, whereas IL-8 may be produced by local cells within the brain.

Background: The process of wound healing is influenced by various factors such as age, hormones, and wound care. Wound care is done to accelerate wound healing which can be done by various methods, one of them is traditional care. Traditional wound care can use medicinal plants. Rhodomyrtus tomentosa is a medicinal plant that has an antioxidant, anti-inflammatory, antitumor and antibacterial content. Thus this study aims to evaluate the effectiveness of the antiseptic solution of the Rodhomyrtus tomentosa leaf extract on wound healing in male Wistar rats. Method: this research is pure experimental research with post test only control group design. Thirty male white rats were divided into five groups, namely negative control, positive control, Rhodomyrtus tomentosa leaf extract 15%, 30%, and 60%. Rhodomyrtus tomentosa leaf extraction was carried out by maceration method with 70% ethano solvent. The extraction results are divided into 3 concentrations (15%, 30% and 60%). The wound healing process was evaluated by measuring the length of the wound manually from 0 to 10 days in each group. Meanwhile, the number of fibroblast cells was calculated through hematoxylin eosin (HE) staining and observed using an Olympus CX41 microscope with a 10x magnification and objective lens magnification in 3 fields. Result: There was a significant difference in the reduction in wound length (p =< 0,000) between the five experimental groups (Rhodomyrtus tomentosa leaf extract solution 15%, 30% and 60%, negative control and positive control. Solution of rhodomyrtus tomentosa leaf extract accelerated the increase in the number of fibroblasts compared to the negative control group (p = 0.003), but did not make a difference (p = 0.403) with the positive control group. Rhodomyrtus tomentosa leaf extraction solution had the same microscopic effect on the number of fibroblasts with a positive control group given 0.9% NaCl solution. Conclusion: There was a significant difference in the number of fibroblasts between all groups, but no difference in wound healing length.

Introduction: Research on the use of papaya and aloevera sap on wound healing each been done, but effects the combination of the two extracts on wound healing in diabetes mellitus condition has not been done.Purpose: This study aims to determine the effect of a gel combination of papaya latex and aloevera flesh extracts on the traumatics ulcer healing process in diabetic rats.Methods: True experimental laboratory research with the post-test-only control group design that performed on 28 male wistar rats which were divided intofour groups: DM and non-DM rat that treatment with povidone iodine and gel combination of papaya latex and aloevera flesh extract. The treatment was done for 7 days with smearing that was given 2x/day. Traumatics ulcer healing seen from the number of fibroblasts that then were analyzed by One WayAnova and post hoc LSD test.Results: The results showed the number of fibroblasts in DM and non-DM rat with povidone iodine were 44.6 and 42.73; in the DM and non-DM rat with a gel combination of papaya latex and aloe vera flesh extracts were 61.10 and 77.03. One Way Anova test resulted p-value = 0.000, it was mean that there were at least a significant difference on the number of fibroblasts in the two groups. Differences of the number fibroblasts was shown in all groups, except in the group DM and non-DM rats with povidone iodine (p = 0.764).Conclusion: Conclusion of the study was the provision of a gel combination of papaya latex and aloevera flesh extracts effect on the traumatics ulcer healing process in diabetic rats.

Background: Oral mucosa ulceration which often occurs usually in the form of white-yellowish spot with concave surface, reddish edge and pain. Based on previous research, Aloe vera process anti-inflammation substance that could help quickening ulceration healing process. This research aims to know the effect of Aloe vera flesh extract on Male wistar rats oral mucosa ulceration in-vivo. Method: this research was quasi experimental research with the post-test only control group design using Male wistar rats as the testing animal. In the research, there were three treatment groups: The first groups which was given aquadest treatment, second groups with Aloe vera flesh extract, and third groups which was given chlorhexidine gluconate 0,2% treatment. The data collecting was based on histopathology observation concerning the increase of fibroblast quantity. Result: The research result based on comparison test among the three groups with One Way Anova showed that on Day 3th, the average quantity of fibroblast didn't have significant difference between the treatment group and control group positive that was p>0,05, meanwhile on Day 7th every group showed significant difference p<0,05. Conclusion: It concluded that Aloe vera flesh extract has influence on the healing of Male wistar rats oral mucosa ulceration as shown by fibroblast increasing quantity.

Abstract Background Tumor necrosis factor-alpha (TNF-α) is a pro-inflammatory cytokine which plays an important role in the pathogenesis of many diseases. Endometriosis is one of the most common gynecological diseases. The purpose of this study was to investigate the association of TNF-α-1031T/C polymorphism with the genetic susceptibility of endometriosis in a European population. Materials and methods In this case-control study, 51 endometriosis patients and 67 healthy control women participated. We used endometrial tissue from the patients and peripheral blood from the healthy women to extract DNA. Polymerase chain reaction (PCR) analysis and the restriction enzyme Bbs I were used to analyze the -1031 T/C polymorphism in the TNF-α gene promoter region. Statistical analysis was performed using Fisher’s exact test. We also calculated the odds ratios. Results In the group of patients, 66.7% of women were detected with the TT genotype, 33.3% with the TC genotype and 0% with the CC genotype while in the control group, 46.3% had the TT genotype, 47.8% had the TC genotype and 6% had the CC genotype. There was a significant association between the TT genotype with endometriosis (p = 0.03). There was no significant deviation from the Hardy-Weinberg equilibrium. Conclusions The TC and CC genotypes appeared more often in the healthy women than the endometriosis patients and this shows that the C allele might have a protective role in endometriosis in the Greek population. Further studies are needed to specify the role of this polymorphism in pathogenesis of endometriosis and the mechanisms that protect the patients from the disease.

The present study, aimed to evaluate the effects of resistance training (RT) on glycemia, insulinemia, insulin sensitivity, insulin signaling (IS), and tumor necrosis factor-α (TNF-α) levels in rats with periodontitis. 40 male Wistar rats were distributed into 4 groups: sedentary control group (SCN), exercised control group (ExCN), sedentary ligature-induced periodontal disease group (SPD), and exercised ligature-induced periodontal disease group (ExPD). 28 days after inducing periodontitis the RT started (14-week). Glycemia, insulin, TNF-α levels, and insulin sensitivity were analyzed using various methods. IS was evaluated by measuring tyrosine phosphorylated pp185 in insulin-sensitive tissues (western blot method). Higher levels of insulin, HOMA-IR, and TNF-α, and a decrease in insulin sensitivity were observed in the SPD group, which also had decreased levels of insulin-stimulated tyrosine phosphorylated pp185 in muscle and adipose tissue, when compared to the other groups. The ExPD group had increased levels of insulin-stimulated tyrosine phosphorylated pp185 compared to the SPD group, but showed no significant difference when compared to the SCN and ExCN groups. RT reversed both the insulin resistance (IR) and the IS alterations in rats with induced periodontitis, and decreased the insulin and TNF-α levels. Therefore, the results of show the importance of RT in preventing or reversing IR in rats with periodontitis.

Background: Photoaging is a premature aging that occurs on skin due to the ultraviolet light exposure that causes the emergence of clinical symptoms, one of which is wrinkle. One kind of material that can prevent photoaging is antioxidant. A single clove garlic has the highest antioxidant activity compared to the other materials. Aim: This research aimed at investigating the effectivity of single clove garlic in inhibiting the clinical symptom of photoaging. Method: This research was an experimental research, utilizing pre-post-test control group design. The used sample were 30 male wistar rats which were divided into 6 experimental groups. All groups were exposed to UV-B light with the amount of 840 mJ/cm2. Control group (P1) was only exposed to UV-B while the other groups were treated with placebo cream (P2), sunblock (P3), 5% garlic cream (P4), 10% garlic cream (P5) and 20% garlic cream (P6) respectively. The clinical symptom in the form of wrinkle was observed by using dermascope and the observations were categorized based on Glogau Scale. The statistical analysis utilized Wilcoxon and Kruskal Wallis test which was followed by Mann Whitney test. Result: The result of the research showed that there was significant difference on the apparent wrinkle on P1 group and P3, P5 and P6. Meanwhile the comparison between P1, P2 and P4 was not significant. Conclusion: The single clove garlic extract can prevent photoaging and has the similar protective effect for the skin as of sunblock.
