Trichoderma rubrum (T. rubrum) is one of the important pathogens because it is the cause of most dermatomycosis. The treatment of Trichophyton rubrum infection is time-consuming and very expensive; it is easy for the infections to reoccur, leading to therapeutic failures, persistence, and chronic infection. These issues have inspired researchers to study natural alternative therapies instead. Cnidium monnieri (L.), as a kind of traditional Chinese medicine, has a variety of pharmacological activities and a wide range of applications, so it has a high potential for researching and economic value. We detected the effect of aqueous extract of C. monnieri (L.) on the activity of T. rubrum by Cell Count Kit-8 assay (CCK-8), and we found that 128 and 256 μg/ml of aqueous extracts of C. monnieri (L.) co-cultured with T. rubrum for 24 h showed the inhibitory effect on T. rubrum. The results of scanning electron microscopy (SEM) and transmission electron microscopy (TEM) confirmed that aqueous extract of C. monnieri (L.) damaged the T. rubrum. At the same time, mass spectrometry screening with T. rubrum before and after the treatment of 256 μg/ml of aqueous extracts of C. monnieri (L.) showed that 966 differentially expressed proteins were detected, including 524 upregulated differentially expressed genes (DEGs) and 442 downregulated DEGs. The most significantly downregulated protein was chitin synthase (CHS); and the results of qRT-PCR and Western blotting demonstrated that the expression level of CHS was downregulated in the 256 μg/ml group compared with the control group. The study showed that the aqueous extract of C. monnieri (L.) could destroy the morphology of mycelia and the internal structure of T. rubrum, and it could inhibit the growth of T. rubrum. The antifungal effect of aqueous extract of C. monnieri (L.) may be related to the downregulation of the expression of CHS in T. rubrum, and CHS may be one of the potential targets of its antifungal mechanism. We concluded that aqueous extract from C. monnieri (L.) may be a potential candidate for antifungal agents.

Abstract Crude oil spills are increasingly likely to occur from drilling, pumping and transportation activities as oil development proceeds at a rapid pace. These spills may occur over the wide range of climatic conditions which obtain in Canada. Little is known of oil toxicity at different temperatures; consequently, laboratory studies were made of the variability of the toxicity of aqueous extracts of a Norman Wells crude oil to freshwater algae over the temperature range 5°C to 35°C. Two unicellular green algae were studied: Chlamydomonas eugametos and Chlorella vulgaris. Their response (measured by cell numbers) varied with temperature and species. Whereas Chlamydomonas eugametos showed a general pattern of growth inhibition by oil at all temperatures with maximum inhibition at 25°C, Chlorella vulgaris showed general growth stimulation by oil with maximum stimulation at 25°C, this temperature was chosen for all further experimentation. All experiments were done using unialgal cultures and sterile technique. Cells were grown in 50 ml of nutrient medium (BBM) in 125 ml Erlenmeyer flasks. Such flasks allow gas exchange and permit loss of volatile hydrocarbons. Aqueous extracts were made by slowly stirring 5% crude oil with the nutrient medium for six hours using a magnetic mixer. The extract was then allowed to sit for two to four hours before the lower fraction was drawn off for use. Experiments were carried out in controlled environment chambers (±2°C) with a twelve hour light-dark cycle. All further experiments used a similar methodology. (Note: Chlamydomonas eugametos experiments were carried out on a rotary shaker at 125 rpm.) An attempt was made to determine the reason for the remarkable stimulation in growth of Chlorella vulgaris #29 at 25°C. This organism has been described in the literature as heterotrophic. Thus three reasons for stimulation seemed possible: 1. heterotrophic uptake of hydrocarbons directly from solution; 2. heterotrophic uptake of organic compounds formed or released by microbial breakdown of hydrocarbons (the aqueous extract of crude was not sterile); or 3. the use of CO2 released to solution by microbial respiration. The original experiment was repeated in the dark at 20°C to determine if stimulation still occurred. It did not, since cells exposed to the aqueous extract decreased in numbers. However, after two weeks the cells were illuminated and even though experimental flasks started off with depleted populations, they outgrew the control cells within two weeks. This suggested that if stimulation was related to heterotrophism, it must, at least in this case, have been the unusual case of photoheterotrophism. The reasons for this stimulation of growth are currently under investigation. Several methods are being employed to investigate the suspected heterotrophism. Experiments will be done to determine whether light energy is essential to the stimulation. Two varieties of Chlorella vulgaris, i.e. #29 and #260 are heterotrophic and autotrophic respectively, are to be used in experiments. Sterile aqueous extracts made by pressure ultrafiltration will be used. These experiments should determine whether algal growth stimulation is related to heterotrophism or whether microbial degradation of hydrocarbons is the real source of stimulation. Although the toxicity of crude oil may be rapidly ameliorated by physical and/or biological phenomena, one must still be aware of the possibility of a large input of organic carbon causing extensive eutrophication. Thus both toxicity and eutrophication will cause a selection, in terms of survival, in a natural environment. It is evident that although an oil spill may not totally destroy an ecosystem, it will certainly alter its natural composition considerably.

The present study was undertaken to establish the diuretic activity of alcoholic and aqueous extract of dried rhizomes of Curcuma amada Roxb in rats. Alcoholic and aqueous extracts of rhizomes were administered to experimental male Wistar rats orally at doses of 250 and 500 mg/kg and compared with furosemide (10 mg/kg) as the reference standard. The parameters measured for diuretic activity were total urine volume, urine electrolyte concentration such as sodium, potassium and chloride have been evaluated. The rats treated with alcoholic and aqueous extract of Curcuma amada in a dose of 250 and 500 mg/kg showed higher urine output when compared to the respective control. Both alcoholic and aqueous extracts have showed a significant dose-dependent increase in the excretion of electrolytes when compared to the control group. The result indicates that alcoholic and aqueous extract is an effective natriuretic and kaliuretic diuretic, which supports the traditional claim about the Curcuma amada Roxb being used as diuretics.

To evaluate the effect of Aqueous extract of Terminalia belliricafruit pulp (AETB) on learning by Hebb William maze model in mice with acute alcohol consumption.Swiss albino mice (n=48) of either sex weighing 20-30g will be divided into eight groups of six mice each. Drugs were given orally after 12 hours of fasting. Group I mice received 10ml/kg of Normal Saline, Group II mice received Piracetam 200mg/kg, Group III received AETB 36mg/kg, Group IV received ethanol 1.5g/kg orally, Group V received ethanol(1.5g/kg )+ piracetam (200mg/kg), Group VI mice received ethanol(1.5g/kg) +AETB(9mg/kg), Group VII mice received ethanol(1.5g/kg) +AETB (18mg/kg), Group VIII mice received ethanol(1.5g/kg) +AETB(36mg/kg). Time taken by the animal to reach the reward chamber from the start chamber (TRC) in Hebb-William maze was used as a parameterto evaluate the learning.Acute alcohol administration showed increase in TRC. Whereas, acute administration of Aqueous extracts of Terminalia belliricafruit pulp showed a decrease in TRC when compared to the control group. The TRC values for the groups that were administered AETB along with acute alcohol administration showed decrease in TRC values compared to the negative control.Current study showed acute alcohol administration caused impairment of thelearning ability in mice. Whereas, acute administration of Aqueous extracts of Terminalia belliricafruit pulp (AETB)caused enhancement of learning. Pre-treatment with AETB before acute alcohol administration indicated protective action of AETB on alcohol affected learning in mice.

Abstract Background: In traditional medicine, the floral diversity permits the inhabitants of North Cameroon to use a great number of plants to fight against Malaria. The aim of this study was to identify plants used in traditional medicine to treat malaria, and to verify the scientific basis for the use of one of these plants in the locality of Gamba.Methods: An Ethnopharmacological survey was carried out on 15 traditional healers. We collected data on use of medicinal plants using questionnaires. Then in-vivo antimalarial activity of the decoctioned and macerated aqueous extracts of khaya senegalensis trunk bark was evaluated. The 4-day suppressive peters test was realised on mus musculus swiss albino mice. On day one, mice were infected with 107 plasmodium berghei parasitized red blood cells through intra-peritoneal inoculation. 2 hours after infestation, mice in batches of 6 were treated orally at a dose of 75, 150.300 mg/Kg for macerated aqueous extract and 65, 120.260 mg/Kg for decoctioned extract daily during 3 days at an administration volume of 10 ml/Kg. An extract was considered (% reduction): Highly active (between 100-90 %); moderate (between 90-50 %); weak (between 50-10 %); Inactive (between 0 %). P-values <0.05 were considered statistically significant.Results: A total of 18 plant species belonging to 12 families were identified for the preparation of 12 recipes. The decocted aqueous extract of khaya senegalensis showed moderate anti-plasmodial activity (% reduction = 52.46%) at the highest dose of 260 mg/kg with p<0.001 compared to the positive control group. The aqueous macerate at doses of 150 and 300mg/kg gave respectively a percentage reduction of parasitaemia of 59.42% and 71.80% and also showed moderate anti-plasmodial activity; with p<0.001 between the different extracts and the positive control (99.18%).Conclusion: In conclusion, extracts of khaya senegalensis showed moderate anti-plasmodial activity. It would therefore be necessary to evaluate the anti-malarial activity in-vivo and the toxicity of the aqueous extracts macerated using other solvents and also test the other plants listed.

Trichophyton rubrum is one of the most common types of dermatophyte, causing superficial skin mycosis in human populations. Although laser treatment of onychomycosis has been proven to be effective in the clinic, the underlying mechanism of the effect of the laser on fungal growth is not clear. The objective of the present study was to observe the ultrastructural changes of Trichophyton rubrum following laser irradiation and compare the transcriptome differences between the laser irradiation group and control group. In the present study, scanning electron microscopy and transmission electron microscopy were used to observe the ultrastructural changes following the laser irradiation of Trichophyton rubrum. We also performed RNA-seq to investigate the effects of laser irradiation on Trichophyton rubrum by comparing the transcriptome pattern with the control. Morphological observation with electron microscopy indicated that laser irradiation resulted in the destruction of the cell membrane system. A significant induction of apoptosis was noted compared with the control group, which was confirmed by the formation of the myeloid body and protein aggregates in the cytoplasm. RNA-seq demonstrated that the expression levels of Acyl-CoA N-acyltransferase and S-adenosyl-L-methionine-dependent methyltransferase were increased in the laser irradiation group. This result indicated that laser irradiation triggered the initiation of the damage repair pathway. In conclusion, the present study suggested that laser irradiation caused physiological injury and therefore inhibited the growth of Trichophyton rubrum.

The anti-diabetic effect of ethanolic and aqueous extracts of Imperata cylindrical  rhizomes was investigated in alloxan-induced diabeties in rats. Diabetes was induced by a single 150 mg/kg intraperitoneal dose of alloxan. Rats were divided into five groups with six rats in each group i.e. the normal control group, diabetic control group, standard group (glibenclamide, 10mg/kg, p.o.), Test-I group (200 mg/kg ethanolic extract) and Test-II group (200 mg/kg aqueous extract). The above concerned groups were inoculated on 21st day. On the last day of the experiment, fasted rats were killed by cervical dislocation. The body weight was measured at the initial day and final day. The blood samples were collected for estimation of glucose. The loss of body weight in control group, but recovery was observed in drug treated group. The serum glucose level was significant increased in diabetic rats. However, significant improvement was observed in treated group. The biochemical parameters such as HDL and proteins level were decreased in the control group but maintained in drug treated group. LDL, cholesterol, triglyceride creatinine and urea were significant increase in control group however, reduced level in drug treated group. The present study concluded that ethanolic and aqueous extracts of I. cylindrical  rhizome showed an appreciable effect in reducing the hyperglycemia and the complications associated with diabetes. However, aqueous extract is found more significant in decreasing blood glucose level in comparison to the ethanolic extract. The study results justify the traditional use of the plant as anti-diabetic.

Pasteurella multocida is a highly versatile pathogen that infects a wide range of animals, including goats, causing pneumonia and hemorrhagic septicemia. Circular RNA (circRNA) is a type of non-coding RNA that plays an important role in regulating cellular metabolism. However, whether and how circRNA is involved in regulating immune responses in the goat lung has not been reported. Thus, this study was designed to examine the function of circRNA in goats infected with Pasteurella multocida. Goats were assigned into one of two groups: an uninfected control group (CK) and an infected group challenged with P. multocida. Compared with the CK group, which remained healthy, the infected goats showed clinical signs of infection, including depression, cough, nasal discharge, and dyspnea, along with elevated body temperature and lesions in the lung. Whole-transcriptome sequencing and small RNA sequencing were then performed using lung samples from goats from each group. A total of 138 circRNA, 56 microRNAs (miRNA), and 2,673 messenger RNA (mRNA) molecules were significantly differentially expressed in the P. multocida-infected group compared with the CK group. Randomly selected differentially expressed circRNA, miRNA, and mRNA molecules (n = 5 per group) were then validated by quantitative reverse-transcriptase polymerase chain reaction analysis. Gene ontology (GO) analysis of the source genes indicated that six immune-related terms were enriched among the differentially expressed cirRNA molecules, including inflammatory response, immune effector process, cell activation involved in immune response, cytokine-mediated signaling pathway, response to endogenous stimulus, and immune response. The corresponding circRNA molecules were then selected for construction of a competitive endogenous RNA network to identify networks that may be involved in the immune response to P. multocida infection. The results indicated that P. multocida HN01 may cause pneumonia and stimulate an immune response in goats via regulation of circRNA expression. This study presents the first comprehensive circRNA profile in response to P. multocida infection in goats, thus, providing a basis for understanding the function of circRNA in the host immune response to P. multocida infection.

Kava roots have been extensively studied in clinical trials as potential candidate anti-anxiety drugs. However, anti-convulsive properties of various tissues of stems of Kava have not been reported to date. The objective of the study was to evaluate the anti-convulsive potential of aqueous extracts prepared from specific tissues of Kava (Piper methysticum) stems in zebrafish, using the PTZ-induced seizure model. The potency of each extract was compared in terms of the intensity of seizure scores and onset time after pre-treating the zebrafish before the PTZ challenge. The results indicate that aqueous extract of Kava stems without peel after 45 min of pre-treatment exhibited anti-convulsive potential at the dose of 50 mg/L. This study provides evidence to the anti-convulsive properties of peeled Kava stems and its potential for investigation and design of candidate anti-convulsive drugs.

Hibiscus sabdariffaL. is a plant of the Malvaceae family, commonly known as roselle.H. sabdariffais known to contain antioxidant, cholesterol-lowering, antiobesity, insulin resistance reduction, antihypertensive, and skin cancer chemopreventive properties. This study evaluated the effects ofH. sabdariffaaqueous extract against cyclophosphamide (CPA, 25 mg/Kg) induced damage to DNA in male Wistar rats by micronucleus test. Samples ofH. sabdariffacalyx were obtained in the municipality of Barra do Garças, Mato Grosso, Brazil. The aqueous extract was prepared by infusion and each animal received a daily dose of 400 mg/Kg by gavage for 15 consecutive days of treatment. The presence of anthocyanins was confirmed by ferric chloride test and phenolic compounds using high-performance liquid chromatography, with emphasis on the identification of rutin. The animals were sacrificed by deepening of anaesthesia to obtain bone marrow and determination of the frequency of micronucleated polychromatic erythrocytes. The group treated with the aqueous extract ofH. sabdariffarevealed a 91% reduction in micronucleus frequency when compared with the positive control group. Under the conditions tested,H. sabdariffaL. presented a protective effect to CPA-induced damage to DNA of the treated animals, and it is a potential candidate as a chemopreventive agent against carcinogenesis.

Cystic echinococcosis (CE) is a cosmopolitan zoonosis caused by the larval stage of Echinococcus granulosus, which affects humans and a wide range of mammalian intermediate hosts. Parasite tetraspanin proteins are crucial for host-parasite interactions, and therefore they may be useful for vaccine development or disease diagnosis. In the present study, the major antigen coding sequence of tetraspanin 11 (Eg-TSP11) from E. granulosus was determined. The results of immunolocalization showed that Eg-TSP11 was mainly located in the tegument of adult worms and protoscoleces. Western blotting analysis showed that the serum from dogs injected with recombinant Eg-TSP11 (rEg-TSP11) could recognize Eg-TSP11 among natural protoscolex proteins. Moreover, the serum from dogs with E. granulosus infection also recognized rEg-TSP11. Serum indirect enzyme-linked immunosorbent assays demonstrated that IgG levels gradually increased after the first immunization with rEg-TSP11 compared with those in the control group. Furthermore, the serum levels of interleukin 4, interleukin 5, and interferon gamma were significantly altered in the rEg-TSP11 group. Importantly, we found that vaccination with rEg-TSP11 significantly decreased worm burden and inhibited segment development in a dog model of E. granulosus infection. Based on these findings, we speculated that rEg-TSP11 might be a potential candidate vaccine antigen against E. granulosus infection in dogs.
