Mineralization of hydrogel biomaterials with calcium phosphate (CaP) is considered advantageous for bone regeneration. Mineralization can be both induced by the enzyme alkaline phosphatase (ALP) and promoted by calcium-binding biomolecules, such as plant-derived polyphenols. In this study, ALP-loaded gellan gum (GG) hydrogels were enriched with gallotannins, a subclass of polyphenols. Five preparations were compared, namely three tannic acids of differing molecular weight (MW), pentagalloyl glucose (PGG), and a gallotannin-rich extract from mango kernel (Mangifera indica L.). Certain gallotannin preparations promoted mineralization to a greater degree than others. The various gallotannin preparations bound differently to ALP and influenced the size of aggregates of ALP, which may be related to ability to promote mineralization. Human osteoblast-like Saos-2 cells grew in eluate from mineralized hydrogels. Gallotannin incorporation impeded cell growth on hydrogels and did not impart antibacterial activity. In conclusion, gallotannin incorporation aided mineralization but reduced cytocompatibility.

We examine the antibacterial activity of Mangifera indica.L against the drug resistant bacterial pathogens. The Indian traditional mango fruit exposed promise in the prevention of diarrhea. However, there is less information existing regarding activity against drug resistance bacteria causing diarrhea. In this study, we performed isolation and identification of bacterial pathogens from diarrheal stool samples and analyzed their antibiotic resistance pattern with three generation antibiotics. Antibiotic activity of mango kernel exhibited remarkable activity from 15.2±0.5mm to 19.0±0.3mm zone of inhibition. MIC of kernel extract was noted as 10 μg /ml. Current study provide the potent antibacterial activity of mango kernel against the drug resistant bacteria. In conclusion, the traditional Indian fruit become the natural remedy for the treatment of diarrhea.

A high molecular weight fraction of a soluble Marsh muscle-relaxing preparation has been shown to contain a calcium-complexing substance. By examining the nature of the competition between this fraction and chelex-100 for calcium at various total calcium concentrations it has been possible to calculate the concentration and calcium stability constant of this calcium-complexing substance. Taking into account dilutions which occur during the preparation of fractions containing this substance its concentration may be estimated at about 2·10-4 in muscle and its calcium stability constant was found to be about 1.5·105 M-1. Preliminary evidence suggests that the calcium-binding substance is a protein.

The serum of patients with obstructive liver disease may contain a high molecular weight form of alkaline phosphatase (high Mr alkaline phosphatase). The presence of this form of alkaline phosphatase is associated with hepatic malignancies. We have investigated the use of anti-alkaline phosphatase monoclonal antibodies which do not bind high Mr alkaline phosphatase in assays for high Mr alkaline phosphatase. Direct immunoprecipitation of liver and bone alkaline phosphatase with solid phase anti-liver alkaline phosphatase antibody (which also reacts with bone alkaline phosphatase) and measurement of the residual supernatant alkaline phosphatase activity led to a precise assay. Intestinal alkaline phosphatase interfered in this assay which, consequently, was of little use in the differential diagnosis of liver disease. Indirect precipitation of liver, bone, placental and intestinal alkaline phosphatase by soluble anti-liver alkaline phosphatase (which reacts with liver and bone alkaline phosphatases), soluble anti-intestinal alkaline phosphatase (which reacts with placental and intestinal alkaline phosphatases) and solid phase anti-mouse IgG led to an assay which, although less precise, showed more promise of being useful clinically.

The proteins of the contractile spasmoneme of Zoothamnium have been examined for comparison with other motile systems. Though capable of calcium-induced contraction, glycerinated preparations of the spasmoneme contain neither actin nor tubulin at levels that can be detected in polyacrylamide gels. Sixty per cent of the protein in sodium dodecyl sulphate gels migrates in a band at a molecular weight of approximately 20,000, consisting largely of 2 similar protein species which are here given the name of spasmins. The amino acid composition of 2 spasmin fractions has been determined by a fluorimetric method. They are rich in Asx, Glx and serine, but have few aromatic amino acids and no cystine or methionine. In calcium-buffered polyacrylamide gels, it was observed that a reduction in the electrophoretic mobility of the spasmins was induced specifically by calcium (but not magnesium) at the same low concentrations as induce contraction. This indicates that the spasmins are calcium-binding proteins which may be involved directly in the calcium-induced contraction of the spasmoneme.

Extracellular alkaline phosphatase produced by a marine Pseudomonas was purified to electrophoretic homogeneity. The molecular weight of the enzyme was estimated to be 100 000. The enzyme had maximal activity at pH 11.5. The enzyme was completely inhibited by 1 mM EDTA. However, divalent cations reversed the enzyme inhibition and their order of effectiveness on the reaction was Zn2+ > Ca2+ > Mn2+ > Mg2+ > Sr2+ > Co2+. The enzyme activity was affected by the species of anion whose order of effectiveness was demonstrated to follow the lyotrophic series, Cl− > Br− > NO3−> ClO4− > SCN−. The activity of phosphatase was accelerated linearly by increased pressure until up to 1000 atm (1 atm = 101.325 kPa), and the enzyme activity at 1000 atm was 3.2 times higher than that at 1 atm.

<p>The study was aimed to look at the effect of different forms of mango kernels (MK) on the shelf life of refrigerated beef sausages over 12 days of cold storage. The (MK) was chemically and microbiologically analyzed. Beef sausages were treated with MK in 3 states, as dry ground (1.5%), an extract (1.5%) and spray MK extract (1.5%) over minced beef of sausages. Two controls were used; BHT 0.02% and no additives. A series of analyses were performed after treatments; thiobarbituric acid reactive substances (TBARS), analysis of color, myoglobin and odor. The results indicated that different forms of MK added to the beef sausages had different effects on its shelf life. Furthermore, the sprayed MK extract has significantly (P ?0.05) lowered metmyoglobin (MMb) and TBARS and increased oxymyoglobin (MbO<sub>2</sub>), odor score and a* (redness) than other forms. The potential effects of the sprayed MK may be due to a cloud of droplets cover the large surfaces of minced beef sausages with efficient extracted antioxidants. MK is source of flavonoids 142mg/g F.W. GAE. The spraying of MK at 1.5% showed an improvement of <em>E. coli</em> from minced beef and beef sausages that were less than 10 cfu g<strong><sup>-1</sup></strong>. Also the concentrations of yeasts and moulds were not detected at day 12 of storage. Hierarchically, sprayed MK extract gave best results than ground MK or MK extract form which shows effective inhibitor of lipid oxidation and microbial growth of beef sausages.</p>

Kasturi (Mangifera casturi) is a typical plant of South Kalimantan which has only been used for consumption because of its distinctive taste and aroma. Mangifera indica, which is in the same genus as Mangifera casturi, shows antioxidant, antibacterial, antifungal, and antidiarrheal activity, so it is suspected that this plant has the same activity. Crude methanol extract from the meat of Casturi mango was fractionated using n-hexane, ethyl acetate, and methanol-water. Testing for antibacterial activity and minimum inhibitory concentration (MIC) were carried out using the disk diffusion method (Kirby-Bauer). The positive control used was Chloramphenicol because it has an antibacterial range that is rather broad and the negative control used was 10% DMSO. The fraction that had the greatest antibacterial activity was the ethyl acetate fraction; with the formation of a clear zone of 7.98mm against Pseudomonas aeruginosa and 7.52mm against Bacillus cereus, with a minimum inhibitory concentration of 2mg/mL. The next test was a cell leakage analysis which revealed an increase in absorbance at MIC 1 and MIC 2. The Gas Chromatography – Mass Spectrometry (GC-MS) results from the ethyl acetate fraction revealed the presence of phenolic, flavonoid, and glycoide content, with the highest compound concentration of 5-Hydroxymethylfurfural. Mangifera indica yang masih satu genus dengan Mangifera casturi menunjukkan aktivitas antioksidan, antibakteri, antijamur, dan antidiare sehingga diduga tanaman ini memiliki aktivitas yang sama. Ekstrak kasar metanol daging buah mangga kasturi difraksinasi dengan menggunakan n-heksan, etil asetat, dan metanol-air. Pengujian aktivitas antibakteri dan kadar hambat minimum dilakukan dengan menggunakan metode difusi cakram (Kirby- Bauer). Kontrol positif yang digunakan adalah Chloramfenikol karena memiliki spektrum antibakteri yang cukup luas dan kontrol negatif yang digunakan adalah DMSO 10%. Fraksi yang memiliki aktivitas antibakteri terbesar pada penelitian ini adalah fraksi etil asetat dengan terbentuknya zona bening sebesar 7,98mm terhadap Pseudomonas aeruginosa dan 7,52mm terhadap Bacillus cereus dengan kadar hambat minimum 2mg/mL. Uji selanjutnya adalah uji kebocoran sel yang menyatakan adanya peningkatan absorbansi pada KHM 1 dan KHM 2. Hasil pemeriksaan Gas Chromatography – Mass Spectrometry (GCMS) dari fraksi etil asetat menyatakan adanya kandungan fenolik, flavonoid, dan glikosida dengan senyawa tertinggi yaitu 5-Hydroxymethylfurfural.
