This current study investigates the facilitative effects and mechanisms of decursin, a major component of Angelica gigas Nakai (AGN), and AGN root extract on hair growth in mice. We perform high-performance liquid chromatography on AGN extract to show it contains 7.3% decursin. Hairs in mouse dorsal skin are shaved distilled in water, 0.15% decursin, and 2% AGN root extract (0.15% decursin in the diluted extract) and topically applied twice a day for 17 days. Hematoxylin and eosin staining are done to examine the morphological changes in the hair follicles. To compare the effects of decursin and AGN extract on inflammatory cytokines in the dorsal skin, Western blot analysis and immunohistochemistry for tumor necrosis factor α (TNF-α) and interleukin (IL)-1β as pro-inflammatory cytokines, and IL-4 and IL-13 as anti-inflammatory cytokines are conducted. The results show that the application of decursin and AGN extract confer effects on hair growth. Hair growth is significantly facilitated from seven days after the treatments compared to that in the control group, and completely grown hair was found 17 days after the treatments. The protein levels and immunoreactivity of TNF-α and IL-1β in this case are significantly decreased, whereas the IL-4 and IL-13 levels and immunoreactivity are significantly increased compared to those in the control group. Additionally, high-mobility group box 1, an inflammatory mediator, is elevated by the topical application of decursin and AGN extract. Taken together, the treatment of mouse dorsal skin with AGE root extract containing decursin promotes hair growth by regulating pro- and/or anti-inflammatory cytokines. We, therefore, suggest that AGN root extract as well as decursin can be utilized as materials for developing hair growth-facilitating treatments.

The advantage in response of Th2 over Th1 is observed in normal pregnancy in peripheral blood. A disturbance of this balance can lead to symptoms of miscarriage and pregnancy loss. The aim of this study was to evaluate the pro- and anti-inflammatory cytokines in sera of women who were diagnosed with missed miscarriage in the first trimester and to compare this systemic immune response to the response in women with normal pregnancy. The study group consisted of 61 patients diagnosed with missed miscarriage. In total, 19 healthy women with uncomplicated first trimester created the control group. Cytokines were determined in the maternal serum by ELISA. The analysis included INF-γ, TNF-α, Il-1β, Il-4, Il-5, Il-6, Il-9, Il-10, Il-13 and TGF-β1. Th1 cytokine levels in the study group reached slightly higher values for INF-γ, Il-1β and slightly lower for IL-6 and TNF-α. In turn, Th2 cytokine levels in the study group were slightly higher (Il-9, Il-13), significantly higher (Il4, p = 0.015; Il-5, p = 0.0003) or showed no differences with the control group (Il-10). Slightly lower concentration involved only TGF-β1. Analysis of the correlation between levels of pro- and anti-inflammatory cytokines resulted in some discrepancies, without showing predominance of a specific immune response. The results did not confirm that women with missed miscarriage had an advantage in any type of immune response in comparison to women with normal pregnancy.

Concentrations of eight different cytokines and the level of expression of CD86 and CD163 macrophages were studied in peritoneal fluid in women with endometriosis. It was found that the concentration of both inflammatory (IL-6, IL-8, TNF-α) and anti-inflammatory cytokines (IL-4) as well as the level of macrophage expression of the proinflammatory marker CD86 and anti-inflammatory marker CD163 increased in women with mild external genital endometriosis (1-2 stage), and did not differ from the control group in women with severe endometriosis (3-4 stage). The content of IL-2, IL-10, CM-CSF and IFN-γ in the peritoneal fluid of women with endometriosis did not differ significantly from the control group. The results of the study indicate that the development of external genital endometriosis may be based on insufficient both inflammatory and anti-inflammatory activity of macrophages in the peritoneal fluid.

The paper analyzes the current literature data and the results of our own researches concerning the state of the cytokine network: pro- and anti-inflammatory cytokines (interleukin (IL) 1α, IL-1β, IL-4, IL-6, IL-10, IL-17 and tumor necrosis factor (TNF) α), α- and β-chemokines, including IL-8 and IL-16, as well as adipokines (leptin and adiponectin) in the peripheral blood of patients with type 2 diabetes (T2D) with normal and increased body weight/obesity. It has been shown that patients with T2D are cha­racterized by an increased content of proinflammatory cytokines (IL-1, IL-6, IL-17, TNFα), α- and β-chemokines in the peripheral blood, including IL-8 and IL-16, as well as leptin with a decrease in adiponectin content. In lean patients (with body mass index (BMI) < 25.5 kg/m2) compared to lean normoglycemic individuals from the control group (BMI < 25.5 kg/m2), there is a small but significant increase in IL-1β, IL-6, IL-17, TNFα and leptin, which, as BMI increases, significantly increases in severe obesity (BMI > 30.0 kg/m2), especially in obese women (BMI > 35.0 kg/m2). Similarly, an increase in proinflammatory cytokines is observed in normoglycemic people, but not as signifi­cant as in T2D. Less clear data were obtained when during determination of the anti-inflammatory cytokines IL-4 and IL-10, which is explained by a significant polymorphism of their genes, and both protective and compensatory effects on pro-inflammatory cytokine rise. In T2D patients, especially those with obesity, there is an increase in the leptin level and a decrease in the adiponectin content. The severity of the course and the percentage of mortality are closely associated with the BMI of patients. The effectiveness of the fight against an increase in the incidence of T2D should be primarily aimed at preventing obesity, and in case of already developed T2D — at reducing concomitant obesity. The analysis of the data presented also suggests that a sharp increase in the content of pro-inflammatory cytokines (so called cytokine storm) observed in patients with T2D and obesity infected with COVID-19, is a consequence of the summation and potentiation of already existing inflammatory process.

During pregnancy, a unique new equilibrium state appears between the systems of the specific and nonspecific mothers immunity. Besides, the cytokine cascade is launched, which includes proinflammatory and anti-inflammatory factors of influence. The balance between these two groups of mediators determines the nature of the course and outcome of the gestation process. The objective: to determine the role of mediators of pro-inflammatory and anti-inflammatory reactions of gestation intercourse in patients with miscarriage. Materials and methods. The main group (the first group) was made up of 153 pregnant women with miscarriage. The control group (the second group) consisted of 25 relatively healthy women with a physiological course of pregnancy and a complcated obstetric and gynecological anamnesis, with one and more physiological births in anamnesis. The concentration of cytokines IL-1 β, IL-6, IL-8, IL-10, TNF- α in the blood and their content in cervical mucus by solid-phase immune-enzyme analysis was determined. Results. Consequences of previous pregnancies having a background of inflammatory complications of genital and extragenital genesis create conditions for long-term persistence of latent infection, including in the uterine cavity and cervical canal, followed by infection of the fetus, and contribute to the development of immune imbalance during gestation, which leads to a cascade of homeostasis disorders with the development of complications of the pregnancy intercourse and perinatal pathology. Thus, the presence of clinical symptoms of the threat of premature abortion occurs in the context of an increase in the concentration of proinflammatory cytokines (IL-6, IL-8, TNF- α and IL-1 β) in serum.Reducing the concentration of IL-10 in non-pregnant women, relative to such in control group, throughout the entire pregnancy in the blood and its content in cervical mucus indicates a violation of the balance of pro– and anti-inflammatory cytokines in the direction of pro-inflammatory reactions and violation of the local immune response. Conclusions. In women with a loss in the first trimester there is a pro-inflammatory activity of the immune response, which is an important pathogenetic factor in the development of abortion in different gestational periods. Целью настоящего исследования было изучение нейроиммунологических особенностей развития алкогольной зависимости, сопровождающейся депрессивными расстройствами, у женщин. В связи с этим был проведён сравнительный анализ содержания в сыворотке крови провоспалительных и противовоспалительных цитокинов и кортизола, как показателя уровня стресса, у женщин с алкогольной зависимостью, с депрессивными расстройствами, а также при их сочетании. Методы. Клиническое обследование, формирование групп для исследования и создание банка биологического материала выполнены на базе отделений аффективных расстройств и аддиктивных состояний и лаборатории клинической психонейроиммунологии и нейробиологии НИИ психического здоровья ФГБНУ «Томского НИМЦ РАН». Клиническая верификация диагноза выполнена психиатрами и наркологами в соответствии с Международной классификацией болезней 10-го пересмотра (МКБ-10). Были исследованы сыворотки крови 58 женщин в возрасте 40-55 лет с установленным диагнозом по МКБ-10: депрессивный эпизод - 16, дистимия - 9, рекуррентное депрессивное расстройство - 10, алкогольная зависимость - 15, зависимость от алкоголя в сочетании с депрессией - 8. Контрольную группу условно здоровых людей, сопоставимую по полу и возрасту, составили 31 женщина, проходивших ежегодную диспансеризацию. Определяли содержание ИЛ-1b, ИЛ-6, ФНО-α, ИЛ-10 и кортизола в сыворотках крови иммуноферментным методом с использованием тест-систем производства «ООО Цитокин» (Санкт-Петербург) и фирмы DBC Diagnostics Biochem Canada Inc, соответственно. Результаты. У женщин с зависимостью от алкоголя наблюдали значимое увеличение провоспалительных (ИЛ-6 и ФНО-α) цитокинов по сравнению с контролем в 2,3 и 1,6 раза, соответственно. Сочетание алкогольной зависимости с депрессией сопровождалось более существенным увеличением содержания ИЛ-1β и ИЛ-6 при сравнении как с контролем, так и с группой женщин с зависимостью от алкоголя без соматической патологии. В сыворотках крови женщин всех обследованных групп по сравнению с контрольной группой условно здоровых женщин было выявлено повышенное содержание кортизола, что свидетельствует о нарушении функции гипоталамо-гипофизарно-надпочечниковой системы при алкогольной зависимости, депрессивных расстройствах, а также о наличии стрессогенных факторов (отмена алкоголя, госпитализация). Заключение. Сочетание алкогольной зависимости и депрессивных расстройств является взаимным отягчающим обстоятельством и неблагоприятным признаком развития как алкогольной зависимости, так и депрессии. Высокое содержание ИЛ-6 может рассматриваться в качестве возможного нейроиммунного биомаркёра сочетанного течения алкогольной зависимости и депрессии, а также может быть использовано в персонализированной профилактике и лечении этой патологии. The aim of this study was to determine neuroimmunological characteristics of the development of alcohol dependence accompanied by depressive disorders in women. For this purpose, serum concentrations of proinflammatory and anti-inflammatory cytokines and cortisol as an indicator of stress intensity were compared in women with alcohol dependence, depressive disorders, and their combination. Methods. Patients underwent a clinical examination, study groups were formed, and a bank of biological material was created at the Department of Affective Disorders, the Department of Addictive States, and the Laboratory of Clinical Psychoneuroimmunology and Neurobiology of the Research Institute of Mental Health of the Tomsk Scientific Research Center. The diagnosis was clinically verified by psychiatrists and narcologists in accordance with the International Classification of Diseases, 10th Revision (ICD-10). Blood serum tests were performed for 58 women aged 40-55 years with the following ICD-10 diagnoses: a depressive episode (n=16), dysthymia (n=9), recurrent depressive disorder (n=10), alcohol dependence (n=15), and alcohol dependence in combination with depression (n=8). The control group of sex- and age-matched without apparent disease healthy people consisted of 31 women who were undergoing a routine annual medical examination. Concentrations of IL-1b, IL-6, TNF-α, IL-10, and cortisol were measured by the enzyme immunoassay with test systems manufactured by Cytokine LLC (St. Petersburg) and DBC Diagnostics Biochem Canada Inc, respectively. Results. In women with alcohol dependence, proinflammatory cytokines (IL-6 and TNF-α) were increased 2.3 and 1.6 times, respectively, compared to the control. The combination of alcohol dependence with depression was associated with a more pronounced increase in IL-1β and IL-6 compared to the control and the group of women with alcohol dependence without somatic pathology. In all examined groups, serum cortisol was higher than in the control group of women without apparent disease, which indicated dysfunction of the hypothalamic-pituitary-adrenal axis in alcohol dependence, depressive disorders, and in presence of stress factors (alcohol withdrawal, hospitalization). Conclusion. Alcohol dependence and depressive disorders are mutually aggravating conditions, and their combination is an unfavorable prognostic sign for both of them. A high concentration of IL-6 can be considered as a possible neuroimmune biomarker for the combination of alcohol dependence and depression and can also be used in personalized prevention and treatment of this pathology.

Our present study focuses on the evaluation of the analgesic, anti-inflammatory and healing activity of Withania frutescens L. The anti-inflammatory result has an inhibition percentage of 78.87% ± 7.08 at 450 mg/kg and 75.14% ± 6.39 at 400 mg/kg and 89.75% ± 3.44 for diclofenac (1%). When applied locally, the 10% cream has an inflammation inhibition of 96.87% ± 5.85 and 76.14% ± 7.88 for the 5% cream with 89.87 ± 6.20 of reference (Indomethacin). The abdominal contractions of rats treated with the root extract are significantly lower than those of the control group that received only physiological NaCl solution, with 41.20 ± 2.30 for the extract and 82.20 ± 5.04 for NaCl and 53.40 ± 4.94 for the reference. The healing activity of the studied extract records a percentage of contraction of about 93.20% ± 3.36 (Extract 10%), 84.50% ± 3.84 (Extract 5%), 48.47% ± 2.15 (control) and 81.88 ± 2.24 for the reference.

Introduction: Obesity is a disorder with low-grade chronic inflammation that plays a key role in the hepatic inflammation and steatosis. Moreover, there are studies to support the role of exosomes in the cellular communications, the regulation of metabolic homeostasis and immunomodulatory activity. Accordingly, we aimed to evaluate the influence of plasma circulating exosomes derived from females with normal-weight and obesity on the secretion of inflammatory cytokines in human liver cells. Methods: Plasma circulating exosomes were isolated from four normal (N-Exo) and four obese (O-Exo) women. The exosomes were characterized and approved for CD63 expression (common exosomal protein marker) and morphology/size using the western blot and TEM methods, respectively. The exosomes were used for stimulation of HepG2 cells in vitro. After 24 h incubation, the protein levels of TNF-α,IL-6, and IL-1β were measured in the culture supernatant of HepG2 cells using the ELISA kit. Results: The protein levels of IL-6 and TNF-α in the cells treated with O-Exo and N-Exo reduced significantly in comparison with control group (P=0.039 and P<0.001 respectively), while significance differences were not found between normal and obese groups (P=0.808, and P=0.978 respectively). However, no significant differences were found between three groups in term of IL-1β levels (P=0.069). Based on the correlation analysis, the protein levels of IL-6 were positively correlated with TNF-α (r 0.978, P<0.001). Conclusion: These findings suggest that plasma circulating exosomes have probably anti-inflammatory properties independently from body mass index and may decrease the secretion of inflammatory cytokines in liver. However, further investigations in vitro and in vivo are needed to address the anti-inflammatory function of N-Exo and O-Exo in human liver cells and/or other cells.

Abstract Background Muscle stem cells (MuSCs) are absolutely required for the formation, repair, and regeneration of skeletal muscle tissue. Increasing evidence demonstrated that tissue stem cells, especially mesenchymal stem cells (MSCs), can exert therapeutic effects on various degenerative and inflammatory disorders based on their immunoregulatory properties. Human mesenchymal stem cells (hMSCs) treated with interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α) were reported to possess anti-inflammatory functions by producing TNF-stimulated gene 6 (TSG-6). However, whether human muscle stem cells (hMuSCs) also possess TSG-6 mediated anti-inflammatory functions has not been explored. Methods The ulcerative colitis mouse model was established by subjecting mice to dextran sulfate sodium (DSS) in drinking water for 7 days. hMuSCs were pretreated with IFN-γ and TNF-α for 48 h and were then transplanted intravenously at day 2 of DSS administration. Body weights were monitored daily. Indoleamine 2,3-dioxygenase (IDO) and TSG-6 in hMuSCs were knocked down with short hairpin RNA (shRNA) and small interfering RNA (siRNA), respectively. Colon tissues were collected for length measurement and histopathological examination. The serum level of IL-6 in mice was measured by enzyme-linked immunosorbent assay (ELISA). Real-time PCR and Western blot analysis were performed to evaluate gene expression. Results hMuSCs treated with inflammatory factors significantly ameliorated inflammatory bowel disease (IBD) symptoms. IDO and TSG-6 were greatly upregulated and required for the beneficial effects of hMuSCs on IBD. Mechanistically, the tryptophan metabolites, kynurenine (KYN) or kynurenic acid (KYNA) produced by IDO, augmented the expression of TSG-6 through activating their common receptor aryl hydrocarbon receptor (AHR). Conclusion Inflammatory cytokines-treated hMuSCs can alleviate DSS-induced colitis through IDO-mediated TSG-6 production.

Currently the TRPV1 (transient receptor potential vanilloid type 1) channel is considered to be one of the main targets for pro-inflammatory mediators including TNF-α. Similarly, the inhibition of TRPV1 activity in the peripheral nervous system affects pro-inflammatory mediator production and enhances analgesia in total. In this study, the analgesic and anti-inflammatory effects of HCRG21, the first peptide blocker of TRPV1, were demonstrated in a mice model of carrageenan-induced paw edema. HCRG21 in doses of 0.1 and 1 mg/kg inhibited edema formation compared to the control, demonstrated complete edema disappearance in 24 h in a dose of 1 mg/kg, and effectively reduced the productionof TNF-α in both doses examined. ELISA analysis of blood taken 24 h after carrageenan administration showed a dramatic cytokine value decrease to 25 pg/mL by HCRG21 versus 100 pg/mL in the negative control group, which was less than the TNF-α level in the intact group (40 pg/mL). The HCRG21 demonstrated potent analgesic effects on the models of mechanical and thermal hyperalgesia in carrageenan-induced paw edema. The HCRG21 relief effect was comparable to that of indomethacin taken orally in a dose of 5 mg/kg, but was superior to this nonsteroidal anti-inflammatory drug (NSAID) in duration (which lasted 24 h) in the mechanical sensitivity experiment. The results confirm the existence of a close relationship between TRPV1 activity and TNF-α production once again, and prove the superior pharmacological potential of TRPV1 blockers and the HCRG21 peptide in particular.

Deer antlers are the only mammalian appendage capable of regeneration. We aimed to investigate the effect of red deer antler extract in regulating hair growth, using a mouse model. The backs of male mice were shaved at eight weeks of age. Crude aqueous extracts of deer antler were prepared at either 4°C or 100°C and injected subcutaneously to two separate groups of mice (n=9) at 1 mL/day for 10 consecutive days, with water as a vehicle control group. The mice skin quantitative hair growth parameters were measured and 5-bromo-2-deoxyuridine was used to identify label-retaining cells. We found that, in both the 4°C and the 100°C deer antler aqueous extract-injection groups, the anagen phase was extended, while the number of BrdU-incorporated cells was dramatically increased. These results indicate that deer antler aqueous extract promotes hair growth by extending the anagen phase and regulating cell proliferation in the hair follicle region.
