The article describes the results of experimental studies aimed at investigating toxic effects of methyl tertbutyl ether [MTBE] and nitrobenzene [NB] according to the criteria of changes in the immune system status of laboratory animals in conditions of cold stress and in conditions of temperature optimum. The research was carried out in the conditions of a subacute experiment (during one month of experiment) on laboratory animals (adult WAG male rats). The NB or MTBE were injected into the stomach at a dosage 1/10 LD50 (70 mg/kg of NB and 500 mg/kg MTBE) thirty times, the experimental animals were kept in two different temperature conditions during four hours five times per week. The animals were divided into 4 groups, six animals in each group. The animals of the first group were subjected to NB or MTBE at an air temperature of 252C (normal ambient temperature). The animals of the second group served as a control group for the first one. The animals of the third group were subjected to NB or MTBE in combination with the lowered air temperature of 4±2℃. The animals of the fourth group were only subjected to the effect of the reduced temperature of 4±20℃, that is, they were a control group for the ones of the third group. The analysis of the results of the experimental data about the features of the immunotoxic action of NB and MTBE under conditions of cold stress indicates the tension of the immune system of the laboratory animals, which is manifested by progressive lymphopenia and a decrease in the phagocytic activity of neutrophils in absorption of foreign agents, especially in the end-period. The immunotoxic effect of NB and MTBE under conditions of temperature optimum indicates a temporary immune system tension, namely leukocytosis and leukopenia at the stage only after 5 and 15 priming. At the end of the priming period, shifts in the immune system's performance under the effect of NB and MTBE under temperature optimum conditions were not observed, which may indicate a positive adaptability of laboratory animals to the action of chemical factors (NB and MTBE) under such temperature conditions. The combined effect of NB and MTBE on the body at the level of their effective concentrations and low temperature causes a detrimental effect on the adaptability of the body. Thus, these chemical compounds at the stage of physiological adaptation, that is before the formation of the pathological process, complicate the formation of specific and nonspecific reactions responsible for the adaptation of the organism to the effect of another factor in the production or environment – a low air temperature. The latter, in turn, creates an additional load on biochemical detoxification systems, leads to their imbalance and changes in the rate of biotransformation of the chemical agent and the speed of development of the toxic process.

Цель исследования - оценка возможности использования интегральных гематологических индексов для выбора оптимальной модели ушивания послеоперационных ран в эксперименте. Методика. Исследование проводили на 297 белых беспородных крысах-самцах массой 190-210 г, которые были разделены на 3 группы, по 99 крыс в каждой. Одна группа являлась контрольной, у второй группы крыс был смоделирован раневой процесс с натяжением брюшной стенки; у третьей группы животных - модель раневого процесса с применением трансплантата нанокомпозитного материала. Определяли следующие интегральные гематологические индексы: индекс Кребса; коэффициент Бредекка; лейкоцитарный индекс; индекс соотношения нейтрофилов и моноцитов; индекс соотношения лимфоцитов и моноцитов; индекс соотношения лимфоцитов и эозинофилов; коэффициент отношения альбуминов к глобулинам; оксипролиновый коэффициент. Результаты. Установлены статистически значимые колебания показателей интегральных гематологических индексов в динамике заживления послеоперационной раны у крыс. Заключение. На основе полученных результатов был сделан вывод, что особенности динамики интегральных гематологических индексов, выявленные на различных моделях раневого процесса, отражают неспецифические и специфические защитные реакции организма в целом, позволяют делать прогноз дальнейшего течения и исхода раневого процесса, а также обеспечивают возможность выбора оптимальной модели ушивания послеоперационной раны. The article presents results of predicting the wound healing process using integral hematological indexes. The aim of the study was to describe the dynamics of integral hematological indexes of wound healing process in rats to predict its outcome. The study included 297 white mongrel male rats weighing 190-210 g, which were divided into three groups, 99 rats in each. One group was a control group; in the second group, a wound healing process with abdominal wall tension was modeled; in the third group, a wound healing process with a graft was modeled. The following integrated hematological indexes were determined: Krebs index; Bredeck index; leukocyte index; neutrophil/monocyte ratio; lymphocyte/monocyte ratio; lymphocyte/eosinophil ratio; albumin/globulin ratio; and oxyproline coefficient. The study showed that changes in values of integral hematological indexes were mostly statistically and clinically significant. We concluded that changes in integral hematological indexes in different models of wound healing reflect nonspecific and specific protective responses of the body as a whole and allow to predict further course and outcome of the wound healing process.

Objective: to develop and justify the application of a new experimental method of the simulation of liver cirrhosis in laboratory animals. Material and methods. The simulation of liver cirrhosis was performed on 11 eugamic white Wistar male rats with the body weight of 203.5 ± 22.2 g (experimental group). The control group consisted of 12 healthy laboratory animals. Acute toxic liver injury resulting in cirrhosis was caused by means of the intraperitoneal administration of 50 % solution of carbon tetrachloride (CCl4) in olive oil on the first day of the experiment at a dosage of 0.1 ml of CCl4 + 0.4 ml of olive oil per 100 g of the body weight of the animals, on the second day of the experiment - 0.3 ml of CCl4 + 0.2 ml of olive oil per 100 g of the body weight of the animals. For synergism and potentiation of the hepatotoxic effect of CCl4, the animals daily had free access to 10% ethanol solution. The duration of the experiment was 65 days. The clinical and laboratory parameters were evaluated, the histological assessment of the preparations was carried out. The obtained data were compared with the same parameters of the control group of the animals. Results. The reproducibility of the model was 81.8% (9 animals). The values of the biochemical blood analysis indicated statistically significant increases in the levels of total bilirubin, serum transaminases (AST, ALT), creatinine, a decrease in the glucose level in the animals of the experimental group. The complex morphological confirmation of liver cirrhosis in progress was obtained. Conclusion. The proposed method of the liver cirrhosis modeling correlate with the values of the biochemical blood analysis, pathological changes in the tissue of the liver and internal organs of liver cirrhosis in humans. With the help of the original model, it is possible to investigate the pathogenesis and effects of various groups of pharmacological drugs on liver cirrhosis and its complications (acute blood loss associated with the syndrome of portal hypertension).

Abstract. Modern conditions of animal husbandry and poultry farming dictate a continuous search for effective means that increase the natural resistance of the organism of farm animals and poultry. The purpose of the study. In this regard, an experimental study was conducted on laboratory animals using a mineral adaptogen of domestic production in order to determine its effectiveness in conditions of artificially induced immunosuppression. The objectives of the study were to determine the immunobiochemical status, subcellular and intra-organ changes in the organs of the immune system at all levels. Research methods. Experimental studies were carried out using generally accepted zootechnical, pathomorphological, histological, immunobiochemical and statistical research methods. Results. Immunobiochemical changes in the body of experimental animals are described. Microscopic changes at the level of tissues and cells in the organs of the immune system at all levels are described. Changes in the organs of the immune system during immunosuppression and against the background of feeding mineral adaptogen are described. Morphological examination of the immune system organs in experimental animals revealed prolonged immunosuppressive changes for 14 days. At the same time, after induced immunosuppression, significant structural changes remain in the central and peripheral organs of the immune system in the form of lymphoid tissue atrophy (in the thymus, both in the organ of central immunogenesis and in the organs of the peripheral immune system – the spleen and lymph nodes and intra-organ lymph formations). During histological studies, the structure of cells and tissues, when using a mineral adaptogen, was more morphologically mature and was in a functionally active state, and dystrophic and necrotic processes were observed in the control group. The conducted studies convincingly prove the effect of mineral adaptogen on increasing natural resistance and its immunoprotective properties. Scientific novelty. For the first time in experimental conditions, a simultaneous assessment of the immunobiochemical parameters of the blood of laboratory animals and a microscopic examination of the organs of the immune system at all levels with morphometric analysis of the data obtained were given. As a result of the conducted studies, the immuno- and organoprotective effect of feeding the mineral adaptogen was proved and the immunomodulatory effect was morphometrically confirmed.

AbstractSerum immunoglobulin (IgG, IgA and IgM) levels were determined in patients with chronic tonsillitis before and one month after tonsillectomy. The preoperative levels of serum IgG, IgA and IgM were significantly higher when compared with the controls. The increase may be due to repeated antigenic stimulation. The post-operative levels for the three immunoglobulins were decreased; however, a significant reduction was observed for IgG only where the mean value was comparable with the control group. The data confirm that tonsillectomy does not disturb the humoral immune system of the body.

Abstract Objectives Several studies have established the ethnobotanical benefits of Pupalia lappacea (PL) in laboratory animals without extensive toxicological evaluation of its safety profiles. Thus, an extensive toxicological investigation of sub-chronic oral administration of the hydroethanol leaf extract of P. lappacea in rodents was carried out in this study. Methods Different groups of rats were treated orally with the extract (10, 50 and 250 mg/kg) daily for 90 consecutive days. The control group received distilled water (10 mL/kg). After 90 days, some rats were left for additional 30 days without treatment for reversibility study. Blood and organs samples were collected for different evaluations at the end of study periods. Results The extract decreased the bodyweights, feeding and water intakes in female rats. PL increased the weights of the liver and kidney in male rats. PL increased the red blood cell (RBC), packed cell volume (PCV), hemoglobin (Hb), triglycerides (TRIG), cholesterol and high density lipoprotein (HDL) contents in rats. PL (250 mg/kg) significantly reduced the sperm motility and serum testosterone level. Cyto-architectural distortions of the testes, liver and spleen were visible. Conclusions The findings showed that P. lappacea is relatively safe at lower doses but cautions should be taken at higher dose.

The aim of the study is to increase the productive qualities of broiler chickens by including ultrafine silicon oxide into main diet. During the experiment, the biological effect of ultrafine silicon oxide on broiler chickens was estab-lished. Use of ultrafine SiO2 particles for poultry feeding contributed to an increase in the number of red blood cells and content of total protein and albumins. By the end of the experiment, the number of red blood cells in birds in-creased by 17.43% (P≤0.001) – in the blood of birds of the first experimental group, 16.51% (P≤0.01) – the second one, 20.80% (P≤0.001) – the third experimental and 21.71% (P≤0.001) – the fourth experimental group, compared with the indicator of the control group. The amount of total protein in blood serum of chickens of the first and the second experimental groups increased by 1.36-1.39 %, in the third and fourth ones there was a significant (P≤0.05) increase by 5.45 and 3.05%, respectively. The blood glucose content of chickens in the experimental groups is higher by 8.04-23.65% compared to this indicator with ones in the control group. During the experiment feed con-sumption per 1 kg gain of live weight decreased: in the first experimental group by 3.00 % in the second by – 0.50 %, the third – 6.00 % the fourth– by 4.50 %, compared to this with the control group. The chicken’s vibrancy of the first the experimental group was higher by 4.77 %, the second – by 6.20 %, the third – by 19.25 % and the fourth– by 11.59% than in the control one. Consequently, when converting the feed energy into the body energy of a broiler chicken, the energy conversion coefficient of the experimental group of poultry is higher than that of the control one by 7.16-21.76 %. Thus, the most optimal dose for further research was determined.

In modern conditions there is a high need to improve methods for establishing the nature of the combined influence of factors using modern approaches for further development of prevention measures against the negative impact of factors on the body. The purpose of the study was to investigate the combined effect of electromagnetic radiation and low temperatures with subsequent determination of the level of contribution of each factor in the formation of biological effects within the experiment by mathematical analysis of data using the method of artificial intelligence. Materials and methods. The subchronic experiment (30 days) was performed on male rats. The animals were divided into groups: combined exposure to electromagnetic radiation (70 kHz, 600 V/m) and reduced temperature (40C), isolated exposure to reduced temperature, isolated exposure to electromagnetic radiation and a control group. The establishment of biological effects was performed at stages 5, 15 and 30 days of the experiment. Changes in the body were assessed by physiological, biochemical and immunological parameters of the blood (30 indicators). Data processing was performed using computational intelligence methods (Neuro-fuzzy system). Results and discussion. The combined effect of the factors was manifested by the following biological effects: intensification of peroxidation processes with simultaneous moderate suppression of antioxidant protection, intensification of lipid metabolism was characterized by dyslipoproteinemia, manifesting in: increase in triglycerides, cholesterol and its fractions. Changes of the immune system were decrease in the ability of neutrophils to phagocytosis at different stages of the experiment, imbalance of the activation of the system of reducing the concentration of C4 at the background of increased intensity of IgM and IgG synthesis. The method of determining the intensity of the factors, developed on the basis of factor and cluster analysis, allowed to determine that during the experiment the ratios between the factors changed, but the greatest contribution was made by reduced temperature by both biochemical parameters (63%) and immunological (53%) at the same time, the level of electromagnetic radiation contribution was 37% and 47%, respectively. Thus, the reduced temperature was the leading factor in the combined effect, which added 60% to the overall biological effect throughout the study, against the effect of electromagnetic radiation for which the level of contribution was only 40%. By means of the factor analysis informative indicators, making it possible to establish the characteristics of biological effects, and therefore leading indicators in formation of biological effects were the increase in concentration of DC with simultaneous decrease in concentration of SH-groups, increase in VLDL and suppression of oxygen-dependent metabolism of neutrophil-test. Conclusion. Thus, the tested mathematical approach allowed to determine the leading role of each of the studied factors under the conditions of their combined influence in different systems, which allows to predict probable appropriate reactions of the whole organism and gives a reasonable approach to the development of preventive measures

     This study was carried out at Animal Farm, College of Veterinary Medicine, Baghdad University from April up to July 2013. The animals were fed on concentrate diet and freely grazed for 3-6 hours/ day at College Field, rams were used in a Latin square design (4*4) and trans located biweekly intervals, respectively to different concentrate diets, the first diet 2% of the body weight which was free from any addition  and considered as a control, the second diet was contain 3% Cuminum cyminum seeds (CU), while the third diet was contain 3% of Ocimum basilicom seeds (B) and the fourth concentrated diet was contain 1.5% of Cuminum cyminum and 1.5% Ocimum basilicom seeds (CU+B). Results revealed that there was significant (P<0.05) difference existed during the first two periods in the body gain compared with other periods, pH of rumen liquor of the control group 6.00±0.24 showed significantly (P<0.05) higher than all other animals during all periods 5.42±0.095, 5.40±0.14 and 5.65±0.11, while the volatile fatty acids were significantly lower in the control animal 8.25±0.75 compared with other animals, ammonia concentration in the rumen liquor, showed higher significant (P<0.05) difference 9.33±0.13 and 9.60±0.43 respectively in the third and fourth period than first period 7.52±0.28, on the other hand bacterial count of the control group was significantly (P<0.05) lower than other groups.

We have recently reported that pretreatment with electroacupuncture (EA) at stomach meridian point 36 (St36) prevents the chronic cold-stress increase in the hypothalamus-pituitary-adrenal axis (HPA), an action that may be under central control. Given that treatment for stress-related symptoms usually begins after onset of the stress responses, the objectives of the present study were to determine the efficacy of EA St36 on HPA hormones when EA St36 is given after stress was initiated, if the results are long lasting, and if blocking the glucocorticoid receptor (GR) using RU-486 had the same effects as EA St36. Adult male rats were placed in 4 groups of animals, 3 of which were exposed to cold and 1 of which was a nontreatment control group. After exposure to the cold stress, 2 groups were treated with either EA St36 or sham-EA, repeated over 10 days. The increase in ACTH and corticosterone observed in stress-only rats was prevented in EA St36 animals, and the effects remained intact 4 days after withdrawal of EA but continuation of cold stress. When the GR was blocked with RU-486, the efficacy of EA St36 remained unchanged. GR blockade did significantly elevate ACTH, which is not seen with EA St36, suggesting that EA St36 does act centrally. The elevated HPA hormones in stress-only rats were associated with a significant increase in depressive and anxious behavior; this was not observed in the stressed EA St36 animals. The results indicate that EA specifically at St36 vs sham-EA is effective in treating chronic poststress exposure.

There are considerable public concerns about the relationship between mobile phone radiation and human health. The present study assesses the effect of electromagnetic field (EMF) emitted from a mobile phone on the immune system in rats and the possible protective role of vitamin D. Rats were randomly divided into six groups: Group I: control group; Group II: received vitamin D (1000 IU/kg/day) orally; Group III: exposed to EMF 1 h/day; Group IV: exposed to EMF 2 h/day; Group V: exposed to EMF 1 h/day and received vitamin D (1000 IU/kg/day); Group VI: exposed to EMF 2 h/day and received vitamin D (1000 IU/kg/day). After 30 days of exposure time, 1 h/day EMF exposure resulted in significant decrease in immunoglobulin levels (IgA, IgE, IgM, and IgG); total leukocyte, lymphocyte, eosinophil and basophil counts; and a significant increase in neutrophil and monocyte counts. These changes were more increased in the group exposed to 2 h/day EMF. Vitamin D supplementation in EMF-exposed rats reversed these results when compared with EMF-exposed groups. In contrast, 7, 14, and 21 days of EMF exposure produced nonsignificant differences in these parameters among all experimental groups. We concluded that exposure to mobile phone radiation compromises the immune system of rats, and vitamin D appears to have a protective effect.
