Abstract: E. coli is part of the normal flora of the human and animal intestine and is commonly non pathogenic. However, one of the serotypes of this bacteria, which is enteropathogenic E. coli (EPEC), can cause primary infection on the intestine such diarrhea.  The growth of pathogenic bacteria in diarrheal patients can be inhibited by Lactobacillus.  Lactobacillus can function as probiotic which can affect the immune system of the digestive canals.  In addition, Lactobacillus also produce mucus  which can act as barrier to the pathogens.  The objective of this study was to reveal the effects of the administration of Lactobacillus on the microscopic features of the mucosa of the intestine of wistar rats infected by Escherichia coli.  This study was a laboratory experimental research employing 16 wistar rats divided into the control group (4 rats) and three treatment groups (12 rats) consisting of 4 rats each.  Results showed that; in group I the histological features were normal, in group II part of the epithelium of the mucosa showed erosion, dilatation of the capillary vessels, and many lymphosites were observed, in group III the epithelium of the mucosa was intact and the number of lymphosites was liitle, and in group IV the surface of the epithelium was intact, the presence of cell regeneration indicated by the increase number of goblet cells and a small number of  lymphocyes. Conclusions: The administration of after infection by E. coli has benefial effects indicated by the improvement of epithelial cells and the absence of denudation of the epithelium of the intestine. Keywords: E.coli, Lactobacillus, intestinal mucosa.   Abstrak: E.coli merupakan flora normal usus halus manusia dan hewan umumnya tidak menyebabkan penyakit.  Namun salah satu serotipe E.coli yaitu E.coli Enteropatogenik (EPEK) bersifat patogen dan dapat menyebabkan infeksi primer pada usus misalnya diare.  Pertumbuhan bakteri patogen pada pasien diare dapat dihambat oleh Lactobacillus. Lactobacillus merupakan probiotik yang akan mempengaruhi sistem imun saluran cerna serta memproduksi mukus yang berfungsi sebagai penghalang saluran cerna terhadap bakteri patogen.  Penelitian ini bertujuan untuk melihat efek pemberian Lactobacillus terhadap gambaran mikroskopis mukosa usus halus tikus wistar yang diinfeksi dengan Escherichia coli.  Penelitian ini merupakan penelitian eksperimental laboratorik.  Subjek penelitian terdiri dari 16 ekor tikur wistar yang dibagi dalam kelompok kontrol (4 ekor) dan kelompok  perlakuan (12 ekor) dibagi dalam 3 kelompok masing-masing 4 ekor.  Hasil penelitian kelompok I dengan gambaran histologik jaringan usus normal, kelompok II sebagian epitel mukosa usus halus terlihat erosi,  ada pelebaran pembuluh darah kapiler, dan banyak limfosit, kelompok III dengan permukaan epitel mukosa yang utuh dan jumlah limfosit sedikit, dan kelompok IV dengan permukaan epitel yang utuh, adanya regenerasi sel ditandai dengan bertambahnya sel goblet, dan sedikit limfosit.  Simpulan: Pemberian Lactobacillus setelah diberikan E.coli memberi efek yang baik berupa perbaikan sel epitel permukaan dan tidak terlihat denudasi epitel permukaan usus halus. Kata Kunci: E.coli, Lactobacillus, mukosa usus halus.

The present study was conducted to investigate the efficacy of Ovsynch and Ovsynch plus protocol in postpartum (day 60) Sahiwal cows (n=18). Animals were randomly divided into three equal groups, viz., Ovsynch group Ovsynch plus group and Control group. Animals of group I (n = 6) were treated with traditional Ovsynch protocol. The animals (n = 6) of group II were treated with Ovsynch plus protocol which consisted of an initial intramuscular injection of eCG (Folligon) @ 250 IU on day 60 postpartum followed 3 days later by GPG (Ovsynch) protocol. In group-III Control, no treatment was given to animals (n = 6). Treated animals were inseminated at a fixed time between 14 and 20 hrs after second GnRH injection, irrespective of estrus detection. Blood samples were collected from each animal on days 50 and 60 postpartum to determine the status of cyclicity in animals based on serum concentrations of progesterone (P4). A third blood sample was collected on the day of prostaglandin treatment to determine the response of first GnRH injection. Four animals each were cyclic, and two were acyclic in both treatment groups. Four animals each responded to first GnRH treatment in both treatment groups. Similarly, two animals each got conceived giving conception rate of 50% (2/4) in each treatment. In the control group, one out of 6 animals got conceived yielding 16.66 % conception rate (1/6) during the study period. It may be thus concluded that Ovsynch and Ovsynch plus protocol may be used during the early postpartum period to improve the reproductive efficiency in postpartum Sahiwal cows.

Summary The effect of aqueous matured stem extract of Opuntia dillenii on selected biochemical parameters in Male Wistar rats was explored. Standard analytical methods were applied. Forty Wistar rats (80-100g) were used in the animal studies, separated into four groups. The control group was solely administered normal feed and saline, group I was administered 100mgkg−1 of the extract, group II received 300mgkg−1 of the extract and group III received 500 mg/kg−1 of the extract. A significant increase (p<0.05) in the activities of alanine aminotransferase (ALT) and alkaline phosphatase was observed in group II and III rats, as compared with the controls. A significant decrease in urea and creatinine concentrations was found only in group III rats against the controls. Also, a significant (p<0.05) decrease in triglyceride, total cholesterol, and low density lipoprotein (LDL)-cholesterol was seen in group II and group III rats when compared with the control. The hematological evaluation revealed a significant (p<0.05) decrease in red blood cell and hemoglobin levels in group III rats when compared with the control. The findings showed both beneficial and toxicological effects of the plant. Hence, for optimal therapeutic benefits, a further toxicological survey could still be carried out perhaps at higher doses.

Background: Bacteria playing a role in periodontitis are Gram-negative anaerobic bacteria that can release endotoxin or lipopolysaccharide (LPS). LPS acts as a stimulus to a variety of host cells that can stimulate expression of pro-inflammatory cytokines in periodontal disease, such as IL-1α, IL-1β, and TNF-α. Increased TNF-α then can lead to periodontal tissue destruction. Furthermore, cassava leaves have many health benefits due to flavonoid organic compound contained, known to possess anti-inflammatory activity are used as a medicine. Purpose: This study aimed to determine the effect of cassava leaf flavonoid extract as a basic ingredient of anti-inflammatory gel on TNF-α expression in Wistar rats suffering from periodontitis induced with Escherichia coli (E. coli) LPS. Method: This study used 24 male Wistar rats. Those rats were divided into six groups. Group 1 consisted of four rats induced with E. coli LPS for 2 weeks, and then decapitation was performed on day 3. Group 2 consisted of four rats induced with E. coli LPS for 2 weeks, and then decapitation was conducted on day 7. Group 3 consisted of four rats induced with E. coli LPS for 2 weeks, treated with the topical cassava leaf flavonoid extract gel (Manihot esculenta) at a concentration of 25%, and then decapitation was performed on day 3. Group 4 consisted of four rats induced with E. coli LPS for 2 weeks, treated with the topical cassava leaf flavonoid extract gel (Manihot esculenta) at a concentration of 25%, and then decapitation was conducted on day 7. Group 5 consisted of four rats induced with E. coli LPS for 2 weeks, treated with the topical cassava leaf flavonoid extract gel (Manihot esculenta) at a concentration of 50%, and then decapitation was performed on day 3. And, group 6 consisted of four rats induced with E. coli LPS for 2 weeks, treated with the topical Cassava leaf flavonoid extract gel (Manihot esculenta) at a concentration of 50%, and then decapitation was conducted on day 7. The topical Cassava leaf flavonoid extract gel was inserted into gingival sulcus on the first right molar of their lower jaw by using a blunted syringe needle. The gel was given two times a day for 7 days. Result: The expression of TNF-α in the control group was more than that in the treatment groups given the Cassava leaf flavonoid extract gel at the concentrations of 25% and 50%. The expression of TNF-α in the treatment groups given the cassava leaf flavonoid extract gel at the concentration of 50% was lower than that in the treatment groups given the cassava leaf flavonoid extract gel at the concentration of 25%. Conclusion: Cassava leaf flavonoid extract gel could be used as an anti-inflammatory gel characterized by a decrease in TNF-α expression in rat models suffering from periodontitis.

Abstract: Liver is the largest organ in the abdominal cavity. As the center of metabolism in the body, liver is potentially damaged by exposure of toxic substances, inter alia carbon tetrachloride (CCl4). Metabolism of carbon tetrachloride (CCl4) produces CCl3 free radicals that can damage the liver. In Indonesia, there are a lot of natural ingredients that have antioxidant properties, such as tomato. Lycopene in tomatoes contains antioxidant compounds that can prevent damages due to free radical. This study aimed to obtain liver histopathological changes of wistar rats fed with tomato juice after being induced of carbon tetrachloride (CCl4). This was an experimental study, using 10 wistar rats which were divided into 4 groups. Group I was the negative control; group II was induced with CCl4 0,05 cc/day and was terminated on day 6; group III was induced with CCl4 0,05 cc/day and was given tomato juice 3 ml/day, and terminated on day 13; group IV was induced by CCl4 0,05 cc/day, given regular pellets, and terminated on day 13. The results showed that group II had histopathological changes of the liver indicating fatty liver, meanwhile group III showed regeneration of nearly all liver cells. Conclusion: Administration of tomato juice after the induction of 3 ml carbon tetrachloride (CCl4) for 7 day showed regeneration of almost all liver cells. Keywords: histopathological changes of the liver, carbon tetrachloride, tomato juice.   Abstrak: Hati merupakan organ terbesar dalam rongga abdomen, dan pusat metabolisme tubuh dengan fungsi yang sangat kompleks dan sangat berpotensi mengalami kerusakan akibat terpapar oleh bahan-bahan toksik, salah satunya yaitu karbon tertraklorida (CCL4). Metabolisme CCl4 menghasilkan radikal bebas CCl3 yang dapat merusak hati. Di Indonesia terdapat  banyak sekali bahan-bahan alami yang mempunyai kandungan antioksidan, salah satunya yaitu tomat. Tomat mengandung senyawa likopen sebagai antioksidan yang dapat mencegah kerusakan jaringan akibat radikal bebas. Penelitian ini bertujuan untuk mendapatkan gambaran histopatologi hati tikus wistar yang diberi jus tomat pasca induksi karbon tetraklorida (CCl4). Metode penelitian ialah eksperimental. Sampel sebanyak 10 ekor tikus wistar yang dibagi dalam 4 kelompok. Kelompok I sebagai kontrol negatif; kelompok II diinduksi CCl4 0,05cc/perhari dan diterminasi hari ke-6; kelompok III diinduksi CCl4 0,05 cc/hari kemudian diberikan jus tomat 3ml/hari;  dan kelompok IV diinduksi CCl4 0,05 cc/hari kemudian diberikan pelet biasa dan diterminasi hari ke-13. Hasil penelitian menunjukkan pada pemberian CCl4 pada tikus wistar selama 5 hari terdapat gambaran morfologik perlemakan sel hati. Pemberian jus tomat dosis 3 ml pasca induksi karbon tetraklorida (CCl4) menunjukkan terjadinya regenerasi pada hampir seluruh sel-sel hati. Simpulan: Pemberian jus tomat dosis 3 ml pasca induksi karbon tetraklorida (CCl4) selama 7 hari menunjukkan regenerasi pada hampir seluruh sel-sel hati. Kata kunci: gambaran histopatologi hati, karbon tetraklorida, jus tomat.

A method for quantitating the conversion of Escherichia coli to colony-forming, cell wall-defective (CWD) bacteria has been developed. The induction frequency, i.e., the percentage of the population recovered as CWD colonies was determined for 20 randomly selected clinical isolates of E. coli under aerobic and anaerobic incubation conditions. Penicillin (1,000 U/ML) was the inducing agent. The 20 strains segregated into three groups. Group I organisms produced CWD colonies with high frequency both aerobically and anaerobically. Grout II organisms showed a much higher induction frequency anaerobically than aerobically. Group III organisms were poor inducers. Thirty percent of the strains were group I, 50% were group II, and 20% were group III organisms. These data indicate that anaerobic conditions enhance the induction and growth of CWD E. coli in the research laboratory and suggest that anaerobic incubation may be important in recovery of medically significant CWD bacteria.

Background and Objectives: The aim of this study was to compare the systemic humoral immune responses, including IgE, IgA, IgG and IgM levels in Balb/c mice administered a probiotic, LPS derived from Escherichia coli (E.coli), and probiot- ic-LPS derived from E. coli. Materials and Methods: Thirty-two male Balb/c mice, 10-12 weeks of age with body weight ranging from 30-40 g were randomly divided into four experimental groups (n=8). The treatment regimens were as follows: Group 1, mice did not receive LPS or probiotic (control group); Group 2, mice received only LPS on the first day; Group 3, mice received probi- otic for 7 days; Group 4, mice received LPS on the first day, and then continued, with probiotic for 7 days. The mice were observed for 8 days, and then, euthanized the next day (day 9). The serum was collected, and the levels of IgE, IgA, IgG and IgM were measured using ELISA. Results: The humoral immune response was higher in the presence of a probiotic compared to that in the control; IgE (9.02 ± 0.58 units/ml, p=0.000), IgA (3.26 ± 0.99 units/ml, p=0.316), IgG (7.29 ± 0.24 units/ml, p=0.000), and IgM (4.01 ± 2.98 units/ml, p=0.505). When administered with LPS E. coli along with probiotic, the humoral immune response was the highest; IgE (10.68 ± 1.63 units/ml, p=0.000), IgA (8.34 ± 1.47 units/ml, p=0.000), IgG (9.96 ± 0.98 units/ml, p=0.000), and IgM (4.31 ± 1.05 units/ml, p=0.319) compared to the control group. Conclusion: Probiotic-LPS derived from E. coli treatment induced a higher humoral immune response (highest IgE, IgA, IgG and IgM levels) compared to treatment with probiotic only.

Doxorubicin-induced cardiotoxicity is the leading cause of morbidity and mortality among cancer survivors. The present study was aimed to investigate the cardioprotective potential of methyl gallate; an active polyphenolic nutraceutical, against doxorubicin-induced cardiotoxicity in Wistar rats. Twenty-four female Wistar rats (150–200 g) were divided into four groups (n = 6) which consist of normal control (group I), doxorubicin control (group II), test-A (group III), and test-B (group IV). Group III and group IV animals were prophylactically treated with methyl gallate 150 mg/kg/day and 300 mg/kg/day orally, respectively, for seven days. Doxorubicin (25 mg/kg; single dose) was administered through an intraperitoneal route to group II, III, and IV animals on the seventh day to induce acute cardiotoxicity. On the 8th day, besides ECG analysis, serum CK, CK-MB, LDH, AST, MDA, and GSH were assayed. Following gross examination of isolated hearts, histopathological evaluation was performed by light microscopy. A significant ( p  < 0.05) cardiac injury, as well as oxidative stress, was observed in doxorubicin control rats in comparison to normal control rats. Methyl gallate at both the doses significantly ( p  < 0.05) reduced doxorubicin-induced ECG changes, dyslipidaemia, and elevation of CK, CK-MB, LDH, AST, MDA and increased GSH level. Methyl gallate reversed the doxorubicin-induced histopathological changes in the heart. The present study revealed that methyl gallate exerts cardioprotection against doxorubicin-induced cardiotoxicity in female Wistar rats by suppressing oxidative stress. Our study opens the perspective to clinical studies for consideration of methyl gallate as a potential chemoprotectant nutraceutical in the combination chemotherapy with doxorubicin to limit its cardiotoxicity.

Objectives: The objective of this study was to find the histologic and motor activity effect of lead on prenatally and postnatally exposed Wistar rats. Methods: In this study, twelve Wistar Rats were used and grouped into four groups of two females and one male. Group I rats served as the control and allowed feed and water freely. The rats in Group II were administered 500ppm of Pb through drinking water from gestation day 8 (GD8) to parturition (GD21). While Group III rats were given 500ppm of Pb in drinking water from postnatal day 1 (PND1) to PND21. The rats in the fourth group (Group IV) were given 500ppm of Pb from GD8 to PND21. Palmer grasp reflex was conducted to assess the motor activity of the rat pups. The animals were then humanely sacrificed and the frontal cortices were isolated for routine histological processing. Results: The histological study has shown normal neurons in the control group while degenerating cells exhibiting karyolysis, pyknosis, karyorrhexis, vacuolation were seen in the lead-treated groups. Group II and Group IV showed considerate deficit in their motor activity while Group III showed mild effect. Conclusion: From this study, lead exposure of Wistar rats at both prenatal and postnatal period of development has effect on the histology of the frontal cortex as well as on their motor activity.

To prevent economic losses due to post-weaning diarrhea (PWD) in industrial pig production, zinc (Zn) feed additives have been widely used, especially since awareness has risen that the regular application of antibiotics promotes buildup of antimicrobial resistance in both commensal and pathogenic bacteria. In a previous study on 179 Escherichia coli collected from piglets sacrificed at the end of a Zn feeding trial, including isolates obtained from animals of a high-zinc fed group (HZG) and a corresponding control group (CG), we found that the isolate collection exhibited three different levels of tolerance toward zinc, i.e., the minimal inhibitory concentration (MIC) detected was 128, followed by 256 and 512 μg/ml ZnCl2. We further provided evidence that enhanced zinc tolerance in porcine intestinal E. coli populations is clearly linked to excessive zinc feeding. Here we provide insights about the genomic make-up and phylogenetic background of these 179 E. coli genomes. Bayesian analysis of the population structure (BAPS) revealed a lack of association between the actual zinc tolerance level and a particular phylogenetic E. coli cluster or even branch for both, isolates belonging to the HZG and CG. In addition, detection rates for genes and operons associated with virulence (VAG) and bacteriocins (BAG) were lower in isolates originating from the HZG (41 vs. 65% and 22 vs. 35%, p &lt; 0.001 and p = 0.002, resp.). Strikingly, E. coli harboring genes defining distinct pathotypes associated with intestinal disease, i.e., enterotoxigenic, enteropathogenic, and Shiga toxin-producing E. coli (ETEC, EPEC, and STEC) constituted 1% of the isolates belonging to the HZG but 14% of those from the CG. Notably, these pathotypes were positively associated with enhanced zinc tolerance (512 μg/ml ZnCl2 MIC, p &lt; 0.001). Taken together, zinc excess seems to influence carriage rates of VAGs and BAGs in porcine intestinal E. coli populations, and high-zinc feeding is negatively correlated with enteral pathotype occurrences, which might explain earlier observations concerning the relative increase of Enterobacterales considering the overall intestinal microbiota of piglets during zinc feeding trials while PWD rates have decreased.

Background: Children require more nutrition as organs grow fast in this age group. Due to abundance of various commercial health supplements in market, parents are often confused about selection of health supplements. Traditional foods are rich source of various nutrients required by growing children. We conducted a study to compare strength, stamina and growth enhancing ability of commercial health supplement and natural health supplement on wistar rats.Methods: Total of 24 wistar rats were included. Animals were divided in four groups. Each group of rats except control group were receiving health supplements. Group I, group II, group III and group IV were receiving health supplement I, health supplement II, natural health supplement and no health supplement (control group) for 60 days. Strength and stamina of wistar rats in each group were analysed using grip strength meter and swim stress test, growth of rats in each group were analysed by measuring body length, tail length and body weight. Group of rat showing maximum growth (body length, tail length and body weight), strength and stamina were analysed.Results: Group II rats showed greater strength and stamina followed by group I, group III and group IV respectively. Group I rats showed maximum growth followed by group II, group III and group IV respectively.Conclusions: In the current study we found that strength and stamina were greater in group II rats followed by group I, group III and group IV respectively also group I rats showed maximum growth  followed by group II, group III and group IV respectively.
