Seagrass is a part of Phanerogamae, commonly has a symbiotic relationship with microbial endophytes. The types of microbes those have ability to produce bioactive compounds with potentially exploited for medical, agriculture and industrial purposes. Antibacterial testing using the modified Kirby-Bauer method. The fungi show a strong antibiotic activity which cultivated statically and Staphylococcus aureus was induced in rice medium for 10 days. The purpose of inducing bacteria to the culture is to stimulate a strong antibiotic activity through Silence Biosynthesis Pathway. Fungal isolates were macerated with 96% of ethanol for 24 hours. Partition process was performed by adding solvents (ethyl acetate, n-hexane, ethanol and water) to get n-hexane, ethanol and water fractions. All fractions were tested their activity against clinical isolates bacteria S. aureus and Escherichia coli. Ten fungal endophytes were isolated from seagrass Thalassia hemprichii and Enhalus acoroides. Two isolates derived from the leaf of both seagrass specimens (E.D.1 and Th.D.1) showed strong antibacterial activity against S. aureus only. Antibacterial activity test of each fraction both active isolates show in water and ethanol fractions. This indicates the active antibacterial compounds of both endophytic fungi have semi-polar and polar characteristics. However, bacterial induction has no effect on their antibacterial activity.Keywords : Endophytic fungi, Thalassia hemprichii, Enhalus acoroides, antibacterial activity Staphylococcus aureus and Escherichia coli.Lamun merupakan tanaman tingkat tinggi yang mempunyai hubungan simbiosis dengan mikroba jamur endofit. Mikroba endofit ini mempunyai kemampuan untuk memproduksi senyawa-senyawa bioaktif dengan potensi yang besar untuk dieksploitasi dan menghasilkan yang bermanfaat di bidang medis, pertanian, dan industri. Isolasi jamur dilakukan mengacu pada penelitian Bara et al (2013). Pengujian antibakteri dilakukan berdasarkan metode Kirby-Bauer yang dimodifikasi. Jamur memperlihatkan aktivitas antibiotik yang kuat dikultivasi statis dan di induksikan bakteri S. aureus dalam media nasi selama 10 hari. Tujuan pemberian bakteri pada kultur yaitu untuk memicu adanya aktivitas antibiotik yang lebih kuat melalui jalur biosintesis senyap (Silence Biosintethic Pathway) pada jamur tersebut. Isolat jamur di maserasi dengan menambahkan etanol 96% selama 24 jam. Proses partisi dengan menambahkan pelarut (etil asetat, n-heksan, etanol dan air) untuk memperoleh fraksi n-heksan, etanol dan air. Tiap fraksi diuji kembali aktivitas antibiotiknya pada bakteri S. aureus dan Escherichia coli. Hasil penelitian ini diperoleh sepuluh isolat jamur dari lamun T. hemprichii dan E. acoroides. Dua isolat daun (E.D.1 dan Th.D.1) menunjukkan aktivitas yang kuat terhadap bakteri S. aureus. Pengujian aktivitas antibakteri tiap fraksi kedua isolat jamur memperlihatkan fraksi air dan etanol yang menunjukkan aktivitas penghambat. Hal ini menunjukkan bahwa senyawa aktif antibakteri kedua jamur endofit berisfat semi polar dan polar. Induksi bakteri tidak memberikan pengaruh terhadap aktivitas antibakteri.Kata Kunci : jamur endofit, Thalassia hemprichii, Enhalus acoroides, aktivitas antibakteri Staphylococcus aureus dan Escherichia coli.

Abstract: The aim of this study is to investigate the antibacterial effect of endophytic fungi originated from the root of mangrove Rhizophora stylosa growing on Mangrove Plantation Area around DAS Simpang 5 Jl. Piere Tendean Manado. The method of this research was using combination of PDA and NA media. Two species of endophytic fungi were extracted from the root of R. stylosa, black mycelium fungi as isolate A and brown mycelium fungi as isolate B. The activities of both isolates were tested against pathogenic bacteria Staphylococcus aureus and Escherichia coli. The result showed the same inhibition zone of isolate A and B to S. aureus, while, isolate A showed bigger the inhibition zone in comparison to isolate B against E. coli. As a conclusion, the endophytic fungi taken from the root of R. stylosa have antibacterial activity toward S. aureus and E. coli. Keywords: endophytic fungi, Rhizophora stylosa, antibacterial, Staphylococcus aureus, Escherichia coli.   Abstrak: Penelitian ini bertujuan untuk melihat adanya efek antibakteri dari jamur endofit yang diambil dari akar bakau Rhizophora stylosa yang ditanam di sekitar Perairan Daerah Aliran Sungai Simpang Lima Jl. Piere Tendean Manado. Metode penelitian dilakukan dengan menggunakan kombinasi media Potato Dextrose Agar (PDA) dan Nutrient Agar (NA). Dari akar bakau R. stylosa diperoleh dua  jenis jamur endofit yaitu isolat A dengan karateristik miselium jamur berwarna hitam dan isolat B yang dengan karakteristik miselium berwarna coklat. Kedua isolat ini selanjutnya diujikan aktivitasnya terhadap bakteri Staphylococcus aureus dan Escherichia coli. Hasil penelitian menunjukkan isolat A dan B memberikan  zona hambat yang sama terhadap bakteri Staphylococcus aureus dan pada bakteri Escherichia coli isolat A menunjukkan diameter zona hambat yang lebih besar dibandingkan isolat B. Kesimpulan, Jamur endofit yang diisolasi dari akar bakau R. Stylosa memiliki efek antibakteri terhadap pertumbuhan bakteri S. aureus dan E. coli. Objective: Colletotrichium capsici endophytic fungi isolated from Jamblang plant (Eugenia cumini  L).  The secondary metabolites of this plant has the potent antibacterial efficacy  as well as diarrheal and anti-diabetic. This research focussed on isolation of the endophytic microbes from branches of Jamblang plants and their secondary metabolites.Methods: Isolation of endophytes were performed in PDA(potato dextrose agar) using direct seed plant. Endophytic fungi isolates with strongest antimicrobial activity against the bacteria Staphylococcus aureus, Escherichia coli were fermented in Potato Dextrose Yeast (PDY) to produce  large scale of the metabolites.  Supernatant was extracted with ethyl acetate solvent. Ethyl acetate extract fractionated by column chromatography (SiO3, n-hexane- ethyl acetate = 50:1 ~ 1:1) and obtained three fractions. Further, agar diffusion method was performed to assess their anti-microbial activity.Results:  Antibacterial  test  results  indicated that fraction III had  the antibacterial  activity Staphylococcus aureus with  inhibition zone diameter  of 10.7 mm but no observed antibacterial activity against Escherichia coli.  Furthermore, identification  by GC-MS showed that compounds present in fraction III was mainly fatty acid and phenolic compounds.Conclusion:  In conclusion, secondary metabolites isolated from Jamblang plants branches contained predominantly fatty acid and phenol related compounds that could be responsible for its potent anti microbial activity. Keywords : Endophytic fungi, Jamblang (Eugenia cumini L.), antimicrobial activity

The aim of this study was to verify the possible interactions between ethanol extracts ofAmburana cearensisA. C. Smith andAnadenanthera macrocarpa(Benth.) Brenan, combined with six antimicrobial drugs against multiresistant strains ofStaphylococcus aureusandEscherichia coliisolated from humans. The antibacterial activity of the extracts was determined using the minimum inhibitory concentration (MIC). The microdilution assay was performed to verify the interactions between the natural products and the antibiotics using a subinhibitory concentration. The activity of amikacin associated with the extract ofAnadenanthera macrocarpaagainst EC 27 was enhanced, demonstrating an MIC reduction from 128 to 4 μg/mL. Among theβ-lactams, no potentiation on its activity was observed, with exception to the antagonism of the natural products with ampicillin againstS. aureus358.

Background: Drug resistant bacteria related health problems are major concern globally and search for newer and most effective antibacterial agents is the urgent need to combat with these challenges. The study was undertaken with the aim of isolating and identifying endophytic fungi associated with Carica papaya and assessing their potential as antibacterial agents.Methods: Carica papaya plants were collected from different locations and endophytic fungi were isolated and characterized phenotypically and genotypically by microscopy, colonial characteristics and ITS gene sequencing, respectively. Antibacterial activity of endophyte was assessed against Escherichia coli, Klebsiella pneumonia, Salmonella Typhimurium, Bacillus cereus, Staphylococcus aureus and Streptococcus pyogenes, using the agar plate diffusion assay method with Ciprofloxacin as a positive control.Result: Thirty four (34) fungal endophytes of two genera Fusarium and Penicillium were recovered from 60 samples of Carica papaya and they showed the antibacterial activity against Staphylococcus aureus. Thus, the endophytic fungi have the potential to be used as an antibacterial agent.

ABSTRACT Endophytic fungi are fungi that live in plant tissues and do not harm these plants. Endophytic fungi can produce compounds that have the potential as antibacterial. This study aimed to examine the antibacterial effects of endophytic fungi isolated from the lemongrass (Cymbopogon citratus) against Staphylococcus aureus and Escherichia coli bacteria. Lemongrass is used to inhibit or kill pathogenic bacteria because it contains essential oils that function as an antifungal and antibacterial against several pathogenic bacteria such as Staphylococcus aureus and Escherichia coli. The laboratory experiment method tested the antibacterial activity of endophytic fungi isolates obtained from lemongrass (Cymbopogon citratus) against Staphylococcus aureus and Escherichia coli using hole / well method. From the results of the study, four endophytic fungi were isolated from the roots and leaves of lemongrass (Cymbopogon citratus). Antibacterial results showed that endophytic fungi extract isolated from lemongrass plants was more effective in inhibiting Staphylococcus aureus compared to Escherichia coli bacteria. In Staphylococcus aureus bacteria the average inhibitory value is 5-9 mm while in Escherichia coli bacteria the average inhibitory value is only 3 mm. Endophytic fungi from lemongrass roots have a better antibacterial effect compared to endophytic fungi from lemongrass leaves. Conclusion, endophytic fungi isolated from the roots and leaves of lemongrass (Cymbopogon citratus) have an antibacterial effect on the growth of S. aureus bacteria and are not effective against E. coli bacteria. Jamur endofit dapat menghasilkan senyawa yang berpotensi sebagai antibakteri. Penelitian ini bertujuan untuk menguji efek antibakteri jamur endofit yang diisolasi dari akar,batang dan daun tumbuhan sereh (Cymbopogon citratus)  terhadap bakteri Staphylococcus aureus dan Escherichia coli. Sereh digunakan untuk menghambat atau membunuh bakteri patogen karena mengandung minyak atsiri yang berfungsi sebagai antijamur dan antibakteri terhadap beberapa bakteri patogen seperti Staphylococcus aureus dan  Escherichia coli.  Metode penelitian secara eksperimen laboratorium menguji aktivitas antibakteri dari isolat jamur endofit  yang diperoleh dari tumbuhan sereh (Cymbopogon citratus) pada bakteri Staphylococcus Aureus dan Escherichia Coli menggunakan metode lubang/sumuran. Dari hasil penelitan diperoleh empat jenis jamur endofit yang diisolasi dari akar dan daun  tumbuhan sereh (Cymbopogon citratus).  Hasil penelitian antibakteri menunjukkan bahwa ekstrak jamur endofit yang diisolasi dari tumbuhan sereh lebih efektif menghambat  bakteri Staphylococcus aureus dibandingkan dengan bakteri Escherichia coli. Pada bakteri  Staphylococcus aureus nilai rata-rata daya hambat yaitu 5-9 mm sedangkan pada bakteri Escherichia coli nilai rata-rata daya hambat yaitu hanya 3 mm. Jamur endofit dari akar sereh memiliki efek antibakteri yang lebih baik dibandingkan dengan jamur endofit dari daun sereh. Kesimpulan, Jamur endofit yang diisolasi dari akar  dan daun sereh (Cymbopogon citratus) memiliki efek antibakteri terhadap pertumbuhan bakteri S. aureus dan tidak efektiv terhadap bakteri E. coli. ABSTRACT                                                                 Endophytic microbes are microbes that live in plant tissues. Endophytic fungi can produce various functional compounds in the form of anticancer compounds, antiviral, antibacterial, antifungal and plant growth hormones. Basil (Ocimum Sanctim L.) is the largest species both in fresh form or for the production of essential oils. Ethanol and methanol extracts, from Basil leaves is stated to have antibacterial activity. This study aimed to examine the presence or absence of antibacterial effects of endophytic fungi isolated from the stem and leaves of basil (Ocimum basilicum L.) against Escherichia coli and Staphylococcus aureus. The method used is an experimental laboratory by testing antibacterial activity. From the research carried out four endophytic fungi isolated from the stems and leaves of the Basil (Ocimum basilicum L.) plant. The four endophytic fungi have an antibacterial effect on the two test bacteria. Endophytic fungus from Basil stem 1.2 by 8mm which is categorized as medium and extract of Basil Stem 4.1 by 7mm which is categorized as medium and Basil Stem 1.1 by 17mm categorized as strong and Basil Leaf 2.1 by 22mm categorized as very strong. The conclusion is that endophytic fungi isolated from the Basil (Ocimum bassilicum L.) plant obtained from Lemoh Uner Village have antibacterial activity. Keywords: Antibacterial, Endophytic Fungi, Basil (Ocimum basilicum L.)  ABSTRAK Mikroba endofit merupakan mikroba yang hidup di dalam jaringan tumbuhan. Kemangi (Ocimum Sanctim L.) merupakan spesies terbesar baik dalam bentuk segar ataupun untuk produksi minyak esensial. Ekstrak etanol, metanol daun Kemangi dinyatakan memiliki aktivitas antibakteri. Penelitian ini bertujuan untuk menguji ada tidaknya efek antibakteri jamur endofit yang diisolasi dari batang dan daun tumbuhan Kemangi (Ocimum basilicum L.) terhadap Escherichia coli dan Staphylococcus aureus. Metode yang digunakan ialah eksperimental laboratorium dengan menguji aktivitas antibakteri. Dari penelitian yang dilakukan diperoleh empat jenis jamur endofit yang diisolasi dari batang dan daun tumbuhan Kemangi (Ocimum basilic um L.). Keempat jamur endofit memiliki efek sebagai antibakteri terhadap kedua bakteri uji. Jamur endofit dari Kemangi batang 1.2 sebesar 8mm yang dikategorikan sedang dan ekstrak Kemangi Batang 4.1 sebesar 7mm yang dikategorikan sedang dan Kemangi Batang 1.1 sebesar 17mm dikategorikan kuat dan Kemangi Daun 2.1 sebesar 22mm dikategorikan sangat kuat. This study aims to examine the antibacterial activity of endophytic fungi isolated from the leaves and stems of the cat's whiskers plant (Orthosiphon aristatus) against Escherichia coli and Staphylococcus aureus bacteria. Cat's whiskers contain essential oils which are used as antibacterial against some pathogenic bacteria such as Staphylococcus aureus and Escherichia coli. The method used is experimentally in the laboratory to test the antibacterial activity of endophytic fungi isolates derived from cat's whiskers (Orthosiphon aristatus) on Escherichia coli and Staphylococcus aureus bacteria using the hole / well method. In the research results obtained four samples of endophytic fungi extract isolated from the leaves and stems of the cat's whiskers plant (Orthosiphon aristatus) . The results of antibacterial research showed that endophytic mushroom extract isolated from cat's whiskers were more effective in inhibiting Escherichia coli bacteria compared to Staphylococcus aureus. In Escherichia coli bacteria the average value of inhibition is 5 mm whereas in Staphylococcus aureus bacteria the average value of inhibition is only 2 mm. So it can be seen that the endophytic fungus from the leaves of cat's whiskers has a better antibacterial effect than the endophytic fungus from the stems of the cat's whiskets. Metode yang dipakai yakni secara eksperimen di laboratorium untuk menguji adanya aktivitas antibakteri dari isolat jamur endofit  yang diberasal dari tumbuhan kumis kucing (Orthosiphon aristatus)  pada bakteri  Escherichia coli dan Staphylococcus aureus dengan menggunakan metode lubang/sumuran. Pada hasil penelitan didapatkan empat sampel ekstrak jamur endofit yang diisolasi dari daun dan batang  tumbuhan kumis kucing (Orthosiphon aristatus). Hasil penelitian antibakteri menunjukkan bahwa ekstrak jamur endofit yang diisolasi dari tumbuhan kumis kucing lebih efektif menghambat  bakteri Escherichia coli dibandingkan dengan bakteri Staphylococcus aureus. Pada bakteri  Escherichia coli nilai rata-rata daya hambat yaitu 5 mm sedangkan pada bakteri Staphylococcus aureus nilai rata-rata daya hambat yaitu hanya 2 mm. Sehingga dapat diketahui bahwa jamur endofit dari daun kumis kucing memiliki efek antibakteri yang lebih baik dibandingkan dengan jamur endofit dari batang kumis kucing. Kesimpulan, Jamur endofit yang diisolasi dari daun  dan batang kumis kucing (Orthosiphon aristatus) memiliki efek antibakteri terhadap pertumbuhan bakteri E. coli dan tidak efektiv terhadap bakteri S. aureus.Kata Kunci:  Tumbuhan kumis kucing (Orthosiphon aristatus), aktivitas antibakteri, Escherichia coli dan Staphylococcu saureus.

Abstract: Endophytic fungi is a species of fungi which life on plant tissue system. Endophytic fungi can be isolated from the roots, stems and leave of the plant. Endophytic fungi can produce a substance potencial to be antibacteria. This research aimed to test the presence of antibacterial effect on an isolated leaf of mangrove plant Sonneratia alba towards Staphylococcus aureus and Escherichia coli. The method that is used for the antibacterial activity test, done by putting mycelia of endophytic fungi in combination media that has been smeared with Staphylococcus aureus and Escherichia coli. Obtained from research that has been made, it conducted two species of endophytic fungi that isolated from mangrove plant leaf Sonneratia alba. Both endophytic fungi has effect as antibacteria against Staphylococcus aureus and Escherichia coli, but the second type of endophytic fungi has more effective and stronger antibacterial effect compare to endophytic fungi type I and positive control.Keywords: antibacterial, endophytic fungi, mangrove Sonneratia albaAbstrak: Jamur endofit adalah jamur yang terdapat dalam sistem jaringan tumbuhan. Penelitian ini bertujuan untuk mengetahui ada tidaknya efek antibakteri jamur endofit daun Sonneratia alba terhadap bakteri Staphylococcus aureus dan Escherichia coli. Metode yang digunakan untuk uji antibakteri dilakukan dengan cara menempelkan miselia jamur endofit pada media agar kombinasi yang telah dioleskan bakteri uji. Dari penelitian yang dilakukan diperoleh dua jenis jamur endofit yang diisolasi dari daun tumbuhan mangrove Sonneratia alba. Kedua jamur endofit memiliki efek sebagai antibakteri terhadap kedua bakteri uji akan tetapi jamur endofit tipe II memiliki efek antibakteri yang lebih baik dibandingkan dengan jamur endofit tipe I dan kontrol positif.Kata kunci: antibakteri, jamur endofit, daun Sonneratia alba

The present study was carried out to determine, for the first time, the chemical composition and antibacterial activity of essential oils derived from the aerial parts of three aromatic plants Thymus algeriensis Boiss & Reut, Rosmarinus officinalis L., and Salvia officinalis L. growing under semiarid conditions. The essential oils were chemically analyzed and identified by gas chromatography (GC) and GC/ mass spectrometry (GC/MS) and their antimicrobial activity was individually evaluated against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa using both agar disk diffusion and agar dilution methods. The major constituents of Thymus algeriensis essential oil were identified as camphor (13.62%), 1,8-cineol (6.00%), borneol (5.74%), viridiflorol (4.00%), and linalool (3.93%). For Rosmarinus officinalis essential oil, 48 compounds were characterized, of which the main constituents were camphor (17.09%), Z-β-ocimene (10.88%), isoborneol (9.68%), α-bisabolol (7.89%), and borneol (5.11%). While, Salvia officinalis essential oil was characterized by β-thujone (16.44%), followed by viridiflorol (10.93%), camphor (8.99%), 1,8-cineol (8.11%), trans-caryophyllene (5.85%), and α-humulene (4.69%) as the major components. Notably, results from antibacterial screening indicated that Thymus algeriensis and Salvia officinalis essential oils exhibited a strong inhibitory effect against both Escherichia coli and Staphylococcus aureus compared to Rosmarinus officinalis essential oil. Further, less activity was recorded against Pseudomonas aeruginosa for the three tested essential oils.

Background: [1, 3, 4]oxadiazolenone core containing chalcones and nucleosides were synthesized by Claisen-Schmidt condensation of a variety of benzaldehyde derivatives, obtained from oxidation of substituted 5-(3/6 substituted-4-Methylphenyl)-1, 3, 4-oxadiazole-2(3H)-one and various substituted acetophenone. The resultant chalcones were coupled with penta-O-acetylglucopyranose followed by deacetylation to get [1, 3, 4] oxadiazolenone core containing chalcones and nucleosides. Various analytical techniques viz IR, NMR, LC-MS and elemental analysis were used to confirm the structure of the synthesised compounds.The compounds were targeted against Bacillus subtilis, Staphylococcus aureus and Escherichia coli for antibacterial activity and Aspergillus flavus, Aspergillus niger and Fusarium oxysporum for antifungal activity. Methods: A mixture of Acid hydrazides (3.0 mmol) and N, Nʹ- carbonyl diimidazole (3.3 mmol) in 15 mL of dioxane was refluxed to afford substituted [1, 3, 4]-oxadiazole-2(3H)-one. The resulted [1, 3, 4]- oxadiazole-2(3H)-one (1.42 mmol) was oxidized with Chromyl chloride (1.5 mL) in 20 mL of carbon tetra chloride and condensed with acetophenones (1.42 mmol) to get chalcones 4. The equimolar ratio of obtained chalcones 4 and β -D-1,2,3,4,6- penta-O-acetylglucopyranose in presence of iodine was refluxed to get nucleosides 5. The [1, 3, 4] oxadiazolenone core containing chalcones 4 and nucleosides 5 were tested to determined minimum inhibitory concentration (MIC) value with the experimental procedure of Benson using disc-diffusion method. All compounds were tested at concentration of 5 mg/mL, 2.5 mg/mL, 1.25 mg/mL, 0.62 mg/mL, 0.31 mg/mL and 0.15 mg/mL for antifungal activity against three strains of pathogenic fungi Aspergillus flavus (A. flavus), Aspergillus niger (A. niger) and Fusarium oxysporum (F. oxysporum) and for antibacterial activity against Gram-negative bacterium: Escherichia coli (E. coli), and two Gram-positive bacteria: Staphylococcus aureus (S. aureus) and Bacillus subtilis(B. subtilis). Result: The chalcones 4 and nucleosides 5 were screened for antibacterial activity against E. coli, S. aureus and B. subtilis whereas antifungal activity against A. flavus, A. niger and F. oxysporum. Compounds 4a-t showed good antibacterial activity whereas compounds 5a-t containing glucose moiety showed better activity against fungi. The glucose moiety of compounds 5 helps to enter into the cell wall of fungi and control the cell growth. Conclusion: Chalcones 4 and nucleosides 5 incorporating [1, 3, 4] oxadiazolenone core were synthesized and characterized by various spectral techniques and elemental analysis. These compounds were evaluated for their antifungal activity against three fungi; viz. A. flavus, A. niger and F. oxysporum. In addition to this, synthesized compounds were evaluated for their antibacterial activity against gram negative bacteria E. Coli and gram positive bacteria S. aureus, B. subtilis. Compounds 4a-t showed good antibacterial activity whereas 5a-t showed better activity against fungi.
