Antioxidant effect of Gundelia Tournefortii Extract on Some Physiological, Biochemical and Histopathological Changes in Male Rats

In this study, experimentally lindane-induced histopathological changes and proliferation and/or apoptosis in germ cells in the rat testes were investigated. A total of 40 healthy fertile 3-month-old male rats were used. Animals were divided into 4 groups, each containing 10 rats. Group 1 (control) was given only pure olive oil, Groups 2, 3 and 4 were administered lindane at 10, 20 and 40 mg/kg/bw, respectively, by gastric gavage for 30 days. Microscopically, degenerative changes were observed in the lindane-treated groups. For proliferative activity PCNA immunolabelling and for germ cells apoptosis TUNEL methods were performed. Although a strong PCNA positivity in the control group was observed, a gradual decrease was noted in the lindane-treated groups especially at higher doses. Significant increases of apoptosis were seen in the lindane-treated groups compared to the control group. A decrease in testosterone concentrations was observed in lindane-treated groups compared to the control group. The study indicates that high-dose lindane intoxication contributes to the suppression of spermatogenesis through a reduction of germ cell proliferation and an increase of germ cell death in rat testes.

The root of pasak bumi (Eurycoma longifolia, Jack) is one of plant from Indonesia known as aphrodisiac. The study about its standardized extract as aphrodisiac was show increasing libido, testosterone level, FSH and LH on male rats. This study was conducted to evaluate testosterone level and histopathological changes of the testes of male rats of standardized extract of pasak bumi root. There are 50 old male rats which ages 3-4 months old divided to five groups. Group I as negative control was administered aquadestilata. Group II as positive control was administered testosterone (Andriol®). Group III, IV and V ware given standardized extract of pasak bumi root at the dose 50, 100 and 200 mg/kg body weight Respectively. The extract was given orally twice a day for six days and forty nine days and then the testes was taken out on 7th and 50th day. The histopathological of the testes was evaluated using quantitative parameter by the presence of testes weight, number and diameter of Leydig cells. Testosterone level was taken on 7th and 50th day used ELISA method. The results were analyzed using parametric test, one way ANOVA (P<0,05), post hoc Dunnet (2-sided) (P < 0,05). The result of this study shows that the standardized extract of pasak bumi root was not effect on histopathological changes of the testes but it could increased the level of testosterone m at the dose 100 and 200 mg/kg body weight.  Key word : Eurycoma longifolia, Jack, aphrodisiac, histopathological testes testosteron

Aluminum is recognized as a public health concern because of its potential toxic effects on human health. Therefore, the present experiment was undertaken to determine the effectiveness of curcumin (CUR) in reducing the hepatotoxicity induced by aluminum chloride (AlCl3) in animals. In this experimental study, forty male rats were allocated to five groups (N = 8), viz. no treatment (control), solvent (DMSO or distilled water), CUR (10 mg/kg B.W.), AlCl3 (10 mg/kg B.W.), and CUR+AlCl3 (each with 10 mg/kg B.W.). Treatments were performed by intra-peritoneal injections for 28 days. On the final day, animals were sacrificed, and liver function markers in blood plasma, hepatic antioxidants, and lipid peroxidation index in liver homogenate were estimated. AlCl3 treatment resulted in a significant increase in plasma AST, ALT, ALP and LDH activities with decreased total protein compared to control. AlCl3 significantly reduced superoxide dismutase (SOD), catalase (CAT) and glutathione (GSH) levels but increased malondialdehyde (MDA) level in the liver compared to control. AlCl3 also caused various histopathological changes in the livers of rats. Curcumin could normalize nearly all these parameters. CUR improved levels of changes in different parameters when was combined with AlCl3. It is concluded that CUR has beneficial effects being able to antagonize AlCl3 toxicity.

Abstract Prevention of infections is a very important issue in treating the burn wounds. The nanosilver dressings have many promising advantages, but absorption of silver ions and its adverse effects to the body were always a question. The aim of this study was to compare Silver serum levels and acute toxic effects of nanosilver on histopathology of organs (lungs, liver, kidney, spleen, and brain) in two types of AgiCoat and Acticoat (nanosilver) dressings on second-degree deep burn in rat. This is an experimental study conducted in our animal laboratory. We divided 24 Sprague–Dawley male rats weighing 300 to 350 randomly into two groups. After anesthesia, a second deep-degree burn was made over dorsal skins of rats by standard method. For group A, Agicoat and, for group B, Acticoat dressings were used. The dressings were changed every 3 days with AgiCoat and Acticoat, respectively. After 14 days, we got blood samples and tissue samples taken from heart, liver, kidneys, spleen, lungs, and brain and a sample from dorsal skin of the rat for histopathological examinations. The results showed that the levels of serum silver in both groups were significantly higher than the standard level (1.22 part per million (PM); AgiCoat, P = .017; Acticoat, P = .000), but there was no significant difference between the groups (P = .551). Examination of the relationship between the level of serum silver and histopathological changes in liver showed that hepatotoxicity of AgiCoat was higher compared with Acticoat and the difference was significant (P = .002). There were no pathological changes in brain, kidneys, spleen, heart, and lungs. Wound healing was faster in Acticoat group. The nanosilver dressings can cause toxicity in liver but not in kidney, brain, spleen, heart, and lungs. Liver pathology and hepatotoxicity were more prominent in AgiCoat group. Wound healing was faster in Acticoat group.

Trichloroethylene (TCE) is an organic solvent used in dry cleaning, metal degreasing, thinner for paints and varnishes, anesthetic agent, and so forth. Human beings are appreciably exposed to TCE vapours by inhalation route. The present study has been undertaken to investigate whether TCE inhalation may also bring about testicular toxic effects. Our results indicate that inhalation of TCE by male rats for 12 and 24 weeks brings about significant reduction in absolute testicular weight, and alters marker testicular enzymes activity associated with spermatogenesis and germ cell maturation, along with marked histopathological changes showing depletion of germs cells and spermatogenic arrest.

Abstract Abamectin (Avermectin B1a), is a natural fermentation product derived from the soil bacterium Streptomyces avermitilis. Abamectin (Avermectin B1a) is widely used as an insecticide, acaricide, and anthelmintic. The present study assessed the effects of repeated subacute and subchronic exposure to the commercial formulation of abamectin (Vertemic, 1.8% EC) in albino male rats. The toxic effects of abamectin were studied. The various biochemical parameters and histopathological changes were noted. A stomach tube was used to orally administer sublethal doses of abamectin suspended in corn oil to the rats. The animals were divided into four groups. Rats of the group T1 were orally administered a sublethal dose of 30 mg/kg body weight (b.wt.) (1/10 LD50) three times a week for 30 days and the animals in group T2 were exposed to 10 mg/kg b.wt. (1/30 LD50) for 210 days, once a week. Two control groups (C1 and C2) were used in parallel studies, where animals were administered a corn oil vehicle. At the end of the study period, blood samples were collected from all groups to measure plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST) activities, and the levels of creatinine and urea. Also, total protein and RNA contents were determined in the liver and kidney tissues. Changes in biochemical parameters were more intense in male rats from group T2 than those reported in group T1. The levels of ALT, AST, urea and creatinine were significantly elevated in rats from group T2 when compared to the control. In group T2, a significant decrease in the levels of total protein and RNA in both the liver and kidneys was observed. Fertility was also significantly reduced in male rats ingesting abamectin in group T2. The number of offspring was significantly reduced. Histopathological changes were more intense in male rats from group T2 than those from group T1. In conclusion, the results of this study demonstrate that subchronic oral administration of abamectin altered some biochemical parameters which correlated with histopathological changes

This study was conducted at the laboratory of histology and anatomy, Faculty of Medical and Health Techniques/Kufa, and laboratory of post Graduate/ Department of biology, Faculty of Science/University of Kufa, The present study was conducted to investigate the effect of Prazosin hydrochloride on some organs in male rats (Rattus norvegicus), about 25  mature male rats with the average body weight of 210-290gram and three months age were randomly divided into four groups (5rats / group). The first group was given orally with distilled water as a control group and the other groups (second, third, and fourth) were also given orally with three doses of Prazosin (25, 50,75 mg /kg. b.wt.) daily for a period of eight weeks.  At the end of the treatment period (eight weeks), rats were sacrificed, blood samples obtained, and organs lung, and spleen. The histopathological changes of lungs in the rats treated with prazosin at dose 25 mg/kg.b.wt for 8-weeks showed emphysema and dilation in some of the alveoli, hemorrhage distributed inside the tissue of the lung, polymorphic nuclear infiltration due to pneumonia, the pulmonary artery revealed degenerative changes in the tunica media structure (smooth muscle) and hyperplasia in the connective tissue around pulmonary artery and alveoli. These symptoms which occur in rats treated with prazosin at dose 50 and 75 mg/kg b.wt. as well as the histopathological changes of rat lung demonstrate severe hemorrhage, emphysema, thickening in the wall of some alveoli, pneumocyte necrosis (pneumocyte type 1 and pneumocyte type 2), and showed exudate among lung tissue. Histopathological changes of Spleen in the rats treated with prazosin at doses(50 and75)mg/kg b.w. for 8-weeks revealed histopathological changes, which represented by proliferation in the white pulp lead to fused white pulp together and destruction of some components of red pulp, stenosis in many splenic venous sinuses, the germinal artery show thickening in the tunica media and stenosis occurs, degenerative change in many nuclei of lymphocytes, and proliferation in the component of the white pulp.
